Preliminary study on the use of near infrared spectroscopy for determination of plasma deuterium oxide in dairy cattle

  • Information of body composition (fat and protein) in living animal is important to determine the nutrients requirement. Deuterium oxide (D2O) dilution techniques, as one of isotope dilution techniques have been useful for the prediction of body composition. However, the determination of D2O concentration is time consuming and complicated. Therefore this study was conducted to develop a new method to predict D2O concentration in plasma using near infrared spectroscopy technique (NIRS). Four dairy cows in early lactation were used. They were fed total mixed ration containing conr silage, timothy hay, and concentrates to make 17.0%CP and 14.0 MJDE/kgDM. Dosing D2O was at week 1,3 and 5 after parturition. After dosing D2O, the blood was collected from hour 0 to 72. Blood samples were then centrifuge at 3,000 rpm for 10 minutes to obtain plasma. D2O concentration was analyzed by gas chromatograph (deuterium oxide analyzable system, HK102, Shokotsusyou) after extracted from plasma by liophilization. Plasma sample was scanned by NIRS using Pacific Scientific (Neotec) model 6500 (Perstorp Analytical, Silver Spring, MD) in the range of wavelength from 1100 to 2500 nm. Calibration equation was developed using multiple linear regression. Sample from one animal (cow #550; n: 74) was used for developing the calibration while the rest three animals were used for validating the equation. The range, R and SEC of the calibration set samples were 135-925 ppm, 0.93 and 48.1 ppm, respectively. Validation of the calibration equation for three individual cows was done and the average of NIR predicted value of D2O at each collection time from three weeks injection showed a high correlation. The range, r and 53 of plasma from cow #474 were 322-840 ppm,0.93 and 53.1; cow #478 were 146-951 ppm,0.95 and 39.8; cow #942 were 313-885 ppm,0.95 and 37.2, respectively. Judgement of accuracy based on ratio of standard deviation and standard error in validation set samples (RPD) for cow #474, #478 and #942 were 2.2,4.3 and 3.4, respectively. The error in application due to the variation between individual was considered smaller than the bias from collection period, however, this prediction can be overcome with correction of standard zero-minute concentration of blood. The results of this preliminary study on the use of NIRS for determination of D2O in plasma showed very promising as shown by a convenient and satisfy accuracy. Further study on various physiological stage of animal should be done.

  • Objective : The aim of this experiments was to investigate the effects of Kyongo Puryu(originated from ${\ll}$Classic on Difficulty${\gg}$), Kyongo Um-gok(originated from ${\ll}$Ling Shu${\gg}$) acupuncture on the Blood Pressure, Cardiomegalic index, and plasma levels of atrial natriuretic peptide in Hypertensive RAT induced by 2K1C. Methods : The effects of Kyongo(L8) Puryu(K7), Kyongo(L8) Um-gok(K10) acupuncture on Blood Pressure in Hypertensive RAT induced by Two Kidney One Clip(2K1C). Results : I. Blood Pressure was decreased significantly after acupuncture of L8 K7. II. Cardiomegalic index decreased significantly after acupuncture of L8 Puryu, L8 K10. IlI. Plasma levels of atrial natriuretic peptide was increased significantly after acupuncture of L8 K7.

Effect of Dietary Taurine on Free Amino Acid Concentrations in Blood and Various Tissues of Cats (식이 내의 타우린 함량이 고양이의 혈액 및 주유조직의 유리 아미노산 농도에 미치는 영향)

  • Taurine concentrations in blood and various tissues were measured in cats fed normal trurine diet(0.15% taurine) and trurine-free diet(0% taurine) for 5 weeks and 12 weeks, respectively. Cumulative body weight gain in young cats fed normal taurine diet and taurine-free diet for the initial 5 weeks were 463$\pm$43g and 383$\pm$53g respectively. Taurine concentration was one to two orders magnitude higher in most tissues than in plasma, among which the liver taurine-free diet for 12 weeks resulted in 98% reduction in plasma taurine concentration, 93% reduction in whole blood tarurine concentration, and 40~90% reduction in tissue taurine concentrations compared to the values for normal taurine diet group. Among the tissues tested, taurine depletion was most profound in heart and kidney, while liver and intestine were relatively resistant to taurine depletion. Whole blood taruined concentration appeared to be a better index for the body taurine status of cats as in more closely reflects the rate of taurine depletion in tissues. Taurine depletion in cats significantly increased the concentrations of essential and non-essential free amino acids in liver, while free amino acid concentrations in other tissues were not affected by taurine depletion.

  • This experiment investigated changes in metabolic rate (MO2), critical oxygen saturation (Scrit), and blood chemistry of olive flounder Paralichthys olivaceus exposed to progressive hypoxia and returned to normoxic water at 20°C. The normoxic standard metabolic rate (SMR) and routine metabolic rate (RMR) were 69.5-83.9 and 70.2-156.4 mg O2 kg-1h-1, respectively based on fish weight. Scrit was 31.0% dissolved oxygen (DO) at 20°C. After returning the fish to 70% DO following exposure to hypoxia (20% DO), MO2 increased two-fold compared to the normoxic SMR and then decreased into the range of the RMR with time. Blood PO2 and plasma lactate decreased significantly after exposure to hypoxia (20% DO) and then increased as ambient oxygen saturation decreased. Cortisol levels increased as ambient oxygen saturation decreased, but the levels decreased rapidly in the range of the normoxic control when the fish were returned to ambient water with 70% DO. Plasma glucose levels increased when the fish were returned to normoxic water after exposure to a progressively more hypoxic condition.

