Effects of Modified Gami-Yiyiren-tang on the Gout Induced ICR Mice (가마의이인탕이 통풍유발 흰쥐에 미치는 영향)

  • In order to obtain practical application of Yiyiren-tang against gout disease, water extract of modified Gami-Yiyiren-tang(GYYT) added with Lonicera japonica is prepared through decoction and freeze drying. The effects were evaluated with blood sample from ICR mice treated with MSU intra-peritoneal injection 1 time a day, 3 days consecutively. The results were as follows. GYYT decreased C-reactive protein level and HDL cholesterol levels. But the creatinine, the LDL cholesterol levels, triglyceride and uric acid levels were not changed were decreased significantly. These results show that GYYT can be used effectively against the urate-related gout disease. But not better than effects of the Gami-daehwangmokdan-tang.

The Effect of Different dietary Patterns (between Non-Vegetarian and Vegetarian) on Protein Nutritional Status during Short-term (식사 Pattern의 차이가 단백질 영양상태에 미치는 영향)

  • The purpose of this study was to describe the protein nutritional status of female collegians between nonvegetarian diet groups(14) and vegetarian diet groups(19). Daily intake, protein were calculated from food direct measurement. Urea/creatinine, muscle mass, fatique conditions and hematological parameter were calculated. Blood samples were analyzed for total protein, albumin ammonia, urea, uric acid, creatinine, BUN contents. The results obtained are summarized as following : 1) On total dietary intake, especially Fe, vitamin C, niacin intake, vegetarian groups were higher than non vegetarian groups. 2) On composition of EAA(essential amino acids), vegetarians were highter than non vegetarian of leucine contents, but were lower lysine and threonine contents. 3) On urea / creatinine excretion, vegetarians were higher than non begetarians. Muscle mass were non signicant, fatique condition, vegetarians were normal state. 4) Non vegetarians were increased serum albumin total protein, uric acid, ammonia but vegetarians were decreased BUN, ammonia after experimental diet intake. 5) In process of time after meal, non vegetarians were increased serum albumin total protein, BUN, but vegetarians were decreased ammonia.

  • In order to obtain practical application of Daehwangmokdan-tang gout disease, water extract of Gami-daehwangmokdan-tang(GDMT) added with Lonicera japonica is prepared through decoction and freeze drying. The effects were evaluated with blood sample from ICR mice treated with MSU intra-peritoneal injection 1 time a day, 3 days consecutively. The results were as follows. GDMT decreased BUN, creatinine and C-reactive protein level in serum significantly. But the HDL and LDL cholesterol levels were not changed in spite of positive tendency in HDL cholesterol. Triglyceride and uric acid levels were decreased significantly too. These results show that GDMT can be used effectively against the urate-related gout disease.

Effects of Chungganhaeju-tang(Qingganjiejiu-tang) on Alcohol Metabolism and Alcoholic Liver Damages (청간해주탕이 알코올대사 및 손상간에 미치는 영향)

  • Objectives: This experiment was conducted to verify the effects of Chungganhaeju-tang(Qingganjiejiu-tang) on the alcohol metabolism and liver functions, by measuring the activity levels of ADH and ALDH, as well as glucose, triglyceride, and BUN. Damage of the liver cells caused by alcohol was determined through the examination of serum levels of AST, ALT, ALP, LDH, and uric acid. Methods: Sprague-Dawley rats were used in this experiment and the rats were divided into control and experiment groups. Chungganhaeju-tang(Qingganjiejiu-tang) extract was orally administered in the experiment group for three weeks. Each group was further classified into two sub-groups, and control group's blood was taken without oral ingestion of alcohol, while the experiment group' s blood was withdrawn after ingestion of alcohol. Evaluation of damage level was done considering the presence of extract and alcohol. Results: In this experiment, Chungganhaeju-tang(Qingganjiejiu-tang) significantly suppressed the activity of ADH which is a precursor enzyme of acetaldehyde, but didn't cause significant changes in the activity of ALDH which is a catabolic enzyme. Decreased glucose level due to alcohol consumption was recovered back to the normal level and increased levels of triglyceride, BUN, AST, ALT, ALP, LDH, and uric acid were significantly reduced. Conclusions: These experiment results suggest that Chungganhaeju-tang(Qingganjiejiu-tang) inhibits the formation of acetaldehyde in the metabolism of alcohol, and affects the recovery of weakened liver functions due to alcohol.

  • Flavin mononucleotide (1,4-butanediamine) Pt(II) complex (7FMN) was synthesized and screened anticancer activity [J. Pharm. Soc. Korea 43(6),762-770 (1999)]. 7FMN have good water solubility and moderate anticancer activiy In this paper cell-cycle specificity and nephrotoxicity were studied. Interaction of DNA with cisplatin and synthesized 7FMN was analyzed by flow cytometry and showed G2 arrest in L1210 cell line. It means that cell-cycle on L1210 was inhibit in S phase by cisplatin and 7FMN. In order to biochemically analyze nephrotoxicity of cisplatin and 7FMN, after injecting each agent intraperitoneally, blood was exsanguinated after 6 hours, 1 day, 3 days and 7 days, respectively. Then, serum was separated from the blood. The serum level of BUN, creatinine and uric acid in cisplatin and 7FMN administated mice (25~35 g, ICR strain, a dose each 8,12 and 16 times of the $IC_{50}$/ value, cisplatin; 7 times) were determined by autochemistry analyzer. In cisplatinadministered mice group, BUN level was elevated than normal control group at 3rd day and repaired at 7th day. In 7FMN administrated group was not elevated. Creatinine and uric acid level were no difference with the normal control group. Therefore synthesized 7FMN is less toxic than cisplatin in nephrotoxiciaty.

