물달개비의 생물학적 방제인자 물달개비바구미의 생활사 및 기주특이성 (Life Cycle and Host Specificity of Tanysphyrus (Tanysphyroides) major Roelofs (Coleoptera : Curculionidae), a Possible Candidate Agent for the Biological Control of Monochoria vaginalis var. plantaginea)

  • 물달개비는 한국의 논에서 문제되는 난방제 잡초중의 하나이다. 물달개비의 생물학적 방제 인자로 물달개비바구미를 선발하였다. 생물학적 방제에 이용하기 위해 2006년과 2007년 계대사육을 실시하여 형태적, 생태적 특징을 조사하였다. 이 종은 1년에 한세대만 경과하며 성충으로 월동하고, 성충은 6월 하순부터 9월 하순까지 발생한다. 물달개비바구미는 알에서 성충까지 $22{\pm}0.7$일이 소요된다. 주요 농작물과 수생식물 60종을 대상으로 기주특이성을 조사 결과 물달개비의 생물학적 방제 곤충으로서의 가능성을 보였다.

  • 종자발아실험과 뿌리에서의 집락 형성실험으로 고추와 토마토의 근권 토양에서 plant growth promoting rhizobacteria(PGPR) 균주를 선발하였다. 그 중 포트실험에서 종자처리와 토양 관주처리 하였을 때 60% 이상 풋마름병 방제효과를 나타낸 SKU-78 균주에 대해서 포장에서의 방제효과 검증에 의해 생물농약으로의 개발 가능성을 확인하였으며, 생화학적 특성조사와 16S rDNA sequence 분석에 의해 SKU-78 균주를 Bacillus sp. SKU-78로 동정하였다.

Biological Control of n Severe Viral Strain Using a Benign Viral Satellite RNA Associated with Cucumber mosaic virus

  • Two strains of Cucumber mosaic virus (CMV) isolated in Kuwait were confirmed their infectivity based on symptomatology and host range on different cultivars of tomato (Lycopersicon esculentum), tobacco(Nicotiana tabacum L.) and squash (Cucurbita pepo). The pattern of symptoms differed for the two CMV strains in tomato and tobacco, showing severe stunting and mosaic symptoms with one strain designated KU2, and almost symptomless with the other strain designated KU1. A satellite RNA 5 (sat-RNA) was found to be associated with the KU1 strain and was characterized as a benign viral satellite RNA. Using reverse transcription and polymerase chain reaction (RT-PCR) with sat-RNA specific primers, an amplified PCR product of about 160bp was determined and analyzed by gel electrophoresis. This naturally occurring benign viral satellite RNA was successfully used as a biological control agent to protect tomato plants against the severe KU2 strain. Tomato plants grown in plant-growth chambers, were preinoculated with KU1 containing the benign viral satellite and then challenge inoculated with the severe KU2 strain at different time intervals. All plants challenged three weeks after preinoculation showed nearly complete protection from subsequent infection by the severe strain. This biological control technology using plant viruses was found protective and could be successfully established sooner after the preinoculation.

  • The Pacific tapeworm Adenocephalus pacifcus (syn. Diphyllobothrium pacificum) is a causative agent of diphyllobothriosis occurred in Pacific coast of South America, mainly in Peru. Source of infections are traditional meal from raw or undercooked marine fish such as "cebiche". We confirmed 3 new cases, one including scolex and the other two headless. A strobila 46 cm long without scolex was discharged from an 8-year-old boy before treatment. Specimens were confirmed morphologically by presence of tegumental protuberances on proglottids and small sized eggs. Partial sequence of cytochrome c oxidase subunit 1 gene was congruent with A. pacificus sequences.

Anti-Melanogenic Activities of Ranunculus chinensis Bunge via ERK1/2-Mediated MITF Downregulation

  • Research on whitening materials using natural alternatives is actively being conducted. The aim of this study was to investigate the in vitro inhibitory effects of Ranunculus chinensis Bunge (RCB) on melanogenesis and associated enzymes, such as tyrosinase, tyrosinase-related protein (TRP)-1, and TRP-2 in B16F10 murine melanoma cells. We found that RCB extract significantly attenuated melanin synthesis and reduced the activity of intracellular tyrosinase, a rate-limiting melanogenic enzyme. Western blot analysis showed that RCB extract decreased the protein expression of tyrosinase and TRP-1. In addition, it significantly decreased the expression of microphthalmia-associated transcription factor (MITF), a key regulator of melanogenesis. Extracellular signal-regulated kinase (ERK) activation has been reported to be involved in the inhibition of melanogenesis. Thus, we investigated whether the hypopigmentary effects of RCB extract were related to the activation of ERK. RCB extract induced ERK phosphorylation in a dose-dependent manner. Furthermore, it markedly inhibited body pigmentation in a zebrafish model. Our results suggest that RCB extract inhibits melanogenesis by activating ERK pathway-mediated suppression of MITF and its downstream target genes, including tyrosinase. Therefore, RCB extract can be used as a whitening agent in the development of functional cosmetics.

