In vitro에 의한 녹용 추출물의 생리 활성 효과 (The Biological Activity of Deer Antler Extract in vitro)

  • Our research objective was to examine the in vitro biological activity of deer antler(Nogyong in Korean) extract, including the antioxidative, nitrite scavenging, and tyrosinase inhibitory effects, as well as the antithrombotic, and angiotensin I converting enzyme(ACE) inhibitory activities. The carbohydrate, protein, fat, and mineral contents of the deer antler were 7.6%, 65.3%, 3.2% and 23.9%, respectively. The electron donating ability(EDA) by the reduction of 2,2'-diphenyl-1-picrylhydrazyl(DPPH) was 67.1%, and the inhibition rate of lipid peroxidation by the thiocyanate method using linoleic acid was 92.1% in 100 mg/ml of extract. The nitrite scavenging effects were pH dependent, and were highest at pH 1.2 and lowest at pH 6.0. The sample inhibition rate against tyrosinase was above 64.0%. The platelet aggregation induced by ADP(adenosine-5'diphosphate) was inhibited up to 51.7%, and the inhibitory effect was dependent on the sample concentration. Lastly, the inhibition rate of ACE was 47.5% in 100 mg/ml of deer antler extract.

생수의 저장기간에 따른 이·취미 및 미생물학적 변화에 관한 연구 (A Study on Odor-Savor and Microbiological Changes of Mineral Water Depending on the Storage Period)

  • As people seek a healthy, wealthy, and quality life more than before, interests in mineral water are picking up and the consumption continues to increase accordingly. In an experiment to examine changes in the water quality after mineral water on the market was kept indoor and outdoor for four weeks, there seemed to be no problem in external appearance. Water kept in an outdoor storage contained more odor-savor than water kept in an indoor storage. Odor was not detected in 90% of the sample, and savor not in 80% of it. As for the mechanism of odor-savor generation, the elution of chemical substances from a container may be taken into consideration. In general, odor and savor have turned out to be in close correlation to each other. The average number of bacteria in the sample mineral water was $200{\pm}10CFU/mL$, and such factors as area of storage and ozone post-treatment did not cause significant effects on the reproduction of common microbes. Hence, it is thought that strict regulations on a production process and reduction of the period of circulation will contribute to customers consumption rates.

  • Objectives It was the aim to examine the association of the thyroid-related hormones with cognitive function, depression, and subjective memory impairment in community-dwelling elders with questionable dementia. Methods The sample consisted of 399 community residents with 'questionable dementia' aged 60 or over in whom serum thyroid-related hormones [thyroid stimulating hormone (TSH) and thyroxine] had been assayed. Cognitive impairment was defined using the Korean version of the Consortium Establish a Registry for Alzheimer's Disease. Depression was diagnosed using the Korean version of Geriatric Depression Scale and subjective memory complaint (SMC) was checked using the subjective memory complaints questionnaire (SMCQ). Age, gender, education, and the presence of apolipoprotein E {\varepsilon}4 were included as covariates. Results There was a significant positive association between verbal fluency test (VFT) score and serum TSH levels (p = 0.01). There was a significant negative association between SMCQ total score and word list memory test (WLMT)(p = 0.002) or word list recall test (WLRT) score (p = 0.013). Conclusions Lower serum TSH levels were associated with semantic memory (VFT), and we found that SMC was associated with episodic memory (WLMT and WLRT) in this sample.

Characterization of Soil Microorganism from Humus and Indigenous Microorganism Amendments

  • This study was conducted to understand the dynamics of microbial communities of soil microorganisms, and their distribution and abundance in the indigenous microorganisms (IMOs) manipulated from humus collected from the forest near the crop field. The soil microorganisms originated from humus and artificially cultured microbial-based soil amendments were characterized by molecular and biochemical analyses. The bacterial population (2 × 106~13 × 106 CFU/g sample) was approximately 100-fold abundant than the fungal population (2 × 104~8 × 104 CFU/g sample). The 16S rDNA and ITS sequence analyses showed that the bacterial and fungal communities in humus and IMOs were mainly composed of Bacillus and Pseudomonas, and Trichoderma and Aspergillus species, respectively. Some of the bacterial isolates from the humus and IMOs showed strong inhibitory activity against soil-borne pathogenic fungi Fusarium oxysporum and Sclerotinia sclerotiorum. These bacteria also showed the siderophore production activity as well as phosphate solubilizing activity, which are requisite traits for biological control of plant pathogenic fungi. These results suggest that humus and IMOs could be a useful resource for sustainable agriculture.

