Growth and carbon storage of black saxaul in afforested areas of the Aralkum Desert

  • This study aimed to determine the growth and carbon storage of planted Haloxylon aphyllum in the Aralkum Desert in Kazakhstan. Six sites afforested in 2000, 2005, 2009, 2010, 2013, and 2017 were selected. The root collar diameter(cm) and height(m) were measured for all H. aphyllum in 30 m×44 m plots. Biomass accumulation (g m-2) and carbon storage(C g m-2) were calculated using allometric equations and the carbon concentration data of Haloxylon species. The diameters varied from 2.5 cm to 4.3 cm and the height varied from 106.2 cm to 223.7 cm. The growth of H. aphyllum was not linearly related to the afforestation year or soil properties. Tree growth might have been influenced by variations in the microclimate, such as temperature, precipitation, and dust storms. The mean total biomass accumulation was 20.57g m-2 and ranged from 2.42 g m-2 to 64.53 g m-2. The mean carbon storage was 9.70C g m-2 and ranged from 1.12 C g m-2 to 30.61 C g m-2. These biomass and carbon storage estimates were smaller than those reported for other Central Asian deserts, but afforestation enabled the generation of vegetative cover and consequently, carbon sequestration in the manmade Aralkum Desert.

Effect of co-inoculation of Brevibacterium iodinum RS16 and Methylobacterium oryzae CBMB20 on the early growth of crop plants in Saemangeum reclaimed soil

  • The objective of this study was to determine the effect of single and co-inoculation of plant growth promoting bacteria (PGPB) on early plant growth in Saemangeum reclaimed soil. Plant growth promoting Brevibacterium iodinum RS16 and Methylobacterium oryzae CBMB20 were inoculated on maize (Zea mays L.) and sorghum-sudangrass hybrid (Sorghum bicolor L.) grown in Saemangeum reclaimed soil. Single and co-inoculation of B. iodinum RS16 and M. oryzae CBMB20 increased plant height, dry biomass accumulation and macro-nutrient accumulation of maize and sorghum-sudangrass hybrid. M. oryzae CBMB20 treatment increased plant height in maize by 41.2% at 30 days after sowing (DAS), shoot dry weight and total dry weight compared to non-inoculated treatment. Macro-nutrient accumulation (N and P) in maize roots was significantly increased with co-inoculation treatment, K and Ca content was significantly increased at B. iodinum RS16 treatment compared to non-inoculated treatment. Macro-nutrient accumulation (P, K, Ca and Mg) in shoot was higher with M. oryzae CBMB20 treatment compared to non-inoculated treatment. In case of sorghum-sudangrass hybrid, co-inoculation treatment showed 33.7% increase in plant height compared to non-inoculated treatment at 30 DAS. M. oryzae CBMB20 treatment increased root dry weight and total dry weight, macro-nutrient accumulation in roots and N, Ca and Mg accumulation in shoot compared to non-inoculated treatment. P and K accumulation in shoot was significantly increased at co-inoculation treatment compared to non-inoculated treatment. This pot culture experiment demonstrated that single and co-inoculation of B. iodinum RS16 and M. oryzae CBMB20 increased the early growth and nutrient accumulation of maize and sorghum-sudangrass hybrid.

Enhancement of Scenedesmus sp. LX1 Biomass Production and Lipid Accumulation Using Iron in Artificial Wastewater and Domestic Secondary Effluent

  • While coupling wastewater treatment with microalgal bioenergy production is very promising, new approaches are needed to enhance microalgal growth and lipid accumulation in wastewater. Therefore, this study investigated the effect of iron on the growth, nutrient removal, and lipid accumulation of Scenedesmus sp. LX1 in both artificial wastewater and domestic secondary effluents. When increasing the iron concentration from 0 to 2 mg/l in the artificial wastewater, the biomass production of Scenedesmus sp. LX1 increased from 0.17 to 0.54 g/l; the nitrogen and phosphorus removal efficiency increased from 15.7% and 80.6% to 97.0% and 99.2%, respectively; and the lipid content was enhanced 84.2%. The relationship between the carrying capacity/maximal population growth rate of Scenedesmus sp. LX1 and the initial iron concentration were also in accordance with the Monod model. Furthermore, when increasing the iron concentration to 2 mg/l in four different domestic secondary effluent samples, the lipid content and lipid production of Scenedesmus sp. LX1 was improved by 17.4-33.7% and 21.5-41.8%, respectively.

