Effect of Medium-chain Triglyceride (MCT) on Growth Performance, Nutrient Digestibility, Blood Characteristics in Weanling Pigs

  • One hundred and twenty weanling pigs in experiment 1 (Exp. 1) ($6.91{\pm}0.99kg$; 21 d of age) and Exp. 2 ($10.20{\pm}1.09kg$; 28 d of age) were used in two 42-d and 35-d experiments to evaluate the effect of medium-chain-triglyceride (MCT) on growth performance, apparent total tract digestibility (ATTD) of nutrients and blood profile. In both of Exp. 1 and Exp. 2, the same dietary treatments were utilized as follows : i) negative control (NC), ii) positive control (PC), NC+antibiotics (40 mg/kg Tiamulin, 110 mg/kg Tylosin, and 10 mg/kg Enramycin, iii) MCT3, NC+0.32% (phase 1, 2 and 3) MCT, and iv) MCT5, NC+0.55% (phase 1), 0.32% (phase 2 and 3) MCT. In Exp. 1, the pigs fed MCT5 diets had higher (p<0.05) ADG compared to NC treatment during the first 2 wk. From d 15 to 28, the ATTD of energy was improved (p<0.05) by MCT3 compared to the PC treatment. No effect has been observed on the blood profiles [red blood cell (RBC), white blood cell (WBC), immunoglobulin-G (IgG), lymphocyte concentration] measured in this study. In Exp. 2, the ADG were increased (p<0.05) by the MCT5 treatment than the PC treatment from d 0 to 14. Pigs fed PC treatment diet had lower ADFI (p<0.05) and better FCR (p<0.05) than NC treatment, whereas no differences were shown between MCT treatments and NC or PC treatment from d 15 to 35 and overall phase. The ATTD of DM and nitrogen were improved (p<0.05) by the effect of MCT5 related to the NC and PC treatment at the end of 2nd and 5th wk. The pigs fed MCT3 had higher (p<0.05) energy digestibility than PC treatment. No effects were seen in the blood profiles we measured (WBC, RBC, lymphocyte and immunoglobulin-G). In conclusion, the addition of MCT in the weanling pigs diet can improve the ADG and digestibility during the earlier period (first 2 wks), but had little effect on the blood characteristics.

Forensic STR Analysis of Mixed Chimerism after Allogeneic Bone Marrow Transplantation

  • Multiplex PCR-based short tandem repeat (STR) analysis is considered as a good tool for monitoring bone marrow engraftment after sex-mismatched allogeneic transplantation and provides a sensitive and accurate assessment of the contribution of both donor and/or recipient cells in post-transplantation specimens. Forensic STR analysis and quantitative real time PCR are used to determine the proportion of donor versus recipient each contained within the total DNA. The STR markers were co-amplified in a single reaction by using commercial $PowerPlex^{(R)}$ 16 system and $AmpFISTR^{(R)}$ $Identifiler^{(R)}$ / $Yfiler^{(R)}$ PCR amplification kits. Separation of the PCR products and fluorescence detection were performed by ABI $PRIS^{(R)}$ 3100 Genetic Analyzer with capillary electrophoresis. The $GeneMapper^{TM}$ ID software were used for size calling and analysis of STR profiles. Extracted DNA was quantified by the $Quantifiler^{TM}$ Human DNA / Y Human Male DNA Quantification Kit The intent of this study was to analyze the ratio of donor versus recipient cells in the post-transplant peripheral blood, spleen, lung and kidney specimens. Specimens were taken from the traffic accident male victim who had been engrafted from bone marrow female donor. Blood and spleen specimens displayed female donor DNA profile. Kidney specimen showed male recipient DNA profile. Interestingly, lung tissue showed mixed profiles. The findings of this study indicate that the forensic STR analysis using fluorescence labeling PCR combined with capillary electrophoresis is quick and reliable enough to assess the ratio of donor versus recipient cells and to monitor the mixed chimeric patterns.

유기농 산양유 사료에 산삼배양근 첨가가 산양유와 혈액 내 진세노사이드 함량 및 조성에 미치는 영향 (Effect of Supplementing Cultured Wild Ginseng Roots in the Diet of Organic Saanen Dairy Goats on Milk Composition and Ginsenoside Profiles in Blood and Milk)

  • The aim of the present study was to determine the effect of dietary cultured wild ginseng root (CWGR) supplementation on goat milk composition and ginsenoside profiles. Sixteen Saanen dairy goats were allocated to two balanced groups based on lactation period, body weight ($38.6{\pm}3.2kg$), and dairy milk yield ($2.85{\pm}1.2kg$), and were kept in separate pens. Goats were fed a total mixed ration (TMR) feed (2.3 kg/d, dry matter basis) and 1.5 g of CWGR powder was supplemented in the experimental diet. The total feeding period was 3 weeks, and milk and blood samples were collected on the last three days of the experimental period. There was no effect of CWGR on daily milk yield and milk composition (fat, protein, lactose, and solid-not-fat). However, the CWGR-treatment group had significantly higher plasma IgG and protein contents than the control group (P < 0.05). Significant amounts of ginsenosides were observed in the milk of the CWGR-treatment group, whereas ginsenosides were not detected in the milk of the control group. In conclusion, dietary CWGR was a useful regimen to produce functional goat milk enriched in ginsenosides.