Application of molecular biology methods to identify species of animal bones excavated from archaeological sites: focusing on the case of Bonghwang-dong, Gimhae

  • People have lived with and used animals for various purposes since the Paleolithic age. Therefore, animal bone research is interesting because it can infer the status of use, determine species, and ascertain the uses of animals that lived at the time. An analysis of ancient DNA was attempted to identify the species of ancient animal bones excavated from an archaeological site. Twelve animal bones from the Geumgwan Gaya period, excavated in Bonghwang-dong, Gimhae, were used in this study. After extracting DNA from the sample, polymerase chain reaction (PCR) gene amplification was performed. Species-specific primers of livestock groups such as pig, cattle, and deer were selected and used. This livestock group was a major source of protein for people who lived on the Korean Peninsula at that time. As a result, 11 sample species were identified. This study is contributes to the restoration of past life information by applying biological technologies to archaeological sites. It is also expected that such analyses of biological remains will ultimately be used to restore historical and cultural information.

Gelatinization and retrogradation characteristics of Korean rice cake in the presence of citric acid

  • The effect of citric acid on rice starch gelatinization and low-temperature (4 ℃) storage was studied in order to produce rice cake with a lower retrogradation rate. A citric acid solution in the ratio of 0, 0.5, 1.0, and 1.5% (w/w) of the water used during production was utilized. The gelatinization properties, gel strength, thermal properties, and texture analysis were evaluated to determine the retrogradation rate. The result showed that acid hydrolysis occurred in samples treated with citric acid. Thus, increasing citric acid decreased gelatinization temperature (58.63±1.98 to 45.84±1.24 ℃). The moduli of elasticity increased with increasing citric acid concentration, indicating an increased gel strength. Thermal analysis of starch showed that the onset, peak, and conclusion temperatures of retrogradation were increased significantly with the storage period and decreased with citric acid concentration. After 72 h of low-temperature storage (4 ℃), the retrogradation rate was lowest in the rice cake with 1.5% citric acid solution, with an increased ratio of 12.01 to 13.60% compared to the control sample, with a ratio of 12.99 to 43.54%. This shows a high retrogradation rate in the control sample. Additionally, sensory properties and retrogradation ratio suggest that the addition of 1.0% citric acid solution during rice cake production is efficient in retarding the retrogradation without an adverse effect on the rice cake modeling and acceptance.

제조용 충전물을 사용한 액체 크로마토그래피의 흡착특성 (Adsorption Characteristics of Liquid Chromatography with Preparative Packings)

  • 제조용 액체 크로마토그래피는 주로 생화학물질에서 유용성분을 분리하는 데 사용된다. 본 연구에서는 제조용 충전물($15{\mu}m$)이 채워진 역상 액체 크로마토그래피를 이용하여 sample size에 따른 흡착특성을 고찰하였다. 시료는 향미제인 5'-GMP이고 이동상은 $KH_2PO_4$ 수용액 20mM과 메탄올을 97:3 (vol.%)으로 혼합하여 사용하였다. 실험결과에 의하면 sample size가 증가해도 체류인자는 거의 일정하였지만 $1{\mu}g$ 이상에서는 peak의 모양이 비대칭성이 되었다. 또한 sample size가 증가함에 따라서 이론단수는 감소하였고 농도가 작은 경우 peak width가 크기 때문에 이론단수는 더 작았다. 본 실험조건에서 5'-GMP는 Freundlich 비선형흡착식에 비교적 잘 일치하였다.

