Volatile Aroma Compounds of Several Domestic Thymus quinquecostatus by Thermal Desorption Gas Chromatograph Mass Spectrometer (열탁착식 가스크로마토질량분석기에 의한 국내 산지별 백리향의 휘발성향기성분)

  • Objective of this research was to investigate the volatile aroma compounds and phenol contents for preservative effects. Aerial part of 5 Korean natured species of thyme located in Jeju alpine, jeju middle mountain, Kyeonggido, Ulleung island, and Gangwondo was analyzed by thermal desorption gas chromatograph mass spectrometer (TD-GC-MSD) method. Jeju middle mountain thyme was relatively high 62 species and has been investigated a high concentration of $7365.22{\mu}g/m^3$ contents according to the quantitative analysis. Total phenol contents containing thymol and carvacrol of Jeju middle mountain thyme were relatively high and showed 35.92%.

  • Effectiveness of transgene transfer into genome is crucially concerned in mass production of the bio-pharmaceuticals using genetically modified transgenic animals as a bioreactor. Recently, the mammary gland has been considered as a potential bioreactor for the mass production of the bio-pharmaceuticals, which appears to be capable of appropriate post-translational modifications of recombinant proteins. The mammary gland tissue specific vector system may be helpful in solving serious physiological disturbance problems which have been a major obstacle in successful production of transgenic animals. In this study, to minimize physiological disturbance caused by constitutive over-expression of the exogenous gene, we constructed new retrovirus vector system designed for mammary gland-specific expression of the hEPO gene. Using piggyBac vector system, we designed to express hEPO gene under the control of mammary gland tissue specific and lactogenic hormonal inducible goat ${\beta}$-casein or mouse Whey Acidic Protein (mWAP) promoter. Inducible expression of the hEPO gene was confirmed using RT-PCR and ELISA in the mouse mammary gland cells treated with lactogenic hormone. We expect the vector system may optimize production efficiency of transgenic animal and reduce the risk of global expression of transgene.

  • By employing a two-stage continuous culture system, some of important physiological parameters involved in cellulase bicsynthesis have been evalua-ted with an ultimate objective of detigning an op-timally controlled tellulase process. Volumetric and specific cellulase productivities obtained were 90 IU/liter/hr and 8IU/g biomass/hr respectively. The maximum specific enzyme productivity observed was 14.8 IU/g hiomass/hr. The optimal dilution rate in the second stage which corresponded to the maximum enzyme productivity was 0.026-0.028 hr$^{-1}$ , and the specific growth rate in the second stage ihat suported maximum specific enzyme productivity was equal to orslightly less than zero. The maintenance coefficients deter-mined for oxygen and for carbon source are M$_{o}$=0.85mmmole/g biomass/hr and M$_{c}$=0.14 mmole hexose/g bio mass/hr respectively. The yield constants determined are; Y(x/o) =32.3g biomass/mole oxygen, Y (x/c) =1.1g bio-mass/g carbon or 0.44g biomass/g hexose, Y(x/n) = 19.6g biomass/g nitrogen for the enzyme produc-tion stage and 12.5g biomass/g nitrogen for the cell growth stage.e.e.

환경농업지구 농촌개발과 관련한 21세기 농촌주거모델 "생태마을"과 유기농업의 국제적 동향

  • There are severl types of Eco-village developed in abvanced countries including urban, suburban and rural style. For the establishment of Eco-village in Korea, it was focussed to introduce the rural style which has been developed inyersively in European countries such as Denmark, England and Germany. As a key cencept for Eco-villge it was explained briefly and discussed on the renewable energy(wind power park, bio-mass and solar energy collector and heating system), eco-architure, methane gas device, living machine (wastewater treatment facility) and organic farming. It was also discussed how important the basic standard and guidelines for organic agriculture to run essential for the system. For this reason major principles of international applicable standard(draft) for organically grown foods by FAO/WHO Codex alimentarius was shortly desidered. as one of very suitable type for Environmental Agriculture Districts Program which Korean govenment is going to establish in the near future. In conculusion it was also suggested that positive participation of organic famers, enthusiastic eneagement of inhabitans and support strategy of govemment /NGO groups might play very important role for successful managment of Eco-village ecologically sustainable after establishment.

Computation of Laryngeal Flow and Sound through a Dynamic Model of the Vocal Folds (동적 성대 모델을 이용한 후두 내 유동 및 음향장에 대한 수치 연구)

  • The present study numerically investigates the glottal airflow characteristics as well as acoustic features of phonation fully coupled with dynamic behavior of vocal folds. The vocal folds are described by a low-dimensional body-covered model characterized by bio-mechanical parameters such as glottal width, vocal folds stiffness, and subglottal pressure. The flow in the vocal tract is modeled as an incompressible, axisymmetric form of the Navier-Stokes equations (INS), while the acoustic field is predicted by the linearized perturbed compressible equations (LPCE). The computed result shows that a two-mass model of vocal folds is sufficient to reproduce temporal variations in oral airflow and glottis motion produced by female speakers. It is also found that i) the glottal width has a significant effect on the amplitude of glottal flow, and thus on the amplitude of acoustic wave in the vocal tract, ii) the vocal fold tension is the main control parameter for the fundamental frequency of phonation, iii) the subglottal pressure plays an appreciable role on reproduction of the self-sustained oscillation of vocal folds, and iv) the strength of pulsating airflow and vortical structures are primarily affected by glottal width and subglottal pressure, and are closely related to pitch, loudness, and voice quality. Finally, more comprehensive explanation about the difference between one- and two-mass models is presented with discussion of effectiveness of vocal folds oscillation and voice quality.

