A comprehensive review of the therapeutic and pharmacological effects of ginseng and ginsenosides in central nervous system

  • Ginseng is one of the most widely used herbal medicines in human. Central nervous system (CNS) diseases are most widely investigated diseases among all others in respect to the ginseng's therapeutic effects. These include Alzheimer's disease, Parkinson's disease, cerebral ischemia, depression, and many other neurological disorders including neurodevelopmental disorders. Not only the various types of diseases but also the diverse array of target pathways or molecules ginseng exerts its effect on. These range, for example, from neuroprotection to the regulation of synaptic plasticity and from regulation of neuroinflammatory processes to the regulation of neurotransmitter release, too many to mention. In general, ginseng and even a single compound of ginsenoside produce its effects on multiple sites of action, which make it an ideal candidate to develop multi-target drugs. This is most important in CNS diseases where multiple of etiological and pathological targets working together to regulate the final pathophysiology of diseases. In this review, we tried to provide comprehensive information on the pharmacological and therapeutic effects of ginseng and ginsenosides on neurodegenerative and other neurological diseases. Side by side comparison of the therapeutic effects in various neurological disorders may widen our understanding of the therapeutic potential of ginseng in CNS diseases and the possibility to develop not only symptomatic drugs but also disease modifying reagents based on ginseng.

Synthesis and Characterization of Amphiphilic Polyurethanes as Coating Materials for Urinary Catheters (요도용 카테타 도포용 양친성 폴리우레탄의 합성 및 분석)

  • The long-term use of indwelling urinary catheters can allow bacterial adhesion to their surfaces, followed by the catheter-associated urinary tract infection. In an attempt to minimize the bacterial adhesion, various amphiphilic polyurethanes (APUs) were synthesized as potential coating materials for urinary catheters. By varying composition of the soft segments such as PEO, PTMO, and PDMS, four different polyurethanes were synthesized. All the APU-coated urinary catheters had the smooth surfaces and showed higher hydrophilicity, compared to the commercial silicone catheters. In particular, the use of APUs with the higher PEG content significantly augmented hydrophilicity and remarkably reduced the total amount of bacteria adhering to the surface. Overall, the APUs prepared in this study provided the promising potential as coating materials for urinary catheters.

Enhanced Production of Succinic Acid by Actinobacillus succinogenes using the Production Medium Supplemented with Recombinant Carbonic Anhydrases (재조합 탄산무수화 효소 첨가 생산배지를 이용한 Actinobacillus succinogenes 유래의 숙신산 생산성 향상)

  • Succinic acid, a representative biomass-derived platform chemical, is a major fermentation product of Actinobacillus succinogenes. It is well known that carbon dioxide is consumed during the succinate fermentation, but the biochemical mechanism behind this phenomenon is not yet understood well. In this study, it was found that the addition of carbonic anhydrase (CA)s into media significantly enhances the succinic acid production by A. succinogenes during the fermentation supplied with carbon dioxide. It is likely that the (bi) carbonate produced by the CA activity from gaseous carbon dioxide is favoured by A. succinogenes for consumption and utilization. Therefore, the $MgCO_3$ requirement could be significantly reduced without compromising the succinate productivity. Furthermore, because of too high price of the commercial carbonic anhydrase, it was undertaken to economically overproduce a cyanobacterial carbonic anhydrase by the use of a recombinant Pichia pastoris. An expression vector system was constructed with the carbonic anhydrase gene PCR-cloned from Cyanobacterium Synechocystis sp., and introduced into P. pastoris for fermentation studies. About 95.9 g/L of succinic acid was produced in the production medium with 30 ppm of carbonic anhydrase, approximately 2 fold higher productivity compared to the parallel process with no supplementation of the enzyme. It is expected that this method can provide a valuable way of overcoming inefficiencies inherent in gas supply during $CO_2$-based bioprocesses like succinic acid fermentation.

Application of an Energy Sensitive CZT Detector to a DXA Type of Bone Densitometer

  • The accuracy of DXA(Dual Energy X-ray Absorptiometry) highly depends on the detection and separation capability of dual energy X-ray X-ray photons. In addition both of scan time and patient exposure are affected by detection efficiency. A CZT detector with a good energy resolution and high detection efficiency was evaluated for the application of bone densitometry. Its performance was compared to a photomultiplier tube with a NaI(T1) scintillator in terms of energy resolution, detection efficiency and the accuracy of bone mineral density measurement. The comparison study was performed with CZT detector and PM tube using DXA equipments(OSTEO Plus, OSTEO Prima, ISOL Technology). The energy spectrum was acquired using MCA(Multi-Channel Analyzer). The used X-ray energy ranged from 20keV to 86keV. The MCA result of the CZT detector showed a slightly sharper energy spectrum than that of NaI(T1). Detection efficiency of the CZT detector at 59.5keV was 1.4 times better. Remarkably the final results of bone mineral density measurements demonstrate only less than 1% difference. The CZT detector appears to have many benefits for the application of bone densitometry. Its excellent energy resolution can enhance the counting accuracy of dual energy X-ray spectrum. Furthermore its compactness in physical dimension and no cooling requirement will be additional benefits for a more compact and accurate bone densitometer.

