기내 검정법을 이용한 국내 과수 화상병 방제제 선발 (In Vitro Screening of Antibacterial Agents for Suppression of Fire Blight Disease in Korea)

  • 국내 과수 화상병은 2015년 사과 및 배에서 발생하여 큰 피해를 주고 있으나, 이에 대한 적절한 방제제는 등록되어 있지 않다. 다만 농촌진흥청은 무름병과 세균점무늬병 등 다른 식물세균병해에 등록되어 있는 살세균제들을 직권등록 시켜놓은 상태이다. 그러나 직권등록이란, 약효는 보증하지 않으면서 약해만을 보증하는 제도로, 국내에서 과수 화상병이 지속적으로 발생되고 있는 상황에서는 적절한 방제제를 선발하는 것이 요구되어지기에 본 연구를 수행하였다. 직권등록 국내 과수 화상병 방제제 중, 각각 4종의 농용항생제와 구리 합성 화학물 그리고 생물학적 방제제 2종에 대한 기내 검정법을 통하여 국내 과수 화상병원세균 억제 및 병징확산 억제효과를 검증하였다. Kasugamycin, Oxytetracycline 및 streptomycin의 항생제는 효과가 우수하였으며, oxolinic acid는 꽃에서의 억제효과는 우수하였지만 신초 및 미성숙과 기내검정에서는 다소 효과가 감소하였다. 4종의 구리 합성 화학물은 항생제만큼의 억제효과는 나타내지 않았지만, 사용가능한 수준으로 조사되었다. 또한 2종의 생물적 방제제는 처리시기 등을고려할 경우, 친환경 재배지에서는 사용 가능할 것으로 판단되었다. 한편 국내 과수 화상병원세균은 선발 방제제 중 항생제 저항성 유전자들을 가지고 있지 않고,화분매개충에 대한 영향도 없는 것으로 조사되었다.

역류성식도염 유발 흰쥐에 대한 이진탕가미방(二陳湯加味方)의 효과 (Effects of Yijintang-gamibang on Reflux Esophagitis Induced by Pylorus and Forestomach Ligation in Rat)

  • Purpose : The object of this study was to observe the suppressive effects of Yijintang-gamibang (YJGMB), Yijintang with Atractylodis Rhizoma, Massa Medicata Fermentata, Hordei Fructus Germiniatus, and Coptidis Rhizoma. YJGMB has been traditionally used in Korean medicine for treating various digestive diseases. We tested it on the rat reflux esophagitis (RE) induced by pylorus and forestomach ligation in rats as compared with omeprazole, a well-known proton pump inhibitor. Method : Three different dosages of YJGMB 200, 100 and 50mg/kg, were orally pretreated once a day for 28 days before pylorus and forestomach ligation. Seven groups, each of 8 rats per group were used in the study. Six hours after pylorus and forestomach ligation, changes of the stomach and esophagus lesion areas, gastric volumes, acid and pepsin outputs, invasive lesion percentages, fundic mucosa and total thicknesses were measured as histomorphometry. The results were compared with omeprazole, antioxidant and proton pump inhibitor, and 30 and 10mg/kg treated groups in which the effects on RE were already confirmed. Results : As results of pylorus and forestomach ligation, marked increases of esophageal and gastric mucosa lesion areas, gastric volumes, acid outputs, pepsin outputs were observed with histopathological changes of RE, such as hemorrhages, ulcerative lesions and edematous changes on the fundic mucosa. However, these pylorus and forestomach ligation-induced RE were dose-dependently inhibited by treatment of 200, 100 and 50mg/kg of YJGMB. YJGMB 200mg/kg showed similar protective effects as compared with 30mg/kg of omeprazole in the present study, and more favorable effects were observed in 50mg/kg of YJGMB treated rats as compared with omeprazole 10mg/kg in the present study. Conclusion : The results obtained in this study suggest that YJGMB has favorable protective effects on the RE induced by pylorus and forestomach ligation. Therefore, it is expected that YJGMB will also show favorable effects on RE corresponding well to the suggestion of traditional Korean medicine. However, more detailed mechanism studies should be conducted in future with the screening of the biological active chemical compounds in herbs.

