바이오 디지털 콘텐츠를 이용한 독성의 분석 (Analysis of toxicity using bio-digital contents)

  • 화학물질은 생체에 들어오면 여러 가지 독성반응을 나타내는데, 독성반응에 따른 유전자 발현을 분석하기 위해 바이오 칩 등을 이용한 신기술이 확산되면서 바이오 디지털 콘텐츠가 다량으로 생성되고 있다. 이 콘텐츠는 그 자체로는 의미가 적고 컴퓨터를 이용한 분석과 보정과정을 거쳐 생물학적으로 의미 있는 값들을 선별하여야 한다. 이런 콘텐츠에는 유전자들의 발현 양상 측정을 목적으로 하는 유전체학(genomics), 유전자의 발현 양상을 측정하는 전사체학(transcriptomics), 단백질의 발현을 측정하는 단백체학(proteomics), 대사체의 발현을 측정하는 대사체학(metabolomics) 등이 있으며, 이를 통칭하여 오믹스(omics)라고 부른다. 오믹스 기술을 독성을 연구하는 분야에 접목한 것이 독성유전체학(toxicogenomics)이며, 이에 대한 콘텐츠를 분석함으로써 독성을 예측하고 독성기전을 규명할 수 있다. 독성분석에 있어서 초기 단계의 분석은 향후 만성독성의 예측에 있어서 중요한 부분을 차지하고 있다. 바이오 디지털 콘텐츠를 이용하여 독성을 예측함에 있어 기존의 방법보다 더 빠르고 정확하게 예측하기 위해서는 많은 정보에 대한 분석기술의 진보가 필요하다. 또, 바이오 디지털 콘텐츠를 이용한 독성예측에 있어서 전체세포보다는 생물학적 현상을 일으키는 특이세포에서 이런 정보를 얻는 것이 중요하다고 생각된다. 또, 향후 바이오 디지털 콘텐츠 분석은 전략적 실험설계에 의한 데이터가 분석되고 축적되어야 하고, 분석알고리즘을 통한 네트워크 분석이 이루어져야 하며, 통합적 데이터 구축을 통해 이루어져야 할 것으로 생각된다.

Ligand Based CoMFA, CoMSIA and HQSAR Analysis of CCR5 Antagonists

  • In this study, we have developed QSAR models for a series of 38 piperidine-4-carboxamide CCR5 antagonists using CoMFA, CoMSIA and HQSAR methods. Developed models showed good statistics in terms of $q^2$ and $r^2$ values. Best predictions obtained with standard CoMFA model ($r^2$ = 0.888, $q^2$ = 0.651) and combined CoMSIA model ($r^2$ = 0.892, $q^2$ = 0.665) with electrostatics and H-bond acceptor parameter. The validity of developed models was assessed by test set of 9 compounds, which showed good predictive correlation coefficient for CoMFA (0.804) and CoMSIA (0.844). Bootstrapped analysis showed statistically significant and robust CoMFA (0.968) and CoMSIA (0.936) models. Best HQSAR model was obtained with a $q^2$ of 0.662 and $r^2$ of 0.936 using atom, connection, hydrogen, donor and acceptor as parameters and fragment size (7-10) with optimum number of 6 components. Predictive power of developed HQSAR model was proved by test set and it was found to be 0.728.

인문치유 연구 -학생 사전 설문조사 분석- (A Study on Humanities Healing - Student Pre-Survey Analysis -)

  • 본 연구는 대학생을 대상으로 한 인문치유의 방법을 연구하고자 사전 설문조사를 실시하였다. 학생 설문조사를 바탕으로 인문치유의 연구 방향을 제시하고, 이에 적합한 인문치유의 방법을 찾고자 한다. 계량적인 분석을 위해 BPS-Modell, 몸(Bio), 정신(Psycho), 삶의 네트망(Sozio)을 중심으로 각 10문항을 구성하였다. 몸(Bio)에 대한 문항에서 50%의 학생들이 자신의 몸과 마음, 의지를 잘 파악하고, 그에 따른 행동을 하고 있다고 하였다, 그러나, 20%의 학생들은 자신의 몸과 마음, 의지에 따른 행동이 맞지 않는 것으로 나타났다. 정신(Psycho)에 대한 문항에서 50% 이상의 학생들이 자신의 내면과 소통이 잘 이루어진다고 하였다. 그러나, 20% 이하의 학생들은 잘 이루어지지 않는 것으로 나타났다. 삶의 네트망(Sozio)에 대한 문항에서 60% 이상의 학생들이 사회적 네트망을 잘 이루고 있다. 그러나, 10% 정도의 학생들은 삶에 대한 태도나 의미가 매우 낮은 것으로 나타났다. 인문학의 궁극적 목적은 '행복'이며, 인간 삶의 목적도 '행복'이다. 인문치유를 통해 안정적인 미래사회를 구현하고, 삶의 질을 높이는 긍정의 힘이 확대되기를 바란다.

