Epidemiological Studies of Rice Blast Disease Caused by Pyricularia oryzae Cavara I. Measurement of the Amount of Spores Released from a Single Lesion (벼 도열병의 역학적 연구 I. 단일병반으로부터 포자이탈량 조사)

  • Four types of spore trap (Kim's original, improved Kim's original, Yoshino's original and mixed type of Kim's and Yoshino's original) were evaluated for their efficacy to "estimate the amount of spores released from leaf blast lesions under the natural conditions. It was found that all four types had one or two defects in allowance for adequate sporulation/release, spore catch or spore counting. Thus, an improved type of spore trap was devised considering that it could cover the defects mentioned above. As a result, newly developed spore trap was quite satisfactory in above mentioned aspects and it could be used for pursuit of spore release phase under the natural conditions.

Expression Patterns of Transposable Elements in Magnaporthe oryzae under Diverse Developmental and Environmental Conditions

  • The genome of the rice blast fungus Magnaporthe oryzae contains several types of transposable elements (TEs), and some TEs cause genetic variation that allows M. oryzae to evade host detection. We studied how five abundant TEs in rice pathogens, Pot3, Pot2, MAGGY, Line-like element (MGL) and Mg-SINE, are expressed under diverse conditions related to growth, development, and stress. Expression of Pot3 and Pot2 was activated in germinated conidia and mycelia treated with tricyclazole. Retrotransposon MAGGY was highly expressed in appressoria and tricyclazole-treated mycelia. MAGGY and Pot2 were also activated during the early and late stages of perithecia development. MGL was up-regulated in conidia and during conidial germination but not during appressorium formation. No noticeable expression of Mg-SINE was observed under most conditions. Our results should help investigate if and how condition-specific expressions of some TEs contribute to the biology and evolution of M. oryzae.

  • Magnaporthe grisea (Hebert) Barr (anamorph: Pyricularia grisea) is a typical heterothallic Ascomycete and the causal agent of rice blast, one of the most destructive diseases on rice (Oryza sativa L.) worldwide. The interactions between cells of the pathogen and those of the host involve a complex of biological influences which can lead to blast disease. The early stages of infection process in particular may be viewed as a sequence of discrete and critical events. These include conidial attachment, gemination, and the formation of an appressorium, a dome-shaped and melanized infection structure. Disruption of this process at any point will result in failure of the pathogen to colonize host tissues. This may offer a new avenue for developing innovative crop protection strategies. To recognize and capture such opportunities, understanding the very bases of the pathogenesis at the cellular and molecular level is prerequisite. Much has been learned about environmental cues and endogenous signaling systems for the early infection-related morphogenesis in M. grisea during last several years. The study of signal transduction system in phytopathogenic filamentous fungi offers distinct advantages over traditional mammalian systems. Mammalian systems often contain multiple copies of important genes active in the same tissue under the same physiological processes. Functional redundancy, alternate gene splicing, and specilized isoforms make defining the role of any single gene difficult. Fungi and animals are closely related kingdoms [3], so inferences between these organisms are often justified. For many genes, fungi frequently possess only a single copy, thus phenotype can be attributed directly to the mutation or deletion of any particular gene of interest.

Invesigation of Functional Roles of a Protein Kinase in a Fungal Plant Pathogen, Magnaporthe oryzae

  • The rice blast disease caused by of Magnaporthe oryzae is one of the most destructive diseases of rice. By the microarray analysis, we profiled expression changes of genes during conidiation and found out many putative genes that are up-regulated. Among those, we first selected MGG_06399 encoding a dual-specificity tyrosine-regulated protein kinase (DYRK), homologous to YAK1 in yeast. To investigate functional roles of MoYAK1, We made ${\Delta}Moyak1$ mutants by homology dependent gene replacement. The deletion mutant showed a remarkable reduction in conidiation and produced abnormally shaped conidia smaller than those of wild type. The conidia form ${\Delta}Moyak1$ were able to develop a germ tube, but failed to form apppressoria on a hydrophobic coverslip. The ${\Delta}Moyak1$ formed appressria on a hydrophobic cover slip when exogenous cAMP was induced, but the appressoria shape was abnormal. The ${\Delta}Moyak1$ also formed appressoria abberent in shape on onion epidermis and rice sheaths and failed to penetrate the surface of the plants. These data indicate that MoYAK1 is associated with cAMP/PKA pathway and important for conidiation, appressorial formation and pathogenic development in Magnaporthe oryzae. Detailed characterization of MoYAK1 will be presented.

