A Co-inhibitory Molecule, B7-H4, Synergistically Potentiates Oral Tolerance by Inducing CD4+CD25+FoxP3+ T Cells

  • Background: A co-inhibitory molecule, B7-H4, is believed to negatively regulate T cell immunity by suppressing T cell proliferation and inhibiting cytokine production. However, the mechanism behind B7-H4-mediated tolerance remains unclear. Methods: Balb/c $(H-2^d)$ mice were fed with dendritic cell line, DC2.4 $(H-2^d)$ every day for 10 days. Meantime, mice were hydrodynamically injected with recombinant plasmid expressing B7-H4 fusion protein (B7-H4.hFc) or hFc via tail vein. One day after last feeding, mice were immunized with allogeneic B6 spleen cells. 14 days following immunization, mice were challenged with B6 spleen cells to ear back and the ear swelling was determined the next day. Subsequently, a mixed lymphocyte reaction (MLR) was also performed and cytokines profiles from the reaction were examined by sandwich ELISA. Frequency of immunosuppressive cell population was assayed with flow cytometry and mRNA for FoxP3 was determined by RT-PCR. Results: Tolerant mice given plasmid expressing B7-H4.hFc showed a significant reduction in ear swelling compared to control mice. In addition, T cells from mice given B7-H4.hFc plasmid revealed a significant hyporesponsiveness of T cells against allogeneic spleen cells and showed a significant decrease in Th1 and Th2 cytokines such as IFN-${\gamma}$, IL-5, and TNF-${\alpha}$. Interestingly, flow cytometric analysis showed that the frequency of CD4+CD25+FoxP3+ Tregs in spleen was increased in tolerant mice given recombinant B7-H4.hFc plasmid compared to control group. Conclusion: Our results demonstrate that B7-H4 synergistically potentiates oral tolerance induced by allogeneic cells by increasing the frequency of FoxP3+ CD4+CD25+ Treg and reducing Th1 and Th2 cytokine production.

H19 Gene Is Epigenetically Stable in Mouse Multipotent Germline Stem Cells

  • Testis-derived germline stem (GS) cells can undergo reprogramming to acquire multipotency when cultured under appropriate culture conditions. These multipotent GS (mGS) cells have been known to differ from GS cells in their DNA methylation pattern. In this study, we examined the DNA methylation status of the H19 imprinting control region (ICR) in multipotent adult germline stem (maGS) cells to elucidate how epigenetic imprints are altered by culture conditions. DNA methylation was analyzed by bisulfite sequencing PCR of established maGS cells cultured in the presence of glial cell line-derived neurotrophic factor (GDNF) alone or both GDNF and leukemia inhibitory factor (LIF). The results showed that the H19 ICR in maGS cells of both groups was hypermethylated and had an androgenetic pattern similar to that of GS cells. In line with these data, the relative abundance of the Igf2 mRNA transcript was two-fold higher and that of H19 was three fold lower than in control embryonic stem cells. The androgenetic DNA methylation pattern of the H19 ICR was maintained even after 54 passages. Furthermore, differentiating maGS cells from retinoic acid-treated embryoid bodies maintained the androgenetic imprinting pattern of the H19 ICR. Taken together these data suggest that our maGS cells are epigenetically stable for the H19 gene during in vitro modifications. Further studies on the epigenetic regulation and chromatin structure of maGS cells are therefore necessary before their full potential can be utilized in regenerative medicine.

키틴 유도체와 하이드록시아페타이트 복합체가 개의 뼈에 미치는 영향 (The Effects of Chitin Derivative and Hydroxyapatite Compound in Canine Bone)

  • The aim of this study was to investigate bioactivities of 50% Chitin - hydroxyapatite (Chitin-H) compound and 50% Chitosan - hydroxyapatite (Chitosan-H) compound in canine bone. Ten healthy mongrel dogs (1-5 years old, 1.7 - 6.9 kg) were used in this study. These compounds had been transplanted into bilateral femur separately, and then the changes of femur were observed through the examinations of hemato-biochemical profiles, radiology, and histological profiles for 42 days. After 3 weeks, expanded radiolucent changes were observed in both areas transplanted the compounds. After 6 weeks, the area transplanted the Chitin-H compound did not observe any changes of bony tissue, while the area inserted the Chitosan-H compound was observed changes of increasing bone formation. In histological examination, infiltrations of inflammatory cells and bone absorptions were observed at both transplanted sites. However an increasing of active osteogenesis was observed at the transplanted site with Chitosan-H compound. In conclusion, Chitosan-H compound had an function of active osteogenesis as compared with Chitin-H compound. From this study, it is indicated that Chitosan-H compound would be used in dogs with severe bone defect.

