Macroalgae as the Source for Environmental Assessment

  • Macroalgal tissues can be used as indicating materials for environmental assessment using several algal biotechnology techniques. As bioassay test organisms, macroalgal tissues are required as an axenic state for suitable biological indicators. Callus formation and blade regeneration under suitable culture conditions are also useful for the tests. Quantitative method using tetrazolium chloride or $alamarBlue^{TM}$ is devised on a rapid assessment of the seaweed viability. The use of RT-PCR especially differential display technique should provide the means for the detection and isolation of the responding genes induced by the environmental stress. Seaweed thriving in more environmental changes might contain more diverse biologically active substances.

Fuzzy Control Method for Balancing Left and Right Cardiac Output in Total Artificial Heart

  • Balancing left/right cardiac output is essential for the automatic control of total artificial hearts(TAH). A fuzzy logic-based control method is presented. We use left atrial pressure( LAP) ann right a'rial pressure( RAP ) as indicators for left/right balancing. The fuzzy controller has four input variables which are measured LAP and RAP and their gradients. Desired variations in left cardiac output(LCO) and right cardiac output(RCO) are cal- culated to keep LAP and RAP within the Physiological limlts. Computer simulations were performed to adjust fuzzy membership functions for variables and verify this control method. Results from simulations showed that LAP and RAP returned to the physiological limits while AoP and PAP stayed within the physiological limits.

Comparative Analysis of Fish Fauna and Community Structures Before and After the Artificial Weir Construction in the Mainstreams and Tributaries of Yeongsan River Watershed (영산강 수계의 본류 및 지천에서 4대강 사업 전.후의 어류분포 및 군집구조 특성 비교)

  • The purpose of this study was to analyze fish fauna and species compositions along with trophic guilds, tolerance indicators, and fish community conditions before weir construction (BWC) and after weir construction (AWC) in the Yeongsan River watershed. Total 45 and 44 fish species were sampled in BWC and AWC, respectively without any distinct differences through weir constructions. Fish fauna analysis revealed that the dominant species was the same, Zacco platypus with 24.3% and 20.8% in BWC and AWC, respectively. However, the subdominant species were Carassius auratus with 8.9% in BWC but Opsarichthys uncirostris amurensis with 20.3%, almost identical that of the dominant species AWC. This phenomenon showed the distinct modification of species composition in the watershed. We sampled the $1^{st}$ class endangered species, Liobagrus obesus in tributary stream as previously reported. Also Culter brevicauda was sampled in the mainstream of Yeongsan River watershed and this was the first sampled record in this watershed. One of the most important features were an increase of exotic species, such as Micropterus salmoides and Lepomis macrochirus, with 3.2% BWC vs. 10.2% AWC as well as the increase in tolerant species with 49.2% BWC vs. 73.7% AWC, indicating ecological degradation through weir construction. Overall, our results indicated that fish fauna and composition analyses showed distinct ecological degradations related to increases of exotic and tolerant species AWC. Further long-term studies of fish monitoring should be conducted in the future to configure existent status of river conditions and to provide key information in order to conserve the healthy ecosystem.

  • This study was carried out to assess problems related to legislative regulations and guidelines concerning some biological assessment systems applied to stream corridor in Korea. We comparatively reviewed the law of stream corridors and the guidelines for master plan concerned, and the law of water quality and health assessment criteria for the aquatic ecosystem concerned. Stream environments were not managed effectively due to the absence of detail regulations and the criteria for stream assessment. A biological assessment system was not equivalently integrated within the management of water resources in process implementation of projects resulting from the dualistic management system for stream corridors in Korea. The current biological assessment system was reflected to mainly physical habitats or only oriented to some aquatic species correlated with water quality. This system was also recognized as part of environment impact assessment based on an intensive survey method of most biological taxa. Conclusively rapid and quantitative assessment techniques based on advanced organisms, such as vegetation, fish and birds, etc. should be urgently provided for considering as representative indicators of stream conditions in Korea.

