We investigated the vegetation structure in Cultural Heritage Site of Odaesan National Park using 52 quadrats for each type of land use to figure out some characteristics of plant biotope. As we classified vegetation communities, they are six groups of communities. distinguished species in two of them are Taraxacum officinal, Erigeron annuus and Poa pratensis which are common in urban areas. Distinguished species in one of them are Potentilla fragarioides var. major which is common in outskirt of forest. And Distinguished species in another 3 communities are Sasa borealis and Quercus mongolica which are common in forest. Using TWINSPAN and DCA, we are able to classify the six communities into 3 types biotope (temple-biotope, slope-biotope, forest-biotope) in Cultural Heritage Site. The dominant species of urban-biotope are Poa pratensis, Artemisia prinseps and that of slope-biotope is Tripterygium regelii. Also the dominant species of forest-biotope are Quercus mongolica, Abies holophylla and Ulmus davidiana var. japonica. We could see more species in slope-biotope than another biotope types. Moreover, in urban-biotope types, we could find many of naturalized plant species.
Our study is purposed to understand effect on spatio-temporal variability of macroinvertebrate community 11 sampling times at 4 sites between two streams (Shinchon stream and Gongji stream in Chuncheun City) from May 2011 to October 2013. In this study, the possible physical factors on spatio-temporal variability of macroinvertebrate community were discussed. After stream improvement project, the effects of anthropogenic disturbance in study sites appeared as increased water temperature more than $30^{\circ}C$ and the difference of water temperature between Shinchon stream and Gongji stream was by maximum $9^{\circ}C$ on 2012. The monsoon rainfall decreased number of species, individuals and biodiversity index of macroinvertebrate community, particularly, in Shincheon stream compare to Gongji stream. Dominant species of macroinvertebrate taxa was caddisfly in Shincheon stream and Diptera in Gongji stream but on August 2013, it was dominated by Diptera, Chironomidae spp. in all study sites. The spatio-temporal variability of macroinvertebrate community in the streams may be useful as bio-indicator influencing anthropogenic factors such as soil erosion (landslide or cultivation) or monsoon rainfall.

Biological health in aquatic environments has been assessed by several approaches using various bio-taxa. Especially, fish has been identified as one of the best ecological indicators for an integrative health assessments in aquatic environments. In this preliminary study, the necropsy-based approach using the Health Assessment Index (Adams et al., 1993; HAI, Blazer, 2000), was employed for the assessments of aquatic environments influenced by effluent water from wastewater disposal plants (WDP) near a urban stream (i.e., Cap-Stream). For the impact analysis from the effluents, we developed 10 metric necropsy-based model including $M_l\;skin,\;M_2\;fin,\;M_3\;thymus,\;M_4\;spleen,\;M_5\;hindgut,\;M_6\;kidney,\;M_7\;liver.\;M_8\;eyes,\;M_9\;gill,\;and\;M_{10}$ pseudobranch. We set up duplicate controls (natural upstream water plus 5 fishes in each control tank) and duplicate treatments (effluent water from the WDP plus 5 fishes in each treatment tank) in the laboratory. Preliminary results showed that HAI values in the controls were zero, indicating an excellent condition by the criteria of HAI, while the HAI values in the treatments ranged between 40 and 150, indicating a poor condition. Also, various health conditions, based on the index of biological integrity (IBI), species richness, tolerance guilds, and trophic guilds supported the HAI approach. The HAI approach, not applied in Korea, not may be used as an surrogate fer ecological health assessment in lotic ecosystems.

In order to establish fundamental data for stream restoration and environmental flow, we investigated fish fauna, community and physical habitat conditions in the Dal Stream of Han River Basin. Field monitoring including fish sampling was conducted from September 2008 to October 2009. A total number of fish caught in the 5 sites was 2,080 representing 9 families 35 species, and 19 species (54.3%) including Acheilognathus koreensis and Iksookimia koreensis were Korean endemic species during the study period. The most frequently found species in number was pale chub (Zacco platypus, 25.7%, n = 632) followed by Coreoleuciscus splendidus (18.0%, n = 375) and Z. koreanus (13.0%, n = 272). The lower reach of Dal Stream was more abundance of species, high diversity, evenness and richness, and lower dominance index than those of the upper reach. According to the dendrogram established at 0.63 level of similarity rate, sampling stations were divided into 2 groups. Most of upper streams in Korea consist of riffles and runs that are repeatedly followed by another one. However, stream channelization and leveling have caused reduction of habitat diversity. Therefore, it is necessary to make an effort on stream rehabilitation with evaluation of physical habitat condition by indicator species in order to maintain biodiversity and perform ecological restoration.

