Objectives : Chekamuiyiin-tang has effects of promoting diuresis, promoting blood circulation, elimating phlegm and strengthening the body's resistance to disease. We experimented with Chekamuiyiin-tang in order to find effects which might possibly cure obesity. Methods : We fed a control group of rats a high fat diet and administered normal saline for 8 weeks. And we fed an experimental group of rats a high fat diet and administered an extract of Chekamuiyiin-tang for 8weeks. Results : The serum total cholesterol level, triglyceride level, free fatty acid level, total lipid level and phospholipid level in the Chekamuiyiin-tang group showed a significant decrease in comparison with the control group. The serum HDL-cholesterol level in the Chekamuiyiin-tang group showed no change in comparison with the control group. The serum LDL-cholesterol level in the Chekamuiyiin-tang group showed a decrease in comparison with the control group, but this result showed no efficacy. The size of the epididymal fat cell and the fat drops area in the hepatic lobule of rats in the Chekamuiyiin-tang group showed a significant decrease in comparison with the control group. Conclusios : According to the above results, Chekamuiyiin-tang has shown to be capable of curing and preventing obesity.
Flavobacterium columnare (F. columnare), the dermotropic Gram negative yellow pigmented bacteria was isolated from different sites of skin ulcerations in the Nile tilapia (Oreochromis niloticus) and Nile catfish (Clarias gariepinus) collected from an earthen pond located at an aquaculture station in Sharkiya Province, Lower Egypt during an acute episode of mass kills during the early summer of 2009. An acute infection with F. columnare was behind the emergent event of mass mortalities among both populations. Many of the Nile tilapias exhibited typical signs of hole - in- the head like lesions from which F. columnare together with the myxosporean spore, Myxobolus tilapiae (M. tilapiae) were retrieved. Most of the cohabitating infected Nile catfishes exhibited severe form of saddle back like ulcer. The identities of the retrieved isolates were confirmed using morphological, biochemical and molecular tools. The research lead us to conclude that the two diverse etiological agents (F. columnare and M. tilapiae) under the triggering effect of the abrupt change in the water quality measures (abrupt rise in the water temperature, ammonia, pH, sharp decrease in dissolved oxygen) have synergized together to induce the above mentioned pathology with the consequent reemergence of fish mass mortalities.
To investigate the mechanism of adriamycin (ADM) resistance in the ADM resistant subline PC-14/ADM, we examined the expressions of p-glycoprotein (P-gp), topoisomerase I (Topo I) and II (Topo II), glutathione-S-transferases (GSTs), tissue transglutaminase (t-TG), epidermal growth factor receptor (EGFR), and E-cadherin and the activity of superoxide dismutase (SOD) in PC-14 and PC-14/ADM cells. There was no change in the cellular levels of P-gp, Topo I, Topo II, and the two isoforms of GSTs. However, SOD activity in PC-14/ADM cells was 2.38 fold higher than that in PC-14 cells. A marked induction of the t-TG expression was also observed in PC-14/ADM cells. In addition to those changes, expressions of EGFR and E-cadherin were down regulated in PC-14/ADM cells. Therefore, molecular modifications such as an increase in SOD activity, induction of the t-TG expression, and down regulation of EGFR and E-cadherin expressions may play important roles in PC-14/ADM cells during the development of ADM resistance.