생물학적(生物學的) 질소고정(窒素固定)에 관(關)한 연구(硏究) III. 수도근(手稻根) 표피미생물(表皮微生物)의 질소고정력(窒素固定力) (Studies on Biological Nitrogen Fixation III. Nitrogen Fixing Activity Associated with Rice Rhizoplane for Varietal Screening by Acetylene Reduction Method)

  • 벼뿌리에서 서식(捿息)하는 질소고정미생물(窒素固定微生物)에 의(依)한 질소고정력(窒素固定力)을 검토(檢討)하기 위(爲)하여 벼육성지역(育成地域)이 상이(相異)한 수종(數種)의 품종(品種)을 공시(供試)하여 Acetylene 환원법(還元法)에 의(依)하여 조사(調査)한 결과(結果)는 다음과 같다. 1. 벼뿌리의 질소고정력(窒素固定力)은 생육시기(生育時期) 및 품종(品種)에 따라 상이(相異)했으며 유수형성기(幼穗形成期) 및 출수기(出穗期)에 최고(最高)로 높은 고정량(固定量)을 보였다. 2. 볏짚시용(施用)은 벼뿌리에 서식(捿息)하는 질소고정미생물(窒素固定微生物)에 의(依)한 질소고정(窒素固定)을 현저(顯著)히 증가(增加)시키지 못했으나 Kataktara, Centry Partna-231, TKM-6, 대구조(大邱租) 및 조동지(趙同知)는 다소(多少) 증가경향(增加傾向)을 나타내었다. 3. 수도(水稻) 1작기간(作期間) 벼뿌리의 질소고정량(窒素固定量)은 밀양(密陽) 23호가 0.55kg으로 제일(第一) 낮았으며 조동지(趙同知)가 2.6kg으로 가장 높았다. 4. 벼뿌리의 질소고정력(窒素固定力)은 뿌리의 무게와 정상관(正相關) 관계(關係)를 나타내지 않았다.

식물병원균에 대한 Bacillus vallismortis 1A 균주의 항진균 활성 (Antifungal Activity of Bacillus vallismortis 1A against Phytopathogen)

  • 항진균성 활성을 가지는 저영양세균을 분리하기 위하여 토양시료는 경북 영양, 충북 제천, 충북 음성, 충북 괴산, 충남 논산 등지의 고추 밭에서 수집하였고 R2A배지를 이용하여 평판희석법으로 9,354여 균주의 저영양세균을 분리하였다. 분리된 저영양세균중 고추역병에 강한 활성을 가지는 1A 균주를 선발 하였으며 16S rDNA 와 표준균주(B. vallismortis, B. mojavensis)를 이용한 생리, 생화학적 실험으로 B. vallismortis로 최종동정 되었다. 1A균주는 Magnaporthe 균을 제외하고 Phytophthora, Collectotrichum, Botrytis, Fusarium 균등에서 폭넓게 강한 활성을 나타내었다. 고추 유묘검정에서 대조구가 89%정도의 발병율을 보인 반면에 1A 처리구에서는 29%의 발병율을 나타내어 고추역병의 길항균으로써의 미생물제제 가능성이 있는 것으로 판단된다.

Identification of Homer1 as a Potential Prognostic Marker for Intrahepatic Cholangiocarcinoma

  • Background: The aim of the present study was to analyze whether Homer1 is a potential prognostic marker for intrahepatic cholangiocarcinoma (ICC). Materials and Methods: The expression of Homer1 in ICC tissue was detected with immunohistochemistry and levels of protein in ICC and paratumor tissues were evaluated by Western blotting. Survival analysis by the Kaplan-Meier method was performed to assess prognostic significance. Results: Homer1 expression was high in 67.4% (58/86) of ICC samples, and there was significant difference between ICC and adjacent noncancerous tissues (p<0.001); high expression was associated with poor histologic differentiation (p=0.019), TNM stage (p=0.014), lymph node metastasis (p=0.040), and lymphatic invasion (p=0.025). On Kaplan-Meier analysis, a comparison of survival curves of low versus high expressors of Homer1 revealed a highly significant difference in OS (p=0.001) and DFS (p=0.006), indicating that high expression of Homer1 was linked with a worse prognosis. Multivariate analyses showed that Homer1 expression was an independent risk factor predicting overall survival[Hazard ratio(HR), 7.52; 95% confidence interval (CI), 2.63-21.47; p=0.002] and disease-free survival (HR, 11.56; 95%CI, 5.17-25.96; p<0.001) in ICC. Conclusions: Homer1 promotes lymphatic invasion and associates with lymph node metastasis and poor prognosis of ICC. The current study shows that Homer1 may be an independent prognostic factor for ICC patients after curative resection, and it provides an important basis for screening/treating high-risk patients.

