The aim of this study was to evaluate the anti-diabetic effects of the water extract of Lespedeza cuneata (LCW) using rat insulinoma (RIN) m5F cells and streptozotocin (STZ)-induced diabetic rats. The effect of LCW on the protection of pancreatic beta cells was assessed using MTT assay, and nitric oxide production was assessed using Griess reagent. STZ-induced diabetic rats were treated with 100 and 400 mg/kg body weight of LCW for 5 weeks. In results, LCW significantly protected cytokine-induced toxicity and NO production, and increased insulin secretion in RINm5F cells. LCW significantly decreased serum blood glucose, thiobarbituric acid reactive substances (TBARS), blood urea nitrogen (BUN) and advanced glycation end products (AGEs) levels, and renal fibronectin expression in STZ-induced diabetic rats. Also, LCW effectively improved BW loss in STZ-induced diabetic rats. Thus, our results suggest that LCW has a beneficial effect on cytokine-induced pancreatic beta cell damage and biomarkers of diabetic complication in hyperglycemic rats.

Ginseng products on the market show high variability in their composition and overall quality. This becomes a challenge for both consumers and health-care professionals who are in search of high-quality, reliable ginseng products that have a proven safety and efficacy profile. The botanical extract standardization is of crucial importance in this context as it determines the reproducibility of the quality of the product that is essential for the evaluation of effectiveness and safety. This review focuses on the well-characterized and standardized ginseng extract, G115, which represents an excellent example of an herbal drug preparation with constant safety and efficacy within the herbal medicinal products. Over the many decades, extensive preclinical and clinical research has been conducted to evaluate the efficacy and safety of G115. In vitro and in vivo studies of G115 have shown pharmacological effects on physical performance, cognitive function, metabolism, and the immune system. Furthermore, a significant number of G115 clinical studies, most of them double-blind placebo-controlled, have reinforced the findings of preclinical evidence and proved the efficacy of this extract on blood glucose and lipid regulation, chronic obstructive pulmonary disease, energy, physical performance, and immune and cognitive functions. Clinical trials and 50 years of presence on the market are proof of a good safety profile of G115.
Tetrahydroisoquinoline (THI) compounds have various pharmacological actions in the cardiovascular system. Recently, we have synthesized 1-${\alpha}$-naphthylmethyl-6,7-dihydroxy-1,2,3,4-tetrahydroisoquinoline, YS 49. In the present study, we evaluated the effect of YS-49 on positive inotropic and chronotropic action using isolated rat heart and on blood pressure and heart rate using anesthesized rabbit. Vasodilating action was also assessed in isolated rat thoracic aorta. YS 49, concentration-dependently relaxed rat aorta precontracted with phenylephrine (PE, 0.3 ${\mu}$M) and high potassium (high K$\^$+/, 65.4 mM). The 50% inhibitory concentration (IC$\sub$50/) of YS 49 in PE-induced and high K$\^$+/-induded contraction was 5.36 ${\mu}$M and 2.52 ${\mu}$M, respectively. In isolated rat atria, YS 49 increased both heart rate and force, and in anesthesized rabbit it decreased blood pressure but increased heart rate. In addition, to know the mechanism of action of the compound, propranolol, nonselective ${\beta}$-antagonist, and phentolamine, ${\alpha}$-blocker, were used. Furthermore, a comparison with the effect of higenamine, trimetoquinol on the vasodilating action in rat thoracic aorta was also made. The action of YS 49 was inhibited by the presence of propranolo, not pentolamine. These results indicate that cardiotonic and vasodilatory action of YS 49 is attributable, at least in part, for ${\beta}$-receptor stimulation.
Objectives : The purpose of this study is to evaluate antioxidant activities and reducing serum alcohol concentration of extract of Lentinus edodes on the alcohol administered rats. Methods : Antioxidant effect was measured by total phenolic compound and DPPH-radical scavenging activity of extract of Lentinus edodes in vitro. Blood alcohol concentration, aldehyde concentration, malondialdehyde concentration, glutathion concentration were measured in vivo. Results : The extract of Lentinus edodes increased DPPH-radical scavenging activity dose-dependently. The water extract with boiling water showed lower antioxidant activity and phenolic content than 70% ethanol extract in vitro. Blood alcohol concentration was significantly reduced by pre-treatment of ethanol extract of Lentinus edodes. The effect was more significant than commercial product used as a positive control. Conclusions : This study suggest that Lentinus edodes can be a potential nature resource for the management of ethanol toxicity although the mechanism of action involved in the treatment remains to be explored.