Development of Economical Fertilizer-Based Media for Mass Culturing of Nannochloropsis oceanica

  • This study was conducted to develop economical agricultural fertilizer media for the mass culturing of Nannochloropsis oceanica. Specific growth rates of N. oceanica cultured with differing concentrations of commercial compounds, urea fertilizers, and trace elements (Zn, Cu, Co, Mo) were compared with the growth rate in f/2 medium. Among the various added trace elements, $CuSO_4{\cdot}5H_2O$ was most effective for high growth of N. oceanica. The main nitrogen source in the agricultural fertilizers was ammonium, which was unsuitable for the growth of N. oceanica. Thus, the fertilizer at a lower concentration infused with $NaNO_3$ as a nitrogen source was more effective than fertilizer at higher concentrations. In this study, the growth of N. oceanica cultured with an agricultural fertilizer medium composed of compound fertilizer (41.7 mg/L), urea fertilizer (34.4 mg/L), $NaNO_3$ (150 mg/L), and $CuSO_4{\cdot}5H_2O$ (0.0588 mg/L) was similar to that of N. oceanica cultured in f/2 medium.

  • RTP(Rapid Thermal Pressing) is to fabricate desired pattern on polymer substrate by pressing patterned mold against the substrate heated around glass transition temperature. For a successful RTP process, the whole process including heating, molding, cooling and demolding should be conducted 'rapidly' as possible. As the RTP process is effective in replicating patterns on flat large surface without causing shape distortion after cooling, it is being widely used for fabricating various micro/bio application components, especially with channel-type microstructures on surface. This investigation finally aims to develop a RTP process machine for mass-producing micro/bio application components. As a first step for that purpose, we intended to examine the technological difficulties for realizing mass production by RTP process. Therefore, in the current paper, 4 kinds of RTP machines were examined and then the RTP process was conducted experimentally for PMMA film by using one of the machines, HEX 03. The micro-patterned molds used for RTP experiment was fabricated from silicon wafer by semi-conduct process. The replicated micro patterns on PMMA films were examined using SEM and the causes of defect observed in the replicated patterns were discussed.

International Movement on Rural Ice-village for 21st Century for an Environmentally Benign Agriculture (친환경농업을 위한 21세기 농촌형 "생태마을"의 국제적 동향)

  • For the development of Eco-village in Korean rural area, it was focussed to introduce the rural style which has been developed intensively in European countries such as Denmark, England and Germany. As the main concept for Eco-village it was explained briefly in the paper on the renewable energy(wind power park, bio-mass and solar energy collector and heating system), eco-architecture, methane gas device, living machine(wastewater treatment facility) and organic farming. It was also discussed how important the basic standard and guidelines for organic agriculture to run Eco-village environmentally soundly as a whole system and why it is so much essential for the system. For this reason major principles of international applicable standard for organically grown foods by FAO/WHO Codex alimentarius was also shortly described. In the paper the Eco-village concept was considered as a suitable model for Environmental Agriculture Districts Project which Korean government is going to establish at rural area in the near future. In conclusion it was suggested that the positive participation of organic farmers, enthusiastic engagement of inhabitants and support strategy of government/NGO groups might play very important role for successful management of Eco-village ecologically benign and economically sustainable after establishment.

Observation of Co-Existing Copepods in the Marine Rotifer Mass Culture Tanks (해산 로티퍼 대량 배양 수조에서 관찰되는 코페포다의 혼재 양상)

  • Microorganisms are contained filtered sea water that is why we often observed co-existing copepod species in the large-scale rotifer culture tanks. In this study, we investigated about taxonomical study of co-existing copepods in Brachionus rotifer mass culture tanks of several seafarming centers at Jeju-do and Pusan, Korea by microscope. Also, we counted number of copeods and density in 10 mL of cultured sea water and distinguished growing patterns and sex. As results, we can identified 4 species copepods, 2 species of Tisbe, 1 species of Tigriopus and 1 species of Apocyclops.

Facile mass production of thermally reduced graphene oxide

  • Mass production of graphene-based materials, which have high specific surface area, is of importance for industrial applications. Herein, we report on a facile approach to produce thermally modified graphene oxide (TMG) in large quantities. We performed this experiment with a hot plate under environments that have relatively low temperature and no using inert gas. TMG materials showed a high specific surface area (430 $m^2g^{-1}$). Successful reduction was confirmed by elemental analysis, X-ray photoelectron spectroscopy, thermogravimetic analysis, and X-ray diffraction. The resulting materials might be useful for various applications such as in rechargeable batteries, as hydrogen storage materials, as nano-fillers in composites, in ultracapacitors, and in chemical/bio sensors.