Effect of Pyridoxal on Pyrazinamide - and its metabolite-induced Hyperuricemia (Pyrazinamide와 그 대사산물이 유발하는 고뇨산 혈증에 미치는 Pyridoxal의 영향)

  • Pyrazinamide, an amide of pyrazinoic acid, is widely used in combination with other drugs for the treatment of tuberculosis. It was attempted to observe the effect of pyridoxal on the pyrazinamide induce hyperuricemia in this study. It was observed that the values of serum transminases were not changed in mice injected pyrazinamide and pyrazinoic acid, respectively (100, 200, and 300mg/kg) compared with control. Blood urate levels were increased in mice treated with these drugs in a dose dependent manner. After pyrazinoic acid was administered intraperitoneally at a dose of 300mg/kg pretreated with 50mg/kg of pyridoxal once daily for 4 days, the blood levels of uric acid and pyrazinoic acid were decreased significantly.

EFFECT OF PANAX GINSENG ON XANTHINE OXIDASE ACTIVITY IN ETHANOL-INDUCED HYPERURICEMIA

  • The influence of Panax ginseng on alcohol-induced hyperuricemia were observed. Changes of uric acid blood levels and hepatic xanthine oxidase activities were studied by means of treating alcohol intoxication with ginseng. It was found that a single dose (4 mg/Kg) of ginseng saponin administered intraperitoneally significantly inhibits the hepatic xanthine oxidase activities and decrease urate blood levels in ethanol-induced hyperuricemic mice. It was also observed that there were some difference in pharmacological aspect between Panax ginseng and allopurinol which is a potent inhibitor of xanthine oxidase from any sources.

  • This study was to investigate the effects of SUNGSIMJIHWANGTANG(SSJHT) on the blood and brain tissues of aged rats. For the experiment, the aged rats were divided into three groups : Non treated group(NC), distilled water fed group(PC), SSJHT fed group(SSJHT). Each group was fed for ten days before administration of scopolamine. Then, we injected scopolamine intraperitoneally to PC and SSJHT group for 5 days. We observed the changes of their blood cell(WBC, RBC, platelet), blood serum(BUN, creatinine, glucose, uric acid), erythrocyte hemolysis, and the activities of cholinesterase and measured the amounts of malondialdehyde in the serum, catalase, and SOD in the brain tissue. The main results of this investigation are as follows. 1. In respect of the number of WBC, SSJHT group exhibited significant increase in comparison with PC. 2. In respect of the amount of creatinine and uric acid in the blood serum, SSJHT group exhibited significant decrease in comparison with PC. 3. In respect of erythrocyte hemolysis, SSJHT group exhibited significant suppression in comparison with PC. 4. In respect of the activity of cholinesterase in the serum, SSJHT group exhibited significant activation in comparison with PC. 5. In respect of the amounts of malondialdehyde in the serum, SSJHT group exhibited significant decrease in comparison with PC. 6. In respect of the activity of catalase in brain tissue, SSJHT group didn't exhibit significant change in comparison with PC. 7. In respect of the activity of SOD in brain tissue, SSJHT group exhibited significant activation in comparison with PC. As a result of this study, SSJHT is expected to have antiaging effect by suppressing the formation of free radicals, the accumulation of antioxidants and further study needs to be carried on about SSJHT.

  • Many interference in blood should be considered for non-invasive blood glucose level monitoring by near-infrared spectroscopy because blood glucose concentration is about 0.1% (w/v) in normal state. In this study, we investigated the influence of other blood components on blood glucose level monitoring by near-infrared spectroscopy. It carried out by newly developed portable type-NIR system (1100∼2200 nm). Spectrum features of NIR diffuse spectral data were investigated for some blood compnents powder such as hemoglobin, blood serum albumin, urea, uric acid, ascorbate, glucose, cholesterol and as adding glucose powder into other blood components powder mixture.   • Objectives: The purpose of this study was to investigate the effect of exterior vascular laser irradiation therapy by live blood analysis. Methods: We analysed the changing forms of the live blood sample with a microscope before and after exterior vascular laser irradiation therapy of the blood. Results: Rouleau of red cells, erythrocyte aggregation of red cells, thrombocyte aggregation, uric acid crystals, red crystals, and protoplasts in blood were decreased significantly after exterior vascular laser irradiation therapy. Conclusions: This study suggests that exterior vascular laser irradiation will have positive effects in eliminating various effete matters in blood and will have efficacy in the treatment and prevention of cardiovascular system disorders and hyperlipidemia, caused by effete matters, or numbness and arthralgia caused by blood stagnancy and blood circulation disorder.