본 연구는 2007년 1월부터 2015년 12월까지 본원에서 쯔쯔가무시병으로 진단을 받고 치료 중인 상태에서 뇌경색이 발생하거나, 뇌경색이 생겨 입원 치료를 받던 중 쯔쯔가무시병이 발견된 16명의 환자를 대상으로 하였다. 급성기 뇌경색의 진단은 뇌자기공명영상 및 뇌자기공명혈관영상으로 하였으며 쯔쯔가무시병의 진단은 PCR (Polymerase chain reaction)로 하였다. 일반적인 뇌경색과 쯔쯔가무시병을 동반한 뇌경색의 차이점을 구별하기 위해 내원시 혈압과 체온을 측정하여 보았다. 일반적으로 급성기 뇌경색에서는 혈압이 올라가는데 흥미롭게도 본 연구에서는 수축기 혈압이 130mmHg 미만인 환자가 12명으로 급성기 뇌경색에서 흔히 보이는 혈압 양상과는 다른 모습을 보였다. 쯔쯔가무시병을 동반한 뇌경색의 특징을 알아보기 위해 발병 위치 및 단일 혹은 다발성 뇌경색 여부를 확인하여 보았는데, 앞순환 동맥 영역의 뇌경색 발생 환자가 13명이었으며 뒤순환 동맥 영역의 뇌경색 발생 환자는 3명이었다. 응고장애를 진단하기 위해 트롬보플라스틴시간(Prothrombin Time, PT), 활성화부분트롬보플라스틴시간(activated partial thromboplastin time, aPTT), D-dimer, 섬유소원(fibrinogen), 섬유소분해산물(fibrin degradation product, FDP)를 기록하였다. 뇌경색시 일반적으로 수치가 증가하는 것으로 알려진 D-dimer의 경우 13명의 환자에서 큰 폭의 증가 소견을 보였다. 섬유소분해산물(FDP)는 15명의 환자에서 큰 폭의 증가 소견을 보였다. 쯔쯔가무시병의 병태생리학적 기전은 혈관염으로 알려져 있는데 이로 인한 대뇌혈관의 내피세포 손상 및 증식이 있을 수 있고 그 과정에서 응고장애가 동반되어 뇌경색이 발생할 수 있다. 또는 내피세포 손상 및 증식이 없더라도 혈관염으로 인한 혈관연축이 발생하여 혈관수축이 오며 뇌경색이 발생 할 수도 있다.

The selective targeting of an integrin ${\alpha}_v{\beta}_3$ receptor using radioligands may enable the assessment of angiogenesis and integrin ${\alpha}_v{\beta}_3$ receptor status in tumors. The aim of this research was to label a peptidomimetic integrin ${\alpha}_v{\beta}_3$ antagonist (PIA) with $^{99m}Tc(CO)_3$ and to test its receptor targeting properties in nude mice bearing receptor-positive tumors. PIA was reacted with tris-succinimidyl aminotriacetate (TSAT) (20 mM) as a PIA per TSAT. The product, PIA-aminodiacetic acid (ADA), was radiolabeled with $[^{99m}Tc(CO)_3(H_2O)_3]^{+1}$, and purified sequentially on a Sep-Pak C-18 cartridge followed by a Sep-Pak QMA anion exchange cartridge. Using gradient C-18 reverse-phase HPLC, the radiochemical purity of $^{99m}Tc(CO)_3$-ADA-PIA (retention time, 10.5 min) was confirmed to be > 95%. Biodistribution analysis was performed in nude mice (n = 5 per time point) bearing receptor-positive M21 human melanoma xenografts. The mice were administered $^{99m}Tc(CO)_3$-ADA-PIA intravenously. The animals were euthanized at 0.33, 1, and 2 hr after injection for the biodistribution study. A separate group of mice were also co-injected with 200 ${\mu}g$ of PIA and euthanized at 1 hr to quantify tumor uptake. $^{99m}Tc(CO)_3$-ADA-PIA was stable in phosphate buffer for 21 hr, but at 3 and 6 hr, 7.9 and 11.5% of the radioactivity was lost as histidine, respectively. In tumor bearing mice, $^{99m}Tc(CO)_3$-ADA-PIA accumulated rapidly in a receptor-positive tumor with a peak uptake at 20 min, and rapid clearance from blood occurring primarily through the hepatobiliary system. At 20 min, the tumor-to-blood ratio was 1.8. At 1 hr, the tumor uptake was 0.47% injected dose (ID)/g, but decreased to 0.12% ID/g when co-injected with an excess amount of PIA, indicating that accumulation was receptor mediated. These results demonstrate successful $^{99m}TC$ labeling of a peptidomimetic integrin antagonist that accumulated in a tumor via receptor-specific binding. However, tumor uptake was very low because of low blood concentrations that likely resulted from rapid uptake of the agent into the hepatobiliary system. This study suggests that for $^{99m}Tc(CO)_3$-ADA-PIA to be useful as a tumor detection agent, it will be necessary to improve receptor binding affinity and increase the hydrophilicity of the product to minimize rapid hepatobiliary uptake.
