Effect of Extraction Time on the Rheological Properties of Sericin Solutions and Gels

  • Recently, silk sericin has attracted the attention of researchers owing to its useful properties as a biomaterial including 1) good wound healing and cell activities, 2) fast gelation character, and 3) high water retention property. In the present study, silk sericin was prepared using different extraction times in hot water and the effect of extraction time on the rheological properties of sericin solutions and gels was examined. It was found that the production yield of sericin increased with extraction time. The shear viscosity of sericin solutions and gels decreased with increasing extraction time due to a decrease in sericin molecular weight. When the sericin solution transformed to a gel, the viscosity increased and the shear thinning behavior was more evident. In addition, the shear stress measurements indicated that the slip between the sericin samples and the measuring plate of the rheometer was increased by the gelation of sericin. The compression strength of sericin gel could be increased remarkably (by more than 100 fold) by preparation using the freezing and thawing method.

  • Dental implants are required to have biomechanical functions and biostability in order to perform authoring, pronunciation, and aesthetic functions in the oral cavity. In terms of biostability, pure titanium for medical have good biostability and no rejection in the alveolar bone. with appropriate strength in terms of strength as well as biocompatibility. In recent years, various surgical methods and devices have been developed to improve the convenience and safety of the procedure. However, as the number of procedures increases, the screw loosening of the abutment screw connecting the artificial root and the abutment There are many reports of artificial root and abutment fracture. Fig. 1 is an example of a case where the upper part of the abutment screw is arbitrarily modified to remove the abutment by the abutment fracture due to the loosening of the abutment screw. The fundamental cause of abduction of the abutment screw is caused by the slight movement due to the lowering of the retention force of the abutment screw. It is necessary to minimize loosening of the abutment screw to avoid problems such as fracture during the period of using the implant. The purpose of this study is to investigate the structure of the abutment screw to prevent the loosening of the abutment screw by forming 0.5mm slot.

Changes in Contents of Ginsenoside Due to Boiling Process of Panax ginseng C.A. Mayer

  • The purpose of the study was to determine a method to use fresh white Korean ginseng in the form of higher intake of medicinal components. Decoction was made at $70^{\circ}C$ and $90^{\circ}C$ in different intervals of time. HPLC (DAD) system was employed to monitor the ginsenosides content in the decoctions and the components were identified by comparing the retention time with that of reference compounds. However, decoction made at $70^{\circ}C$ in 72 hrs possessed higher amount of total ginsenosides ($209.6{\mu}g/mL$) content where considerable amount of bioactive ginsenosides like Rg3, Rb2, Rb1 and Rg1 were accumulated. Overall, it can be concluded that the fresh white Korean ginseng decoction made in 72 hrs at $70^{\circ}C$ would be useful for the health and other medicinal approach of ginseng.

Structure and properties of silk sericin obtained from different silkworm varieties

  • Recently, many researchers have studied silk sericin because of its high water retention, good wound healing, good cyto-compatibility, and blood-glucose- and cholesterol-lowering effects. Although sericin film can be used in wound dressing and cosmetic packs, its poor mechanical properties have prevented its use in industrial fields. In the present study, sericin was obtained from different silkworm varieties, and the effect of silkworm variety on the structure and properties of sericin was examined. Except for a small difference in serine content, no significant difference in sericin was noted among the silkworm varieties. In addition, silkworm variety almost had no effect on solution viscosity, implying that it does not influence the molecular weight of sericin. Mechanical properties of sericin film were strongly affected by silkworm variety. Wonwon 126 showed the best mechanical properties, while N74 and Geumgwangju displayed the worst properties.

