Unique epithelial expression of S100A calcium binding protein A7A in the endometrium at conceptus implantation in pigs

  • Objective: S100A7A, a member of the S100 protein family, is involved in various biological processes, including innate immunity, antimicrobial function, and epithelial tumorigenesis. However, the expression and function of S100A7A in the endometrium during the estrous cycle and pregnancy are not well understood in pigs. Therefore, this study determined the expression and regulation of S100A7A at the maternal-conceptus interface in pigs. Methods: We obtained endometrial tissues from pigs throughout the estrous cycle and pregnancy, conceptus tissues during early pregnancy, and chorioallantoic tissues during midto late pregnancy and analyzed the expression of S100A7A in these tissues. We also determined the effects of steroid hormones, estradiol-$17{\beta}$ ($E_2$) and progesterone, and interleukin-$1{\beta}$ (IL1B) on S100A7A expression in endometrial tissues. Results: We found that S100A7A was expressed in the endometrium during the estrous cycle and pregnancy in a pregnancy status- and stage-dependent manner and was localized to endometrial luminal epithelial (LE) and superficial glandular epithelial cells with strong intensity in LE cells on day 12 of pregnancy. Early stage conceptuses and chorioallantoic tissues from day 30 to term pregnancy also expressed S100A7A. The expression of S100A7A was increased by $E_2$ and IL1B in endometrial tissues. Conclusion: S100A7A was expressed at the maternal-conceptus interface at the initiation of implantation in response to conceptus-derived estrogen and IL1B and could be a unique endometrial epithelial marker for conceptus implantation in pigs. These findings provide an important insight into the understanding of conceptus-endometrial interactions for the successful establishment of pregnancy in pigs.

A Basic Study on the Variation of Temperature Characteristics for Attenuation Coefficient and Sound Velocity in Biological Tissues

  • This study is concerned with the temperature dependence characteristics of ultrasound parameters in biological tissues, which are basic on the noninvasive deep body temperature estimation. Used parameters are ultrasonic attenuation coefficient and sound velocity In order to accomplishment our purpose, several signal processing methods were used. Attenua4iorl coefficient was estimated by spectral difference method and sound velocity was estimated by P-P method. And we also examined these methods through a series of IN VITRO experi mentis that used tissue-mimicking phantom samples and biological tissue samples. In order to imitate the biological soft tissue two kinds of phantom samples are used, one is agar phantom sample which is composed of agar, graphite, N-propyl alcohol and distilled water, and the other is fat phantom sample which is composed of pure animal fat. And the ultrasound transmission mode and reflection mode experiments are performed on the pig's spleen, kidney and fat. As a result, it is found that the temperature characteristics are uniform in case of phan- tom samples but not in biological tissues because of complicate wave propagation within them. Consequently, the possibility of temperature measurement using ultrasound on biological tissue is confirmed and its results may contribute to the establishment of reference values of internal temperature measurement of biological tissues.

Temperature Compensation of Complex Permittivities of Biological Tissues and Organs in Quasi-Millimeter-Wave and Millimeter-Wave Bands

  • This study proposes a temperature compensation method of the complex permittivities of biological tissues and organs. The method is based on the temperature dependence of the Debye model of water, which has been thoroughly investigated. This method was applied to measured data at room temperature for whole blood, kidney cortex, bile, liver, and heart muscle. It is shown that our method can compensate for the Cole-Cole model using measured data at 20 $^{\circ}C$, given the Cole-Cole model based on measured data at 35 $^{\circ}C$, with a root-mean-squared deviation of 3~11 % and 2~6 % for the real and imaginary parts of the complex permittivities, respectively, among the measured tissues.

  • A novel nonlocal model with one thermal relaxation time is presented to investigates the thermal damages and the temperature in biological tissues generated by laser irradiations. To obtain these models, we used the theory of the non-local continuum proposed by Eringen. The thermal damages to the tissues are assessed completely by the denatured protein ranges using the formulations of Arrhenius. Numerical results for temperature and the thermal damage are graphically presented. The effects nonlocal parameters and the relaxation time on the distributions of physical fields for biological tissues are shown graphically and discussed.