  • Expressed sequence tags (EFTs) are the partial segments of cDNA produced from 5 or 3 single-pass sequencing of cDNA clones, error-prone and generated in highly redundant sets. Advancement and expansion of Genomics made biologists to generate huge amount of ESTs from variety of organisms-human, microorganisms as well as plants, and the cumulated number of ESTs is over 5.3 million, As the EST data being accumulate more rapidly, it becomes bigger that the needs of the EST analysis tools for extraction of biological meaning from EST data. Among the several needs of EST analyses, the extraction of protein sequence or functional motifs from ESTs are important for the identification of their function in vivo. To accomplish that purpose the precise and accurate identification of the region where the coding sequences (CDSs) is a crucial problem to solve primarily, and it will be helpful to extract and detect of genuine CD5s and protein motifs from EST collections. Although several public tools are available for EST analysis, there is not any one to accomplish the object. Furthermore, they are not targeted to the plant ESTs but human or microorganism. Thus, to correspond the urgent needs of collaborators deals with plant ESTs and to establish the analysis system to be used as general-purpose public software we constructed the pipelined-EST analysis system by integration of public software components. The software we used are as follows - Phred/Cross-match for the quality control and vector screening, NCBI Blast for the similarity searching, ICATools for the EST clustering, Phrap for EST contig assembly, and BLOCKS/Prosite for protein motif searching. The sample data set used for the construction and verification of this system was 1,386 ESTs from human intrathymic T-cells that verified using UniGene and Nr database of NCBI. The approach for the extraction of CDSs from sample data set was carried out by comparison between sample data and protein sequences/motif database, determining matched protein sequences/motifs that agree with our defined parameters, and extracting the regions that shows similarities. In recent future, in addition to these components, it is supposed to be also integrated into our system and served that the software for the peptide mass spectrometry fingerprint analysis, one of the proteomics fields. This pipelined-EST analysis system will extend our knowledge on the plant ESTs and proteins by identification of unknown-genes.

202종(種) 한약재(韓藥材)의 $IFN-{\gamma}$ 분비조절에 관한 비교연구 (Comparative study of 202 herbs on interferon-gamma secretion)

  • Objects : This study has been carried out to assess the effects of the variable herbs on $IFN-{\gamma}$ secretion in the mouse spleen cell. Methods : 202 kinds of herb extracts were used to evaluate the $IFN-{\gamma}$ secretory distinction by each $1{\mu}g/ml$ and $10{\mu}g/ml$ density of water. All experimental herbs were grouped by oriental herbalogical method. But each herb had its independent variables. Results : The secretions increased in 20% of all herbal water. The density differences also make different effects on the secretion of $IFN-{\gamma}$. The secretion of IFN-${\gamma}$ inclosed in some kinds of herbs of IFN-${\gamma}$. It has representatively increased in Imperaetae Rhizoma(白茅根) of $1{\mu}g/ml$ and Notopterygii Rhizoma(羌活)of $10{\mu}g/ml$. $IFN-{\gamma}$ incresed in 12 kinds of heybs of both densities. The secretion of $IFN-{\gamma}$ decreased in some kinds of herbs of $IFN-{\gamma}$. It has representatively decreased in Moutan Radicis Cortex(蘇丹皮) of $1{\mu}g/ml$ and Angelicae Radix(富歸尾) of $10{\mu}g/ml$. $IFN-{\gamma}$ decreased in 18 kinds of herbs of both densities. In t oriental herb group, The secretion of $IFN-{\gamma}$ increased in Bang-Hyang-Hwa-Sup group(芳香化濕藥), He-Pyo group(解表藥), I-Su-Sam-SuP group(利水渗) The secretion of $IFN-{\gamma}$ decreased in Gu-Chung group(驅蟲藥), An-Sin group(安神藥). Su-Sap group(收澁) On-Li group(溫裏藥), I-Gi group(利氣藥). Conclusions : The result of this study will not only broaden applications of oriental medicine to biological therapy, but also form the basis of oriental medical therapy to find out the meaning of oriental classification.

