Monitoring the carbon dioxide concentration in arterial blood is vital for the evaluation and prevention of pulmonary disease. Yet, domestic pure arterial blood carbon dioxide sensor technologies are not being developed, instead all sensors are imported. In this paper, we develop a real time monitoring system for arterial blood partial pressure of carbon dioxide($pCO_2$) gas from the wrist by using a carbon micro-heater. The micro-heater was fabricated with a thickness of 0.3 ${\mu}m$ in order to collect the carbon dioxide under the skin. The micro-heater has been designed to perform temperature compensation in order to prevent damage to the skin. Two clinical trials of the system were undertaken. As a result, we demonstrated that a portable, transcutaneous carbon dioxide analysis($TcpCO_2$) device produced domestically is possible. In addition, this system reduced the analysis time significantly. Carbon films could reduce the unit price of these sensors by replacing the gold film used in foreign models. Also, we developed a real time monitoring system which can be used with optical biosensors for medical diagnostics as well as gas sensors for environmental monitoring.
Chemical modification of magnetic nanoparticles(MNPs) with functional polymers has recently gained a great deal of attention because of the potential application of MNPs to in vivo and in vitro biotechnology. The potential use of MNPs as capturing agents and sensitive biosensors has been intensively investigated because MNPs exhibit good separation-capability and binding-specificity for biomolecules after suitable surface functionalization processes. In this work, we demonstrate an efficient method for the surface modification of MNPs, by combining surface-initiated polymerization and the subsequent conjugation of the biologically active molecules. The polymeric shells of non-biofouling poly(poly(ethylene glycol) methacrylate)(pPEGMA) were introduced onto the surface of MNPs by surface-initiated, atom transfer radical polymerization(SI-ATRP). With biotin as a model of biologically active compounds, the polymeric shells underwent successful post-functionalization via activation of the polymeric shells and bioconjugation of biotin. The resulting MNP hybrids showed a biospecific binding property for streptavidin and could be separated by magnet capture.
Recently, the wearable computing technology with bio-sensors has been rapidly developed and utilized in various areas such as personal health, care-giving for senior citizens who live alone, and sports activities. In particular, the wearable computing equipment to measure vital signs by means of digital yarns and bio sensors is noticeable. The wearable computing devices help users monitor and manage their health in their daily lives through the customized healthcare service. In this paper, we suggest a system for monitoring and analyzing vital signs utilizing smart healthcare clothing with bio-sensors. Vital signs that can be continuously acquired from the clothing is well-known as unstructured data. The amount of data is huge, and they are perceived as the big data. Vital sings are stored by Hadoop Distributed File System(HDFS), and one can build data warehouse for analyzing them in HDFS. We provide health monitoring system based on vital sings that are acquired by biosensors in smart healthcare clothing. We implemented a big data platform which provides health monitoring service to visualize and monitor clinical information and physical activities performed by the users.

Photonic crystal structures have been received considerable attention due to their high optical sensitivity. One of the techniques to construct their structure is the dip-pen lithography (DPN) process, which requires a nano-scale resolution and high reliability. In this paper, we propose a two dimensional photonic crystal array to improve the sensitivity of optical biosensor and DPN process to realize it. As a result of DPN patterning test, we have observed that the diffusion coefficient of the mercaptohexadecanoic acid (MHA) molecule ink in octanol is much larger than that in acetonitrile. In addition, we have designed and fabricated optical waveguides based on the mach-zehnder interferometer (MZI) for application to biosensors. The waveguides were optimized at a wavelength of 1550 nm and fabricated according to the design rule of 0.45 delta%, which is the difference of refractive index between the core and clad. The MZI optical waveguides were measured of the optical characteristics for the application of biosensor.

Gene fusions were constructed by the transcriptional fusion of the dnaK promoter of pseudomonas sp. DJ-12 or E. coli to the lux or luc marker gene. The dnaKp-DJ::luxCDABE bioluminescent fusion in the biosensor using the Pseudomonas sp. DJ-12 dnaK promoter exhibited about 5-fold more extensive response to ethanol than that of dnaKp-EC::luxCDABE. The bioluminescent response of the dnaK-DJ::luc fusion to ethanol was much weaker than those of the other fusions. The biosensor harboring the dnaKp-DJ::luCDABE fusion was examined for its bioluminescence production based on exposure to aromatic compounds, such as biphenyl, 4-chlorobiphenyl (4CB), 4-hydroxybenzoate (4HBA), and catechol. In particular, the bioluminescence produced by the dnaKp-DJ::luxCDABE fusion was most sensitive to 1 mM biphenyl and 4CB when exposed for 80 min, and the responses were also very strong to other aromatics. Therefore, the biosensor bearing the dnaKp-DJ::luxCDABE fusion would appear to be the most useful for the detection of aromatics and other pollutants.