생물무기 특성과 사례연구 (Characteristics of Biological Agent and relavent case study)

  • 생물테러에 사용되는 생물무기는 세균과 바이러스, 리켓치아, 곰팡이 등을 가공하여 만들어지며, 화학작용제, 방사능작용제와 함께 대량살상무기(Weapons of Mass Destruction, WMD)로 분류되어진다. 생물무기는 다른 대량살상무기들에 비하여 개발과 생산의 용이성, 저렴한 비용, 은닉살포가 가능하다는 운용상 이점들이 존재한다. 본 논문은 생물무기 개발이 이루어진 역사와 현존하는 생물학적 위협에 대해 분석하고, 생물작용제의 물리적 특성과 인체내 감염기작을 기초로 생물무기 공격으로 인한 사회적 영향을 예측하였다. 스벨드롭스크 탄저균유출사고, 아랄스크 두창발생, 2001년 탄저테러 등 생물무기관련 역사적 사례들에서 사고 인지형태, 작용제 오염확산범위와 피해자의 질병특성을 분석한 결과, 생물테러는 공격 즉시 인지가 어렵고, 광범위한 확산이 가능하여 대규모 노출자를 발생시키며, 호흡기관련 중증질환을 일으켜 보건 의료기관에 막대한 부담을 일으킬 것이라는 사실을 발견하였다. 이를 토대로 생물테러에 효과적으로 대응하기 위해서 요구되는 보건대응역량을 제시하였다. 효과적인 증후군감시체계 및 관련질병 보고체계를 운영하여 신속히 이상징후를 파악해야하며, 실험실진단체계를 통해 병원체의 확진을 실시하여야한다. 또한 보건기관은 역학조사 시 수사기관과의 공조를 통해 잠재적노출자를 명확히 규명하여야 하며, 마지막으로 인적 물적 자원을 효율적으로 운용하여 집단예방조치가 효과적으로 이루어지도록 되어야할 것이다. 이러한 보건대응역량을 지속적으로 강화한다면 생물테러위협에 성공적으로 대응할 수 있을 것이다.

경화제 변화에 따른 WLP(Wafer Level Package)용 신규 Epoxy Resin System의 경화특성 (Cure Properties of Novel Epoxy Resin Systems for WLP (Wafer Level Package) According to the Change of Hardeners)

  • The curing characteristics of naphthalene type epoxy resin systems according to the change of curing agent were investigated to develop a new next-generation EMC(Epoxy Molding Compound) with excellent warpage characteristics, low thermal expansion, and excellent fluidity for WLP(Wafer Level Package). As epoxy resins, DGEBA, which are representative bisphenol type epoxy resins, NE-16, which are the base resins of naphthalene type epoxy resins, and NET-OH, NET-MA, and NET-Epoxy resins newly synthesized based on NE-16 were used. As a curing agent, DDM (Diamino Diphenyl Methane) and CBN resin with naphthalene moiety were used. The curing reaction characteristics of these epoxy resin systems with curing agents were analyzed through thermal analysis experiments. In terms of curing reaction mechanism, DGEBA and NET-OH resin systems follow the nth curing reaction mechanism, and NE-16, NET-MA and NET-Epoxy resin systems follow the autocatalytic curing reaction mechanism in the case of epoxy resin systems using DDM as curing agent. On the other hand, it was found that all of them showed the nth curing reaction mechanism in the case of epoxy resin systems using CBN as the curing agent. Comparing the curing reaction rate, the epoxy resin systems using CBN as the curing agent showed a faster curing reaction rate than them with DDM as a hardener in the case of DGEBA and NET-OH epoxy resin systems following the same nth curing reaction mechanism, and the epoxy resin systems with a different curing mechanism using CBN as a curing agent showed a faster curing reaction rate than DDM hardener systems except for the NE-16 epoxy resin system. These reasons were comparatively explained using the reaction rate parameters obtained through thermal analysis experiments. Based on these results, low thermal expansion, warpage reduction, and curing reaction rate in the epoxy resin systems can be improved by using CBN curing agent with a naphthalene moiety.
