Human umbilical cord blood cells(HUCBC) are rich in mesenchymal progenitor cells, endothelial cell precursors and hematopoietic cells. HUCBC have been used as a source of transplantable stem and progenitor cells. However, little is known about survival and development of HUCBC transplantation in the CNS. Estrogen has a neuroprotective potential against oxidative stress-induced cell death so has an effect on reducing infarct size of ischemic brain. We investigated the potential use of HUCBC as donor cells and tested whether estrogen mediates intravenously infused HUCBC enter and survive in ischemic brain. PKH26 labeled mononuclear fraction of HUCBC were injected into the tail vein of ischemic OVX rat brain with or without $17\beta$-estradiol valerate(EV). Under fluorescence microscopy, labeled cells were observed in the brain section. Significantly more cells were found in the ischemic brain than in the non-ischemic brain. HUCBC transplanted into ischemic brain could migrate and survive. Some of cells have shown neuronal like cells in hippocampus, striatum and cortex tissues. These result suggest that estrogen reduces ischemic damage and increases the migration of human umbilical cord blood cells. Stem cell therapy using mesenchymal stem cells(MSCs) transplantation have been paid attention because of their powerful proliferation and pluripotent differentiating ability. Although umbilical cord blood (UCB) is well known to be a rich source of hematopoietic stem cells with practical and ethical advantages, the presence of mesenchymal stem cells (MSCs) in UCB has been controversial and it remains to be validated. In this study, we examine the presence of MSCs in UCB harvests and the prevalence of them is compared to that of endothelial progenitor cells. For this, CD34+ and CD34- cells were isolated and cultured under the endothelial cell growth medium and mesenchymal stem cell growth medium respectively. The present study showed that ESC-like cells could be isolated and expanded from preterm UCBs but were not acquired efficiently from full-terms. They expressed CD14-, CD34-, CD45-, CD29+, CD44+, CD105+ cell surface marker and could differentiate into adipogenic and osteogenic lineages. Our results suggest that MSCs are fewer in full-term UCB compared to endothelial progenitor cells.
It is important to coordinated interaction among neurons, astrocytes and endothelial cells to maintain the function of brain. To study their regulatory mechanisms in vitro system, the co-culture system among the isolated cells from brain may be needed. However, the method for purifying brain microvascular endothelial cells (BMEC) far culture have not established yet. In this study, the proper culture methods of mice cells using two different strains, CD1 and C57BL6, to obtain the pure and plentiful endothelial cells were described. The flatted-round forms of CD1 endothelial cells grew on the collagen-IV coating plates, while the purified cells from C57 mice preferred type collagen-I dishes for their growth. Both cells displayed anti-PECAM-1 (CD31) and von Willebrand Factor immune-reactivity. These results indicated that different coating materials not only improve attachment of isolated cells but also promoting growth of cells, suggesting that this method of purifying murine Brain microvascular endothelial cells (BMEC) provides a suitable model to investigate blood-brain-barrier (BBB) properties within neurovascular unit in vitro.
Background: It is reported that the percentage of smudge cells in the blood smear could be a prognostic indicator in chronic lymphocytic leukemia. However, the clinical significance of smudge cells in other hematological malignancies, solid tumors or non-malignant diseases is less clear. Hence, this study was conducted to survey the clinical significance of smudge cells in hematological cancers and other disorders. Materials and Methods: From January to November, 2015, the clinical data of patients who received blood examination with differential counts for clinical purpose and were found to have smudge cells in the peripheral blood film in Far Eastern Memorial Hospital were selected. The percentage of smudge cells and patient outcomes were evaluated for further univariate and survival analyses. Results: A total of 102 patients with smudge cells in their blood smears were included. Smudge cells were frequently presented in out-of-hospital cardiac arrest (OHCA; n=30), infections (n=23), hematological cancers (n=23) and solid cancers (n=10). There was no relationship between the percentage of smudge cells and the patient mortality in all diseases (OR: 1.08, 95% CI: 0.47-2.48, P=1.000) as well as the OHCA group (OR: 1.91, 95% CI: 0.38-9.60, P=0.694). It was observed that in patients with all cancers with the percentage of smudge cells less than 50% had a lower mortality rate in comparison with those who had the percentage of smudge cells of 50% or more (OR: 22.29, 95% CI: 2.38-208.80, P<0.001). Additionally, it was seemingly that patients with smudge cells of 50% or more had a lower survival rate than those with smudge cells less than 50% in all cancers with follow-up at 2-month intervals, but without statistical significance (P=0.064). Conclusions: Our survey indicated that in all cancers, those who had higher percentage of smudge cells were prone to have poor outcomes when compared with the subjects with lower percentage of smudge cells. This finding was quite different from the results of previous studies in which the race-ethnicity of most study populations was non-Asian; hence, further investigations are required. Besides, there was no apparent association of the percentage of smudge cells with patient outcomes in all diseases, including OHCA.