Heavy metal contaminants on the aquatic environment are of interest because they can have severe effects on economy and public health. Recently, the studies for monitoring of heavy metals try to do on aquatic system to assess safety and health of ecosystem by heavy metals. Thus, biological responses were investigated on Korean fairy shrimp Branchinella kugenumaensis exposed to cadmium (Cd) or copper (Cu) for long-periods (30 days). The survival rate decreased significantly (p<0.05) on B. kugenumaensis exposed to Cd and Cu at all concentrations. Especially, the highest decrease was observed at the relatively high concentration of Cd and Cu (p<0.01) and the response by Cd exposure was at dose-dependent. The growth rates were also decreased significantly (p<0.05) on B. kugenumaensis exposed to Cd and Cu for at all concentrations. Then, the reproduction rate, numbering cyst, was decreased significantly (p<0.01) on B. kugenumaensis after Cd or Cu exposures. Long exposure of the relatively high concentration Cd and Cu can have severe effects on the reproduction, while exposures of Cd and Cu can not have effects on sex ratios of B. kugenumaensis. Additionally, asymmetric telson deformity was only observed after Cd exposure. Therefore, these results suggest that B. kugenumaensis is a sensitive bio-indicator of heavy metal exposure and these biological responses of B. kugenumaensis give important information for long-term monitoring on aquatic ecosystem.

To evaluate biochemical safety of gejang distributed in Korea, 33 samples were collected and analyzed for indicator microorganisms, pathogen, parasite, pH, volatile basic nitrogen(VBN), salinity, 3-MCPD, histamine, and synthetic food colors. Total plate counts of gejang were the range of 3 to 7 log cfu/g and coliform were the range of ND to 4 log cfu/g. Vibrio parahaemolyticus was detected from 4 samples, and Escherichia coli(1 log cfu/g) and copepoda were detected from 1 sample among those. pH and VBN value of samples were the range of 6.69 to 8.10 and 21.99 to 94.55 mg%, respectively. The ranges of salinity concentration for ganjang chamgejang, ganjang ggokgejang, and ganjang dolgejang were $11.6{\sim}32.6%,\;2.8{\sim}20.5%$, and $11.6{\sim}13.3%$, respectively. But, the range of salinity concentration of yangnum gejang was $2.6{\sim}9.4%$, which was lower than ganjang gejang. Besides, 3-MCPD, histamine and synthetic food colors were not detected. From the results, most of gejang was appropriate for Korea national standard for food regulation, but still biological hygiene control for raw materials, processing, and distribution should be improved.

To identify a solution for the restricted availability of healthy lungs and the high risk of immune rejections following organ transplantation, tissue engineering techniques for culturing lungs have been studied by many research groups. The most promising method for culturing lungs is the utilization of a bio-scaffold that was prepared using harvested organs from human donors or other animals by removing their original cells. In this study, a pulsatile perfusion pump was used to alleviate the cell removal effect with the high fluid-dynamic power of the perfusion stream during the decellularization process, while other conventional studies focused on chemical methods to identify efficient detergents. The purpose of this study was to analyze the developed device by using energy equivalent pressure (EEP), which is an indicator of pulsatility, to understand the characteristics of pulsatile energy transmitted according to the load size by using the artificial model and compare it with the measured EEP. The pulsatility of the device can be estimated with the concept of fluid-dynamic energy during a particular constant time period or fluid-dynamic power represented as EEP and EEP increment. Because the measured EEP of perfusion flow during decellularization can be changed by the amount of fluid leakage and the degree of clogging in the capillary vessels, EEP should be measured to determine whether the decellularization is progressing without problems. The decrement of EEP caused by the high perfusion resistance was observed from some experimental results that were obtained with artificial models. EEP can be used to monitor the decellularization process after analyzing the varying EEP according to the amount of load. It was confirmed that the EEP was maintained at a high level in the experiment using the harvested lungs from 12-13-week-old rats. In addition, it was confirmed that the cell removal time was faster than when continuous perfusion was performed. In this study, pulsatile power delivered to the lungs was measured to monitor the process of cell removal, and it serve as the evidence for efficient decellularization.
In this study, to produce Flammulina velutipes mushroom liquid spawn efficiently and effectively the effects of explosive aeration (supplying air with tiny bubbles) of the liquid culture medium on carbon dioxide concentration and residual sugar content in the medium on carbon dioxide concentration and residual sugar contentin the medium were measured. Carbon dioxide concentrations were measured at the outlet of the incubator. On the third day the explosive aeration greatly increased mycelial growth of the liquid spawn, and carbon dioxide concentration also greatly increased but decreased after 5 days. Free sugar contents in the liquid culture consistantly decreased up to 7 days and thereafter was not detected. The weight of the mycelia were maintained similar levels after 3 days. Total nitrogen content in the liquid medium constantly decreased during the 11days of explosive aeration. The content of free sugars in 7 days of culture was the lowest level, thus the inoculum incubated for 6~7 days was thought to be the most effective. Carbon dioxide concentration measurement at the outlet of the container during the liquid spawn incubation required low cost but was efficient to estimate the degree of mycelial growth to be used as a simple indicator.