In relations to cold acclimation, experiment was carried out to understand the seasonal changes in reserve substances of the mulberry. The shoot barks and leaves of three mulberry varieties(Kaeryangppong, Shinilppong and Yongcheonppong) were sampled, after that their reserve substances were analyzed. The cold hardiness of mulberry was investigated by DTA(Differential Thermal Analysis) method. To increase cold hardiness, gibberellin(100 ppm), kinetin(100 ppm) and Jambi 8 were sprayed on the mulberry leaves. After spraying, falling of the leaves of Yongcheonppong occured earier than the other varities. After the first frost, all of treatments except gibberellin were entirely fallen. Growth regulator extended the leaves fallen. After spraying, water of the shoot barks was not showed difference in the content among the treatments, but amino acid, carbohydrate and soluble protein increased from September to October. Starch content of the shoot barks and leaves was maximum in October, but thereafter decreased during wintering stage. In Shinilppong, Jambi 8 spray increased cold hardiness by 1-2$^{\circ}C$ more than no spray. It was concluded that the cold hardiness of the mulberry in midwinter is closely related to the reserve substances with spraying Jambi 8 on the mulberry leaves.
Objectives : This study aimed to elucidate the effects of TBE on hyperlipidemia. Methods : We studied the effects of TBE on hyperlipidemia through gene expressions related with lipid metabolism and serum triglyceride as well as total and HDL-cholesterol levels, and perceived histological changes. Results : The present studies demonstrate that TBE can reduce the rise in plasma cholesterol and TG levels induced by a high-cholesterol diet and also reverse pre-established hypercholesterolemia and hypertriglycemia. In the TBE group total cholesterol levels decreased, TG levels decreased, but HDL-cholesterol levels also decreased. In the analysis of absolute and relative liver weight, TBE inhibited the weight gain induced by a high-cholesterol diet. In the histological observations, lipid droplet and apoptotic change in the TBE treated group were less compared with the control group. In the serum biochemical analysis, a difference of serum AST and ALT changes among groups was not shown, but TG and total cholesterol levels were less and HDL level decreased compared with the control group. In the gene expression related with TG and cholesterol metabolism, DGAT decreased slightly but ACAT decreased more as compared with control and Lipidil groups. Conclusion : From this study, we can infer that TBE possesses a hypolipidemic effect by inhibiting the intestinal absorption and storage of exogenous and endogenous cholesterol.
In order to investigate a pathogenesis of liver damage induced by skin burn, thermal injury was induced by scald burn on entirely dorsal surface in rats (total body surface area 30%) except for inhalated injury. At 5 and 24 h after scald bum, biochemical assay and morphological changes in skin tissue, serum and liver tissue were examined. The effects of bum injury on the levels of glutathione, lipid peroxide and on the activities of oxygen free radical generating and scavenging enzymes have been determined in association with observing of histologic and ultrastructural changes, measuring the protein concentration in plasma, and counting the number of intravascular polymorphonuclear leukocytes. The activity of xanthine oxidase, an enzyme of oxygen free radical generating system. was elevated (p<0.01) in serum, but not in skin and in liver tissue. Futhermore, thermal injury decreased not only the protein concentration in plasma but also the number of leukocytes, that indicates induction of edema formation with protein exudation and inflammation by neutrophil infiltration into the internal organs. These data suggest that acute dermal scald burn injury leads to liver damage, that is related to elevation of xanthine oxidase activity in serum. Xanthine oxidase may be a key role in the pathogenesis of liver damage induced by skin burn.
The object of this study is to investigate the toxicity of S-biol (d-allethron-d-transchrysanthemate) and the mode of action between other synthetic pyrethroid insecticides and S-biol in rats. Rats were treated daily with S-biol (25 mg/kg, 50 mg/kg, 100mg/kg) by oral administration for 5 weeks. The experimental results were summerized as follows: Biochemical parameters such as LDH and Glucose in serum were much more increased in control groups. No significant hematological change excepts for MCHC in rats treated with S-biol 100 mg/kg were observed in all groups compared to control groups. In animals treated with S-biol for 4∼5 weeks, the levels of cytochrome P-450 in the liver were significantly increased. In renal microsomal fractions, however, no significant changes of cytochrome P-450 contents were observed. The activitis of ATPase in groups treated with S-biol (50 mg/kg, 100 mg/kg) were decreased compared to those in other groups. TBA values and activities of glucose-6-phosphatase in the liver were increased a little in the groups treated with S-biol (100 mg/kg) for 5 weeks. The activities of cholinesterase in hepatic and serum were not significantly changed in all groups but slightly decreased in animals treated with high dose of S-biol (100 mg/kg). The activities of carboxylesterase in serum and in the liver were slightly increased but not significantly changed.
