L-Glycine Alleviates Furfural-Induced Growth Inhibition during Isobutanol Production in Escherichia coli

  • Lignocellulose is now a promising raw material for biofuel production. However, the lignin complex and crystalline cellulose require pretreatment steps for breakdown of the crystalline structure of cellulose for the generation of fermentable sugars. Moreover, several fermentation inhibitors are generated with sugar compounds, majorly furfural. The mitigation of these inhibitors is required for the further fermentation steps to proceed. Amino acids were investigated on furfural-induced growth inhibition in E. coli producing isobutanol. Glycine and serine were the most effective compounds against furfural. In minimal media, glycine conferred tolerance against furfural. From the $IC_{50}$ value for inhibitors in the production media, only glycine could alleviate growth arrest for furfural, where 6 mM glycine addition led to a slight increase in growth rate and isobutanol production from 2.6 to 2.8 g/l under furfural stress. Overexpression of glycine pathway genes did not lead to alleviation. However, addition of glycine to engineered strains blocked the growth arrest and increased the isobutanol production about 2.3-fold.

Draft Genome Assembly and Annotation for Cutaneotrichosporon dermatis NICC30027, an Oleaginous Yeast Capable of Simultaneous Glucose and Xylose Assimilation

  • The identification of oleaginous yeast species capable of simultaneously utilizing xylose and glucose as substrates to generate value-added biological products is an area of key economic interest. We have previously demonstrated that the Cutaneotrichosporon dermatis NICC30027 yeast strain is capable of simultaneously assimilating both xylose and glucose, resulting in considerable lipid accumulation. However, as no high-quality genome sequencing data or associated annotations for this strain are available at present, it remains challenging to study the metabolic mechanisms underlying this phenotype. Herein, we report a 39,305,439 bp draft genome assembly for C. dermatis NICC30027 comprised of 37 scaffolds, with 60.15% GC content. Within this genome, we identified 524 tRNAs, 142 sRNAs, 53 miRNAs, 28 snRNAs, and eight rRNA clusters. Moreover, repeat sequences totaling 1,032,129 bp in length were identified (2.63% of the genome), as were 14,238 unigenes that were 1,789.35 bp in length on average (64.82% of the genome). The NCBI non-redundant protein sequences (NR) database was employed to successfully annotate 11,795 of these unigenes, while 3,621 and 11,902 were annotated with the Swiss-Prot and TrEMBL databases, respectively. Unigenes were additionally subjected to pathway enrichment analyses using the Gene Ontology (GO), Kyoto Encyclopedia of Genes and Genomes (KEGG), Cluster of Orthologous Groups of proteins (COG), Clusters of orthologous groups for eukaryotic complete genomes (KOG), and Non-supervised Orthologous Groups (eggNOG) databases. Together, these results provide a foundation for future studies aimed at clarifying the mechanistic basis for the ability of C. dermatis NICC30027 to simultaneously utilize glucose and xylose to synthesize lipids.

Identification and Characteristics of Lactic Acid Bacteria Isolated from Shellfishes (패류로부터 젖산 세균의 분리 및 특성)

  • Lactic acid is an important product arising from the anaerobic fermentation by lactic acid bacteria (LAB). It is used in the pharmaceutical, cosmetic, chemical, and food industries as well as for biodegradable polymer and green solvent production. The poly lactic acid (PLA) is an important material for bio-plastic manufacturing process. For PLA production by new LAB, we screened LAB isolates from shellfish. A total of 28 LAB were isolated from various shellfishes. They were all Gram positive, oxidase and catalase negative. Based on API 50CHL kit, 7 strains among the 28 isolates were identified as Lactobacillus plantarum, 6 strains as Lactobacillus delbrueckii, 5 strains as Leuconostoc mesenteroides, 3 strains as Lactobacillus brevis, 2 strains as Lactococcus lactis, 1 strain as Lactobacillus salivarius, 1 strain as Lactobacillus paracasei, 1 strain as Lactobacillus pentosus, 1 strain as Lactobacillus fermentum and 1 strain as Pediococcus pentosaceu. Also, we examined the amount of total lactic acid produced by these new strains by HPLC analysis with Chiralpak MA column. One strain E-3 from Mytilus edulis was indentified as Lactobacillus plantarum and found to produce 20.0 g/L of D-form lactic acid from 20 g/L of dextrose. Further studies are underway to increase the D-lactic acid production by E-3.

  • Many new functional materials have been studied for efficient production and storage of energy. Many new materials such as sodium-based and sulfide-based materials have been proposed for energy storage, but research on Li batteries is still dominant. Due to the influence of environmental concerns regarding nuclear energy, interest in and research on fusion power are steadily increasing. For the commercialization of nuclear fusion, a design standard based on a considerable level of physical analysis and modeling is proposed. Nevertheless, limitations of existing materials in nuclear fusion environments limit practical applications. Tritium propagation material for continuous fusion reaction is one of the core materials, and therefore research on this material is being carried out intermittently. The key material for Li-based energy storage and tritium generation is the functional material Li-M-O. In this review, a structural description of functional Li-M-O system materials and technical trends for its applications are introduced.