  • Recently, the problem of air pollution is drawing attention as a social problem worldwide. Particularly, fine dust is the biggest issue among air pollutants, and it is analyzed that fine dust is generated from air pollutants such as burning fossil fuels such as petroleum or coal, or exhaust gases from automobiles. In addition, yellow dust originating from China adjacent to Korea flows into the Korean Peninsula in a western wind, causing the concentration of fine dust to deteriorate. Fine dust is a harmful substance to the human body such as asthma or respiratory disease, and awareness of the risk is also increasing to a degree designated as a primary carcinogen. In this study, as a method for improving the indoor air quality, the Properties of the matrix according to the mixing ratio of powdered activated carbon based on blast furnace slag are reviewed. The flexural strength and compressive strength were measured, and a fine dust concentration measurement experiment will be conducted later.

Analysis of the Distribution of Rice Blast Pathogens in High-Altitude North Korea Border Areas and Domestic Rice Cultivars (고위도 북한 접경지역과 국내 벼도열병균 레이스 분포 분석)

  • To explore the distribution and the resistance reaction of rice blast pathogens that may occur in North Korea, rice blast pathogens in the North Korean border regions of Dandong and Yeon-gil in China and the North Korean border region of Cheorwon in South Korea were analyzed. In addition, comparative analysis was conducted with rice blast pathogen in Suwon and Jeonju, inland regions of South Korea. Resistance reactions above average were observed in monogenic rice lines (IRBLzt-T, IRBL9-W, IRBL20-IR24, and IRBLta-CP1) in Jeonju, Suwon, and Cheorwon from 2018 to 2020. In Dandong and Yeon-gil, the monogenic lines IRBLz5-CA, IRBL12-M, and IRBL19-A consistently showed resistance reactions for three years. Notably, IRBL19-A exhibited strong resistance. Race distribution analysis in South Korea indicated a shift from KI to KJ dominance from 2018 to 2020, while in the North Korean border regions of Dandong and Yeon-gil, the KI race was dominant in 2021 and 2022. The race distribution of rice blast pathogens in China's North Korean border regions differed significantly from that in South Korea.

Current Status and Future Prospect of Plant Disease Forecasting System in Korea (우리 나라 식물병 발생예찰의 현황과 전망)

  • Disease forecasting in Korea was first studied in the Department of Fundamental Research, in the Central Agricultural Technology Institute in Suwon in 1947, where the dispersal of air-borne conidia of blast and brown spot pathogens in rice was examined. Disease forecasting system in Korea is operated based on information obtained from 200 main forecasting plots scattered around country (rice 150, economic crops 50) and 1,403 supplementary observational plots (rice 1,050, others 353) maintained by Korean government. Total number of target crops and diseases in both forecasting plots amount to 30 crops and 104 diseases. Disease development in the forecasting plots is examined by two extension agents specialized in disease forecasting, working in the national Agricul-tural Technology Service Center(ATSC) founded in each city and prefecture. The data obtained by the extension agents are transferred to a central organization, Rural Development Administration (RDA) through an internet-web system for analysis in a nation-wide forecasting program, and forwarded far the Central Forecasting Council consisted of 12 members from administration, university, research institution, meteorology station, and mass media to discuss present situation of disease development and subsequent progress. The council issues a forecasting information message, as a result of analysis, that is announced in public via mass media to 245 agencies including ATSC, who informs to local administration, the related agencies and farmers for implementation of disease control activity. However, in future successful performance of plant disease forecasting system is thought to be securing of excellent extension agents specialized in disease forecasting, elevation of their forecasting ability through continuous trainings, and furnishing of prominent forecasting equipments. Researches in plant disease forecasting in Korea have been concentrated on rice blast, where much information is available, but are substan-tially limited in other diseases. Most of the forecasting researches failed to achieve the continuity of researches on specialized topic, ignoring steady improvement towards practical use. Since disease forecasting loses its value without practicality, more efforts are needed to improve the practicality of the forecasting method in both spatial and temporal aspects. Since significance of disease forecasting is directly related to economic profit, further fore-casting researches should be planned and propelled in relation to fungicide spray scheduling or decision-making of control activities.