  • The purpose of this study to examine lipid profiles and lipoprotein (a) concentrations and identify the risk factors of type 2 diabetes mellitus patients with macrovascular complications. The subjects were consisted of 618 out patients with type 2 diabetes mellitus from beginning of March through the end of April in 2001, who visited at the endocrinology department at Kangnam St. Mary's Hospital of Catholic University in Seoul. The patient's clinical laboratory data and the occurrence of chronic complications of diabetes were assessed at medical record review. The data were analyzed using for t-test. chi-square test and logistic regression. The results were as follows : 1. There were no significant differences in age. duration of diabetes. body mass index, sex, fasting blood glucose, 2-hour postprandial blood glucose and $HbA_{1}c$ level between macrovascular complication group and non macrovascular complication group. 2. There were significant differences in the level of total cholesterol. triglyceride. HDL(High density lipoprotein) cholesterol, LDL(Low density lipoprotein) cholesterol and lipoprotein(a) between macrovascular complication group and non macrovascular complication group. 3. Significant factor associated with macrovascular complication in the logistic regression best gut model was HDL cholesterol.

Effects of Humic Acid and Blueberry Leaf Powder Supplementation in Feeds on the Productivity, Blood and Meat Quality of Finishing Pigs

  • The objective of this study was to determine effects of humic acid (HA) and blueberry leaf powder (BLP) supplementation in pig feed on productivity, blood profiles, and meat quality characteristics of longissimus muscle. The experimental design included six treatments: 1) CON, no addition; 2) T1, BLP 0.1%; 3) T2, BLP 0.2%; 4) T3, humic acid 2%; 5) T4: humic acid 2%+BLP 0.1%; and 6) T5: humic acid 2%+BLP 0.2%. HA and BLP supplementation in pig feed significantly increased average daily feed intake (ADFI) values (p<0.05). HA supplementation in pig feed had beneficial effects in lipid profiles without altering feed efficiency rate (FER). HA and BLP co-supplementation in pig feed decreased pH in longissimus thoracis (p<0.05). In addition, sensory characteristics were enhanced when pig feed was supplemented with HA and BLP without causing adverse effects in meat quality. Taken together, addition of HA and BLP in pig feed may produce functional meat products.

채소혼합음료의 섭취가 남자 성인의 혈압, 항산화 효소 및 혈장지질 양상에 미치는 영향 (The Effect of Green Vegetable Drink Supplementation on Blood Pressure, Erythrocyte Antioxidant Enzyme Activities and Plasma Lipid Profiles of Korean Male Subjects)

  • 흡연은 체내에서 활성산소중(ROS)을 증가시키어 혈액내 지질과 지단백의 변화, 산화된 LDL에 의한 내피세포의 손상 등을 유발하므로 심혈관질환 발병의 주요 위험인자로 여겨지고 있으며, 과도한 DNA의 산화는 발암원인이 될 수도 있기 때문에 흡연으로 인한 산화적손상의 방어기전으로는 항산화 영양상태의 개선이 제시되고 있다. 본 연구에서는 비흡연자 20명과 흡연자 19명을 대상으로 채소혼합음료를 8주간 매일 240 mL씩 보충 섭취시킨 후, 혈압과 항산화효소 활성도, 지질양상에 대한 개선효과를 관찰하였다. 채소혼합음료 섭취 후에 대상자의 수축기혈압은 흡연자 5.3 mmHg(5.8% 감소), 비흡연자 6.8 mmHg(7.5% 감소)로 유의적으로 감소하였으며, 이완기혈압은 흡연자에게서 3.9 mmHg(3.1% 감소) 감소하였다. 적혈구 내 항산화효소인 SOD활성은 채소혼합음료 섭취 후에 두 군 모두 증가하였으나, GSH-Px, catalase 활성은 두 군 모두 변화를 보이지 않았다. 혈장지질 양상은 채소혼합음료 섭취 후에 비흡연자의 혈장 TC, LDL-C이 감소하였고 흡연자에서는 혈장 TC수준이 감소하였다. 한편, 간 기능을 살펴본 결과, 채소혼합음료 섭취 후에 비흡연군에서는 GOT, GPT, 흡연군에서는 GPT가 각각 감소하였다. 본 연구에서 대상자에게 채소혼합음료를 섭취케 한 후 혈압, 항산화 효소, 혈장지질 양상 및 간 기능수치 등 여러 지표의 개선효과를 관찰할 수 있었는데 이로부터 하루에 240mL씩의 채소혼합음료의 섭취가 흡연자뿐만 아니라 비흡연자의 산화적손상과 관련된 질병을 예방하기 위한 건강에 유익한 방법 중 하나로 제시될 수 있을 것으로 사료된다. 앞으로는 새로이 개발된 건강기능식품의 생리효과를 검증하기 위한 실험을 수행하고자 할 경우, 정상인뿐 아니라 혈압, 지질양상 및 간 기능 등 각 항목의 고위험자를 대상으로 하는 연구도 광범위하게 수행되어야 하리라고 본다.

