Glycolipids produced by Pseudomonas aeruginosa YPJ-80 were characterized by chromatographic and spectorscopic techniques as a mixture of two rhamnolipids. For recovery of glycolipids from the culture broth, various isolation methods including ultrafiltration, adsorption and solvent extraction were compared. Ultrafiltration showed the best results in terms of glycolipids recovery. Further purification for spectroscopic analysis was carried out by adsorption chromatography and preparative thin layer chromatography. From the spectroscopic analysis, such as IR spectroscopy. FAB-MS, 1H-NMR and 13C-NMR and hydrolysis analysis, the glycolipids were identified as L-${\alpha}$-rhamnopyranosyl-${\beta}$-hydroxydecanoly-${\beta}$-hydroxydecanoate and 2-O-${\alpha}$-L-rhamnopyranosyl-${\alpha}$-L-rhamnopyranosyl-${\beta}$-hydroxydecanoyl-${\beta}$-hydroxydecanoate. Monorhamnolipid and dirhamnolipid lowered the surface tension of water to 28.1 mN/M and 29.3 mN/m, respectively.
Anaerobiospirillum succiniciproducens requires expensive complex nitrogen sources such as yeast extract and polypeptone for its growth and succinic acid production. It was found that A. succiniciproducens was able to grow in a minimal medium containing glucose when supplemented with corn steep liquor (CSL) as the sole complex nitrogen source. The concentration of CSL had a significant effect on the glucose consumption by A. succiniciproducent. When 10-15 g/L of CSL was supplemented, cells were grown to an OD(sub)600 of 3.5 and produced 17.8 g/L succinic acid with 20 g/L glucose. These results are similar to those obtained by supplementing yeast extract and polypeptone, thereby suggesting that succinic acid can be produced more economically using glucose and CSL.
The solid-state anaerobic digestion (SS-AD) has promoted the development and application for biogas production from biomass which operate a high solid content feedstock, as higher than 15% of total solids. However, the digested byproduct of SS-AD can be used as a fertilizer or as solid fuel, but it has serious problems: high moisture content and poor dewaterability. The organic residue from SS-AD has to be improved to address these problems and to make it a useful alternative energy source. Hydrothermal carbonization was investigated for conversion of the organic residue from the SS-AD of livestock waste to solid fuels. The effects of hydrothermal carbonization were evaluated by varying the reaction temperatures within the range of $180-240^{\circ}C$. Hydrothermal carbonization increased the calorific value through the reduction of the hydrogen and oxygen contents of the solid fuel, in addition to its drying performance. Therefore, after the hydrothermal carbonization, the H/C and O/C atomic ratios decreased through the chemical conversion. Thermogravimatric analysis provided the changed combustion characteristics due to the improvement of the fuel properties. As a result, the hydrothermal carbonization process can be said to be an advantageous technology in terms of improving the properties of organic waste as a solid-recovered fuel product.

MCM-41 촉매를 이용하여 일본 낙엽송의 열분해 바이오 오일의 촉매 개질 반응을 수행하였다. MCM-41 촉매는 바이오 오일의 불안정성의 원인중 하나인 산소를 $H_2O$, CO, $CO_2$의 형태로 제거하여 개질 전의 오일보다 더 안정한 오일을 생성하였다. MCM-41 촉매는 무촉매 반응에 비해 경제적 가치가 높은 페놀류 화합물의 생성량을 증가시켰다. 특히 Si-MCM-41 촉매에 비해 산점의 양이 많은 Al-MCM-41의 경우 촉매 활성이 더 우수하였다. 또한 촉매와 일본 낙엽송을 직접 혼합한 것보다, 바이오 오일이 촉매층을 통과했을 때 더 나은 개질 결과를 얻을 수 있었다.
