Production of Recombinant Proteins as Immuno-Analytical Markers of Genetically-Modified Organisms (GMO)

  • Marker proteins of genetically-modified organisms (GMO) and their antibodies were prepared and characterized as major components of an analytical system. We selected two GMO markers, neomycin phosphotransferase II and 5- enolpyruvylshikimate-3-phosphate synthase, and produced them from E. coli employing genetic recombination technology. After purification, their structural conformation and binding affinities to the respective antibodies were characterized. The results showed that the recombinant proteins were identical with commercially obtained reference proteins. We further used them as immunogens to raise polyclonal antibodies capable of discriminating GMO containing protein from non-GMO. Well-characterized marker proteins and antibodies will be valuable as immunoreagents in constructing analytical systems such as biosensors and biochips to measure quantities of GMO.

Bio-marker Detector and Parkinson's disease diagnosis Approach based on Samples Balanced Genetic Algorithm and Extreme Learning Machine (균형 표본 유전 알고리즘과 극한 기계학습에 기반한 바이오표지자 검출기와 파킨슨 병 진단 접근법)

  • A novel Samples Balanced Genetic Algorithm combined with Extreme Learning Machine (SBGA-ELM) for Parkinson's Disease diagnosis and detecting bio-markers is presented in this paper. Proposed approach uses genes' expression data of 22,283 genes from open source ParkDB data base for accurate PD diagnosis and detecting bio-markers. Proposed SBGA-ELM includes two major steps: feature (genes) selection and classification. Feature selection procedure is based on proposed Samples Balanced Genetic Algorithm designed specifically for genes expression data from ParkDB. Proposed SBGA searches a robust subset of genes among 22,283 genes available in ParkDB for further analysis. In the "classification" step chosen set of genes is used to train an Extreme Learning Machine (ELM) classifier for an accurate PD diagnosis. Discovered robust subset of genes creates ELM classifier with stable generalization performance for PD diagnosis. In this research the robust subset of genes is also used to discover 24 bio-markers probably responsible for Parkinson's Disease. Discovered robust subset of genes was verified by using existing PD diagnosis approaches such as SVM and PBL-McRBFN. Both tested methods caused maximum generalization performance.

Kidney protective potential of lactoferrin: pharmacological insights and therapeutic advances

  • Kidney disease is becoming a global public health issue. Acute kidney injury (AKI) and chronic kidney disease (CKD) have serious adverse health outcomes. However, there is no effective therapy to treat these diseases. Lactoferrin (LF), a multi-functional glycoprotein, is protective against various pathophysiological conditions in various disease models. LF shows protective effects against AKI and CKD. LF reduces markers related to inflammation, oxidative stress, apoptosis, and kidney fibrosis, and induces autophagy and mitochondrial biogenesis in the kidney. Although there are no clinical trials of LF to treat kidney disease, several clinical trials and studies on LF-based drug development are ongoing. In this review, we discussed the possible kidney protective mechanisms of LF, as well as the pharmacological and therapeutic advances. The evidence suggests that LF may become a potent pharmacological agent to treat kidney diseases.

Identification of a Third Haplotype of the Sequence Linked to the Restorer-of-fertility (Rf) Gene and Its Implications for Male-Sterility Phenotypes in Peppers (Capsicum annuum L.)

  • Cytoplasmic male sterility (CMS), one of the most important traits in crop breeding, has been used for commercial seed production by $F_1$ hybrid cultivars of pepper (Capsicum annuum L.). To develop reliable molecular markers for allelic selection of the Restorer-of-fertility (Rf) gene, which is known to be a major determinant of pollen fertility restoration in peppers, a sequence of approximately 10 kb flanking an RAPD fragment closely linked to the Rf locus was obtained by genome walking. A homology search revealed that this sequence contained an LTR retrotransposon and a non-LTR LINE-like retrotransposon. Sequencing of this Rf-linked region to search for polymorphisms between a dominant and recessive allele revealed 98% nucleotide sequence identity between them. A third polymorphic haplotype of the Rf-linked sequence, which has 94-96% nucleotide sequence identity with the two previously isolated haplotypes, was identified among a large number of breeding lines. Utilizing polymorphic sequences in the haplotypes, PCR markers were developed for selection of particular haplotypes and used to examine the distribution of the haplotypes in diverse breeding lines, cultivars, and C. annuum germplasms. Surprisingly, the third haplotype was the predominant type in C. annuum germplasms, while its frequency in $F_1$ hybrid cultivars was relatively low. Meanwhile, analysis of breeding lines whose Rf allele genotypes and male-sterility phenotypes were already known revealed that the third haplotype was mainly present in exotic breeding lines that cause unstable male-sterility when combined with sterile cytoplasms.

Carbon Nanotube DNA Bioconjugates as Nano-Bio Markers (탄소 나노튜브와 DNA와의 결합을 통한 나노-바이오 마커 응용)

  • Carbon nanotubes exhibit strong fluorescence emissions in the region of near infrared regions where most biomolecules are transparent. Such signals are highly sensitive to environment variations as well as adsorption of specific biomolecules. In this research, single walled carbon nanotubes(SWNTs) are assembled with different types of DNAs and used to target specific types of DNAs. Dot blot investigations and corresponding raman spectroscopy observations demonstrated excellent selectivity of carbon nanotube-DNA bioconjugates. The results show possibility of using SWNT as generic nano-bio markers for precise detection of different kinds of genes.

CD11b as a Biomarker for Canine Systemic Inflammatory Response Syndrome and Sepsis (개 전신성염증반응증후군 및 패혈증의 진단적 표지자로서 CD11b의 활용)

  • The aim of this study is to investigate neutrophil activation markers among canine ICU (intensive care unit) control, systemic inflammatory response syndrome (SIRS) and sepsis. These markers include WBC (white blood cells), platelets counts, blood film examination (neutrophilic band to segmentation ratio and neutrophilic degenerative changes), and flow cytometric analysis (CD11b expression of neutrophils). As a result, the mean CD11b fluorescence intensity of neutrophils and the neutrophilic degenerative change scores were both significantly higher in sepsis group (P<0.05). In addition, mortality was also found to be correlated with the up-regulation of CD11b expression in circulating neutrophils. This study demonstrates that CD11b expression of neutrophils could be more a reliable biomarker to predict prognosis in ICU patients than traditional blood film examination according to this study.