Viruses Associated with Fig Mosaic Disease in Different Fig Varieties in Montenegro

  • Symptoms of fig mosaic disease have been noticed on leaves of fig (Ficus carica) for several decades, in Montenegro. In 2014, leaf samples were collected from trees of six fig cultivars in a plantation located in the main fig-producing area of Montenegro, to study the disease. After RNA isolation, samples were tested by RT-PCR for detection of nine fig viruses and three viroids. Four viruses were detected: fig leaf mottle-associated virus 1 (FLMaV-1), fig mosaic virus (FMV), fig mild mottle-associated-virus (FMMaV) and fig badnavirus 1 (FBV-1). Most of the viruses were present in mixed infections. The amplicons of the viruses were directly sequenced from both directions. A BLAST search of these sequences revealed sequence identities with their closest counterparts at GenBank of 92, 97, 92 and 100%, for FLMaV-1, FMV, FMMaV and FBV-1, respectively. Different responses in symptom expression due to the various virus combinations detected have been demonstrated. Variety $Su{\check{s}}ilica$ had the least symptom expression, with only one virus (FBV-1) found. Considering that the production of figs in Montenegro is increasing and has a substantial relevance in this geographic location, the results indicate that more attention should be given to improving the phytosanitary condition of fig trees in the country.

Survey of Major Disease Incidence of Rice in Korea during 1999-2008 (1999-2008년도 벼 주요 병해 발생 개황)

  • During the period from 1999 to 2008, we surveyed the major diseases incidence of rice in Korea. Leaf blast showed 0.85% of the diseased leaf area in 2000 and then consistently decreased to 0.02% by 2008. However, panicle blast irregularly appeared by the years depending on weather conditions for disease development from heading to milk stage of rice. Diseased lesion height of sheath blight had the lowest (10.1%) incidence in 2001 and the highest (21.4%) incidence in 2007. A negative corelation ($r=-0.88^*$) was measured between diseased lesion height of sheat blight and sunshine hours from mid- to late August. In Chunbuk, Gyeongbuk, and Gyeongnam provinces, brown spot occurrence declined since 2001, however, in Chungnam, Jeonbuk, and Jeonnam provinces, it increased since 2005. False smut drastically decreased after showing the highest incidence of 7.59% in panicles in 2000. The correlation coefficient between diseased panicles of false smut and rainfall during the late August was calculated as $0.94^{**}$. Bacterial leaf blight severely increased in the chronically infested fields in Jeonnam, Jeonbuk, Gyeongnam, and Chungnam province since 2003.

Investigation on Disease Incidence and Yield of Rice Cultivars for Use in Processing of Eco-friendly-grown Germinated Brown Rice (친환경 발아현미 생산에 적합한 병 저항성 및 생산성이 우수한 벼 품종선발)

  • In order to select the optimum rice cultivars for the use in processing of eco-friendly-grown germinated brown rice (GBR), disease incidences and yields in rice cultivars bred in National Institute of Crop Science were investigated in eco-friendly paddy fields (Gokseong, Jeonnam Province, Korea) during the years 2009-2011. The incidences of rice sheath blight and blast on the cultivar Samgwang were higher than those on one reference cultivar Chucheong, but much lower than those on the other reference cultivar Gosihikkali. The cultivars Keunnun and Haiami selected as special rices showed disease incidences and yields similar to the reference rice cultivars Chucheong and Gosihikkali. These results indicated that the cultivars Samkwang, Keunnun, and Haiami can be selected as rice cultivars for the use of in processing of eco-friendly grown GBR because their disease incidences and yields in Gokseong may be similar to the reference cultivars.