Metabolic engineering has become a new paradigm for the more efficient production of desired bioproducts. Metabolic engineering can be defined as directed modification of cellular metabolism and properties through the introduction, deletion, and modification of metabolic pathways by using recombinant DNA and other molecular biological tools. During the last decade, metabolic flux analysis(MFA) has become an essential tool fur metabolic engineering. By MFA, the intracellular metabolic fluxes can be quantified by the measurement of extracellular metabolite concentrations in combination with the stoichiometry of intracellular reactions and mass balances. The usefulness and functionality of MFA are demonstrated by applying to metabolic pathways in E. coli. First, a large-scale in silico E. coli model is constructed, and then the effects of carbon sources on intracellular flux distributions and succinic acid production were investigated on the basis of the uptake and secretion rates of the relevant metabolites. The results indicated that succinic acid yields increased in order of gluconate, glucose and sorbitol. Acetic acid and lactic acid were produced as major products rather than when gluconate and glucose were used carbon sources. The results indicated that among three carbon sources available, the most reduced substrate is sorbitol which yields efficient succinic acid production.
In this paper miniaturized disposable micro/nanofluidic components applicable to bio chip, chemical analyzer and biomedical monitoring system, such as blood analysis, micro dosing system and cell experiment, etc are reported. This system includes various microfluidic components including a micropump, micromixer, DNA purification chip and single-cell assay chip. For low voltage and low power operation, a surface tension-driven micropump is presented, as well as a micromixer, which was implemented using MEMS technology, for efficient liquid mixing is also introduced. As bio-reactors, DNA purification and single-cell assay devices, for the extraction of pure DNA from liquid mixture or blood and for cellular engineering or high-throughput screening, respectively, are presented.
본 연구에서는, 높은 함수율을 가진 음식물류 폐기물을 Bio-dying 공법을 이용하여 처리하였을 때 수분과 유기물의 변화정도를 분석하였으며 음식물류 폐기물 처리 시 온도와 염도가 Bio-dying 공법의 운전특성에 미치는 영향을 평가하였다. 또한, Bio-drying 후 분해산물의 SRF(Solid Refuse Fuel)로서의 가치 평가를 위한 펠렛 제조의 최적조건 도출 및 SRF의 발열량 분석을 통한 고형연료로서의 가치를 평가하였다. 그 결과, 1일 투입량에 따른 온도, $CO_2$ 농도, 유기물 제거율과 무게 감소율 값을 분석하였다. 유기물 제거율과 무게 감소율은 투입량 2.4 kg/day에서 각각 86%, 68%로 최대값을 나타내었다. 이를 통해 최적 음식물류 폐기물 투입량은 2.4 kg/day라 판단되었다. 펠렛 성형 결과, 원료 함수율이 10~25% 이내에서 펠렛 제조가 가능하고, 외형유지와 강도면에서 가장 좋은 품질결과를 보여준 함수율 25%가 가장 적합하다고 판단되었다. Bio-drying 후 분해산물의 SRF 고위발열량은 3,500 kcal/kg 이상의 값을 나타내었다.
This study aims to propose a performance-based design method of a novel passive base isolation system, BIO isolation system, which is inspired by an energy dissipation mechanism called 'sacrificial bonds and hidden length'. Fragility functions utilized in this study are derived, indicating the probability that a component, element, or system will be damaged as a function of a single predictive demand parameter. Based on PEER framework methodology for Performance-Based Earthquake Engineering (PBEE), a systematic design procedure using performance and fragility objectives is presented. Base displacement, superstructure absolute acceleration and story drift ratio are selected as engineering demand parameters. The new design method is then performed on a general two degree-of-freedom (2DOF) structure model and the optimal design under different seismic intensities is obtained through numerical analysis. Seismic performances of the biologically inspired (BIO) isolation system are compared with that of the linear isolation system. To further demonstrate the feasibility and effectiveness of this method, the BIO isolation system of a 4-storey reinforced concrete building is designed and investigated. The newly designed BIO isolators effectively decrease the superstructure responses and base displacement under selected earthquake excitations, showing good seismic performance.
This study is a basic research on the functional performance review for the use of environmentally friendly construction materials as lapilli-bio block. The neutralization processed Mt. Backdusan and Mt. Hallasan lapilli-bio block were identified vegetation characteristics and water purification properties. As a result, the adsorption of root was more favorable, depending on the material properties of many voids Lapilli. And if the neutralization processed lapilli-bio blocks maintain a long-term immersion conditions in contaminated water was found to be expected from the improvement effect of SS, T-N and T-P. However, field trials and continuous studies are expected to be identified by the use of lapilli-bio block.
