Cadmium is considered one of the most toxic, non biodegradable heavy metal for the human and animals. The purpose of the present study was to investigate the changes in biochemical parameters of blood and accumulation of cadmium in various tissue caused by various levels of dietary cadmium chloride ($CdCl_2$) in broiler chicks. $CdCl_2$ was administered through drinking water to broiler chicks. In spectral analysis, $CdCl_2$ treatment caused a significant increase in Glutamate pyruvate transaminase (GPT), creatinine and uric acid levels in all treated groups. Intriguingly, the GPT, creatinine, and uric acid levels were significantly higher at 75 mg/kg as compared to the groups treated with high doses (100, 125 and 150 mg/kg) of $CdCl_2$. Atomic Absorption Spectrophotometer (AAS) was used for the determination of Cd accumulation in kidney, liver and Breast muscles. AAS analysis revealed that Cd accumulation is increased in breast muscles as compared to liver and kidney at higher doses of Cd than 75 mg/kg.

Obesity increases the risk of mortality and morbidity because it results in hypertension, heart disease, and type 2 diabetes. Therefore, there is an urgent need for pharmacotherapeutic drugs to treat obesity. We performed a screening assay using natural products with anti-adipogenic properties in 3T3-L1 cells and determined that tschimganidine, a terpenoid from the Umbelliferae family, inhibited adipogenesis. To evaluate the anti-obesity effects of tschimganidine in vivo. Mice were fed either a normal chow diet (NFD) or a high-fat chow diet (HFD) with or without tschimganidine for 12 weeks. Treatment with tschimganidine decreased lipid accumulation and adipogenesis, accompanied by reduced expression of adipogenesis and lipid accumulation-related factors. Tschimganidine significantly increased the phosphorylation of AMP-activated protein kinase (AMPK) and decreased that of AKT. Depletion of AMPK relieved the reduction in lipid accumulation resulting from tschimganidine treatment. Moreover, tschimganidine administration drastically reduced the weight and size of both gonadal white adipose tissue (WAT) and blood glucose levels in high-fat diet-induced obese mice. We suggest that tschimganidine is a potent anti-obesity agent, which impedes adipogenesis and improves glucose homeostasis. Tschimganidine can then be evaluated for clinical application as a therapeutic agent.
This study was performed to measure the effect of carotenoid polarity on absorption and Pigmentation in blood, muscle, and skin of laying hens. Carotenoids used in this study and Polarity were ${\beta}$-8-Apo-carotenoic acid ethyl ester(ACAEE) > astaxanthin > canthaxanthin > ${\beta}$-carotene. The chickens used in this study were 61∼78 weeks old ISA brown laying hens. Experiment #1 was designed to measure the effect of carotenoid level on the accumulation of carotenoids in carcass of laying hens after feeding for 6 weeks. D-carotene was accumulated in skin only at a detectable level when it was fed at 300 mg/kg feed. The skin was pigmented as yellow when it was measured by colorimeter. The concentration of ${\beta}$-carotene in blood was proportional to that in the feed. Pigmentation of muscle by 9-carotene was not effective. Canthaxanthin significantly increased redness of the skin(p<0.05). However, canthaxanthin did not pigment muscle. The level of canthaxanthin in the blood and skin increased as the concentration in feed increased. ACAEE at 200 and 300 mg/kg feed significantly increased yellowness of the skin(p<0.05). At all levels of ACAEE used($\geq$50 mg/kg feed) the b values of colorimeter increased. With increases in the contents of ACAEE, the concentration of ACAEE in the blood and skin increased. Compared to ${\beta}$-carotene, ACAEE and canthaxanthin were absorbed 9- and 3-fold more into the blood, respectively. The concentration of ACAEE and canthaxanthin in the skin was 1/10 of those in the blood. The lower were the concentrations of carotenoids in the feed, the higher were the absorption rates(from feed to blood and from blood to skin) The results indicated that the higher was the polarity of carotenoids, the more effective were the absorption and pigmentation. In experiment #2, the effect of carotenoid levels of feed on the accumulation of carotenoids in each body part of laying hens was determined. The colorimeter values for redness and yellowness significantly increased when canthaxanthin was fed at $\geq$50 mg/kg feed(p<0.05). Breast and thigh were not affected by feeding of canthaxanthin at the levels used. The L values of muscle but not the a and b values were significantly affected by feeding at $\geq$200 mg/kg feed for wings and breasts, respectively. The yellowness of skin and muscle significantly increased when ACAEE was fed at $\geq$ 100 and $\geq$ 200 mg/kg feed, respectively(p<0.05).