  • Molecular diagnostics consists of three processes, which are a sample pretreatment, a nucleic acid amplification, and an amplicon detection. Among three components, sample pretreatment is an important process in that it can increase the limit of detection by purifying nucleic acid in biological sample from contaminants that may interfere with the downstream genetic analysis such as nucleic acid amplification and detection. To achieve point-of-care virus detection system, the sample pretreatment process needs to be simple, rapid, and automatic. However, the commercial RNA extraction kits such as Rneasy (Qiagen) or MagnaPure (Roche) kit are highly labor-intensive and time-consuming due to numerous manual steps, and so it is not adequate for the on-site sample preparation. Herein, we have developed a rotary microfluidic system to extract and purify the RNA without necessity of external mechanical syringe pumps to allow flow control using microfluidic technology. We designed three reservoirs for sample, washing buffer, and elution buffer which were connected with different dimensional microfluidic channels. By controlling RPM, we could dispense a RNA sample solution, a washing buffer, and an elution buffer successively, so that the RNA was captured in the sol-gel solid phase, purified, and eluted in the downstream. Such a novel rotary sample preparation system eliminates some complicated hardwares and human intervention providing the opportunity to construct a fully integrated genetic analysis microsystem.

Cs-137 감마선에 대한 생물학 연구용 시료의 흡수선량 결정에 관한 연구 (Absorbed Dose Determination for a Biological Sample Irradiated by Gamma Rays from a Cs-137 Source)

  • 본 연구에서는 생물학 연구용 방사선 조사기(Gamma Irradiator, Chiyoda Technol Co., Japan)에서 방출되는 Cs-137 감마선에 대하여 선량자료 구축을 위한 선량평가를 수행하였다. 대상 시료는 직경 3 cm인 구형의 물로 가정하였으며 공기커마로부터 선량계산 체계에 따라 흡수선량을 결정하였다. 측정값의 평가를 위하여 이론적 계산 및 몬테칼로 계산(MCNPX)을 이용하여 상호 비교하였다. 측정한 공기커마는 선원과 200 cm 이내에서 이론적 계산과 3.1% 이내로 일치하였다. 최종 결정된 흡수선량은 몬테칼로 계산과 100 cm와 200 cm에서 각각 1.9% 및 3.7%의 차이를 보였다. 본 결과는 비록 단편적이지만 생물 실험용 자료구축을 위한 기초 결과로서 타당하며 추후 다양한 형태의 시료 조건에 대해서 연구를 진행해야 할 것이다.

압출떡의 유통기한 연장을 위한 LED 조사의 Bacillus cereus 억제 효과 및 LED의 배열에 따른 빛의 조사 패턴 시뮬레이션 (Effect of LED light on the inactivation of Bacillus cereus for extending shelf-life of extruded rice cake and simulation of the patterns of LED irradiation by various arrays of LEDs)

  • 본 연구에서는 460 nm 파장의 청색 가시광선을 압출 떡의 표면에 조사하기 위하여 장치의 최적 디자인을 시뮬레이션을 통하여 확인하고 청색광이 압출 떡의 표면에서 식품 위해균인 B. cereus group에 미치는 영향을 확인하였다. LED 장치에서 광원모듈의 세 가지 배열(centered, cross, evenly spaced) 및 광원과 샘플 표면 사이의 거리(22, 32, 42 mm)에 따른 조사 면적에서 빛의 세기 패턴을 시뮬레이션을 통하여 계산하고, Petri factor를 통하여 균일도를 확인하였다. LED 배열의 균일도는 evenly spaced 배열에서 가장 균일한 패턴을 보였으며, 광원과의 거리가 32 및 42 mm일 경우 Petri factor가 0.9 이상으로 높은 균일도를 나타내었다. 떡볶이 떡에 LED 청색광을 조사한 경우 24 h 후 균 수가 초기 균 수에 비하여 감소하였으며, LED를 조사하지 않은 대조군에서는 초기 균 수가 증가하여 1.21 log CFU/g의 차이를 보였다. LED 조사 시 광원과 샘플의 거리가 증가할수록 Petri factor는 증가하나 감균 효과가 낮아지는 결과를 나타내었다. 따라서 Petri factor가 0.9 이상임을 만족하는 evenly spaced 배열의 32 mm 거리가 떡의 유통기한 연장을 위한 LED 장치의 디자인에 적합함을 확인하였다.