숙취를 해결하기 위해서는 숙취를 직접적으로 유발하는 물질인 아세트알데하이드의 조절이 가장 중요한 인자로 작용한다. 숙잠을 함유하는 복합물인 시험식품(CKDHC)의 섭취로 혈중 아세트알데하이드 농도가 대조식품 대비 통계적으로 유의하게 감소하여 숙취개선 효능을 확인하였다. 특히, 30분부터 각 시간별(mg/L), 혈중 아세트알데하이드 농도-시간곡선하 면적(AUC), 최고 혈중 아세트알데하이드 농도(Cmax)의 감소를 통해 30분부터 빠른 숙취개선 효능을 확인하였다. 본 연구의 주원료로 사용된 숙잠은 Ji 등(2016a)에 따르면 18종의 아미노산 및 무기질을 모두 함유하며, 고단백 식용곤충으로 간에 좋은 주요 아미노산인 Alanine, Arginine 등을 많이 함유하여 간의 효소활성에 긍정적인 영향을 줄 수 있음을 알수있다. 본 연구결과를 통해 숙잠을 함유하는 시험식품이 숙취해소제로 작용할 수 있으며, 특히 음주 전 섭취시 아세트알데하이드를 빠르게 분해하여 30분부터 숙취 개선에 효과가 있음을 입증하였다.
본 연구는 성인 흡연자를 대상으로 NKF 정제를 단기 (4,000 U를 1회 투여)와 장기(2,000 U를 매일 1회 4주 투여) 실험을 통하여 NKF의 혈전용해 개선 및 항 동맥경화효과를 검증하였다. 임상대상자는 20세${\sim}$31세의 건강한 성인으로 하루 흡연량이 하루 1갑 이상인 고혈압 또는 항 혈전증 약을 섭취하지 않는 사람을 선정하여 무작위로 placebo군과 NKF군으로 분류하여 이중맹검법으로 실험을 하였다. NKF와 placebo 정제는 (주)HK바이오텍에서 제공하였다. 단기시험은 placebo군(10명)과 NKF군(29명)을 대상으로 NKF 섭취전과 섭취 1, 2, 4시간에 혈전용해효과를 검토하였고, 장기시험은 placebo군 10명과 NKF군 15명에게 NKF 섭취 전과 섭취 후 매주 혈전용해효과 및 혈압개서효과를 검토하였다. 단기시험에서 fibrin clot 용해능은 placebo군에 비해 NKF군에서는 섭취 후 2시간에 유의적으로 증가하여 4시까지 그 활성이 유지되었다. ECLT도 NKF 섭취 후 시간이 경과함에 따라 placebo군에 비해 현저하게 감소되었으며 aPTT는 섭취 후 4시에 섭취 전에 비해 12.5% 지연시키는 것으로 나타났다. 장기시험에서는 수축기와 이완기 혈압이 placebo군에서는 섭취 4주에 섭취 전에 비해 2.8%와 3.5%로 증가하였고 NKF군에서는 각각 1.3%와 1.0% 감소하여 두 군 모두에서 유의차는 없었다. Fibrin clot 용해능은 NKF군에서 1주에 증가하여 4주에도 높은 수준으로 유지되었고, ECLT는 NKF군은 섭취 전에 비해 1주에 현저하게 감소되어 4주 후까지 유사한 수준으로 유지하여 NKF의 효과를 볼 수 있었지만, placebo군에서는 효과가 없었다. 따라서 NKF는 PT와 aPTT를 개선하여 전 처리 기간 placebo군에 비해 혈액응고시간을 유의적으로 지연시켰다. 또한 NKF 섭취는 TG/HDL-cholesterol을 감소시켜 항 동맥경화 지수를 상승시키는 효과가 있었다. 이상의 결과로 미루어 볼 때 단기와 장기 임상시험에서 NKF는 fibrin clot 용해능을 증가시키고 ECLT를 감소시키며 PT와 aPTT를 지연시켜 혈전 용해능을 상승시키며, TG/HDL-cholesterol을 낮추어 혈중 지질수준을 개선하고 동맥경화를 예방하는데 유익하게 작용할 것으로 사료된다.
본 시험은 인삼부산물 급여 유무에 따른 돼지 생산성, 혈액, 도체 및 육질 특성에 관하여 조사하였다. 시험 동물은 2원 교잡종(LandraceYorkshire) 육성-비육돈 24두를 공시하였으며 개시 체중은 65.81±2.02kg이였다. 기초 사료를 대조구(CON)로 설정하였고, 처리구(GBP)은 인삼 부산물을 기초사료의 lupin 2.5%로 대체하였다. 시험구는 완전임의 배치법으로 암·수 동일한 비율로 각 처리구당 4마리씩 3반복 실시하였다. 생산성에서 일당사료섭취량은 GBP가 CON 보다 유의적으로 낮았다(P<0.0001). 혈장의 생화학적 조성에서는 total protein, blood urea nitrogen, glucose, albumin, calcium 그리고 inorganic phosphate는 각각 GBP가 CON 보다 유의적으로 높은 함량을 나타내었다. 도체 및 육질 특성은 GBP 및 CON간에 유의적인 차이가 나타나지 않았다. 돈육의 총 ginsenoside 함량은 GBP가 CON 보다 유의적으로 높은 함량을 나타내었고 TBRAs는 저장 6일과 13일에 GBP가 CON 보다 유의적으로 낮았다. 이상의 시험 결과를 종합하면, 인삼부산물 급여는 혈장의 화학적 조성의 변화를 가져왔고, 돈육의 ginsenoside의 함량을 증가시키고 지방산패도를 감소시켰는데 이 사실은 인삼부산물이 돼지의 사료첨가제로 이용될 수 있는 가능성을 나타낸다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