공기부상반응조에서 체류시간과 반송율에 의한 유기물질 및 질소제거 향상에 관한 연구 (Improvement of Organics and Nitrogen Removal by HRT and Recycling Rate in Air Lift Reactors)

  • This study was performed to evaluate the air lift reactors (ALR) by variations of HRT and recycling rate. Air lift reactor was composed of bioreactor and clarifier above it. To remove organic matters and nitrogen through the formation of microbic film and filtration, bio-filter reactors were filled with clay, glass, bead, waste plastic, respectively. Influent wastewater was fed to biofilter reactor, and effluent wastewater from bio-filter reactor was injected ALR again, instead of adding external carbon source. Effluent BOD concentration was satisfied with lower than 10 mg/L in recycling rate 100% regardless of the variation of HRT and the kinds of media materials. In HRT 4 hr, recycling rate 100%, BOD removal efficiency rate was from about 85 to 90%, COD removal efficiency rate was higher than 90%. Effluent TN concentration was satisfied with less than 20 mg/L, if HRT was maintained by over than 6 hr regardless of recycling rate and media materials. Over than HRT was 4 hr, microbes concentration in air lift reactor was maintained over than 2,500 mg/L constantly, not sensitive to environmental condition, and organic removal was effective as it was higher.

Biological Synthesis of Baicalein Derivatives Using Escherichia coli

  • Two baicalein derivatives, baicalin and oroxylin A, were synthesized in this study. These derivatives exhibit diverse biological activities, such as anxiolytic and anticancer activities as well as memory enhancement. In order to synthesize baicalin from aglycon baicalein using Escherichia coli, we utilized a glycosyltransferase that regioselectively transfers glucuronic acid from UDP-glucuronic acid to the 7-hydroxy group of baicalein. To increase baicalin productivity, an araA deletion E. coli mutant, which accumulates UDP-glucuronic acid, was used, and ugd, which converts UDP-glucose to UDP-glucuronic acid, was overexpressed. Using these strategies, approximately $720.3{\mu}M$ baicalin was synthesized from $1,000{\mu}M$ baicalein. Oroxylin A was then synthesized from baicalein. Two O-methyltransferases (OMTs), ROMT-15 and POMT-9, were tested to examine the production of oroxylin A from baicalein. E. coli harboring ROMT-15 and E. coli harboring POMT-9 produced reaction products that had different retention times, indicating that they are methylated at different positions; the structure of the reaction product from POMT-9 was consistent with oroxylin A, whereas that from ROMT-15 was 7-O-methyl baicalein. Using E. coli harboring POMT-9, approximately 50.3 mg/l of oroxylin A ($177{\mu}M$) was synthesized from 54 mg/l baicalein ($200{\mu}M$).

Molecular Cloning and Characterization of Bacillus cereus O-Methyltransferase

  • Biotransformation is a good tool to synthesize regioselective compounds. It could be performed with diverse sources of genes, and microorganisms provide a myriad of gene sources for biotransformation. We were interested in modification of flavonoids, and therefore, we cloned a putative O-methyltransferase from Bacillus cereus, BcOMT-2. It has a 668-bp open reading frame that encodes a 24.6-kDa protein. In order to investigate the modification reaction mediated by BcOMT-2, it was expressed in E. coli as a His-tag fusion protein and purified to homogeneity. Several substrates such as naringenin, luteolin, kaempferol, and quercetin were tested and reaction products were analyzed by thin layer chromatography (TLC) and high performance liquid chromatography (HPLC). BcOMT-2 could transfer a methyl group to substrates that have a 3' functional hydroxyl group, such as luteolin and quercetin. Comparison of the HPLC retention time and UV spectrum of the quercetin reaction product with corresponding authentic 3'-methylated and 4'-methylated compounds showed that the methylation position was at either the 3'-hydroxyl or 4'-hydroxyl group. Thus, BcOMT-2 transfers a methyl group either to the 3'-hydroxyl or 4'-hydroxyl group of flavonoids when both hydroxyl groups are available. Among several flavonoids that contain a 3'- and 4'-hydroxyl group, fisetin was the best substrate for the BcOMT-2.