The toxicity and histopathologic changes of diazinon (O, O-diethy-O-(2-isopropyl-6-methyl-4-pyrimidinyl)phosphorothioate) was investigated in rat. Rat was treated with diazinon (100 mg/kg/day) by oral administration for 12 days. The experimental results were summarized as follows. Biochemical parameters such as ALT, AST, LDH and glucose in serum were significantly increased and hematological parameters such as Hb, Hct and PLT in blood were slightly increased in treated groups. Also the activities of serum cholinesterase were very significantly decreased in treated groups. In the histopathological changes, the normal lobular architectural pattern of the liver was well preserved in all treated groups. However, vacuolation or fatty change were represent in hepatocytes. Sections of liver from rats treated with diazinon for 3 and 6 days contained slight lipid infiltration in the form of small droplets randomly distributed that were graded minimal (+) or moderate (++) compared with the respective control condition. After 9 days, there were numerous small and large vacuoles in the terminal hepatic venule and perilobular areas of many serial sections of these rats indicative of fatty infiltration which were graded moderate (++) and severe (+++). After 12 days, fatty infiltration progressed periportal tract areas and graded severe (+++) and very severe (++++) in experimental groups. Note absence of cellular necrosis or inflammation.
Nutritional and health status was assessed in the 86 healthy elderly women who aged 65 through 96 and resided in nursing home in chonbuk area. Nutritional status was determined by dietary intake, anthropometric and hematological indices and hair elements. Correlation analysis among nutritional indices were performed to identify the factors which related specifically to nutritional status of the elderly. Nutrients intake below two-thirds of the RDA were Ca, riboflavin and niacin. Especially mean Ca intake was only 51.9% of RDA and most of nutrients intake were decreased as theri age increased. Height, body weight and mid upper arm circumference were decreased with age but W/H ratio did not differ among age groups. These data suggested that body fat accumulation did not change with age but lean body mass was decreased with age in the elderly. Waist circumference was positively correlated to dietary fat intake, and body weight, BMI, waist and hip circumferences and total body fat were positively correlated with serum LDC/HDL ratio. It could be concluded anthropometric indices were good indicator that reflect the lipid nutritional status in elderly women. Hematological indices, Hb, RBC, MCH, albumin, globulin, transferrin levels, belonged to normal ranges of aged women. However, the lowest Hb level showed in the oldest-old group(aged 85 and over years group). The avalilavilty of hair analysis to measure nutritional status was tested. There were not found any significant correlations between many kinds of nutritional indices and hair elements except hair Zn content which was positively correlated with BMI.
Objective : It is well known that changes in cerebral hemodynamics occur after traumatic brain injury (TBI). Osmo-regulation in the brain is important for maintaining a constant milieu in the central nervous system. Nevertheless, to our knowledge, early osmolarity changes after minor head injury have not been studied until now. Methods : In this study, serum osmolarity was measured in 99 patients with minor head trauma. As a control group, blood samples were drawn from 99 patients who had a minor trauma in an extremity. Serum osmolarity was estimated using a fully automatic biochemical autoanalyzer within the first 3 hours after the trauma. Results : The mean serum osmolarity levels were $286.08{\pm}10.1\;mOsm/L$ in the study group and $290.94{\pm}5.65\;mOsm/L$ in the control group (p<0.001). However, after age adjustment between the study and control groups, this statistical significance was found to be valid only for patients over 30 years of age. Conclusion : It was noted that serum osmolarity levels decrease in the first 3 hours following minor head trauma in patients over 30 years of age. Further studies into this area could provide guidance for the management/treatment of elderly patients.
