Manufacturing Protein-DNA Chip for Depigmenting Agent Screening (전사인자 저해제 통한 미백제 탐색용 단백질 칩 제작)

  • An attempt was made to develop a proteinchip for screening of MITF (microphthalmia transcription factor) inhibitor. Binding of MITF to E-box causes transcription of several pigmenting genes including tyrosinase gene. We investigated binding of MITF and its DNA binding site (E-box) using a protein-DNA chip with various detection methods including flurorescence (Cyt3). SPR (surface plasmon resonance) and SPRi (surface plasmon resonance imaging). A fusion protein (MITF-Maltose Binding Protein) was attached on the glass plate by chemical modification. An inhibitory synthetic DNA oligomer, artificially designed based on the E-box sequence, inhibited the binding of MITF and E-box. These results showed the potentials of flurorescence-based MITF protein chip as a microarray for high throughput screening (HTS) system of depigmenting agents.

In situ Recovery of hGM-CSF in Transgenic Rice Cell Suspension Cultures (형질전환 벼 현탁세포 배양에서 hGM-CSF의 in situ Recovery 연구)

  • Production of foreign proteins by transgenic plant cell cultures has several advantages such as post-translational modification, low risk of product contamination and low-cost production and purification. However, target proteins are degraded by extracellular proteases existing in the media. A solution to this problem is the use of perfusion culture and ion exchange chromatography for the application of integrated bioprocess using in situ recovery. With this method, production of human granulocyte-macrophage colony-stimulating factor (hGM-CSF) was investigated in this study. First, optimization of cell concentration during the induction phase for the production of hGM-CSF was examined. As cell concentration increased, the level of hGM-CSF was decreased due to the presence of extracellular proteases. Induction using sugarfree media produced 33% more hGM-CSF. The effects of pH on the binding of hGM-CSF to cationic and anionic exchange resins were also investigated. In terms of stability, optimal pH was found to be 5~7. In the case of using buffer exchange when CM-Sepharose was used as a cationic exchange resin, optimal pH for binding was 4.8 and adsorption yield was 77%. When DEAE-Sepharose was used as an anionic exchange resin, it was 5.5 (74%). Without buffer exchange, optimal pH was 4.6 (84%). From these results, an integrated bioprocess using in situ recovery with simultaneous production and separation of foreign protein in transgenic plant cell suspension cultures was found to be feasible.

Enhanced Sialylation of Albumin-erythropoietin by Biphasic Cultivation in CHO Cells (CHO 세포의 2단계 배양을 통한 Albumin-erythropoietin의 시알산 증대)

  • In glycoprotein, Terminal sialic acid residues of N-linked glycan are imperative things because they prevent the recognition from asialoglycoprotein-receptor that affect the half-life of glycoproteins. So establishment of culture process for enhancing sialic acid is important to maximize sialic acid contents of glycoprotein. In this study, we investigated effects of biphasic culture of Chinese hamster ovary (CHO) cells producing albumin-erythropoietin to increase sialylation. Biphasic cultures were performed with shift of $CO_2$ concentrations and temperatures at day 5 when viable cell density was decreased and sialidase was started to be released by cell lysis. The examined temperature set points were 33, 35 and $37^{\circ}C$ respectively and the $CO_2$ concentration was 1, 5, 10 and 15%. We confirmed that sialidase activity was the lowest in biphasic culture that was shifted from normal culture condition to 1% of $CO_2$ and $33^{\circ}C$ on day 5. However, the temperature and concentration of $CO_2$ have little effect on activity of ${\alpha}2,3$-sialyltransferase. Also, sialic acid contents were enhanced 1.13-fold higher than that in control culture. In conclusion, Biphasic cultivation in CHO cells led to inhibition of sialidase activity and increases of sialylated glycan.

  • Traditional semiconductor process control has been performed through statistical process control techniques in a constant process-recipe conditions. However, the complexity of the interior of the etching apparatus plasma physics, quantitative modeling of process conditions due to the many difficult features constraints apply simple SISO control scheme. The introduction of the Advanced Process Control (APC) as a way to overcome the limits has been using the APC process control methodology run-to-run, wafer-to-wafer, or the yield of the semiconductor manufacturing process to the real-time process control, performance, it is possible to improve production. In addition, it is possible to establish a hierarchical structure of the process control made by the process control unit and associated algorithms and etching apparatus, the process unit, the overall process. In this study, the research focused on the methodology and monitoring improvements in performance needed to consider the process management of future developments in the semiconductor manufacturing process in accordance with the age of the APC analysis in real applications of the semiconductor manufacturing process and process fault diagnosis and control techniques in progress.

Electrochromic Properties of Li+-Modified Prussian Blue (리튬이온이 첨가된 프루시안 블루의 전기변색 특성 연구)

  • The durability problem of Prussian blue in non-aqueous $Li_+$-based electrolytes has been due to the degradation of the Prussian blue electrode matrix during the insertion/extraction processes by $Li_+$. In this work, we designed and synthesised the Prussian blue without reducing the electrochromic performance in non-aqueous $Li_+$-based electrolytes. Prussian blue was electrodeposited on a glass which has ITO coating, and the coating solution is a mixture solution of $FeCl_3\;and\;K_3Fe(CN)_6$ with deionized water added HCl, KCl, and LiCl, respectively. The durability of Prussian blue was evaluated by an in-situ transmittance measurement during a continuous and pulse potential cycling test, and measured by electroactive layer thickness due to evaluating the degradation.