  • E3 ligase BRUTUS (BTS), a putative iron sensor, is expressed in both root and shoot tissues in seedlings of Arabidopsis thaliana. The role of BTS in root tissues has been well established. However, its role in shoot tissues has been scarcely studied. Comparative transcriptome analysis with shoot and root tissues revealed that BTS is involved in regulating energy metabolism by modulating expression of mitochondrial and chloroplast genes in shoot tissues. Moreover, in shoot tissues of bts-1 plants, levels of ADP and ATP and the ratio of ADP/ATP were greatly increased with a concomitant decrease in levels of soluble sugar and starch. The decreased starch level in bts-1 shoot tissues was restored to the level of shoot tissues of wild-type plants upon vanadate treatment. Through this study, we expand the role of BTS to regulation of energy metabolism in the shoot in addition to its role of iron deficiency response in roots.

Fabrication of multi-well platform with electrical stimulation for efficient myogenic commitment of C2C12 cells

  • To engineer tissue-like structures, cells are required to organize themselves into three-dimensional networks that mimic the native tissue micro-architecture. Here, we present agarose-based multi-well platform incorporated with electrical stimulation to build skeletal muscle-like tissues in a facile and highly reproducible fashion. Electrical stimulation of C2C12 cells encapsulated in collagen/matrigel hydrogels facilitated the formation 3D muscle tissues. Consequently, we confirmed the transcriptional upregulations of myogenic related genes in the electrical stimulation group compared to non-stimulated control group in our multi-well 3D culture platform. Given the robust fabrication, engineered muscle tissues in multi-well platform may find their use in high-throughput biological studies drug screenings.

Structural and Quantitative Expression Analyses of HERV Gene Family in Human Tissues

  • Human endogenous retroviruses (HERVs) have been implicated in the pathogenesis of several human diseases as multi-copy members in the human genome. Their gene expression profiling could provide us with important insights into the pathogenic relationship between HERVs and cancer. In this study, we have evaluated the genomic structure and quantitatively determined the expression patterns in the env gene of a variety of HERV family members located on six specific loci by the RetroTector 10 program, as well as real-time RT-PCR amplification. The env gene transcripts evidenced significant differences in the human tumor/normal adjacent tissues (colon, liver, uterus, lung and testis). As compared to the adjacent normal tissues, high levels of expression were noted in testis tumor tissues for HERV-K, in liver and lung tumor tissues for HERV-R, in liver, lung, and testis tumor tissues for HERV-H, and in colon and liver tumor tissues for HERV-P. These data warrant further studies with larger groups of patients to develop biomarkers for specific human cancers.

Analysis of Differentially Expressed Genes Between Leaves and Grain Tissues of Three Wheat Cultivars

  • Wheat is a very important crop as a food source worldwide, but gluten in wheat causes a variety of allergic reactions. Previous studies have developed ${\omega}-5$ gliadin deleted O-free, known as the central antigen of WDEIA (wheat-dependent exercise-induced anaphylaxis). In this study, we performed RNA sequencing on the grains and leaves of the allergic-reduced species O-free and their cultivars, Keumkang and Olgeuru, to analyze differentially expressed genes (DEG) based on different cultivars and tissues. Tissues of all species were biologically repeated three times. We used bowtie2 version 2.3.5.1 to get sequence data from RNAseq and used cufflinks and Tophat programs to find DEG. When comparing leaf and grain tissues, a total of 1,244 DEGs were found in the leaf tissues while only 563 DEGs were found in the grain tissues. As a result of gene ontology analysis of differentially expressed genes, the leaf tissues were mostly included in the "catalytic activity" part of molecular function, "metabolic process" part of biological process, and "membrane" part of cell component. The grain tissues were mostly included in the "metabolic process" part of biological process, "binding" and "catalytic activity" part of molecular function, and "membrane, cell, cell part" parts of cell component. Based on these results, we present information on the differentially expressed genes of the three cultivars of leaves and grains. This study could be an important basis for studying the characteriztion of O-free.