치은섬유아세포와 치주인대섬유아세포의 interleukin-6 분비 및 세포성장에 미치는 단핵구세포주와 섬유아세포의 세포간 접촉작용 (Effects of Direct Cell Contact Between Monocytes and Fibroblasts on the Interleukin-6 Production and Cell Proliferation of Human Gingival and Peri - odontal Ligament Fibroblasts)

  • In order to reveal immunopathogenesis of periodontal tissue destruction, it is important to clarify the molecular mechanism of trafficking and retention of activated leukocytes, including monocytes/macrophages. Gingival fibroblasts may be involved in the regulation of inflammatory cell accumulation in the extravascular periodontal connective tissues via cytokine production and surface expression of adhesion molecules. In this study, it was investigated the molecular basis for the adhesive interactions between monocytes and fibroblasts such as peri-odontal ligament fibroblast(PDLF), human gingival fibroblast(HGF), and human dermal fibroblast(HDF). First, it was examined the evidence whether monocyte-fibroblast cell contact may cause signal transduction in fibroblasts. Being directly in contact with fixed human monocyte cell line THP-1, or U937, upregulation of IL-6 production, $TNF-{\alpha}$ mRNA expression and increased cell proliferation could be seen for fibroblasts. IL-6 production induced by monocyte- fibroblast coculture were further increased when fibroblasts had been pretreated with $IFN-{\gamma}$ or $IL-1{\beta}$ , and monocytes with LPS. Next, it was examined the expression of ICAM-1 which has been known to be involved in accumulation and activation of leukocytes in inflammatory diseases such as periodontitis. ICAM-1 was upregulated up to 10-fold on PDLF, HGF, and HDF by exposure to $IFN-{\gamma}$ or $IL-1{\beta}$. Furthermore, anti-ICAM-1 monoclonal antibody clearly blocked cocultureinduced IL-6 production by fibroblasts, suggesting that $ICAM-1/{\beta}_2$integrin pathway is involved in periodontal fibroblastmonocyte interaction. Overall, these findings provide evidence that periodontal fibroblasts could be involved in the accumulation and retention of monocytes/macrophages in periodontal inflammatory lesion at least in part by ICAM-1 expression. In addition, periodontal fibroblast-monocyte interaction could cause activation signals in fibroblasts intracellularly which result in cytokine production and cell proliferation. Thus, periodontal fibroblasts are speculated to play an important role in immunoregulation and tissue destruction in chronic periodontal diseases by interaction with monocytes/macrophages.

국산 낙엽송으로 제조한 에코우드포트(Eco Wood pots)의 식물 생장 효과에 관한 연구 (A Study on the Effect of Plants Growth on Eco Wood Pots)

  • The purpose of this study was to develop wood pots for create Optimum environment of plant growth using unused wood. to prove this, we examined the effects of cycle of water supply and nutrient concentration in wood pots on plant germination rate and growth factors (leaf number, stem diameter and length). The results are as follows. 1) The growth rate was higher at once of 2 days watering period. This suggests that the growth of the plants was better than that of the less water because the larch pots itself has the water retention capacity inside. 2) Germination rate and growth rate were better than other treatment groups when the concentration of nutrient solution was 0.5%. 3) Nitrogen, available phosphoric acid, and potassium showed higher contents than the nutrient - treated soil at 0.5% concentration of nutrient solution. This indicates that the nutrient solution absorbed from the larch affected the soil and plant growth in the inside.

배합사료내 사료첨가제가 조피볼락(Sebastes schlegeli) 치어의 성장, 사료 이용성 및 체조성에 미치는 영향 (Effect of Dietary Additives on Growth, Feed Utilization, and Body Composition of Juvenile Rockfish Sebastes schlegeli)

  • The effects of dietary supplementing natural additives [yacon (YC), ginger (GG), and blueberry (BB)] on growth, feed utilization, and body composition of juvenile rockfish were compared to those of ethoxyquin (EQ), which is commonly used to prevent rancidity of fish feed. A total of 480 fish were distributed randomly into 12 50-L flow-through tanks (40 fish/tank). Four experimental diets were prepared: the control diet (Con) with 0.01% EQ, and the YC, GG and BB diets containing 1% YC, GG, and BB instead of wheat flour. Each diet was assigned randomly to triplicate tanks of fish, which were hand fed to satiation twice daily for 8 weeks. Weight gain of fish fed the YC diet was higher than that of fish fed all other diets. Feed efficiency (FE), protein efficiency ratio (PER), and protein retention (PR) in fish fed the YC diet were higher than those in fish fed the other diets. Whole-body chemical composition of the fish was affected by the additive sources. In conclusion, dietary supplementation with GG effectively improved weight gain and feed utilization (FE, PER, and PR) of juvenile rockfish.
