Organic Solvent-Tolerant Esterase from Sphingomonas glacialis Based on Amino Acid Composition Analysis: Cloning and Characterization of EstSP2

  • Organic solvent-tolerant (OST) enzymes are widely applied in various industries for their activity and stability in organic solvents, for their higher substrate solubility, and for their greater stero-selectivity. However, the criteria for identifying OST enzymes largely remain undefined. In this study, we compared the amino acid composition of 19 OST esterases with that of 19 non OST esterases. OST esterases have increased the ratio of Ala and Arg residues and decreased the ratio of Asn, Ile, Tyr, Lys, and Phe residues. Based on our amino acid composition analysis, we cloned a carboxylesterase (EstSP2) from a psychrophilic bacterium, Sphingomonas glacialis PAMC 26605, and characterized its recombinant protein. EstSP2 is a substrate specific to p-nitrophenyl acetate and hydrolyzed aspirin, with optimal activity at $40^{\circ}C$; at $4^{\circ}C$, the activity is approximately 50% of its maximum. As expected, EstSP2 showed tolerance in up to 40% concentration of polar organic solvents, including dimethyl sulfoxide, methanol, and ethanol. The results of this study suggest that selecting OST esterases based on their amino acid composition could be a novel approach to identifying OST esterases produced from bacterial genomes.

Effects of Defect Factors of Combine Header on Cutting Speed of Combine Header, Feeding Depth of Straw, and Cylinder Speed of Thresher (콤바인 예취부의 고장요인이 예취날의 평균 속도, 반송 두께, 탈곡통의 회전 속도에 미치는 영향)

  • The purpose of this study is to analysis effects of defect factors of combine header for cutting speed of combine header, feeding depth of straw, and cylinder speed of thresher. Measurement system for defect factors was consists of sensors to monitor the combine operation and I/O interface to convert the signals. Cutting speed of combine header, feeding depth of straw, cylinder speed of thresher were measured and analyzed. The data were collected from three paddy field during rice harvesting. The tests were conducted at different grounding speeds, lug troubles, and cutter condition. The one way ANOVA and the multiple comparison tests were performed. The results showed that the measured data were useful to monitor the defect factors of combine during harvesting. The faults conditions of grounding speeds, lug troubles, and cutter conditions affected cutting speeds, feeding depths and cylinder speeds of the combine. The data seem to be useful to analysis the faults conditions of combine header.

Consumers' Choice for Fresh Food at Online Shopping in the Time of Covid19

  • Purpose: This study aims at investigating consumers' choice in online food purchasing behavior and the impact on repurchase for fresh food delivery which has recently shown rapid growth in Korea. The study focuses on the user experience factors af fecting satisfaction and intention to continuously use the online food market. Research design, data and methodology: The survey was conducted by 309 people who had purchased fresh food online, and the analysis was conducted using SPSS and AMOS. Structural Equation Modeling was used for the analysis for the verification of hypotheses. The factors that consumers value when ordering fresh food delivery services were defined as system quality, service quality, commodity quality, brand characteristics, and economics from the preceding study and the relationship between satisfaction and willingness to repurchase was verified. Results: When consumers purchase fresh food online, system quality, product quality, brand characteristics, and economics have had a significant impact on satisfaction. Meanwhile, of the five optional attributes of consumers, only economic efficiency has been verified to have a statistically significant impact on repurchase intentions. Conclusions: The results of the study suggested factors that consumers consider important when ordering fresh food online, providing basic data for companies to develop related strategies.

Optimization of Input Parameters by Using DOE for Dynamic Analysis of Bio-inspired Robotic Fish 'Ichthus' (생체모방형 물고기 로봇 '익투스'의 동적 해석을 위한 DOE를 이용한 입력파라미터 최적화)

  • Recently, there is a rising interest on studying bio-inspired robotic fish because of real fish's great maneuverability and high energy efficiency. However, the researches about the robotic fish have not been done so much and there are still lots of problems to use them in the real environment such as in the river. This paper describes a bio-inspired robotic fish 'Ichthus' which is developed in KITECH and has 3 DOF propulsive mechanism. We develop the dynamic motion equation of 'Ichthus' in the underwater environment and analyze response characteristics of 'Ichthus' according to the input parameters of tail fin's amplitude and oscillation frequency. Then we propose control parameters at the various velocities. These parameters are useful to increase energy efficiency and it can be used when the fish robot moves in the real environment, for example, we can propose proper amplitude and oscillation frequency when the fish robot passes through the narrow space between obstacles.

Feeding habits of juvenile Chaenogobius annularis in an eelgrass bed in the coastal water of Geoje, Korea (경남 거제 연안 잘피밭에 출현하는 점망둑 (Chaenogobius annularis) 치어의 식성)

  • The feeding habits of juvenile Chaenogobius annularis were examined based on gut content analysis of 281 individuals, collected from May to June 2011 in the coastal water of Geoje, Korea. Copepods were the main source of food for C. annularis, which constituted 61.6% in IRI. Barnacle larvae was the second largest dietary component. Graphical analysis of the diet composition showed that C. annularis was a specialized predator characterized by strong individual feeding specialization. Both small and large size class of C. annularis mainly consumed harpacticoida and cirripedia cypris.