  • With the advances in modern diagnostic expertise for cancer, certain approaches allowing scanning of the complete genome and the proteome are becoming very useful for researchers. These high throughput techniques have already proven power, over traditional detection methods, in differentiating disease sub-types and identifying specific genetic events during progression of cancer. This paper introduces major branches of omics-technology and their applications in the field of cancer. It also addresses current road blocks that need to be overcome and future possibilities of these methods in oncogenic detection.

  • The marine ecosystem represents a vast and dynamic array of bio-resources attributed with its huge diversity and considered as potential untapped reservoirs for the development of functional foods for future health markets. Basically, marine microorganisms, sponges, algae, invertebrates such as crustaceans and mollusks along with marine fish species can be considered as marine bio-resources, which can be utilized to obtain different health benefits for humans, directly or after processing. Most of the bio-molecular components, such as lipids and proteins from these marine bio-resources, which can be extracted in large scale using the modern and advanced biotechnological approaches, are suitable drug candidates for the pharmaceutical industry as well as functional food ingredients for the food industry. Moreover, the furtherance of high throughput molecular biological techniques has already been incorporated with identification, mining and extraction of molecular components from marine bio-resources. In this review, potential marine bio-resources with respect to their extractable bio-molecules were described in details, while explaining the present and prospective methods of identification and extraction, which are integrated with advanced techniques in modern biotechnology. In addition, this provides an overview of future trends in marine biotechnology.

Development of Grilled-type Shrimp Flavor by Maillard Reaction and Sensory Evaluation

  • A grilled-type shrimp flavor was developed through the Maillard reaction to reduce or mask fish odor or off-flavor in seafood. Model systems were created by using enzymatic hydrolysate of shrimp and adding precursors to increase flavor quality and stability. Amino acid precursors such as cysteine and methionine, sugar precursors such as glucose, xylose, ribose, and sucrose, and one particular compound of glucosamine were tried and their flavor qualities were tested by sensory evaluation. Also, the optimum reaction condition was investigated using the pH values of pH 5, 6, 7, and 8 with reaction times of 1 hr, 2 hr and 3 hr after the best precursors were determined. The best condition of the precursors for grilled-type shrimp flavor was the mixtures of methionine, threonine, xylose, and glucosamine. The optimum reaction condition was at pH 8.0 and 2 hr reaction time.

유기물(有機物)(BIO-COM) 시용(施用)이 답토양(畓土壤)의 이화학적(理化學的) 성질(性質)과 수도수량(水稻收量)에 미치는 영향(影響) (The Effects of Organic Matter (BIO-COM) Application on the Soil Physico-Chemical Properties and Rice Yields)

  • 1. 두 토양(土壤) 모두에서 무비구(無肥區)에 비하여 BIO-COM, 퇴비 및 N-P-K 비료시용은 모두 수량증수효과(收量增收效果)를 보였으나, 그 중 BIO-COM 효과가 가장 크며 Humic acid 처리는 오히려 수량을 감소시켰다. 2. 특히 BIO-COM은 삼요소(三要素) 무시용구(無施用區)에서는 4% 시용수준(施用水準)까지, 그리고 삼요소(三要素) 병행(竝行) 시용구(施用區)는 2% 수준까지 수량증가(收量增加)를 보였다. 3. 수도수량(水稻收量)은 수량구성요소중(收量構成要素中) 주당수수와 고도(高度)의 유의성(有意性) 있는 정(正)의 상관관계(相關關係)를 보였으나 천립중(千粒重)과는 부(負)의 상관(相關)을 보였다. 4. BIO-COM 시용은 수도재배후(水稻栽培後) 토양의 pH, EC, OM, T-N, Av-P, Ex-Ca, Ex-Mg, 그리고 $Av-SiO_2$ 함량을 증가시켰다. 5. BIO-COM 시용은 전생육(全生育) 시기별(時期別) 초장, 분얼수, 생물중 및 건물중을 증가시켰으며, 또한 이들 상호간에는 고도의 유의성 있는 정의 상관관계를 보였다. 6. 두 토양 공히 처리에 따라 수도체중의 엽록소함량(葉綠素含量) 및 질소, 인산, 칼슘, 마그네슘함량이 증가되었다. 7. 토양의 pH, 전기전도도, 유기물, 전질소, 치환성칼슘, 마그네숨 함량과 수량간에는 유의성 있는 정의상관을 보였다. 8. 최고 분얼기, 출수기의 수도체중의 질소, 인산함량과 수량간에는 고도의 유의성 있는 정의상관율 보였다.