Landscape Scale Ecosystem Assessment Modelling Using Spatial Pattern Analysis of GIS: A Case Study of Yongin, Korea (GIS 공간유형분석 모형을 이용한 경관 규모 생태계의 평가기법)

  • The objective of this study were to develop landscape scale ecosystem assessment model, and apply the model for the assessment of the state and change of ecosystem of the study area, Yongin, Korea. Since natural ecosystem of the site has been deteriorated significantly during recent extensive residential development, it is essential to correctly assess ecosystem of the study site. Traditional ecosystem assessment mainly utilizing intensive field survey requires high cost, but the outcome rarely represents spatial pattern of the regional ecosystems. Ecosystem assesment of landscape scale based on landscape ecology can resolve most of the shortfalls of the traditional approach. The research method can be summarized as follows. First, extensive literature review on such topics as spatial pattern of ecosystem, ecosystem assessment of landscape scale, ecological analysis was carried out. Second, a model for the ecosystem assessment of landscape scale emphasizing spatial pattern of ecosystem was developed. This model evaluates three indicators; ecological integrity and biological diversity, watershed integrity, and landscape resilience of 11 watersheds in the study area. Finally, ecological assessment utilizing two sets of indicators, enhancement of and disturbance of ecosystem stability, was carried out. This assessment method is based on Environmental Monitoring and Assessment Program´s Landscape component(EMAP-L) of EPA(1994). The results of this study are as follows. First, the ecosystem assessment of landscape scale of the study area of Yongin, Korea, showed that escosystems of Tanchun01 and Chungmichun01 watersheds had the worst state in the study site in 1991. On the other hand, the ecosystems of Jinwechun01, Kyunganchun02, and Bokhachun01 watersheds had the most stable ecosystem in 1991. Second, ecosystems of Tanchun01, Shingal reservoir, and Kyunganchun01 watersheds were evaluated to be the worst state in the study site in 1996. And, ecosystems of Jinwechun01 and Gosam reservoir watersheds had the most stable ecosystem. Third, ecosystem of Tanchun01 watershed which incudes Suji residential development project site changed the most drastically between 1991 and 1996. The ecosystem of the watershed the most drastically deteriorated due to it´s proximity to Seoul and Bundang new town.

The Effect of Oral Administration of Herbal Medicines on Memory in Alzheimer's Disease Animal Models: A Review of Animal Study Reports Published in Korea (알츠하이머병 유발 동물모델에서 한약제재 경구투여가 기억에 미치는 영향에 대한 국내 연구보고 고찰)

  • Objectives: The objective of this study was to review the effect of oral administration of herbal medicines on the improvement of memory in Alzheimer's disease animal model reported in Korean domestic journals. Methods: The Korean databases (Koreantk, KISS) were searched with memory as a popular search term. During the searches, only animal study reports were reviewed. Data of animal models, intervention, observation methods of measuring indicators were extracted from the databases. Results: Typically, 36 articles were reviewed. Twenty-two studies used scopolamine to induce Alzheimer's disease, 24 studies used complex herbal medicines, and 12 studies used simple herbal medicines. Polygalae Radix and Acori Rhizoma were the most frequently used herbal medicines to improve memory in Alzheimer model. To evaluate the effect of herbal medicines, 36 studies used macroscopy, 16 studies used molecular biological analysis, 21 studies used biochemical analysis, 15 studies used histological analysis, and 11 studies used hematological analysis. Each study showed significant improvement with respect to memory indicators. Conclusions: Overall, the results suggest that treatment employing herbal medicines is an effective option to treat memory impairment in Alzheimer's disease.

Evaluating the Soil Salinity of Reclaimed Wastewater Irrigation in Paddy Plots using the Soil-Water-Atmosphere-Plant Model and Water Management Response Indicators (하수처리수의 재이용에 따른 SWAP 모형과 물관리반응지수를 이용한 논에서의 토양염분 평가)

  • 본 연구에서는 하수재이용에 따른 논에서의 토양염분 변화를 추정하기 위하여 경기도 화성시 수원환경사업소 인근에 위치한 병점지구를 대상으로 수문순환과 작물성장과의 관계를 고려한 SWAP (Soil-Water-Atmosphere-Plant) 모형을 이용하여 평가하였다. 실험에 사용한 관개용수는 지하수 (TR#1), 하수처리장 방류수+여과+UV (TR#3)로 분류하여 모형에 사용하였다. 유입관개수의 EC (Electrical Conductivity)는 지하수 관개수인 TR#1이 다른 처리구에 비해 작았고, TR#3의 경우 0.442~0.698 dS $m^{-1}$의 범위를 보였다. 모형의 보정과 검정을 위해서 대상지구에 FDR (Frequency Domain Reflection)을 설치하여 토양수분함유량과 염분농도를 토심에 따라 일단위로 모니터링 하였다. 토심 (50, 100, 140 cm)에 따른 토양함수량의 RMSE는 검정기간 중 TR#1에서 0.003~0.064 $cm^3\;cm^{-3}$, TR#3에서 0.001 $cm^3\;cm^{-3}$ 범위를 보여 주었고, 토양염분의 보정기간 중 토심별 RMSE는 TR#1에서 0.018~0.037 dS $m^{-1}$, TR#3에서 0.004~0.014 dS $m^{-1}$ 범위를 보여 적용성이 있는 것으로 나타났다. 토양내의 염분수지 분석 결과, 토양에서의 염분저장량이 (-)로 나타나 토양내로 침출되는 것으로 나타났으며, WMRI (Water Management Response Indicators)을 이용한 분석 결과, 높은 침투능으로 인하여 토양에서의 염분 집적 영향은 낮은 것으로 평가되었다.