혐기성 부착미생물의 초기성장에 미치는 영향인자에 관한 연구 (Laboratory Study on the Factors Affecting on Initial Anaerobic Biomass Development)

  • Laboratory-scale investigation into initial anaerobic biofilm development was carried out by circulating mixed liquor from a steady-state anaerobic reactor through silicone tubing and then rerurning the mixed liquor to the reactor. The wall of the silicone tubing was the surface upon which anaerobic biofilm accumulation or development was monitored. Methanogenic bacteria accumulation was monitored by F$_{420}$ fluorescence (picomoles F$_{420}$/cm$^{2}$) of the extracted biofilm material. Biofilm accumulation was measured by the increase in COD of the extracted material ($\mu $g COD/cm$^{2}$). Experiments were conducted for 25 days, and biofilm analyses were performed at 5 days intervals. The results indicated that the initial rates of methangen and anaerobic biofilm accumulation increased with increasing organic loading rate and higher initial rates were observed for 15 days than 15 day liquid HRT or SRT. When the initial rates were plotted against the corresponding mixed liquor volatile suspended solids the difference between the results at the two HRT's became much less significant. Thus, the concentration of mixed liquor volatile suspended solids was found to be a very important parameter affecting initial anaerobic biofilm development. The ratio of methanogens to anaerobic biofilm was also investigated. The results showed that the ratio remained constant through the 25 days of each experiment and for high organic loading rates. Based on the results of this research, a reduction, a reduction of start-up period of anaerobic fixed film reactors might be achieved by maintaining a high organic loading and a large concentration of anaerobic microorganisms in the mixed liquor during the start-up period.

충주지역(忠州地域)의 자작나무와 가래나무 조림지(造林地)의 물질생산(物質生産)에 관(關)한 연구(硏究) (Biomass and Net Primary Production of Betula platyphylla and Juglans mandshurica Plantations in Chungju Area)

  • 충청북도 충주지역에서 식재된 22년생 자작나무와 24년생 가래나무 인공림의 지상부 현존생물량 및 연순생산량을 분석하기 위하여 임분별 9주씩 총 18주의 표본목을 선정 벌목하였다. 수식 $Wt=aD^b$를 사용하여 추정한 자작나무와 가래나무의 지상부 총 현존생물량은 자작나무림에서 79.33t/ha와 가래나우림에서 67.47t/ha으로 가래나무림보다 연령이 작은 자작나무림에서 높은 현존생물량을 보였다. 부위별 구성비는 자작나무림과 가래나무림 모두에서 수간목질부, 가지, 수피, 그리고 잎의 순으로 높았다. 총 연순생산량은 자작나무림이 9.92t/ha, 가래나무림이 11.79t/ha으로 총 현존생물량이 많았던 자작나무림보다 가래나무림에서 높게 나다났는데, 이 결과는 잎을 제외한 수간목질부, 수피, 소지 그리고 가지에서의 연순생산량이 가래나무림에서 높았기 때문이다.

Direct Fermentation of Potato Starch in Wastewater to Lactic Acid by Rhizopus oryzae

  • The fungal species of Rhizopus oryzae 2062 has the capacity to carry out a single stage fermentation process for lactic acid production from potato starch wastewater. Starch hydrolysis, reducing sugar accumulation, biomass formation, and lactic acid production were affected with variations in pH, temperature, and starch source and concentration. A growth condition with starch concentration approximately 20 g/ L at pH 6.0 and 30$^{\circ}C$ was favourable for starch fermentation, resulting in a lactic acid yield of 78.3%∼85.5% associated with 1.5∼2.0 g/L fungal biomass produced in 36 h of fermentation.