ADHESIVENESS EVALUATION OF ACTIVATED PLATELET USING Arg-Gly-Asp-Phe(RGDF)-IMMOBILIZED SURFACE

  • The adhesion of activated and normal platelets to fibrinogen requires the receptor binding site of GPIIb/IIIa. These recognition sites exists in the A ${\alpha}$ chain(RGDS at 572-575 and RGDF at 95-98) and the carboxy-terminal of ${\gamma}$ chain (HHLGGAKQAGDV at 400-411) of fibrinogen. In this study, we developed RGDF-immobilized surface to detect the unctional state of platelet. RGDF-immobilized surface was prepared on the glass using photolithographic technology. Platelet adhesion to RGDF-immobilized surface was observed by staining platelets with mepacrine using a fluorescence microscope using mepacrine. Using the RGDF peptide of fragment E, we observed that the platelets pretreated with PGE1 interacted incompletely with RGDF-immobilized surface, whereas ADP activated platelets interacted with the surface extensively. These results show that the distinct selectivity of RGDF-immobilized micro-patterned surface can be used to detect the unctional state of platelets.

Surface-enhanced infrared detection of benzene in air using a porous metal-organic-frameworks film

  • Infrared (IR) spectroscopy is a powerful technique for observing organic molecules, as it combines sensitive vibrational excitations with a non-destructive probe. However, gaseous volatile compounds in the air are challenging to detect, as they are not easy to immobilize in a sensing device and give enough signal by themselves. In this study, we fabricated a thin nanocrystalline metal-organic framework (nMOF) film on a surface plasmon resonance (SPR) substrate to enhance the IR vibration signal of the gaseous volatile compounds captured within the nMOF pores. Specifically, we synthesized nanocrystalline HKUST-1 (nHKUST-1) particles of ca. 80 nm diameter and used a colloidal dispersion of these particles to fabricate nHKUST-1 films by a spin-coating process. After finding that benzene was readily adsorbed onto nHKUST-1, an nHKUST-1 film deposited on a plasmonic Au substrate was successfully applied to the IR detection of gaseous benzene in air using surface-enhanced IR spectroscopy.

Influence of Reaction Parameters on Biocrude Production from Lipid-extracted Microalgae using Hydrothermal Liquefaction (열수액화를 이용한 미세조류 추출잔사로부터 바이오원유 제조에 대한 반응인자의 영향)

  • Hydrothermal liquefaction of lipid-extracted Tetraselmis sp. feedstock containing 80 wt.% water was conducted in a batch reactor at different temperatures (300, 325, and $350^{\circ}C$) and reaction times (5, 10, 20, 40, and 60 min). The biocrude yield, elemental composition and higher heating value obtained at various reaction conditions were used to predict the optimum conditions for maximizing energy recovery of biocrude with good quality. A maximum energy recovery of 67.6% was obtained at $325^{\circ}C$ and 40 min with a high energy density of 31.8 MJ/kg and lower contents of nitrogen and oxygen. Results showed that reaction conditions of $325^{\circ}C$, 40 min was most suitable for maximizing energy recovery while at the same time achieving improved quality of biocrude.

  • Selenium-containing peptide (Selenium peptide) was produced by autolysis of Saccharomyces cerevisiae which was cultured in inorganic selenium-supplemented medium. Selenium peptide showed antioxidant activity and protective effects on UVB irradiated human fibroblast. Minimal toxicity of selenium peptide was observed whereas selenium nitrate exhibited cell toxicity as low as $10^{-9}\;M$. Selenium peptide also increased human fibroblast growth, procollagen type I and also decreased MMP-1 (matrix metalloprotease-1). This result showed the potential of selenium peptide as a nontoxic antioxidant.

Preparation and Characterization of Palladium Nanoparticles Supported on Nickel Hexacyanoferrate for Fuel Cell Application

  • Nickel hexacyanoferrate supported palladium nanoparticles (Pd-NiHCF NPs) were synthesized and studied for oxygen reduction reactions in direct methanol fuel cell. The NiHCF support was readily synthesized by a comixing of $Ni(OCOCH_3)_2$ and equimolar $K_3[Fe(CN)_6]$ solution into DI water under rigorous stirring. After the preparation of NiHCF support, Pd NPs were loaded on NiHCF via L-ascorbic acid reduction method at $80^{\circ}C$. Pd-NiHCF NPs were electrochemically active for oxygen reduction reaction in 0.1 M $HClO_4$ solution. X-ray absorption near edge structure analysis was conducted to measure the white line intensity of Pd-NiHCF to verify the OH adsorption. As a comparison, carbon supported Pd NPs exhibited same white line intensity. This study provides a general synthetic approach to easily load Pd NPs on porous coordination polymers such as NiHCF and can provide further light to load Pd based alloy NPs on NiHCF framework.

Isolation of New Microalga, Tetraselmis sp. KCTC12236BP, and Biodiesel Production using Its Biomass (신규 미세조류 Tetraselmis sp. KCTC12236BP의 분리 및 이를 이용한 바이오디젤 제조)

  • The microalgae have been studied for a source of biodiesel production. It is important to select the microalgae, which grows rapidly in local environmental conditions such as temperature range and ingredient of local seawater. The aim of this study was isolating microalga, which has rapid growth rate and high FAME contents in wide temperature ranges, for microalgal offshore cultivation in Korea, one of the country with four distinct seasons. Firstly, we had isolated a green microalga, Tetraselmis sp. KCTC12236BP, which has faster growth rate in low temperature (5 and $10^{\circ}C$) than Tetraselmis suecica and Dunaliella tertiolecta LB999 from Young Heung Island, Incheon, Korea. This microalga was cultivated in outdoor circulated tank photobioreactor (CT-PBR). As a result, this microalga could grow in wide temperature ranges (6 to $29^{\circ}C$), outdoors. After that, the biomass was recovered, and 13.2 g biodiesel could be acquired from 110 g dry biomass. These results indicate that the isolated microalga, Tetraselmis sp. KCTC12236BP is proper to biodiesel production using outdoor cultivation in Korea for all seasons.