Proposition of a New Implantable Acoustic Sensor Based on Technology Evaluation of Fully Implantable Hearing Aids (완전 이식형 보청기 기술 평가 기반의 새로운 이식형 음향센서 제안)

  • Key components of implantable hearing aids are consist of an acoustic sensor that collect external sound by suppressing the body noise, a signal processor module for compensation algorithm of hearing loss, and a output transducer which has tiny size but have high efficiency, respectively. In the partial implantable hearing aids, technologies of transducer and signal processor are so matured that can be applied not too much difficulty. However, due to the difficulties in implantable acoustic sensor technology, such as minimization of masticatory sound and damage of sensor's membrane from external impact, practical use of fully implantable hearing aids have not successful so far. In this paper, we have proposed a novel implantable acoustic sensor which has trans-tympanic structure, and is verified that the proposed method can be very useful for fully implantable hearing aids by cadaveric experiments.

Effects of Elicitors on Seedling Growth, Total Polyphenol and Chlorophyll Content and Antioxidant Activity of Barley (Hordeum vulgare L.)

  • This study is focused on the evaluation of growth parameters, total polyphenol content (TPC), chlorophyll content as well as the DPPH (1,1-diphenyl-2-picryhydrazyl) free radical scavenging activity of young barley seedling (YBS) affected by elicitation. Salicylic acid (SA), methyl jasmonate (MJ), amino acid liquid fertilizer (ALF) and microbial metabolism activator (MMA) were used. Elicitation was conducted for two times and various concentrations were used in this study. The result revealed that, MJ 1 ml/L treated-YBS gave the longest seedling length of 1.33 cm, followed by the ones treated with SA 1.38 mg/L and ALF 2 ml/L, respectively. ALF 3 ml/L treatment gave the highest fresh weight of 10 seedlings, followed by MJ 5 ml/L and SA 13.8 mg/L treatment with 1.56 g, 1.55 g and 1.53 g respectively. SA 138.12 mg/L elicitor treated-YBS gave the highest Chl a, Chl b content of $8.57{\mu}g/mg$ and $3.83{\mu}g/mg$, respectively while the highest carotenoid content was found in MJ ml/L treatment with $1.62{\mu}g/mg$. Among elicitor treated-YBS, SA showed better TPC. The highest TPC was found in SA 1.38 mg/L treatment with 18.82 mg/g TAE. Likewise, SA 1.38 mg/L showed the highest DPPH free radical scavenging activity among all the treatments. However, the lowest TPC was found in ALF 1ml/L treated-YBS with 9.46 mg/g TAE, which was even lower than the control (14.31 mg/g TAE).

Electrochemical Synthesis of Red Fluorescent Silicon Nanoparticles

  • Herein, we report on the preparation of red fluorescent Si nanoparticles stabilized with styrene. Nano-sized Si particles emit fluorescence under UV excitation, which could be used to open up new applications in the fields of optics and semi-conductor research. Unfortunately, conventional methods for the preparation of red fluorescent Si nanoparticles suffer from the lack of a fully-established standard synthesis protocol. A common initial approach during the preparation of semi-conductors is the etching of crystalline Si wafers in a HF/ethanol/$H_2O$ bath, which provides a uniformly-etched surface of nanopores amenable for further nano-sized modifications via tuning of various parameters. Subsequent sonication of the etched surface crumbles the pores on the wafer, resulting in the dispersion of particles into the solution. In this study, we use styrene to occupy these platforms to stabilize the surface. We determine that the liberated silicon particles in ethanol solution interact with styrene, resulting in the substitution of Si-H bonds with those of Si-C as determined via UV photo-catalysis. The synthesized styrene-coated Si nanoparticles exhibit a stable, bright, red fluorescence under excitation with a 365 nm UV light, and yield approximately 100 mg per wafer with a synthesis time of 2 h. We believe this protocol could be further expanded as a cost-effective and high-throughput standard method in the preparation of red fluorescent Si nanoparticles.