  • This paper presents development of a management system for the health and diseases of the elderly. This study aimed to the promotion of the u-healthcare industry and to the increase in its competitive power by developing and expanding a system for managing the health and diseases of the elderly, in cooperation with u-healthcare companies. The study is underway through the following four substudies. In the first substudy titled development of the bio-signal collection and analysis technology using smart media, a technology that supports the collection of bio-signals in the elderly using portable terminals. In the second substudy titled development of the patient-specific healthcare platform expansion and enhancement technology, a technology is being developed for making medical decisions and taking measures based on the results of the processing of the collected bio-signals. In the third substudy titled development of the N-screen based healthcare contents open service technology, a technology is being developed to provide information on health, diseases, and medicine to platforms. In the fourth substudy titled development of the oriental medicine diagnosis and analysis technology for senile diseases. This study is expected to help ensure an excellent workforce and new technologies in the healthcare sector using smart phones, and to help reduce medical expenses by improving the health of citizens.

A Study on the Reliability of an Ultrasonic Measurement Device(SDM) (연조직두께 측정기구(SDM)의 재현성에 관한 연구)

  • The aim of this study was to analyze the reliability of an ultrasonic device(SDM) measuring soft tissue thickness in relation to tooth position, and to find factors which can influence the reliability. The results showed that 1. measurement error was the largest in the maxillary second molar position and the smallest in the mandibular central incisor position. 2. in a box whisker plot, the difference between two measurements was most widely distributed in molar positions. 3. in Pearson correlation analysis, the relationship between two measurements was the highest in the maxillary lateral incisor position and, the lowest in the maxillary second molar, mandibular first and second premolar position. 4. a stepwise multiple regression analysis could explain the difference of two measurements with various independent variables in 29.7% (P<0.0001). Gingival thickness was the only variable influencing the measurement difference in a statistically significant level(P<0.0001). It can be concluded that its high reliability, ease to use and patient comfort justified the application of the SDM in measurement of soft tissue thickness.

Quantitative Analysis of Leuconostoc mesenteroides and Lactobacillus plantarum Populations by a Competitive Polymerase Chain Reaction

  • A multiplex competitive polymerase chain reaction (PCR) method was developed for the rapid identification and quantification of Leuconostoc mesnteroides and Lactobacillus plantarum populations which are the key microorganisms in kimchi fermentation. The strain-specific primers were designed to selectively amplify the target genes encoding 165 rRNA of L. plantarum and dextransucrase of L. mesenteroides. There was a linear relationship between the band intensity of PCR products and the number of colony forming units of each model organism. The PCR quantification method was compared with a traditional plate-counting method f3r the enumeration of the two lactic acid bacteria in a mixed suspension culture and also applied to a real food system, namely, watery kimchi. The population dynamics of the two model organisms in the mixed culture were reliably predictable by the competitive PCR analysis.

  • Isolation of oils from leaves of Juniperus phoenicea and Juniperus oxycedrus was obtained by steam distillation extraction method. The compositions of essential oils (EOs) were studied by means of GC-MS and GC-FID, using the internal standard method and relative response factors. Around ninety eight compounds were determined in total, representing 98.25 g/100 g of EO of J. phoenicea and 98.48 g/100 g of EO of J. oxycedrus, respectively. The volatile leaf oils were dominated by the terpenic hydrocarbon fractions (79.87 g/100 g) and (61.27 g/100 g) characterized by high contents of α-pinene (64.6 g/100 g) and (54.0 g/100 g) in J. phoenicea and J. oxycedrus, respectively, as the main component. Also, the enantiomeric distribution of α-pinene, sabinene, camphene, δ-3-carene, β-pinene, limonene, linalool, terpinen-4-ol, bornyl acetate, and borneol in both oils is presented for the first time.

Characterization of γ-Aminobutyric acid(GABA) produced by a lactic acid bacterium from button mushroom bed

  • ${\gamma}$-Aminobutyric acid(GABA) is a four carbon non-protein amino acid that has several well-known physiological functions, such as a postsynaptic inhibitory neurotransmitter in the brain and induction of hypotensive and tranquilizer effects. A lactic acid bacterium was isolated from button mushroom bed, which is showing high GABA productivity by TLC or HPLC analysis. The strain was identified as Lactobacillus hilgardii by analysis of 16S rDNA gene sequence. When the maximum production of GABA by L. hilgardii was investigated with various concentration of monosodium glutamate, the yield of GABA reached to be 53.65 mM at 1% mono sodium glutamate (MSG) in flask cultivation. A Glutamate decarboxylase (GAD) enzyme, which was known to convert MSG to GABA, was purified from a cell-free extract of L. hilgardii and the molecular weights of purified GAD was estimated to 60,000 by SDS-PAGE. The optimum pH and temperature of GAD were at pH4.6 and at $37^{\circ}C$, respectively. The GAD activity was increased by the addition of sulfate ions such as ammonium sulfate, sodium sulfate and magnesium sulfate, indicating that the increase of hydrophobic interaction causes the increase of GAD activity.
