Reversible DNA Information Hiding based on Circular Histogram Shifting (순환형 히스토그램 쉬프팅 기반 가역성 DNA 정보은닉 기법)

  • DNA computing technology makes the interests on DNA storage and DNA watermarking / steganography that use the DNA information as a newly medium. DNA watermarking that embeds the external watermark into DNA information without the biological mutation needs the reversibility for the perfect recovery of host DNA, the continuous embedding and detecting processing, and the mutation analysis by the watermark. In this paper, we propose a reversible DNA watermarking based on circular histogram shifting of DNA code values with the prevention of false start codon, the preservation of DNA sequence length, and the high watermark capacity, and the blind detection. Our method has the following features. The first is to encode nucleotide bases of 4-character variable to integer code values by code order. It makes the signal processing of DNA sequence easy. The second is to embed the multiple bits of watermark into -order coded value by using circular histogram shifting. The third is to check the possibility of false start codon in the inter or intra code values. Experimental results verified the our method has higher watermark capacity 0.11~0.50 bpn than conventional methods and also the false start codon has not happened in our method.

A Study on the Core Management Competencies of Ventures Formed by Entrepreneur's Incubator Organizations and Startup Experience: Focusing on the Biomedical Industry in Korea (창업가의 배태조직과 창업경험이 형성하는 창업기업의 핵심경영자원에 관한 연구: 한국의 바이오메디컬 산업을 중심으로)

  • This study explored the core management competencies of ventures formed by the entrepreneur's incubator organization and startup experience in the biomedical industry in Korea. An in-depth interview was conducted with 13 entrepreneurs of biomedical ventures. Based on the previous literature, the core management competencies of the ventures, which are influenced by the incubator organization and startup experience, are classified into 'technical competency', 'organization management competency', 'network competency' and 'market pioneering competency'. Analysis of the in-depth interview has revealed 18 factors influencing the formation of the core management competencies of ventures. Qualitative factors that were not addressed by the previous empirical studies were identified in this study. These include 'confidence in technology development', 'way of performing R&D', 'organizational culture' etc. This study is characterized by its scarcity as a qualitative study that deals with the entrepreneurs' prior experience. In addition, this study categorize the core management competencies which are formed by entrepreneurs' incubator organization and startup experience as four factors. This result is expected to be useful in future research.

Conical Diffuser Design and Hydraulic Performance Characteristics in Bioreactor Using Empirical and Numerical Methods (원뿔형 산기관 설계와 생물반응조에서 수력학적 운전특성에 관한 실험 및 해석)

  • In this study, we develop a highly efficient conical-air diffuser that generates fine bubble. By inserting a sufficient number of aerotropic microorganisms with dissolved oxygen from an air diffuser and minimizing the air-channel blockages within the air diffuser, we expect to improve the efficiency and durability of the decomposition process for organic waste. To upgrade the conventional air diffuser, we perform experiments and numerical analysis to develop a conical-type that generates fine bubble, and which is free from nozzle blockage. We complement the air-diffuser design by numerically analyzing the internal air-flow pattern within the diffuser. Then, by applying the diffuser to a mockup bioreactor, we experimentally and numerically study the bubble behavior observed in the diffuser and the 2-phase fluid flow in the bioreactor. The results obtained include statistics of the cord length and increased velocity, and we investigate the mechanisms of the fluid-flow characteristics including bubble clouds. Throughout the study, we systemize the design procedures for the design of efficient air diffusers, and we visualize the fluid-flow patterns caused by bubble generation within the mockup bioreactor. These results will provide a meaningful basis for further study as well as the detection of oxygen transfer and fluid-flow characteristics in real-scale bio-reactors using sets of air diffusers.