본 연구는 추출용매에 따른 레몬밤 추출물의 중성지방 조절에 대한 논문으로 3T3-L1 지방전구세포를 이용해 지방세포로 분화유도한 뒤 lipid accumulation, triglyceride contents, PKA, HSL, perilipin, ATGL 및 CGI-58 단백질 발현을 확인하였다. 실험결과 네가지 조건의 레몬밤 추출물 중 물 추출 조건인 MOW100 추출물만 lipid accumulation과 triglyceride contents의 유의적 억제 효능을 나타내었으며, MOE70, MOE50, MOE50 추출물은 lipid accumulation과 triglyceride contents의 감소 효과가 없거나 유의적 차이를 나타내지 않는 것이 확인되었다. MOW100 추출물의 세포내 지방 축적 억제는 지방세포 내 lipolysis를 조절하는 효소 조절을 통해 나타난 결과로 판단된다. 따라서 레몬밤 물 추출물은 cAMP-PKA를 활성화시키고, lipolysis를 조절하는 효소의 상승작용을 통해 세포내 중성지방을 조절함으로써 혈중 중성지방을 개선하는 후보소재로의 사용이 가능할 것으로 사료된다.
Objectives The purpose of this study is to understand formation courses of the ten types of LBP (十種腰痛) in Korean medicine through reviewing classic literatures. Methods We summarized sentences describing syndrome differentiation of LBP directly in Uibujeonrok (醫部全錄) and Donguibogam (東醫寶鑑), and then organized similarities and differences among diagnostic factors described in the classic literatures. Results In most of the classics LBP was classified according to the cause but the causes varied depending on the classic literatures. Cheonkeumbang (千金方) tried to suggest a reasonable classification of LBP in a relatively early age. In Dangyesimbeop (丹溪心法) the causes of LBP were divided into 6 factors; qi movement stagnation (氣鬱), dampness-heat (濕熱), kidney deficiency (腎虛), static blood (瘀血), sprain (挫閃) and phlegm accumulation (積痰). It had a lot of influence on the classic literatures published later. Donguibogam was also influenced by the Dangyesimbeop and the ten types of LBP in Donguibogam was similar to the information on the classification shown in Uihakipmun (醫學入門) and Uijongpildok (醫宗必讀). Conclusions We verified universality of the ten types of LBP; kidney deficiency, phlegm-retained fluid (痰飮), food accumulation (食積), sprain, static blood, wind (風), cold (寒), dampness (濕), dampness-heat and qi (氣).
Tumor localizing properties of $^{67}Ga$ citrate and $^{169}Yb$ citrate were studies with sarcoma 180 bearing mice. To compare precisely the accumulation of $^{67}Ga$ citrate and $^{169}Yb$ citrate in tumor tissue itself, $^{67}Ga$ and l63yb were administered simultaneously as a mixture of same chemical form of citrate. The yield of $^{169}Yb$ labeled citrated and the of radiopharmaceuticals purity was identified more than 90% by chromatography on silica gel plates. $^{67}Ga$ and $^{169}Yb$ were injected into mice and the mice were killed at 3, 24, and 48 hours following intraperitoneal injection of the radiopharmaceuticals. The retention value of $^{67}Ga$ and $^{169}Yb$ in tumor was similar but they differ from each other markedly in normal tissue distribution. $^{169}Yb$ citrate is cleared up from blood more rapidly than $^{67}Ga$ citrate, and a high tumor to blood ratio is achieved shortly after injection.