A Position based Kinematic Method for the Analysis of Human Gait

  • Human joint motion can be kinematically described in three planes, typically the frontal, sagittal, and transverse, and related to experimentally measured data. The selection of reference systems is a prerequisite for accurate kinematic analysis and resulting development of the equations of motion. Moreover, the development of analysis techniques for the minimization of errors, due to skin movement or body deformation, during experiments involving human locomotion is a critically important step, without which accurate results in this type of experiment are an impossibility. The traditional kinematic analysis method is the Angular-based method (ABM), which utilizes the Euler angle or the Bryant angle. However, this analysis method tends to increase cumulative errors due to skin movement. Therefore, the objective of this study was to propose a new kinematic analysis method, Position-based method (PBM), which directly applies position displacement data to represent locomotion. The PBM presented here was designed to minimize cumulative errors via considerations of angle changes and translational motion between markers occurring due to skin movements. In order to verify the efficacy and accuracy of the developed PBM, the mean value of joint dislocation at the knee during one gait cycle and the pattern of three dimensional translation motion of the tibiofemoral joint at the knee, in both flexion and extension, were accessed via ABM and via new method, PBM, with a Local Reference system (LRS) and Segmental Reference system (SRS), and then the data were compared between the two techniques. Our results indicate that the proposed PBM resulted in improved accuracy in terms of motion analysis, as compared to ABM, with the LRS and SRS.

Pretreatment and enzymatic saccharification process of rapeseed straw for production of bioethanol

  • This study was conducted to evaluate the yield of bio-ethanol produced by separate hydrolysis and fermentation (SHF) with the pretreated rapeseed straw (RS) using crude enzyme of Cellulomonas flavigena and Saccharomyces cereviase. Crude enzyme of C. flavigena showed enzymatic activity of 14.02 U/mL for CMC 133.40 U/mL, for xylan 15.21 U/mL, for locust gum and 15.73 U/mL for rapeseed straw at pH 5.0 and $40^{\circ}C$, respectively. The hemicellulose contents of RS was estimated to compromise 36.62% of glucan, 43.20% of XMG (xylan + mannan + galactan), and 2.73% of arabinan by HPLC analysis. The recovering ratio of rapeseed straw were investigated to remain only glucan 75.2% after 1% $H_2SO_4$ pretreatment, glucan 45.44% and XMG 32.13% after NaOH, glucan 44.75% and XMG 5.47% after $NH_4OH$, and glucan 41.29% and XMG 41.04% after hot water. Glucan in the pretreatments of RS was saccharified to glucose of 45.42 - 64.81% by crude enzyme of C. flavigena while XMG was made into to xylose + mannose + galactose of 58.46 - 78.59%. Moreover, about 52.88 - 58.06 % of bio-ethanol were obtained from four kinds of saccharified solutions by SHF using S. cerevisiae. Furthermore, NaOH pretreatment was determined to show the highest mass balance, in which 21.22 g of bio-ethanol was produced from 100 g of RS. Conclusively, the utilization of NaOH pretreatment and crude enzyme of Cellulomonas flavigena was estimated to be the best efficient saccharification process for the production of bio-ethanol with rapeseed straw by SHF.

  • Rapid and accurate diagnosis of diseases is very important for appropriate treatment of patients. Recent advances in molecular-level interaction and detection technologies are upgrading the clinical diagnostics by providing new ways of diagnosis, with higher speed and accuracy. In particular, DNA microarrays can be efficiently used in clinical diagnostics which span from discovery of diseaserelevant genes to diagnosis using its biomarkers. Diagnostic DNA microarrays have been used for genotyping and determination of disease-relevant genes or agents causing diseases, mutation analysis, screening of single nucleotide polymorphisms (SNPs), detection of chromosome abnormalities, and global determination of posttranslational modification. The performance of DNA-microarray-based diagnosis is continuously improving by the integration of other tools. Thus, DNA microarrays will play a central role in clinical diagnostics and will become a gold standard method for disease diagnosis. In this paper, various applications of DNA microarrays in disease diagnosis are reviewed. Special effort was made to cover the information disclosed in the patents so that recent trends and missing applications can be revealed.

