선복화 에탄올 추출물의 급성 독성 연구 (Safety Evaluation of Ethanol Extract of Inulae Flos : Single-dose Oral Toxicity Study in Mice)

  • Objectives : This experiment was designed to assess the single oral toxicity of Ethanol Extract Inulae Flos (IF) ethanol extracts. IF is one of the important herbs to remove phlegmy which is the viscous turbid pathological product that can accumulate in the body, causing a variety of diseases. Nevertheless, there has been a lack of research on the pharmacology toxicity of IF. Methods : In this study, IF was orally administered to 5 weeks ICR mice as an oral dose of 2,000 or 3,000 or 5,000 mg/kg. The condition of the mice was observed for 14 days and their weights were measured every two days. Results : None of the mice died for 14 days. The abnormal clinical symptoms and anatomical signs of toxicity were not found in any treatment groups. The gain of net body weight was observed. There was also no significant difference in the organ weight. The serum biochemistry and hematological analysis showed a decrease in BUN, red blood cells, white blood cells and platelets although within the normal ranges. Conclusions : These results suggest that the 50% lethal dose of IF is more than 5,000 mg/kg. This could be thought that IF is a safe drug without acute toxicity and side effects. However, IF showed some weight loss and change in blood test, so it will need to be careful when using it for high doses.

  • According to the literatual study on the external medical trearment of Menorrhalgia and Leucorrhea for gynecologic condition, the results were as follows. 1. Using external medical treatment for Menorrhalgia is to insert vagina theraphy, fumigation theraphy, to wash vagina theraphy, to insert anus theraphy, to apply hot pack theraphy and use with suppository such as BANSUKHWAN(礬石丸), SASANGZASAN(蛇床子散), KUMBONGHYUNGJU(金鳳衡珠) 2. Using external medical treatment for Leucorrhea is washing and fumigation on vagina, to wash vagina, to insert vagina, cleansing theraphy and use with Suppository such as YONGYEOMGO(龍鹽膏), KAMISASANGJASAN(加味蛇床子散), SASANGSACHUNGSEJE(蛇床子洗劑). 3. ANGELICAE GIGANTIS RADIX (當歸) is in mostly general use for external medical treatment of Menorrhalgia and Leucorrhea, in that order ANSU SEMEN(杏仁), EVODIAE FRUCTUS(吳茱萸), TORILIS FRUCTUS (蛇床子), CORYDALIS TUBER(玄胡索), CINIAMOMI CORTEX(肉桂), CARYOPHYLLI FLOS(丁香), ALUMEN(枯白礬), AQUILARIAE LIGNUM (沈香). 4. The efficacy of medicines to use external medical treatment is as follow to help circulation of blood, to warm spleen and stomach, to warm blood, to warm uterus and remove cold, to remove heat and dry moisture, to down heat-product, to contract bloodvessel, to counteract poison and destory virus, to make energy and blood. 5. KUMBONGHYUNGJU, made same size as cherry and shape vagina medicine, use for Menorrhalgia, menstural irregularity, Leucorrhea, it has the efficacy as follow to remove cold and dry moisture, to warm uterus, to help circulation and romove pain, to down moisture-heat.

생쥐에 있어서 약물의 혈액-뇌 관문 투과성 평가를 위한 간편한 in vivo 방법 (The Simple in Vivo Evaluation Method for Blood-Brain Barrier Permeability of Drugs in Mice)

  • This study compared the permeability of $[^3H]taurine,\;[^3H]phenylalanine,\;and\;[^3H]oxytocin$ through the blood-brain barrier (BBB) in mice and rats with common carotid artery perfusion (CCAP) method that modified internal carotid artery perfusion (ICAP) method. External carotid artery (ECA) was cannulated with coagulating pterygopalatine artery (PPA) in ICAP method, while CCA was cannulated without coagulating PPA in CCAP method. Also, for evaluation of BBB permeability of drugs in mice and rats, we used intravenous injection technique. The results of CCAP method in mice at a perfusion flow-rate of 2 ml/min, the brian volume of distribution $(V_D)$ of $[^{14}C]sucrose,\;[^3H]taurine,\;[^3H]phenylalanine,\;and\;[^3H]oxytocin$ were similar to the result of ICAP method in rats at perfusion flow rate of 4 ml/min. The area under the plasma concentration-time curve and brain uptake of $[^3H]taurine$ by intravenous injection technique, were $65.5{\pm}9.7%ID^*min/ml\;and\;0.515{\pm}0.093%ID/g$, respectively, in mice, and the corresponding values were $8.00{\pm}0.03%ID^*min/ml\;and\;0.052{\pm}0.003%ID/g$ in rats. But the BBB permeability surface-area product of $[^3H]taurine$ was similar between mice and rats. In conclusion, the CCAP method in mice was simple, fast and comparable to ICAP method in rats for drug permeability through the BBB.