Effect of Feeding High Forage Diets with Supplemental Fat on Blood Metabolites, Rumen Fermentation and Dry Matter Digestibility in Dairy Cows

  • Fifty mid-lactation Holstein cows were used in a six-week feeding trial to study effects of high-forage, and high-fat diets on blood constituents, rumen fermentation and dry matter digestibility. Cows were divided into 10 replicates, each consisting of five cows. Each cow was assigned to a control (diet 1) or one of the four experimental diets (high-forage (75%), high-fat (7.5%) (diet 2); high-forage. medium-fat (5.0%) (diet 3); medium forage (65%), high-fat (diet 4); medium-forage, medium-fat (diet 5)), or a control diet containing about 50% forage and 2% fat. All diets were isonitrogenous (17.7% crude protein). The forage mixture consisted of 20% alfalfa hay, 40% alfalfa haylage, and 40% corn silage. Supplemental fat included 80% rumen-protected fat and 20% yellow grease. A non-significant difference was observed in concentrations of blood glucose for cows on different experimental and control diets. Plasma nonesterified fatty acids (NEFA) were higher in cows consuming experimental diets than those consuming the control diet. However, differences in NEFA concentrations in the plasma of cows consuming diets with different forage and fat levels were not significant. Rumen pH, concentration of volatile fatty acids (VFA) in rumen contents, and dry matter digestibility of control and experimental diets, and diets with different levels of forage and supplemental fat did not differ significantly.

  • The effects of Miyairi (Clostridium butyricum) on blood constituents and growth of cultured freshwater fishes, such as carp, tilapia and israeli carp were studied and then the following results were obtained : 1) All fishes produced the most efficient growth in 0.1% Miyairi-treated group. 2) Blood constituents of Miyairitreated group compared with those of contrul gruop showed no changes in RBCs. hemoglobin and hemato crit. and the increase in plasma protein and glucose. and the decrease in plasma cholesterol. 3) GOT level and GPT level in blood of Miyairi-treated group were almost equal to those in blood of control group.

Oxidatvive Stress in Rat Model of Preeclampsia and Clinical Correlates

  • There are growing evidences suggesting a pivotal role of oxidative stress in the pathophysiology of preeclampsia. We investigated oxidative stress in the rat model of preeclampsia, and in clinical cases. Pregnant female rats were injected intraperitoneally with deoxycorticosterone acetate (DOCA) and given 0.9% saline as drinking water during their pregnancy. We assessed plasma $F_2-isoprostane(8-iso-PGF_{2{\alpha})$ and malondialdehyde (MDA) in a rat model, and the same markers in the plasma of maternal blood and fetal cord blood in pregnant women with preclampsia. Blood samples from the umbilical arteries and veins were collected separately. The concentrations of MDA were increased in the preeclampsia groups of animal and humans, compared with the control group; it was significantly increased in the umbilical artery and vein of the preeclampsia group. The concentrations of $F_2-isoprostane$ were elevated in the preeclampsia groups of animal and humans, compared with the control group, and the increase in $F_2-isoprostane$ concentration was prominent in the umbilical vein than umbilical artery of the preeclampsia group. Therefore, it appears that the placenta has an important role in the pathophysiology of preeclampsia, and the $F_2-isoprostane$ of the umbilical vein may serve as a relatively reliable marker for ischemic/hypoxic injury to the fetus during the perinatal period.

Blood Lead Level of Families and Environmental Factors (가족 구성원의 혈중 납농도에 영향을 미치는 환경요소)

  • Objectives : To estimate the correlation of blood lead level of families and environmental factors and infer its influence on blood lead levels in the Korean urban area in Seoul Korea. Methods : The study subjects comprised 499 men and 489 women from 366 families and we analyzed the blood lead level using induced coupled plasma/mass spectroscopy and had interviews. Results : The mean blood lead level of men was 3.00 $\mu\textrm{g}$/dL. The stastical analysis of this study used the tool of frequency rate and t-test between blood lead levels of families and environmental factors. Especially father's smoking and dusty workplace environment showed significancy to blood lead levels. Conclusion : There was positive relationship of blood lead level to several environmental factors. These facts show outdoor environment is more influential than home environment.

  • The miniaturization and integration are trend of modern blood analyses. Micro-Bio-Fluidics plays an important role in a micro blood analysis system. In this paper, analysis and design technology for blood analysis system is presented. Numerical simulations of a blood flow in micro separator and reservoir are conducted. As a result, we suggest on-chip micro separator, which performed plasma separation from whole blood in micro channels.

  • Effects of titanium dioxide nanoparticles on blood coagulation and platelet aggregation were investigated using diluted whole blood and platelet rich plasma (PRP) solution prepared from human and rat blood, respectively. Blood coagulation was monitored by using a whole blood impedance aggregometer and titanium dioxide nanoparticle (10, 20, and 40 ppm) did not show any effect on the coagulation both in human and in rat blood. When platelet aggregation was measured by turbidometric method after addition of titanium dioxide nanoparticles to PRP solution with final concentrations of 1, 5 and 10 ppm. no aggregation was observed.