  • The European Iron Fertilization Experiment EIFEX studied the growth and decline of a phytoplankton bloom stimulated by fertilising $10km^2$ in the core of a mesoscale $(80{\times}120km)$ cyclonic eddy south of the Antarctic Polar Front with about 2 times 7 tonnes of iron sulphate. The phytoplankton accumulation induced by iron fertilization did not exceed $3{\mu}g\;chl\;a\;l^{-1}$ despite a draw down of $5{\mu}M$ of nitrate that should have resulted in at least double to triple the amount of phytoplankton biomass assuming regular Redfield-ratios for draw down after phytoplankton growth in the Southern Ocean. During EIFEX the fertilized core of the mesoscale eddy evolved to a hotspot for a variety of small and medium sized mesozooplankton copepods. In contrast to copepods, the biomass of salps (Salpa thompson)) that dominated zooplankton biomass before the onset of our experiment decreased to nearly extinction. Most of the species of the rnosozooplankton community showed extremely hiか feeding rates compared to literature values from Southern Ocean summer communities. At the end of the experiment, massive phytoplankton sedimentation reached the sea floor at about 3800m water depth.

Physiological and biochemical responses of elevated ozone on Pterocarpus indicus under well-watered and drought conditions

  • Seedlings of Pterocarpus indicus were grown in both well-watered and drought stress conditions in phytotron. Seedlings grown under well-watered and drought stress conditions were exposed to either combined or without ozone of 200 ppb for one month. First, the physiological responses to elevated ozone levels indicated a decreased biomass. The seedlings grown in arid soil and exposed to ozone showed less biomass than those grown in arid soil but not exposed to ozone. Moreover, all the seedlings except the well-watered and unexposed ones showed a significantly lower photosynthetic rate ($P_N$) over time. However, with the accumulation of ozone injuries, the antioxidant enzyme activities increased overall. In the study results, when exposed to ozone, the well-watered seedlings exhibited more antioxidative enzyme activity than did the seedlings grown in arid soil. Generally, P. indicus in arid soil suffered less damage from elevated ozone than did the well-watered plants.

Mid-term (2009-2019) demographic dynamics of young beech forest in Albongbunji Basin, Ulleungdo, South Korea

  • Background: The stem exclusion stage is a stage of forest development that is important for understanding the subsequent understory reinitiation stage and maturation stage during which horizontal heterogeneity is formed. Over the past 11 years (2009-2019), we observed a deciduous broad-leaved forest in the Albongbunji Basin in Ulleungdo, South Korea in its stem exclusion stage, where Fagus engleriana (Engler's beech) is the dominant species, thereby analyzing the changes in the structure (density and size distributions), function (biomass and species richness), and demographics. Results: The mean stem density data presented a bell-shaped curve with initially increasing, peaking, and subsequently decreasing trends in stem density over time, and the mean biomass data showed a sigmoidal pattern indicating that the rate of biomass accumulation slowed over time. Changes in the density and biomass of Fagus engleriana showed a similar trend to the changes in density and biomass at the community level, which is indicative of the strong influence of this species on the changing patterns of forest structure and function. Around 2015, a shift between recruitment and mortality rates was observed. Deterministic processes were the predominant cause of tree mortality in our study; however, soil deposition that began in 2017 in some of the quadrats resulted in an increase in the contribution of stochastic processes (15% in 2019) to tree mortality. The development of horizontal heterogeneity was observed in forest gaps. Conclusions: Our observations showed a dramatic shift between the recruitment and mortality rates in the stem exclusion stage, and that disturbance increases the uncertainty in forest development increases. The minor changes in species composition are likely linked to regional species pool and the limited role of the life-history strategy of species such as shade tolerance and habitat affinity. Our midterm records of ecological succession exhibited detailed demographic dynamics and contributed to the improvement of an ecological perspective in the stem exclusion stage.

Optimization of Aconitine Production in Suspension Cell Cultures of Aconitum napellus L.

  • Aconitine alkaloids produced from cell suspension cultures of Aconitum napellus for the first time. The effects of various culture conditions on cell biomass and aconitine accumulation in cell suspension cultures were investigated. Suspension cell cultures of A. napellus were established by transferring callus tissues from leaf explants onto liquid MS medium supplemented with $1\;mg/l$ NAA and $0.1\;mg/l$ kinetin. Among the culture media tested, MS medium had a pronounced effect on cell growth and aconitine accumulation. The maximum dry cell weight was obtained at inoculum size of 3 g (FCW) per flask and in MS medium supplemented with 5% sucrose after 8 weeks. The addition of salicylic acid (SA) and yeast extract (YE) in the MS medium enhanced aconitine accumulation. Using a proper combination of culture condition and supplements, aconitine content could reach 0.043% (dry weight basis), that was $2.5{\sim}3$ fold higher that detected in control cultures.