암 환자들을 대상으로 하는 항암치료법은 나노입자, 폴리머 중합체, 지질, 리포솜 등을 치료 전달체로 이용하여 항암치료를 진행하는 방법들이 주로 활발하게 사용되고 있다. 이러한 전달체는 항암 치료제를 직접 암세포로 정확하게 표적 운반하는 정확성, 정확하게 운반한 후 선택적으로 항암치료제를 방출해야하는 유출제어, 다른 일반 세포들을 약물로부터 보호하는 기능 등을 동시에 가지고 있어야 하지만, 대부분 항암약물의 독성에 기인한 부작용이 발생하고 있다. 겸형적혈구는 암세포주변 혈관세포와 멤브레인 표면에 존재하는 리셉터 사이에의 점착성이 존재하며 추가적 생화학처리 없이 암세포 주변에 표적화가 가능함을 보인다. 또한, 암세포 주변의 혈관의 구조적 변화특성은 겸형적혈구의 중합화 반응을 증가시킨다. 따라서 본 논문에서는 겸형적혈구를 이용한 새로운 항암치료법의 효과를 정량적 혈관분석 방법을 통해 제시하고자 한다.
To ascertain the effects of hair dyeing application on oxidative stress in human, a mixture of permanent black colored hair dye with the same amount of oxidant containing 6% hydrogen peroxide was used. A hair dyeing with contacting the scalp (conventional dyeing) and a hair dyeing with 3 to 4mm away from the scalp (alternative dyeing) were applied to each 15 young healthy women. Blood was taken from the brachial vein at two sampling times, just before and 6 hours alter the hair dyeing, and antioxidant enzyme activities and antioxidant contents were measured in red blood cells. After dyeing, malondialdehyde(MDA) contents for conventional dyeing group was shown to a tendence of more increased than alternative dyeing group. After dyeing, reduced glutathione (GSH) contents for conventional dyeing group was shown to a tendence of more decreased than alternative dyeing group. After dyeing, superoxide dismutase (SOD) and catalase (CAT) activities were significantly decreased in conventional dyeing group (p < 0.01), however, SOD and CAT activities were not significantly decreased in alternative dyeing group. After dyeing, there was no significant decrease in glutathione peroxidase (GPX) activity both for conventional dyeing group and alternative dyeing group. Therefore, after dyeing, the degree of oxidative stress in red blood cells for alternative dyeing group was appeared to be lower than conventional dyeing group.
In this paper, a simple nondestructive technology is used to investigate unstained biological blood cells in three dimensions (3D). The technology employs a reflective phase-only spatial light modulator (SLM) for displaying the phase hologram of the object being tested, and a Fourier lens for its reconstruction. The phase hologram is generated via superposing a digital random phase on the 2D image of the object. The phase hologram is then displayed by the SLM with 256 grayscale levels, and reconstructed by a Fourier lens to present the object in 3D. Since noise is the main problem in this method, the windowed Fourier filtering (WFF) method is applied to suppress the noise of the reconstructed object. The quality of the reconstructed object is refined and the noise level suppressed by approximately 40%. The technique is applied to objects: the National Institute of Standards (NIS) logo, and a film of unstained peripheral blood. Experimental results show that the proposed technique can be used for rapid investigation of unstained biological blood cells in 3D for disease diagnosis. Moreover, it can be used for viewing unstained white blood cells, which is still challenging with an optical microscope, even at large magnification.