A Quantitative Study of the Quality of Deconvolved Wide-field Microscopy Images as Function of Empirical Three-dimensional Point Spread Functions

  • In this work, for the first time, the quality of restoration in wide-field microscopy images after deconvolution was analyzed as a function of different Point Spread Functions using one deconvolution method, on a specimen of known size and on a biological specimen. The empirical Point Spread Function determination can significantly depend on the numerical aperture, refractive index of the embedding medium, refractive index of the immersion oil and cover slip thickness. The influence of all of these factors is shown in the same article and using the same microscope. We have found that the best deconvolution results are obtained when the empirical PSF utilized is obtained under the same conditions as the specimen. We also demonstrated that it is very important to quantitatively check the process' outcome using several quality indicators: Full-Width at Half-Maximum, Contrast-to-Noise Ratio, Signal-to-Noise Ratio and a Tenengrad-based function. We detected a significant improvement when using an indicator to measure the focus of the whole stack. Therefore, to qualitatively determinate the best deconvolved image between different conditions, one approach that we are pursuing is to use Tenengrad-based function indicators in images obtained using a wide-field microscope.

The Analysis of Intervention Studies for Patients with Metabolic Syndrome (대사성증후군 중재연구 논문 분석)

  • Purpose: This study was to analyze the trend of research on intervention for patients with metabolic syndrome. Method: Using Pubmed, Medline, and CINAHL search engines, a randomized controlled trial(RCT) researching titles such as 'metabolic syndrome', 'intervention', 'lifestyle modification', or 'community-based' were collected. A total of 16 researches were analyzed based on the guidelines of the National Cholesterol Education Program - Adult Treatment Panel(NCEP-ATP III). Results: 1) The total period of the intervention was from 12 to 24 weeks, the frequency was 3 to 5 times per week, and the duration of each session was from 45 to 60 minutes. The types of intervention included exercise, diet, and medication. Among these types, diet was performed most frequently. 2) The outcomes of the intervention was measured with physical aspects such as anthropometric measures, body composition, or biological markers. No studies have evaluated psychosocial outcomes such as quality of life. 3) In terms of effectiveness of the intervention, anthropometric indicators, body composition, or serological markers showed positive effects, whereas results on endothelial or urine indicators were inconsistent. Conclusion: Methodological research developing comprehensive therapeutic lifestyle modification programs and intervention studies are needed for patients with metabolic syndrome. In addition, effects should be evaluated with multidimensional perspectives.

Bacteriocin ("Vulnificin") Typing of Vibrio vulnificus

  • Vibrio vulnificus, a halophilic vibrio is an estuarine gram-negative bacteria that is associated with severe and frequently fatal wound infections and life-threatening septicemia. Bacteriocins are defined as antibacterial substance produced by various species of bacteria which are usually active against closely related organisms. Bacteriocins have found widespread application in epidemiological studies as specific markers of bacteria. It was proposed by Ha et al. (1990. J. Korean. Soc. Microbiol. 25: 586.) to give the bacteriocins produced by V. vulnificus the name "vulnificins". In the present study, a total of 72 strains of V. vulnificus isolated from patients and oysters were subjected to screen potential producers and indicators of vulnificin, applying ultraviolet induction method. Sensitivity of several strains of Serratia marcesans, Pseudomonas aeruginosa, Shigella flexneri, Salmonella typhi and Yersinia enterocolitica to vulnificins were also examined out. All the tested strains of V. vulnificus produced vulnificins active against indicator strains with various different inhibitory patterns. The spectrum of vulnificin activity and sensitive spectrum of indicator strains were considerably broad. Interestingly, almost all strains of S. marcescens, P. aeruginosa, Salmonella sp., Shigella sp. and Y. enterocolitica tested were sensitive to 1-7 vulnificin(s). Taken together, the present study demonstrated that all of the isolates of V. vulnificus produced vulnificins and that 8 good vulnificin producers and 10 good indicators were detected. These strains can be employed efficiently for establishing vulnificin typing scheme of V. vulnificus and for the detection of bacteriocinogeny and sensitivity in V. vulnificus. Biological role of vulnificin remains to be further elucidated.