Inhibitors of apoptosis: expression and regulation in the endometrium during the estrous cycle and at the maternal-conceptus interface during pregnancy in pigs

  • Objective: Caspase-mediated apoptosis plays a crucial role in the regulation of endometrial and placental function in females. Caspase activity is tightly controlled by members of the inhibitors of apoptosis proteins (IAPs) family. However, the expression and regulation of IAPs at the maternal-conceptus interface has not been studied in pigs. Therefore, we determined the expression of IAP family members baculovirus IAP repeat-containing 1 (BIRC1) to BIRC6 at the maternal-conceptus interface in pigs. Methods: We obtained endometrial tissues from pigs at various stages of the estrous cycle and pregnancy, conceptus tissues during early pregnancy, and chorioallantoic tissues during mid- to late pregnancy and analyzed the expression of IAPs. Furthermore, we determined the effects of the steroid hormones estradiol-17β (E2) and progesterone on the expression of IAPs in endometrial explant tissue cultures. Results: During the estrous cycle, BIRC2 and BIRC5 expression varied cyclically, and during pregnancy, endometrial BIRC1, BIRC2, BIRC3, BIRC4, and BIRC5 expression varied in a stage-specific manner. Conceptus and chorioallantoic tissues also expressed IAPs during pregnancy. The BIRC2 and BIR3 mRNAs were localized to luminal epithelial cells, and BIRC4 proteins to glandular epithelial cells in the endometrium. Exposure of endometrial tissues to E2 increased the expression of BIRC6, while progesterone increased the expression of BIRC1, BIRC4, and BIRC6 in a dose-dependent manner. Conclusion: These results indicated that IAPs were expressed in the endometrium during the estrous cycle and at the maternal-conceptus interface during pregnancy in a stage-specific manner. In addition, steroid hormones were found to be responsible for the expression of some IAPs in pigs. Together, the results suggested that IAPs may play important roles in endometrial and placental functions by regulating caspase action and apoptosis at the maternal-conceptus interface.

카드뮴 노출이 붕어(Carassius auratus) 조직에 미치는 영향 (Effects of Cadmium Exposure on Tissues of Carassius auratus)

  • 본 연구는 Cd에 노출시킨 붕어를 통하여 각 조직에 축적된 Cd 양과 항산화효소 활성을 통해 생리적인 변화 및 아가미와 신장 조직에서의 미세구조 변화를 조사하였다. Cd을 처리한 실험군 붕어 조직 내 Cd의 함량은 아가미 조직에서 가장 높았고 근육 조직에서 가장 낮았다. 또한 모든 조직에서 Cd의 함량은 노출기간이 길어질수록 증가하여 노출 20일에 가장 높았다. 항산화효소의 활성은 아가미와 근육 조직에서 높았으며, 신장과 간 조직에서 낮았다. 모든 조직에서 SOD의 활성이 가장 높았으며, SOD와 CAT의 활성은 10일에 높은 활성을 보이고 감소하는 유사한 결과를 보였다. Cd에 20일간 노출시킨 붕어의 아가미 조직은 대조군 아가미 조직에 비하여 노출기간이 길어질수록 새변 사이에 점액세포의 수가 증가하였으며, 이차새변에서는 부종, 상피세포의 박리와 새변과 새변이 융합한 형태도 관찰되었다. 또한 아가미 조직의 미토콘드리아와 핵에서는 막의 손상이 확인되었다. 신장 조직은 대조군에서 보우만 주머니 안에 사구체가 꽉 찬 정상구조였으나 노출기간이 길어짐에 따라 사구체가 수축하여 보우만 주머니 안의 공간이 넓은 형태로 관찰되었다.