Beneficial Cardiovascular Effects Of Hydroalcoholic Extract From Crocus Sativus In Hypertension Induced By Angiotensin II

  • Objectives: Angiotensin II (AngII), a major product of renin-angiotensin system (RAS) has important role in induction of hypertension and antihypertensive effect of several medicinal plant was mediated by effect on this agent. Therefore, this study examined the possible effect of hydroalcoholic extract of Crocus sativus (C. sativus) on hypertension induced by AngII. Methods: Six groups (n = 6) of rats were used as follow: 1) Control, 2) AngII (300 ng/kg), 3) Losartan (Los, 10 mg/kg) + AngII and 4-6) C. sativus extract (10, 20 & 40 mg/kg,) + AngII. The femoral artery and vein were cannulated for recording cardiovascular parameters and drugs administration, respectively. All drugs were injected intravenously (i.v). Los and all doses of C. sativus injected 10 min before AngII. Systolic blood pressure (SBP), mean arterial blood pressure (MAP) and heart rate (HR) were recorded throughout the experiment and those peak changes (${\Delta}$) were calculated and compared to control and AngII. Results: AngII significantly increased ${\Delta}MAP$, ${\Delta}SBP$ and ${\Delta}HR$ than control (P < 0. 01 to P < 0.001) and these increments were significantly attenuated by Los. All doses of C. sativus significantly reduced peak ${\Delta}MAP$, ${\Delta}SBP$, and ${\Delta}HR$ than AngII group (P < 0. 05 to P < 0.001). In addition, peak ${\Delta}MAP$, ${\Delta}SBP$ in doses 10 and 20 were significant than Los + AngII group (P<0.05 to P< 0.01) but in dose 40 only MAP was significant (P<0.05). Peak ${\Delta}HR$ in all doses of C sativus was not significant than Los+ AngII. Conclusion: Regarding the improving effect of the C. sativus extract on AngII induced hypertension, it seems that this ameliorating effect partly mediated through inhibition of RAS.

Vascular rinsing and chilling carcasses improves meat quality and food safety: a review

  • Rinse & Chill® technology (RCT) entails rinsing the vasculature using a chilled isotonic solution (3℃; 98.5% water and a blend of dextrose, maltose, and sodium phosphates) to rinse out the residual blood from the carcass. Infusion of pre-chilled solutions into intact animal carcasses immediately upon exsanguination is advantageous in terms of lowering the internal muscle temperature and accelerating chilling. This technology is primarily used for purposes of effective blood removal, favorable pH decline, and efficient carcass chilling, all of which improve meat quality and safety. Although RCT solution contains some substrates, the pre-rigor muscle is still physiologically active at the time of early postmortem and vascular rinsing. Consequently, these substrates are fully metabolized by the muscle, leaving no detectable residues in meat. The technology has been commercially approved and in continuous use since 2000 in the United States and since 1997 in Australia. As of January 2022, 23 plants have implemented RCT among the 5 countries (Australia, US, Canada, New Zealand, and Japan) that have evaluated and approved RCT. All plants are operating under sound Sanitation Standard Operation Procedures (SSOP) and a sound Hazard Analysis Critical Control Point (HACCP) program. No food safety issues have been reported associated with the use of this technology. RCT has been adapted by the meat industry to improve product safety and meat quality while improving economic performance. Therefore, this review summarizes highlights of how RCT technically works on a variety of animal types (beef, bison, pork, and lamb).

조직 혈액응고인자에 대한 식용버섯류의 저해활성 (Inhibitory Activity of Edible Mushrooms on the Tissue Thromboplastin (Tissue Factor))