Background: This study was conducted to evaluate the usefulness of the BACTEC MGIT (Mycobacterium Growth Indicator Tube) 960 system for mycobacteria culture and immunochromatographic assay to identify Mycobacterium tuberculosis (MTB) in positive MGIT culture. Methods: Mycobacteria-culture-positive cases were retrospectively analyzed from December 2010 to July 2011. The detection rates and the recovery times of the mycobacteria between the Ogawa media and the MGIT were compared. An immunochromatographic assay (ICA) (SD BIO-LINE) was also performed in the positive MGIT culture for identification, and the results were compared with those of the Ogawa media in the Korea National Tuberculosis Association. Results: Among the 261 patients (M:F, 168:93; mean age, $61.6{\pm}17.16$ yrs), 450 specimens (sputa, 365; bronchial washing, 61; and pleural effusion, 24) were found positive with mycobacteria. Mycobacteria were grown both on the MGIT and Ogawa media in 310 cases (68.9%); only on the MGIT in 115 cases (22.6%); and only on the Ogawa media in 25 cases (5.5%) (p<0.05). The recovery time was $28.2{\pm}8.9$ days in the Ogawa media and $11.1{\pm}5.8$ days in the MGIT (p<0.05). Among the 127 cases from the positive MGIT culture, all 92 cases that were confirmed as MTB cases bythe Korea National Tuberculosis Association were identified as MTB by ICA, with 100% sensitivity. Conclusion: MGIT increases the detection rate and shortens the recovery time of mycobacteria in clinical respiratory specimens, and the TB Ag MPT64 kit using ICA is useful in identifying MTB in a positive MGIT culture.
This study was designed to investigate the effects on anti-oxidative activities and increasing extraction yield of Aralia elata Cortex by gamma irradiation. Electron spin resonance (ESR) analysis as physical techniques for irradiation identification of Aralia elata showed that a pair of peak appeared on a space of 6.0 mT at the left and right of symmetric unspecific central ESR spectrums, confirming that the plant was gamma-irradiated. The optimum extracting conditions for preparing gamma irradiated samples from Aralia elata Cortex were to extract with 50% ethanol for 15 hrs after 10 kGy irradiation. DPPH scavenging activity and ABTS radical cation inhibitory activity of the water and 50% ethanol extracts from non irradiated and irradiated Aralia elata Cortex was very high as over 80% and 98%, respectively, at tested low concentration of $50{\mu}g/mL$. Antioxidant protection factor (PF) as anti-oxidation indicator of lipophilic compounds showed a very high level of activity as 2.18~2.78 PF. As for TBARs, water and ethanol extracts showed high level. Increase of TBARs inhibitory activity of water extracts was not shown by gamma-ray irradiation but ethanol extracts showed slight increasement of TBARs inhibitory activity with 10 kGy gamma-ray irradiation. These results shown confirmed increasement of extraction yield for phenolic compounds and anti-oxidative activity from Aralia elata. Thus, the treatment of gamma-irradiation can be used a way to amplify a solubility for biological active compounds and anti-oxidative activity in plants.
