Biosurface Organic Chemistry: Interfacial Chemical Reactions for Applications to Nanobiotechnology and Biomedical Sciences

  • In this review, the field of biosurface organic chemistry is defined and some examples are presented. The aim of biosurface organic chemistry, composed of surface organic chemistry, bioconjugation, and micro- and nanofabrication, is to control the interfaces between biological and non-biological systems at the molecular level. Biosurface organic chemistry has evolved into the stage, where the lateral and vertical control of chemical compositions is achievable with recent developments of nanoscience and nanotechnology. Some new findings in the field are discussed in consideration of their applicability to nanobiotechnology and biomedical sciences.

Relationship between Phosphorus Release and Intracellular Storage Polymer Synthesis by Phosphorus Accumulating Organisms (인축적 미생물의 인방출과 세포내 저장물질 합성관계)

  • Biological phosphorus removal is characterized by complex interactions between different intracellular components of energy as PHA. Therefore, fundamental understanding of the behavior of the intracellular components and their influence on the removal of phosphorus is essential before control strategies to stabilize the proper process. The purpose of this study is to investigate relationship between release of phosphorus and synthesis of intracellular storage polymer. Mass of stored intracellular storage polymer was 21.2 mg PHA/L, 28.8 mg PHA/g MLSS. And phosphorus release/intracellular storage polymer synthesis rate was 1.8545 mg stored polymer/mg Phosphate. In the aerobic phase, mass of PAOs synthesis is 49.37 mg PAOs/L. And PAOs fraction was 6.7-6.9%. Thus intracellular storage polymer synthesis by PAOs is calculated as 493mg PHA/g PAOs.

Intensified Low-Temperature Fischer-Tropsch Synthesis Using Microchannel Reactor Block : A Computational Fluid Dynamics Simulation Study (마이크로채널 반응기를 이용한 강화된 저온 피셔-트롭쉬 합성반응의 전산유체역학적 해석)

  • Fischer-Tropsch synthesis reaction converts syngas (mixture of CO and H2) to valuable hydrocarbon products. Simulation of low temperature Fischer -Tropsch Synthesis reaction and heat transfer at intensified process condition using catalyst filled single and multichannel microchannel reactor is considered. Single channel model simulation indicated potential for process intensification (higher GHSV of $30000hr^{-1}$ in presence of theoretical Cobalt based super-active catalyst) while still achieving CO conversion greater than ~65% and $C_{5+}$ selectivity greater than ~74%. Conjugate heat transfer simulation with multichannel reactor block models considering three different combinations of reactor configuration and coolant type predicted ${\Delta}T_{max}$ equal to 23 K for cross-flow configuration with wall boiling coolant, 15 K for co-current flow configuration with subcooled coolant, and 13 K for co-current flow configuration with wall boiling coolant. In the range of temperature maintained (498 - 521 K), chain growth probability calculated is desirable for low-temperature Fisher-Tropsch Synthesis.

t10,c12 Conjugated Linoleic Acid Upregulates Hepatic De Novo Lipogenesis and Triglyceride Synthesis via mTOR Pathway Activation

  • In mice, supplementation of t10,c12 conjugated linoleic acid (CLA) increases liver mass and hepatic steatosis via increasing uptake of fatty acids released from adipose tissues. However, the effects of t10,c12 CLA on hepatic lipid synthesis and the associated mechanisms are largely unknown. Thus, we tested the hypothesis that gut microbiota-producing t10,c12 CLA would induce de novo lipogenesis and triglyceride (TG) synthesis in HepG2 cells, promoting lipid accumulation. It was found that treatment with t10,c12 CLA ($100{\mu}M$) for 72 h increased neutral lipid accumulation via enhanced incorporation of acetate, palmitate, oleate, and 2-deoxyglucose into TG. Furthermore, treatment with t10,c12 CLA led to increased mRNA expression and protein levels of lipogenic genes including SREBP1, ACC1, FASN, ELOVL6, GPAT1, and DGAT1, presenting potential mechanisms by which CLA may increase lipid deposition. Most strikingly, t10,c12 CLA treatment for 3 h increased phosphorylation of mTOR, S6K, and S6. Taken together, gut microbiota-producing t10,c12 CLA activates hepatic de novo lipogenesis and TG synthesis through activation of the mTOR/SREBP1 pathway, with consequent lipid accumulation in HepG2 cells.