A Method for Absolute Determination of the Surface Areal Density of Functional Groups in Organic Thin Films

  • To develop a methodology for absolute determination of the surface areal density of functional groups on organic and bio thin films, medium energy ion scattering (MEIS) spectroscopy was utilized to provide references for calibration of X-ray photoelectron spectroscopy (XPS) or Fourier transformation-infrared (FT-IR) intensities. By using the MEIS, XPS, and FT-IR techniques, we were able to analyze the organic thin film of a Ru dye compound ($C_{58}H_{86}O_8N_8S_2Ru$), which consists of one Ru atom and various stoichiometric functional groups. From the MEIS analysis, the absolute surface areal density of Ru atoms (or Ru dye molecules) was determined. The surface areal densities of stoichiometric functional groups in the Ru dye compound were used as references for the calibration of XPS and FT-IR intensities for each functional group. The complementary use of MEIS, XPS, and FT-IR to determine the absolute surface areal density of functional groups on organic and bio thin films will be useful for more reliable development of applications based on organic thin films in areas such as flexible displays, solar cells, organic sensors, biomaterials, and biochips.

PCL/TiO2 Nanoparticle 3차원 지지체 제조 및 특성 평가 (Fabrication and Characterization of PCL/TiO2 Nanoparticle 3D Scaffold)

  • Polycaprolactone(PCL)은 생분해성 고분자로 인장강도, 신장률, 충격강도 등의 기계적 물성이 우수하다. $TiO_2$ (titanium dioxide) nanoparticle은 친수성으로 밀도가 높고 생체적합성이 우수하다. 본 연구에서는 PCL과 $TiO_2$(titanium dioxide) nanoparticle을 이용하여 salt-leaching방법으로 3차원 다공성 지지체를 제작하였다. 제작한 지지체를 FESEM, FTIR, TGA, 압축강도 측정 등을 통해 물성을 분석하였다. $TiO_2$ nanoparticle에 의해 물흡수도와 팽윤도는 감소하였으나 압축강도는 증가하였다. CCK-8 assay를 통해 세포의 증식률을 확인한 결과, $TiO_2$ nanoparticle에 의한 세포 독성은 없는 것으로 확인되었다. 이러한 연구결과는 PCL/$TiO_2$ nanoparticle 지지체의 생체재료로 사용가능성을 제시하였다.

Thromboprophylaxis in Abdominoplasty: Efficacy and Safety of a Complete Perioperative Protocol

  • Background Venous thromboembolism, a spectrum of diseases ranging from deep venous thrombosis to pulmonary embolism, is a major source of morbidity and mortality. The majority of cases described in plastic surgery involve abdominoplasty. Risk assessment and prophylaxis are paramount in such patients. General recommendations were recently developed, but the evidence in the literature was insufficient to prepare exhaustive guidelines regarding the medication, dosage, timing, or length of the prophylaxis. Methods A thromboprophylaxis protocol was developed for patients undergoing abdominoplasty. The protocol consisted of preoperative, intraoperative, and postoperative measures. Enoxaparin was administered as chemoprophylaxis in selected patients. The study involved 253 patients. The patients were analyzed for age, body mass index, enoxaparin dosage, risk factors, and complications. Results Deep venous thrombosis was documented in two cases (0.8%). No pulmonary embolism occurred. Three patients (1.2%) presented mild subcutaneous abdominal hematoma within the first postoperative week that spontaneously resorbed with neither aesthetic nor functional complications. Two patients (0.8%) presented severe hematoma requiring surgical re-intervention for drainage and hemostasis revision. Statistical analysis showed no significant correlation between enoxaparin dosage and hematoma (P=0.18) or deep venous thrombosis (P=0.61). Conclusions The described thromboprophylaxis protocol proved to be effective in the prevention of thrombotic events, with an acceptable risk of hemorrhagic complications. Furthermore, it provides new evidence regarding the currently debated variables of chemoprophylaxis, namely type, dosage, timing, and length.

Cloning and Expression of a Rice cDNA Encoding a Lls1 Homologue of Maize

  • A cDNA (Oslls1) encoding Lls1-homologue of maize was isolated from cDNA library of rice (Oryza sativa cv. Ilpum). The 2,138 bp of full length Oslls1 clone contains an open reading frame of 1,623 nucleotides encoding 575 amino acid residues. The deduced amino acid sequence of Oslls1 has a high level of homology with chlorophyll a oxygenases of Arabidopsis thaliana (67%) and Marchantia polymorpha (65%). Southern blot analysis of genomic DNA indicates the existence of a small gene family for Oslls1 in the rice genome. The expression of Oslls1 mRNA was induced in leaves and germinating seeds. Treatment of $H_2O$$_2$significantly down-regulated Oslls1 expression. The expression of Oslls1 mRNA was consititutively down-regulated in the blm, a rice mutant exhibiting spontaneous necrotic lesions. These results suggest that this Oslls1 gene may be involved incell death mechanisms in the blm mutant of rice.