Augmented Reality to Localize Individual Organ in Surgical Procedure

  • Objectives: Augmented reality (AR) technology has become rapidly available and is suitable for various medical applications since it can provide effective visualization of intricate anatomical structures inside the human body. This paper describes the procedure to develop an AR app with Unity3D and Vuforia software development kit and publish it to a smartphone for the localization of critical tissues or organs that cannot be seen easily by the naked eye during surgery. Methods: In this study, Vuforia version 6.5 integrated with the Unity Editor was installed on a desktop computer and configured to develop the Android AR app for the visualization of internal organs. Three-dimensional segmented human organs were extracted from a computerized tomography file using Seg3D software, and overlaid on a target body surface through the developed app with an artificial marker. Results: To aid beginners in using the AR technology for medical applications, a 3D model of the thyroid and surrounding structures was created from a thyroid cancer patient's DICOM file, and was visualized on the neck of a medical training mannequin through the developed AR app. The individual organs, including the thyroid, trachea, carotid artery, jugular vein, and esophagus were localized by the surgeon's Android smartphone. Conclusions: Vuforia software can help even researchers, students, or surgeons who do not possess computer vision expertise to easily develop an AR app in a user-friendly manner and use it to visualize and localize critical internal organs without incision. It could allow AR technology to be extensively utilized for various medical applications.

Feasibility of Improving the Accuracy of Dose Calculation Using Hybrid Computed Tomography Images: A Phantom Study

  • Purpose: Kilovoltage computed tomography (kV-CT) is essential for radiation treatment planning. However, kV-CT images are significantly distorted by artifacts when a metallic prosthesis is present in the patient's body. Thus, the accuracies of target delineation and treatment dose calculation are inevitably lowered. We evaluated the accuracy of the calculated doses using an image restoration method with hybrid CT, which was introduced in our previous study. Methods: A cylindrical phantom containing four metals, namely, silver, copper, tin, and tungsten, was scanned using kV-CT and megavoltage CT to produce hybrid CT images. We created six verification plans for three head and neck patients on kV-CT and hybrid CT images of the phantom and calculated their doses. The actual doses were measured with film patches during beam delivery using tomotherapy. We used the gamma evaluation method to compare dose distribution between kV-CT and hybrid CT with three gamma criteria, namely, 3%/3 mm, 2%/2 mm, and 1%/1 mm. Results: The gamma pass rates decreased as the gamma criteria were strengthened, and the pass rate of hybrid CT was higher than that of kV-CT in all cases. When the 1%/1 mm criterion was used, the difference in gamma pass rates between them was up to 13%p. Conclusions: According to our findings, we expect that the use of hybrid CT can be a suitable approach to avoid the effect of severe metal artifacts on the accuracy of dose calculation and contouring.

Effects of starvation-induced negative energy balance on endoplasmic reticulum stress in the liver of cows

  • Objective: Endoplasmic reticulum (ER) stress engages the unfolded protein response (UPR) that serves as an important mechanism for modulating hepatic fatty acid oxidation and lipogenesis. Chronic fasting in mice induced the UPR activation to regulate lipid metabolism. However, there is no direct evidence of whether negative energy balance (NEB) induces ER stress in the liver of cows. This study aimed to elucidate the relationship between the NEB attributed to feed deprivation and ER stress in bovine hepatocytes. Methods: Blood samples and liver biopsy tissues were collected from 6 non-lactating cows before and after their starvation for 48 h. The blood non-esterified fatty acids (NEFA), β-hydroxybutyric acid (BHBA) and glucose level were analyzed. Real-time quantitative polymerase chain reaction and Western blotting were used to explore the regulation of genes associated with UPR and lipid metabolism. Results: The starvation increased the plasma BHBA and NEFA levels and decreased the glucose level. Additionally, the starvation caused significant increases in the mRNA expression level of spliced X-box binding protein 1 (XBP1s) and the protein level of phosphorylated inositol-requiring kinase 1 alpha (p-IRE1α; an upstream protein of XBP1) in the liver. The mRNA expression levels of peroxisome proliferator-activated receptor alpha and its target fatty acid oxidation- and ketogenesis-related genes were significantly upregulated by the starvation-mediated NEB. Furthermore, we found that the mRNA expression levels of lipogenic genes were not significantly changed after starvation. Conclusion: These findings suggest that in the initial stage of NEB in dairy cows, the liver coordinates an adaptive response by activating the IRE1 arm of the UPR to enhance ketogenesis, thereby avoiding a fatty liver status.