  • 식용버섯류를 대상으로 혈액응고계를 활성화시키는데 주된 역할을 하는 것으로 알려진 단백질인 tissue factor에 대한 저해활성을 시험관내에서 측정하였으며 몇 가지 유기용매를 이용하여 추출분획을 조제하고 이들의 TF에 대한 저해활성을 비교 검색하여 다음과 같은 결과를 얻었다. 검색된 버섯류의 메탄올 추출물들은 모두 TF에 대한 저해활성을 나타내었으며 특히 표고버섯은 비교적 강한 저해활성을 나타내었고 양송이버섯과 영지버섯 역시 TF 저해활성을 갖는 것으로 나타났다. 양송이버섯의 메탄올 추출물의 $IC_{50}$은 in vitro에서 $1.30\;{\mu}g$이었고 영지버섯은 $4.32\;{\mu}g$, 표고버섯은 $0.25\;{\mu}g$으로 각각 조사되었다. 유기용매를 이용하여 용매분획을 실시하고 각 용매분획의 TF 저해활성을 측정한 결과 표고버섯의 에틸아세테이트 분획에서 강한 TF 저해활성이 확인되었고 영지버섯의 에틸아세테이트 분획에서도 TF에 대한 저해활성이 나타났으며 표고버섯의 에틸아세테이트 분획에서 얻은 결정성 물질 역시 TF를 저해하는 활성을 나타내었다. 모든 버섯의 물층에서는 활성이 확인되지 않았으며 영지버섯의 에틸아세테이트 분획의 $IC_{50}$$107\;{\mu}g$, 표고버섯의 경우 $770\;{\mu}g$의 값을 나타냈고 표고버섯의 에틸아세테이트 분획으로부터 얻은 결정성물질의 $IC_{50}$은 10.2 mg이었다.

Evaluation of FSP (Fermented Soy Protein) to Replace Soybean Meal in Weaned Pigs: Growth Performance, Blood Urea Nitrogen and Total Protein Concentrations in Serum and Nutrient Digestibility

  • A total of one hundred and forty four weaned pigs with an average BW of $8.09{\pm}0.05$ kg were used in a 28 day study to investigate the effects of fermented soy protein on growth performance, blood urea nitrogen and total protein concentrations in serum and nutrient digestibility in weaner pigs. Pigs were blocked by initial body weight and randomly allocated to one of four dietary treatments in a randomized complete block design. There were six replications per treatment. Dietary treatments included: SBM (corn-soybean meal basal diet), F 5, 10 and 15 (fermented soy product was used at 5, 10 and 15% to replace soybean meal in basal diet, respectively). ADG (average daily gain) and ADFI (average daily feed intake) were not affected (p>0.05) by dietary treatments during the entire 4-wk study period. There were linear increments in feed efficiency (p<0.01) as the dietary FSP level increased during the entire feeding period. No significant differences were observed for dry matter and nitrogen digestibility during the experimental period (p>0.05). Digestibilities of histidine, lysine and methionine were increased as the FSP level increased (linear effect, p<0.05, p<0.01). Among non-essential amino acids, alanine, glutamic acid, serine, tyrosine and total non essential amino acid digestibilities were increased linearly (p<0.05, p<0.01). There were quadratic effects in protein digestibility (p<0.05). Total amino acid digestibility of the F15 diet was improved compared with the F5 diet (p<0.05). There were no significant differences in fecal consistency score among the treatments (p>0.05). At the end of experiment, BUN (blood urea nitrogen) concentration was increased as the FSP level increased (linear effect, p<0.01) and total protein concentration was lowest (p<0.05) for pigs fed the SBM diet among treatments. In conclusion, the feeding of 10 or 15% FSP to nursery pigs improved feed efficiency, amino acid digestibility and blood urea nitrogen and total protein concentrations in blood.

자발성 고혈압 흰쥐에서 찐콩과 청국장 분말의 혈압 및 지질대사 개선 효과 (Improving Effect of Powders of Cooked Soybean and Chongkukjang on Blood Pressure and Lipid Metabolism in Spontaneously Hypertensive Rats)