목 적 : 조직적합 항원의 불일치로 인하여 골수이식을 할 수 없는 경우에 점점 더 제대혈이 사용되고 있다. 그러나 제대혈의 조혈모세포의 수가 적기 때문에 이를 증가시킬 대책이 필요한 바, 여러 성장인자를 조합하여 체외증폭하여 말초혈의 체외증폭과 비교하였다. 방 법 : 저자들은 제대혈 및 말초혈로부터 분리한 CD34+ 세포를 혈청이 아닌 배양체에서 체외 증폭하여 비교하였다. Miltenyi 방법으로 분리한 CD34+는 조혈성장인자들과 함께 체외 증폭 시켰다. 증폭 당일, 4일 후, 7일 후 및 14일에 증폭된 세포를 가지고 burst-forming units of erythrocytes (BFU-E), colony-forming units of granulocytes and monocytes (CFU-GM) 및 colony-forming units of megakaryocytes (CFU-Mk)의 생성 능력을 알아보았다. 결 과 : 말초혈에 비하여 제대혈로부터 분리한 CD34+ 세포의 증폭 능력이 2배로 컸다. 체외에서7일 및 14일 동안 증폭된 제대혈이 더 많은 BFU-E를 생성하였고, 4일 및 7일 동안 증폭된 제대혈이 더 많은 CFU-Mk를 생성하였다. 결 론 : MGDF, FL 및 IL-3를 포함한 성장인자의 자극 하에서 제대혈의 체외 증폭이 더 많은 BFU-E 및 CFU-Mk를 생성하였으므로, 이를 이용한 체외 증폭을 시도하는 것의 가능성을 시사하고 있다.
In the view of oriental medicine, the liver is the general of the army in its function of protecting against the enemy. So this concept is very closely associated to the immunological function. Its relations with immunological function are as follows. 1. The liver produces most of the proteins and converts them with hepatocytes, composes 80% in total reticuloendothelial system with Kuffer cells & endothelial cells and has typical structure of sinusoidal vessels closely related with the blood system. 2. The liver plays an important role in innate immunity with Kuffer cells as well as with the molecules that the liver produces, related to complementary systems. 3. In the embryonic period, the liver is associated with immune associated cell growth and their maturation. After birth, it is associated with removing old red blood cells and with systematically modulating immune system through hormone metabolism. 4. The liver controls the autoimmune disease resulting from immune complex by removing molecules like immune complex. 5. In the processing of blood 19A from the digestive system, the liver has an important role in protecting the body from unnecessary immune responses. 6. In the oriental medical view, liver plays a major role in the immune function by storing blood and dispersing stagnated hepatoqi with the help of the kidneys and spleen.
The locomotor responses of human peripheral blood neutrophils and lymphocytes were measured by the change from spherical to polarized shapes in the presence of endotoxins (lipopolysaccharide, LPS) of enteric pathogens: S. dysenteriae type 1, V. cholerae Inaba 569B, S. typhimurium, and K. pneumoniae. We reported earlier that these endotoxins are chemotactic factors for the neutrophils since they stimulated cell polarization within a few minutes of incubation. Endotoxins had an inhibitory effect upon neutrophil phagocytosis of opsonized yeast and the cells engulfed fewer yeasts. Interestingly, endotoxins increased neutrophil adhesion to clean glass surfaces, but stimulated the cells to exhibit increased random locomotion (chemokinesis) through cellulose nitrate filters and show an enhanced ability to reduce nitroblue tetrazolium (NBT) dye. Unlike neutrophils, lymphocytes direct from blood do not show polarized morphology towards chemotactic factors but the cells acquire locomotor capacity during 24-72 h culture with mitogens such as phytohemagglutinin (PHA), phorbol myristate acetate or concanavalin A. Stimulation of blood lymphocytes with endotoxins did not induce cell polarization in short-term but long-term culture resulted in an increase in the proportion of polarized cells that acquired locomotor morphologies. The majority of these cells were identified as esterase negative B-lymphocytes that migrated through filters. Despite the optimum time of incubation for each of these cell types being different, we found that lymphocytes respond to much lower concentrations of endotoxins than the neutrophils. These findings suggest that endotoxins of enteric pathogens modulate the functions of human blood neutrophils and lymphocytes.