Evaluation of the Anti-thrombosis Activities of the Aerial Parts of Sageretia thea (상동나무 지상부의 항혈전 활성)

  • Recently, thrombotic diseases have become rapidly more prevalent due to Westernized lifestyles and high-fat diets. In this study, the anti-thrombosis activities of the aerial parts of Sageretia thea were evaluated using ethanol extracts of the leaf (ST-L), branch (ST-B), and fruit (ST-F), and their anti-coagulation, platelet aggregation inhibition, and hemolytic toxicity were assessed. In comparison to the ST-F extract, the ST-B exhibited 6.7 times more polyphenol content, and the ST-L had 2.7 times more total flavonoid content. The ST-L and ST-B extracts showed stronger inhibitions of thrombin, prothrombin, and blood coagulation factors than aspirin, berry extracts, or commercial oriental herbs. Furthermore, ST-L and ST-B showed superior platelet aggregation-inhibitory activities than aspirin. The ST-F extract demonstrated only minor anti-thrombosis effects, and none of the extracts showed hemolysis against red blood cells up to 1 mg/ml. Phenolic acid and flavonoid analysis of the ST-L and ST-B extracts showed abundant rutin, isoquercitrin, and astragalin as the major active compounds. Further research on the anti-thrombotic activity of isoquercitrin, a rare flavonoid from quercetin, is necessary. This is the first report of isoquercitrin in Sageretia thea, and our results suggest that ST-L and ST-B extracts could therefore developed as anti-thrombosis agents.

Stalk Rot Caused by Fusarium thapsinum on Sorghum at Organic Paddy-upland Rotation System in Korea (유기농 답전윤환지 Fusarium thapsinum에 의한 수수 줄기썩음병)

  • In late June 2016, stalk rot symptoms were observed on five vatieties of sorghum (Sorghum bicolar) at organic paddy-upland rotation system in Anseong city, Korea. The initial symptom on stalk surfaces was red color with a dark red spot lesion. A fungus was isolated from the initial lesion, and cultured on potato dextrose agar. Size of microconidia mostly extend to $5-19{\times}2-{\mu}m$ in culture, with 0-1 septa and macroconidia extend to $29-52{\times}3-4{\mu}m$ with 4-6 septa. Pathogenicity was investigated using conidial suspension spray to seedling of sorghum. After 3 days of inoculation, the dark red lesion was produced on stalks. On the basis of mycological characteristics, pathogenicity, and internal transcribed spacer (ITS) rDNA sequence analysis, this fungus was identified as Fusarium thapsinum. This is the first report of stalk rot on sorghum caused by F. thapsinum in Korea.

Incidence of Major Plant-parasitic Nematodes in Main Producing Areas of Strawberry in Korea (국내 딸기 시설재배 주산단지 토양 내의 주요 식물기생선충 감염 현황)

  • To investigate the incidence of plant-parasitic nematodes from plastic-house fields of strawberry, 170 soil samples were collected from the main producing area of strawberry in Korea from 2014 to 2015. Based on the investigation, root-lesion nematode (RLN; Pratylenchus spp.) and root-knot nematode (RKN; Meloidogyne spp.) are the most important pests in plastic-house fields of strawberry with 57% and 36% incidences, respectively. Among the collected fields, 96 and 63 fields were infected RNL and RKN, respectively with 44 fields (38.8%) being detected at both nematode. High degree of incidence of both nematodes were observed in the regions of Jinju, Gyeonnam Province, Korea. For identification of species level, phylogenetic and distance analysis were performed using RLN and RKN isolates generated from the regions of Jinju. The results indicated that the RLN and RKN isolates were Pratylenchus vulnus and Meloidogyne hapla, respectively. This study showed that P. vulnus and M. hapla could be most dominant species in plastic-house fields of strawberry. This data will provide basic knowledge for development of effective control strategies.

Proteome Analysis of Chicken Embryonic Gonads: Identification of Major Proteins from Cultured Gonadal Primordial Germ Cells

  • The domestic chicken (Gallus gallus) is an important model for research in developmental biology because its embryonic development occurs in ovo. To examine the mechanism of embryonic germ cell development, we constructed proteome map of gonadal primordial germ cells (gPGC) from chicken embryonic gonads. Embryonic gonads were collected from 500 embryos at 6 day of incubation, and the gPGC were cultured in vitro until colony formed. After 7-10 days in cultured gPGC colonies were separated from gonadal stroma cells (GSCs). Soluble extracts of cultured gPGCs were then fractionated by two-dimensional gel electrophoresis (pH 4-7). A number of protein spots, including those that displayed significant expression levels, were then identified by use of matrix-assisted laser desorption/ionization-time of flight (MALDI-TOF) mass spectrometry and LC-MS/MS. Of the 89 gPGC spots examined, 50 yielded mass spectra that matched avian proteins found in on-line databases. Proteome map of thistype will serve as an important reference for germ cell biology and transgenic research.