  • 자발성 고혈압 흰쥐(SHR)에서 찐콩과 청국장 분말이 혈압 및 지질대사에 미치는 효과를 조사하여 가열처리 및 발효과정을 거친 콩제품의 항동맥경화 효과를 평가하고자 하였다. 식 이의 단백질 급원으로써 카제인, 찐콩 분말, 그리고 청국장분말을 단백질 함량으로 각각 18% 되게 첨가한 semi-pu-rified diet를 SHR(n=10, 숫컷)에 6주간 공급한 후 혈압, 혈장과 간의 지질농도, 간의 HMG-CoA reductase 활성 및 분변의 담즙산 배설능을 분석하였다. 그 결과, 카제인 대조군에 비교하여 찐콩과 청국장 섭취군의 체중증가량은 차이를 보이지 않았으며, 식이효율은 오히려 높게 나타났다(p<0.05). 찐콩 및 청국장의 장기간 섭취는 자발성 SHR의 가령에 따른 혈압상승을 효과적으로 억제하였으며(p<0.05) 청국장은 찐콩보다 우수한 혈압 강하효과를 보였다. 혈중 콜레스테롤 함량은 실험군간에 유의적인 차이가 없었으나 중성지방의 농도는 대조군에 비해 찐콩 섭취군에서 다소 낮았으며 청국장 섭취군에서 유의적으로 낮았다(p<0.05). LDL-콜레스테롤 농도는 대조군에 비해 찐콩 및 청국장 섭취군에서 유의적으로 낮았으며 항동맥경화인자로 알려진 HDL-콜레스테롤 농도는 유의적으로 높게 나타났다(p<0.05). 따라서 동맥경화 지수는 대조군에 비해 찐콩과 청국장군에서 유의적으로 낮았으며, 청국장군에서 가장 낮은 동맥경화지수를 보였다(p<0.05). 간의 총지질과 중성지방 함량은 대조군에 비해 찐콩 및 청국장 섭취군에서 유의적으로 낮게 나타나(p<0.0.5) 혈장에서의 결과와 일치하였다. 간의 HMG-CoA reductase 활성은 찐콩과 청국장군에서 유의적으로 낮았다 (p<0.05). 분변 중 총지질량은 세 군간에 유의적인 차이가 없었으며 콜레스테롤 배설량은 찐콩군에서 가장 높았으며, 분변으로의 담즙산 배설능은 청국장군에서 유의적으로 높았다(p<0.05). 이상의 결과는 단백질 급원으로 카제인을 대체한 찐콩과 청국장의 섭취는 동맥경화 위험인자인 혈압과 고지혈증을 개선함으로써 순환기계질환을 예방하는데 도움을 줄 수 있음을 제시하였다.

Effect of Saccharomyces cerevisiae Fermentation Product on Lactation Performance and Lipopolysaccharide Concentration of Dairy Cows

  • To evaluate lactation performance and changes in plasma and fecal lipopolysaccharide (LPS) concentrations in response to the supplementation of Saccharomyces cerevisiae fermentation product (SC), two dairy farms were selected. On each farm, 32 cows in early to mid lactation (21 to 140 DIM) were blocked by parity and days in milk (DIM), and randomly assigned to one of the two treatments within block (Control or 56 g SC/cow/d). Effect of SC on lactation performance (daily) and changes in blood and fecal LPS level were examined on d 0 and 28 of supplementation. The results showed that SC supplementation increased lactation performance of dairy cows on both farms. On Farm 1, milk production, 3.5% fat corrected milk (FCM), and yield of milk fat and protein were greater (p<0.01) for cows supplemented with SC. Supplementation of SC increased percentage milk fat (p = 0.029) from 81 to 110 DIM. There was no significant effect (p>0.05) of SC supplementation on percentage of milk protein, dry matter intake and feed efficiency. On Farm 2, cows supplemented with SC had a greater (p<0.05) milk yield, percentage of milk fat and milk protein, yield of milk fat and protein, 3.5% FCM and feed efficiency. Supplemental SC had no effect on LPS concentrations in feces (p>0.05) while it trended to reduce (p = 0.07 or 0.207) the concentration in plasma. The results indicate that supplemental SC can increase lactation performance of dairy cattle and has potential for reducing plasma LPS concentration.

Effects of calcium intake, milk and dairy product intake, and blood vitamin D level on osteoporosis risk in Korean adults: analysis of the 2008 and 2009 Korea National Health and Nutrition Examination Survey

  • This study was performed to determine the effects of dietary calcium (Ca) intake, milk and dairy product intake, and serum vitamin D level on bone mineral density. The survey data from the 2008-2009 Korea National Health and Nutrition Examination Survey (KNHANES) for adults (3,819 males, 5,625 females) aged > 20 years were examined; osteoporosis was defined according to the standards for Asian populations (T-score < -2.5). The risk for osteoporosis significantly decreased as Ca intake increased; this effect persisted (quartile 4 vs. quartile 1 of Ca intake: odds ratio [OR] 0.66; 95% confidence interval [CI]: 0.50-0.87) even after adjustment for gender, age, and other factors (body mass index, serum vitamin D, menstruation, female hormone intake, menopausal status, and the number of days per week of muscular strength exercise). Additionally, the risk for osteoporosis significantly decreased as the Ca/P ratio increased (quartile 4 vs. quartile 1: OR 0.76; 95% CI: 0.58-0.98). The degree of risk was 0.96 (0.66-1.38) in those who consumed < 1 portion of milk or dairy products daily, and 0.71 (0.53-0.96) in those who consumed > 1 portion per day, compared with those who had zero intake. The risk for osteoporosis significantly decreased as the serum 25(OH) vitamin D level increased. From these results, we advocate an increase in Ca, milk, and dairy product intake, and that serum 25(OH) vitamin D levels be maintained within the normal range, for the maintenance of bone health and the prevention of osteoporosis in adults.