  • The transition of interconnection metal from aluminum alloy to copper has been introduced to meet the requirements of high speed, ultra-large scale integration, and high reliability of the semiconductor device. Since copper, which has low electrical resistivity and high resistance to degradation, has different electrical and material characteristics compared to aluminum alloy, new related materials and processes are needed to successfully fabricate the copper interconnection. In this review, some important factors of multilevel copper damascene process have been surveyed such as diffusion barrier, seed layer, organic additives for bottom-up electro/electroless deposition, chemical mechanical polishing, and capping layer to introduce the related issues and recent research trends on them.

Preparation of Polypropylene Grafted Polystyrene Sulfonic Acid Membranes for DMFCs in Supercritical CO2 (초임계 이산화탄소 함침을 이용한 연료전지용 폴리스타이렌/폴리프로필렌 복합막의 제조)

  • The composite membranes were made by grafting using supercritical carbon dioxide (scCO2) impregnation and polymerization procedures. The membranes were synthesized by changing amount of monomer. The polypropylene grafted polystyrene sulfonic acid (PP-g-pssa) membranes were characterized with various methods. The morphology and structure of PP-g-pssa membranes were analyzed with scanning electron microscopy (SEM) and energy dispersive spectrometer (EDS). As amount of monomer was increased, ion conductivity, cell performance was increased and methanol permeability was decreased. However PP-g-pssa membranes with 1.5g monomer and over had similar values of methanol permeability, ion conductivity and cell performance.

Adsorption characteristics of the sericite and diatomite for ammonia gas (견운모와 규조토에 대한 암모니아 기체의 흡착특성)

  • The feasibility of the use of porous fossil diatoms for indoor air pollution control was investigated via the characterization of physical and chemical properties. The fossil diatoms were observed by SEM(Scanning Electron Microscope). Diatomite had well-distributed pores below 5 nm and relatively large surface area compare to sericite. However, no porosity in sericite was found. Results showed that diatomite had better performance than sericite in respect to porosity and large surface area. But diatomite which is thermally treated at $950^{\circ}C$ has no porosity and low surface area because of combustion of fossil diatoms or calcination of inorganic oxide at high temperature, and has poor adsorption capability of ammonia gas. In conclusion, porous diatomite has relatively high performance to adsorb noxious chemical compounds, such as ammonia gas and VOCs.

  • Several studies regarding depolymerization of polycarbonate waste to get the essential monomer, bisphenol A, have been reported in recent years. However, those methods have some environmental safety problems of using highly toxic organic solvents as well as product separation problem due to the use of alkali catalyst. In this study, we proposed the combination of glycolysis and methanolysis to depolymerize the polycarbonate waste. Glycolysis reaction reached at the reaction equilibrium after about 180 minat 473.15K and dissolution of the polycarbonate was found to be a rate controlling step of the reaction. The yield of BPA was improved with the aid of combination of glycolysis and methanolysis. The methanolysis was carried out at a temperature range of $303.15K{\sim}363.15K$ and MeOH/PC molar ratio $0.5{\sim}3$. The yield of BPA had a maximum at 1.0 MeOH/PC molar ratio and increased with the reaction temperature.

Effect of Electrochemical Redox Reaction on Biochemical Ammonium Oxidation and Chemical Nitrite Oxidation

  • A modified graphite felt electrode with neutral red (NR-electrode) was shown to catalyze the chemical oxidation of nitrite to nitrate under aerobic conditions. The electrochemically oxidized NR-electrode (EO-NR-electrode) and reduced NR-electrode (ER-NR-electrode) catalyzed the oxidation of $1,094{\pm}39$ mg/l and $382{\pm}45$ mg/l of nitrite, respectively, for 24 h. The electrically uncharged NR-electrode (EU-NR-electrode) catalyzed the oxidation of $345{\pm}47$ mg/l of nitrite for 24 h. The aerobic bacterial community immobilized in the EO-NR-electrode did not oxidize ammonium to nitrite; however, the aerobic bacterial community immobilized in the ER-NR-electrode bioelectrochemically oxidized $1,412{\pm}39$ mg/l of ammonium for 48 h. Meanwhile, the aerobic bacterial community immobilized on the EU-NR-electrode biochemically oxidized $449{\pm}22$ mg/l of ammonium for 48 h. In the continuous culture system, the aerobic bacterial community immobilized on the ER-NR-electrode bioelectrochemically oxidized a minimal $1,337{\pm}38$ mg/l to a maximal $1,480{\pm}38$ mg/l of ammonium to nitrate, and the community immobilized on the EU-NR-electrode biochemically oxidized a minimal $327{\pm}23$ mg/l to a maximal $412{\pm}26$ mg/l of ammonium to nitrate every two days. The bacterial communities cultivated in the ER-NR-electrode and EU-NR-electrode in the continuous culture system were analyzed by TGGE on the $20^{th}$ and $50^{th}$ days of incubation. Some ammonium-oxidizing bacteria were enriched on the ER-NR-electrode, but not on the EU-NR-electrode.