The gene ADH encoding NAD-dependent alcohol dehydrogenase from Bacillus stearothennophilus was cloned and overexpressed as a GST fusion protein at a high level in Escherichia coli. The expressed fusion protein was purified simply by glutathione affinity chromatography. GST fusion protein was then cleaved by thrombin, while soluble enzyme was further purified by glutathione affinity chromatography. The recombinant enzyme had the same elctrophoretic mobility as the native enzyme from Bacillus stearothennophilus. The recombinant enzyme catalyzed the oxidation of a number of alcohols and exhibited high activities towards secondary alcohols. The $K_m\;and\;V_{max}$ values of the recombinant enzyme for ethanol were 5.11 mM and 61.35 U/mg, respectively. Pyridine and imidazole notably inhibited the enzymatic activity. The activity of the recombinant enzyme optimally proceeded at pH 9.0 and $70^{\circ}C$. The midpoint of the temperature-stability curve for the recombinant enzyme was approximately $68^{\circ}C$, and the enzyme was not completely inactivated even at $85^{\circ}C$. The recombinant enzyme showed a high resistance towards denaturing agents (0.05% SDS, 0.1 M urea). Therefore, due to its stability and relatively broad substrate specificity, the recombinant enzyme could be utilized in bio-industrial processes and biosensors.
Many proteins consist of distinctive domains that can act independently or cooperatively to achieve a unique function. As these domains evolve from a naturally existing repertoire of functional domains, this implies that domain organization is an intrinsic element involved in building the complex structure and function of proteins. Thus, identifying functional domains would appear to be critical to the elucidation of questions related to protein evolution, folding, and the engineering of hybrid proteins for tai- lored applications. However, the simple application of "Lego-like assembly" to the engineering of hybrid proteins is an oversimplification, as many hybrid constructs lack structural stability, usually due to unfavorable domain contacts. Thus, directed evolution, along with computational studies, may help to engineer hybrid proteins with improved physico-chemical properties. Accordingly, this paper introduces several approaches to functional hybrid protein engineering that potentially can be used to create modulators of gene transcription and cell signaling, and novel biosensors to analyze biological functions in vivo.
We have developed an electromagnetic generator to bury in subcutaneous area or abdominal cavity of the birds. As we can't use a solar battery, it is extremely difficult to supply a power for subcutaneous implantation such as biosensors under the skin due to the darkness environment. We are aiming to test the antigen-antibody reaction to confirm an avian influenza. One solution is a very small generator with the electromagnetic induction coil. We attached the developed coil to chickens and pheasants and recorded the electric potential generated as the chicken walked and the pheasant flew. The electric potential generated with physical simulator is equal to or exceeds the 7 V peak-to-peak at maximum by 560/min of flapping of wings. Even if we account for the junction voltage of the diode (200 mV), efficient charging of the double-layer capacitor is possible with the voltage doubler rectifier. If we increase the voltage, other problems arise, including the high-voltage insulation of the double-layer capacitor. For this reason, we believe the power generated to be sufficient for subcutaneous area of birds. The efficiency, magnetic 2 mm in length and coil 15mm in length, if axial direction is rectified, the magnetic flux density given to the coil could calculated to 7.1 % and generated power average 0.47mW. The improvements in size and wire insulation are expected in the future.
The principle and applications of quartz crystal sensors based on the three basic concepts for mass, viscosity, and viscoelastic changes are reviewed. In the general discussion the basic principle of quartz crystal and realization of a resonant frequency-resonant resistance diagram are described in detail. As examples of their applications, gas sensing with a carbon-coated quartz crystal, determination of the blood coagulation factor, an electrochemical analysis and crystallization analysis are reported. The possibility of developing new biosensors and chemical sensors is discussed on the basis of these results.
The recombinant fluorescent chinese hamster ovary (CHO) cell line was developed and optimized through this study for biomonitoring system. This cell line, called KFC-A10, contains recombinant plasmid(pKCFG) constructed in this study for detecting toxic conditions (Mitomicyn C, EDCs, ${\gamma}-ray$, etc.). It is known that c-Fos is involved in proliferation and differentiation of the signal transduction and overexpression of this gene can lead cell to death under the toxic conditions including apoptosis status. Therefore, pKCFG which has the c-fosSRE::GFP is induced by toxic chemicals, especially DNA damage agents and apoptotic chemicals, and produces green fluorescence protein(GFP) under these toxic conditions. Through the characterization of KFC-A10 using fluorescent assays of GFP, it was shown that KFC-A10 cell line had a manifest GFP expression pattern due to various toxicants especially mitomycin C, ${\gamma}-ray$ and bisphenol A.