한약재 물 추출물의 Monoamine Oxidase B의 활성 억제 및 항산화 활성 검색 (Screening on Monoamine Oxidase B Inhibitor and Antioxidant Activity from the Water Extracts of Medicinal Plants)

  • This present study was designed to screen medicinal plants for the treatment of brain diseases such as Parkinson's disease or Alzheimer's disease. The inhibitory activity of monoamine oxidase B (MAO-B) was investigated in the water extracts of 56 species traditional medicines. Among the tested medicinal plants, E. lathyris, R. palmatum, F. rhynchonphylla, E. caryophyllata, E. pekinensis and H. syriacus were showed the strong inhibitory activity against MAO-B. Therefore, MAO-B inhibitory activity of 6 traditional medicine extracts in the different concentration (2.5, 6.5 and 12.5 ㎍/ml) was determined. The inhibitory effect of MAO-B was detected with dose dependently in 6 traditional plants extracts. E. caryophyllata and R. palmatum were showed the highest inhibitory activity, the MAO-B inhibitory activity at 2.5㎍ of herbal extract being 58% and 52%, respectively. The water extracts of 6 species were tested on antioxidant activity using radical scavenging effects against ABTS/sup +/. The water extracts of R. palmatum, E. caryphyllata, E. pekinensis and H. syriacus were showed strong antioxidant capacity at 20 ㎍ concentration. Among the 56 medicinal plants investigated, the water extracts of R. palmatum and E. caryphyllata were showed significant antioxidant capacity and MAO-B inhibiory activity. Therefore, R. palmatum and E. caryphyllata are expected to ameliorate the clinical symptoms in Parkinson's disease due to significant MAO-B inhibition and radical scavenging effect.

  • 작물들의 염분에 대한 반응을 알아보기 위해 200mM의 NaCl이 들어있는 영양액을 공급하는 사경재배를 통하여 작물들의 내염성을 비교하였고,간척지 토양에 파종하여 작물들의 출현율과 초기생육을 관찰하였다. 작물종 중 내염성이 강한 종은 목화, 보리, 밀, 옥수수 등으로 이들은 200mM의 NaCl이 함유된 영양액에서 고사하지 않고 생육이 가능하다. 그러나 녹두, 완두, 팥, 콩과 갚은 콩과작물이 염분에 특히 약하였다. 작물을 간척지 토양에 파종했을때 건물중의 피해가 가장 심하였고, 출현율, 초장 순으로 염분에 대한 반응 정도가 작았다. 또한 염분 정도가 증가함에 따라 작물체의 엽수도 감소하였다. 공시 작물들의 초장이 50% 저해 되는 염분 농도를 추정해 볼 때 화본과의 벼, 보리, 옥수수는 각각 22.6, 56.9, 40.4ds/m의 EC에서 초장 50% 저해되었고, 콩과 작물인 녹두와 콩은 각각 22.6과 21.7ds/md의 EC에서 초장이 50% 감소됨을 추정할 수 있었다.

  • 우리나라 염해지의 식생 현황과 식물의 염분에 대한 내성 정도를 알아보기 위해 본 실험을 수행한 결과는 다음과 같다. 간척지를 비롯한 염해지의 주요 우점초종은 명아주과의 나문재, 칠면초, 해흥나물, 퉁퉁마디와 국화과의 갯개미취 등이었으며 탈염이 진행된 곳에는 부들, 갈대, 갯꾸러미풀 등의 화본과 식물이 우점하였다. 염해지의 우점 초종은 양액에 NaCl을 200mM로 첨가하여 실시한 사경재배에서도 강한 내염성을 나타내어 간척지의 식생과 이들의 내염성과는 일치성을 나타냈다. 내염성 식물인 명아주과의 나문재, 해흥나물, 가는갯는쟁이, 칠면초와 갯개미취 등이 내염성이 강한 식물종이었으며, 마디풀과의 개여뀌, 큰개여뀌, 털여뀌와 국화과의 별꽃아재비, 진득찰 등은 내염성이 매우 약한 종으로 나타났다. 갯벌 간척 후 탈염이 진행됨에 따라 초기에는 명아주과의 나문재, 해흥나물, 칠면조 등이 우점하고 이후에는 갯개미자리, 갯질경, 갯골풀, 갯꾸러미풀 등이 우점하게되고 이후에는 물대, 갈대 등이 우점하게될 것으로 추정되었다.