Characterization of Interfacial Adhesion of Cu-Cu Bonding Fabricated by Thermo-Compression Bonding Process (열가압 접합 공정으로 제조된 Cu-Cu 접합의 계면 접합 특성 평가)

  • Four-point bending tests were performed to investigate the interfacial adhesion of Cu-Cu bonding fabricated by thermo-compression process for three dimensional packaging. A pair of Cu-coated Si wafers was bonded under a pressure of 15 kN at $350^{\circ}C$ for 1 h, followed by post annealing at $350^{\circ}C$ for 1 h. The bonded wafers were diced into $30\;mm\;{\times}\;3\;mm$ pieces for the test. Each specimen had a $400-{\mu}m$-deep notch along the center. An optical inspection module was installed in the testing apparatus to observe crack initiation at the notch and crack propagation over the weak interface. The tests were performed under a fixed loading speed, and the corresponding load was measured. The measured interfacial adhesion energy of the Cu-to-Cu bonding was $9.75\;J/m^2$, and the delaminated interfaces were analyzed after the test. The surface analysis shows that the delamination occurred in the interface between $SiO_2$ and Ti.

Ecological Changes of the Larix kaempferi Plantations and the Restoration Effects Confirmed from the Results (일본잎갈나무조림지의 생태적 변화와 그 결과를 통해 확인된 복원 효과)

  • Developmental process of vegetation over years after afforestation was analyzed in the Japanese larch (Larix kaempferi) plantations with different stand ages. The height and diameter of Japanese larch increased rapidly until 24 years after afforestation and tended to be blunted thereafter. The density of Japanese larch was similar with each other in the 8 and 17 years old stands but was reduced greatly in the 24 years old plantation and changing little thereafter. The floristic composition of the Japanese larch plantation did not show any clear trend depending on stand ages. The differences of species composition among plantation themselves with different ages were bigger than those between plantations and the reference stands, Mongolian oak (Quercus mongolica) stands. Japanese larch plantations showed a trend of succession to native vegetation dominated by Mongolian oak based on the results of analysis of frequency distribution by diameter class of major tree species. Species richness and diversity of all plantations were higher than those of the reference vegetation. As were shown in the above mentioned results, it can be evaluated that Japanese larch plantations practiced in the level of the functional restoration achieved successful restoration based on the floristic composition similar to the reference vegetation, the successional trend toward native vegetation, and higher species diversity compared with the reference vegetation.

Isolation and Characterization of Bacillus licheniformis SC082 Degrading Fibrin and Chitin from Shrimp Jeot-Gal (새우젓으로부터 혈전과 chitin 분해능을 지닌 균주 Bacillus licheniformis SC082의 분리 및 특성)

  • Shrimp Jeot-Gal is a popular traditional Korean fermented seafood and has been used for seasoning. We isolated a bacterium showing strong extra-cellular fibrinolysis and chitinase activity from shrimp Jeot-Gal and the strain was designated SC082. SC082 was identified as Bacillus licheniformis by 16S rRNA sequence homology search. B. licheniformis SC082 exhibited optimum temperature, pH, and salt concentration at $37^{\circ}C$, pH 7.0, and 6%, respectively. Substrate specificity of the culture supernatant from B. licheniformis SC082 was detected in fibrin, skim milk, and chitin plate. The fibrinolytic activity was highly maintained up to $50^{\circ}C$ at a pH of 7.0 for 3 hr and was stable up to pH 9.0 at $37^{\circ}C$ for 3 hr. The chitinase activity was remarkably induced by addition of 1.0% colloidal chitin and the pH and temperature optima of the enzyme were 5.0 and $45^{\circ}C$, respectively. In sodium dodecyl sulfate-polyacrylamide gel electrophoresis and zymogram analysis, this strain produced three fibrinolytic isozymes and two chitinase isozymes. The approximate molecular weights of the putative fibrinolytic enzymes were 23.0, 62.0, and 72.0 kDa and those of the chitinases were 62.0 and 55.0 kDa, respectively. The antioxidant activity of SC082 was also measured by using 2,2-diphenyl-l-picryl-hydrazyl (DPPH) free radical. The DPPH radical scavenging was slightly increased in a dose-dependent manner.