Metallothionein (MT), a small protein molecule which can bind or release metal ions, is involved in the regulation of cellular metal homeostasis. This study was investigated the accumulation of cadmium in blood, tissue (liver, kidney and brain), and the effect of cadmium on several key genes (MT-I, MT-II, ZnT-1) in zinc metabolism in the mouse. Mouses weighing 20∼25 g were randomly assigned to control and cadmium treated group (Cd group). Cd group was intraperitoneally injected with cadmium 2, 4, 8 mg/kg and control group was administerd with saline. Mouses of each group were sacrificed by decapitation 4 hours after the administration of cadmium. Cadmium contents in blood, liver, kidney and brain were increased by a dose-dependent manner. Accumulation of cadmium was mainly occurred in liver and kidney. Induction of MT-I and MT-II protein was increased, but ZnT-1 expression was decreased in a dose-dependent manner by the treatment of 2∼8 mg/kg cadmium. These results suggested that cadmium can be transported to brain and alter the expression of several key genes in zinc homeostasis.
The suppressive effects of glutathione-enriched Saccharomyces cerevisiae FF-8 strain (FF-8 GY) on alcoholinduced hepatotoxicity have been studied. FF-8 GY (256 mg/L) from the fermentation at a large scale bioreactor was used. Either of 5% FF-8 GY or 5% commercial glutathione-enriched yeast extract (GYE) with or without 30% alcohol was tested with rats for 4 weeks. FF-8 GY and GYE were found to reduce those alcohol-elevated serum alanine aminotransferase (ALT), alkaline phosphatase (ALP), and lactate dehydrogenase (LDH) activities. Blood alcohol and acetaldehyde were also decreased by FF-8 GY and GYE. Interestingly, FF-8 GY drastically increased both hepatic alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) activities in comparison to GYE group, thus FF-8 GY would be more effective in blood alcohol and acetaldehyde reduction. Attenuated lipid droplet accumulation in hepatocytes was observed in both FF-8 GY and GYE when alcohol stimulated the accumulation. Therefore, FF-8 GY may be useful to protect liver from alcohol-induced hepatotoxicity.
The purpose of this study was to observe the microscopic change of salivary gland tissues, which is the cause of xerostomia in diabetic condition: for this target the author injected STZ 0.1ml/100gm b.w. on rat to produce diabetes, and than observed microscopic change in submandibular gland through the histopathologic method, obtaining as follows : 1. All of the experimental specimens suffered diabetes after injection of STZ, but the blood glucose level was irregular. 2. There were not interrelationship between the blood glucose level and microscopic change on salivary gland tissues. 3. The salivary gland changed after diabetes initiation in lapse of times; after 14 days,suffered severe destruction, however after 17 days, it was regenerated. 4. Salivary glands showed congested, destructive acini cells, and hyperplastic ductal cells as well as salivary gland duct-like structures. 5. Then were accumulation of fat granules within the cytoplasm of the acini cells on mucous gland in diabetic condition. 6. According to insulin injection, there were no more changes on salivary gland tissues, even in the accumulation of fat granules. 7. Histological changes of the serous gland were obvious more than the mucous gland in this experimental condition.
The possible protective effects of highly zinc-containing yeast Saccharomyces cerevisiae, FF-10 strain, isolated from tropical fruit rambutan on acute alcoholic liver injury in rats were evaluated. Zinc concentration in this strain was 30.6mg%. The activities of serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), and $\gamma$-glutamyl transpeptidase ($\gamma$-GTP) were highly increased when alcohol was treated, relative to the normal rats. Also, a highly significant increase in the blood alcohol and acetaldehyde levels by alcohol treatment was observed. Administration of FF-10 strain markedly prevented alcohol-induced elevation of the activities of serum ALT, AST, and $\gamma$-GTP, and the levels of blood alcohol and acetaldehyde, and these reduced levels reached to that of normal rats. As compared with alcohol treated control rats, the FF-10 strain supplementation showed highly decreased the triglyceride concentration in serum. Alcohol treatment induced the marked accumulation of small lipid droplets, hepatocytes necrosis, and inflammation, but FF-10 strain administration attenuated to alcohol-induced accumulation of small lipid droplets and hepatocyte necrosis in the liver. Therefore, the current finding suggests that zinc-enriched yeast FF-10 strain isolated from tropical fruit rambutan may have protective effect against alcohol-induced hepatotoxicity.