Influences of Phytoncide Supplementation on Growth Performance, Nutrient Digestibility, Blood Profiles, Diarrhea Scores and Fecal Microflora Shedding in Weaning Pigs

  • A total of 140 weaning pigs ((Landrace${\times}$Yorkshire)${\times}$Duroc, BW = $6.47{\pm}0.86$ kg) were used in a 5-wk growth trail to determine the effects of phytoncide supplementation on growth performance, nutrient apparent total tract digestibility (ATTD), blood profiles, diarrhea scores and fecal microflora shedding. Pigs were assigned randomly by BW into 5 treatments, dietary treatments were: i) NC, basal diet; ii) PC, NC+0.05% tylosin; iii) EO, NC+0.1% essential oil; iv) PP, NC+0.2% PP (phytoncide with 2% citric acid), and v) PA, NC+0.2% PA (phytoncide). Each treatment had 7 replicate pens with 4 pigs per pen. All pigs were housed in pens with a self-feeder and nipple drinker to allow ad libitum access to feed and water throughout the experimental period. During 0 to 2 wks, supplementation with essential oil and PA decreased (p<0.05) G/F compared with the other treatments. During 2 to 5 wks, supplementation with PA led to a higher (p<0.05) G/F than the other treatments. At 2 wk, ATTD of dry matter (DM) and gross energy (GE) in EO treatment were decreased (p<0.05) compared with NC treatment. Dietary PC treatment improved (p<0.05) ATTD of DM and E compared with the CON group, and PA and PP treatments showed a higher (p<0.05) ATTD of E than that in NC treatment. Pigs fed phytoncide (PA and PP) had a greater (p<0.05) ATTD of DM than those of NC and EO treatments at 5 wk. Moreover, supplementation with phytoncide elevated (p<0.05) the concentration of immunoglobulin (IgG) in blood at 2 wk. The inclusion of EO, PP and PA treatments showed a greater (p<0.05) amount of fecal Lactobacillus compared with CON group. However, no difference (p>0.05) was observed in diarrhea scores among treatments. In conclusion, phytoncide can elevate feed efficiency, nutrient digestibility, and improve the fecal Lactobacillus counts in weaning pigs. Our results indicated that the phytoncide could be used as a good antibiotics alternative in weaning pigs.

여자노인에서 항산화 비타민 보충이 혈장 지질 개선에 미치는 영향 (Effects of Antioxidant Supplementation on Lipid Profiles in Elderly Women)

  • Antioxidant vitamin supplementation focuses one's attention on the prevention of age-related diseases. This study was conducted to investigate the antioxidant status and lipid profiles and to look into the antioxidant vitamin supplementation that affects lipid metabolism in 20 elderly non-smoking Korean women (placebo group: n = 6, vitC suppl: n = 7, vitE suppl: n = 7). Age, height, weight, muscle, percent of fat and WHR were not significantly different among the groups, however $\%$ of fat was above $33\%$ and WHR was above 0.9. And blood pressure of the placebo group was 131.7/81.7 (border line hypertension), that of vitamin C supplement was 141.4/87.1 (hypertension) and that of vitamin E supplement was 151.4/92.9 (hypertension). Although nutrient intakes of all groups were poor, antioxidant status (blood vitamins C, E, A, and beta-carotene) and lipid profile (TG, total-cholesterol, VLDL-cholesterol, LDL-cholesterol, HDL-cholesterol) were normal. For nutritional intervention, the vitamin C supplement group received L-ascorbic acid 1,000 mg, and vitamin E supplement group received d-alpha-tocopherol 400IU for 4 weeks, showing the effects of vitamin E supplementation. Response total cholesterol of HDL-cholesterol (T-Chol/HDL) in vitamin E supplement group was significantly decreased from 4.3 to 3.2. And response LDL-cholesterol of HDL-cholesterol (LDL/HDL) in the vitamin E supplement group was also significantly decreased from 2.6 to 1.7. In addition, after the adjustment for plasma lipids (TG, total cholesterol), plasma vitamin A levels in vitamin E supplement group were significantly increased from 7.89 mg/g to 14.91 mg/g. And systolic blood pressure in vitamin E supplement group was significantly reduced. These results suggested that vitamin E supplementation affects the lipid profiles and blood pressure in elderly non-smoking women. So various nutrition programs must be implemented against age-related diseases and further studies are needed regarding sorts and amounts of antioxidant nutrients and supplementation periods.