Enhanced Synthesis of Active rPA in the Continuous Exchange Cell-free Protein Synthesis [CECF] System utilizing Molecular Chaperones (분자 샤페론을 사용한 연속확산식 무세포단백질 발현 시스템에서의 재조합 Plasminogen Activator의 효율적 발현)

  • In this report, we describe that the use of GroEL/GroES-enriched S30 extract remarkably enhances the solubility and enzymatic activity of cell-free synthesized rPA, which requires the correct formation of 9 disulfide bonds for its biological activity. We found that the stable maintenance of redox potential is necessary, but not sufficient for the optimal expression of active rPA. In a control reaction without using additional molecular chaperones, most of the rPA molecules were aggregated almost instantly after their expression and thus failed to exhibit the enzymatic activity. However, by the use of GroEL/GroES-enriched extract, combined with IAM-treatment, approximately $30{\mu}g/ml$ of active rPA was expressed in the cell-free synthesis reaction. This result not only demonstrates the efficient production of complex proteins, but also shows the control and flexibility offered by the cell-free protein synthesis system.

A novel nicotinoyl peptide, nicotinoyl-LVH, for collagen synthesis enhancement in skin cells

  • A novel Nicotinoyl fused peptide, Nicotinoyl-LVH, was synthesized by solid phase peptide synthesis method, purified, and tested in cultured skin cells. Nicotinoyl-LVH enhanced the expression level of collagen mRNA and its fragments in fibroblasts. These data show that this novel Nicotinoyl peptide is a promising biomaterial in the anti-aging functional cosmetic market.

Mycorrhizal Formations and Seedling Growth of Pinus desiflora by in vitro Synthesis with the Inoculation of Ectomycorrhizal Fungi

  • The mycelia were directly isolated from eight species of fungal basidiocarps, confirmed to the ectomycorrhiza in the roots from the fields(forestry); Suillus bovinus, Paxillus involutus, Lactarius hysginus, Russula fragilis, Lepista nuda, Lyophyllum shimeji, Tricholoma matsutake, and Russula integra. The mycelia were pure-cultured with several transferring in various agars, and inoculated to the roots of pine(Pinus densiflora) seedling by in vitro method. After ten months growth under artificially aseptic conditions, all pine seedlings inoculated were stimulated at the growth-height, whereas those not inoculated were nearly dead. Also, the ramifications of ectomycorrhizal pine roots formed in the synthetic in vitro systems and were various according to the different mycelia. Synthesis of ectomycorrhiza were clearly confirmed in ten months growth, but not distinguished at this moment. It was clearly proved that the mycelia isolated caused the ectomycorrhizae in the roots of pine seedlings.

  • For the synthesis of new antifungal agents, We have synthesized four new ketoconazole derivatives were synthesized by the reaction of cis-[2-(2,4-dichlorophenyl)-2-(1H-imidazol-1-yl)-1,3-dioxolan-4-yl]methane sulfonate with isolated fig compounds. These compounds were showed strong antifungal activity against C, albicans ATCC 10231. C, utilis. S, cerevisiae ATCC 9763. A and niger ATCC 9029. Among them, sample No.(13) showed potent inhibition activity. Generally, other samples showed biological activity in vitro test. The above results showed the possibility of the development of new antifungal agents.

Effect of microwave irradiation on lipase-catalyzed reactions in ionic liquids

  • Microwave-assisted organic synthesis has gained a remarkable interest over the past years because of its advantages - (i) rapid energy transfer and superheating, (ii) higher yield and rapid reaction, (iii) cleaner reactions. Ionic liquids are well known for their unique properties such as negligible vapor pressure and high thermal stability. With these properties, ionic liquids have gained increasing attention as green, multi-use reaction media. Recently, ionic liquids have been applied as reaction media for biocatalysis. Lipase-catalyzed reactions in ionic liquids provide high activity and yield compared to conventional organic solvents or solvent free system. Since polar molecules are generally good absorbent to microwave radiation, ionic liquids were investigated as reaction media to improve activity and productivity. In this study, therefore, the effect of microwave irradiation in ionic liquids was investigated on lipase catalyzed reactions such as benzyl acetate synthesis and caffeic acid phenethyl ester synthesis. Comparing to conventional heating, microwave heating showed almost the same final conversion but increased initial reaction rate (3.03 mM/min) compared to 2.11 mM/min in conventional heating at $50^{\circ}C$.