노년기 여성들의 혈장 타우린의 농도와 혈장 호모시스테인 및 혈청 지질농도와의 상관관계 (Correlations of Plasma Taurine, Homocysteine and Serum Lipid Levels of Elderly Women)

  • This study was conducted to assess blood components caused by metabolic syndrome increasing in postmenopausal elderly women. The blood samples of these subjects were analyzed to investigate the correlation of plasma taurine levels and plasma homocysteine levels, and serum lipid profiles. The subjects were 33 elderly women($72.8{\pm}4.4$ years). Their mean height, weight and BMI were $150.5{\pm}5.7\;cm$, $57.5{\pm}6.3\;kg$ and $25.4{\pm}2.5\;kg/m^2$. 16 women of this study subjects have been chronic diseases such as hypertension or diabetes. Their fasting blood glucose was $98.2{\pm}24.0\;mg/d{\ell}$, and their plasma total cholesterol (TC), HDL-C, LDL-C, triglyceride(TG) were $216.5{\pm}29.9$, $52.1{\pm}10.7$, $145.7{\pm}27.9$ and $141.2{\pm}59.6\;mg/d{\ell}$, respectively. Their blood lipid profiles were higher than the standard levels of metabolic syndrome, thus these levels of lipid profiles may play a role as risk factors on the elderly person. Plasma taurine level of the subjects was $278.5{\pm}48.1\;{\mu}mol/{\ell}$, and their plasma homocysteine level was $12.8{\pm}2.9\;{\mu}mol/{\ell}$. The concentration of plasma vitamin $B_{12}$ was significantly decreased by aging(p<0.05). The correlation of plasma homocysteine and plasma folate showed significantly negative(p<0.05). Thus, the decreased levels of plasma vitamin $B_{12}$ and folate by aging might affect on the plasma homocyteine concentration acting as a risk factor of cardiovascular diseases for elderly person. The correlation of plasma taurine and hemoglobin, and their platelet showed significantly positive(p<0.05). In conclusion, the diet on the elderly person is one of the important factors to prevent their health from chronic diseases. This study recommends that well balanced diets are needed for elderly person to keep their health and prevent from metabolic syndrome.

Eating habits, obesity related behaviors, and effects of Danhak exercise in elderly Koreans

  • The aims of this study were to evaluate obesity-related dietary behaviors and to determine long-term exercise effects on obesity and blood lipid profiles in elderly Korean subjects. A total of 120 subjects, aged 60-75 yr, were recruited, and obesity-related dietary behaviors were determined. An exercise intervention was conducted with 35 qualified elderly females for 6 months, and body composition and blood lipids were measured 6 times at 4 week intervals. At baseline, mean BMI ($kg/m^2$) was 24.8 for males and 23.1 for females. The females had better eating habits than the males and were more concerned with reading nutrition labels on food products (P < 0.001); they also preferred convenience foods less than the male subjects (P < 0.05). Obese individuals were more likely than overweight or normal weight individuals to misperceive their weight (P < 0.001). Those with a high BMI responded feeling more depressed (P < 0.01), lacking self-confidence (P < 0.01), and feeling isolated (P < 0.01) as well as having more difficulty doing outdoor activities (P < 0.01). After exercise, body fat (%) and WHR were significantly reduced (P < 0.05), while body weight and BMI were also decreased without statistical significance. Total cholesterol and blood HDL were significantly improved (207.1 mg/dl vs. 182.6 mg/dl, HDL: 45.6 mg/dl vs. 50.6 mg/dl, P < 0.05). Other benefits obtained from exercise were improvements in self-confidence (26.4%), movement (22.6%), stress-relief (18.9%), and depression (13.2%). In conclusion, elderly females had better eating habits and were more concerned with nutrition information and healthy diets compared to elderly males. However, misperceptions of weight and obesity-related stress tended to be very high in females who were overweight and obese, which can be a barrier to maintain normal weight. Long-term Danhak practice, a traditional Korean exercise, was effective at reducing body fat (%) and abdominal obesity, and improved lipid profiles, self-confidence, and stress.
