Characterization of D-Glucose ${\alpha}$-1-Phosphate Uridylyltransferase (VldB) and Glucokinase (VIdC) Involved in Validamycin Biosynthesis of Streptomyces hygroscopicus var. limoneus KCCM 11405

  • Aminocyclitol antibiotic validamycin A, a prime control agent for sheath blight disease of rice plants, is biosynthesized by Streptomyces hygroscopicus var. limoneus. Within the validamycin biosynthetic gene cluster, vldBC forms an operon of vldABC with vidA, the gene encoding 2-epi-5-epi-valiolone synthase. Biochemical studies, employing the recombinant proteins from Escherichia coli, established VldB and VldC as D-glucose $\alpha$-1-phosphate uridylyltransferase and glucokinase, respectively. This finding substantiates that the validamycin biosynthetic gene cluster harbors genes encoding the enzymes for UDP-glucose formation from glucose. Therefore, we propose that validamycin biosynthesis employs its own catalysts to generate UDP-glucose, but not depending on the primary metabolism.

Safety Evaluation of Filamentous Fungi Isolated from Industrial Doenjang Koji

  • A few starters have been developed and used for doenjang fermentation but often without safety evaluation. Filamentous fungi were isolated from industrial doenjang koji, and their potential for mycotoxin production was evaluated. Two fungi were isolated; one was more dominantly present (90%). Both greenish (SNU-G) and whitish (SNU-W) fungi showed 97% and 95% internal transcribed spacer sequence identities to Aspergillus oryzae/flavus, respectively. However, the SmaI digestion pattern of their genomic DNA suggested that both belong to A. oryzae. Moreover, both fungi had morphological characteristics similar to that of A. oryzae. SNU-G and SNU-W did not form sclerotia, which is a typical characteristic of A. oryzae. Therefore, both fungi were identified to be A. oryzae. In aflatoxin gene cluster analysis, both fungi had norB-cypA genes similar to that of A. oryzae. Consistent with this, aflatoxins were not detected in SNU-G and SNU-W using ammonia vapor, TLC, and HPLC analyses. Both fungi seemed to have a whole cyclopiazonic acid (CPA) gene cluster based on PCR of the maoA, dmaT, and pks-nrps genes, which are key genes for CPA biosynthesis. However, CPA was not detected in TLC and HPLC analyses. Therefore, both fungi seem to be safe to use as doenjang koji starters and may be suitable fungal candidates for further development of starters for traditional doenjang fermentation.

  • '봄내엽1호' 들깨 품종은 강원대학교 농장에서 2006년에서 2008년에 걸쳐 파종과 계통선발을 통해 육성하였다. 품종의 특성검정은 2006년부터 2008년까지 들깨의 적정 재배조건 아래서 3회에 걸쳐 조사하였다. '봄내엽1호' 들깨 품종은 대조 품종보다 경장이 크고, 화방군수와 화방군당삭수가 많으며 잎색은 대조품종에 비해 진한 녹색을 띤다. 정유 성분 분석 결과 '봄내엽1호' 들깨잎의 주요성분으로는 Perilla ketone, Beta-caryophyllene, Myristicin인 것으로 나타났다.

방울토마토의 여름재배시 적정수경재배방식 구명 (The investigation of Appropriate Hydroponic System for Cherry Tomatoes in Summer Season)

  • 방울토마토에 적합한 수경재배방식에 관한 연구 결과, 담액수경에서 주경의 기부직경, 엽장, 엽폭, 화방길이 등의 생육이 가장 좋았으며, 암면재배와 NFT는 유사하였다. 개화시기와 과실의 착색일에 대하여는 각 방식마다 차이를 보이지 않았으나, 과실의 수확량은 담액수경에서 많았다. 건물율, 유기산, 경도, 당도 등은 담액수경에서 낮았고, 당산비와 vitamin C는 높아 담액수경에서 품질이 약간 우수한 것으로 나타났다. 따라서, 방울토마토의 여름재배에서는 담액수경방식이 바람직한 것으로 평가되었다.

가뭄 대응형 지하수 개발 우선순위 선정을 위한 농촌용수구역의 유형 분석 (Classifying Agricultural Districts for Prioritizing Groudwater Development Area based on Correlation and Cluster Analysis)

  • In this study, we analyzed the characteristics of 511 agricultural districts through statistical data, and classify these districts as the vulnerable area to drought through correlation and cluster analysis. The criteria for classification was related to ground-water recharge, irrigation water demand, and water supply. As a result, 8 types of agricultural districts were extracted. For example, the type 1 indicated the high priority area for ground-water development, thus the districts which were classified as type 1 showed ground-water use was less than 80 % of maximum capacity, and irrigation water supply was only 37.5 % and 76.5 % of irrigation water demand in upland and paddy field, respectively. As a result, 44 of 511 districts were classified as type 1.36 districts (types 5-8) were areas where groundwater development is limited. The results of this study are expected to provide useful information for establishing the direction of the rural area development project in connection with the revitalization of policy of people return to rural area.

Analysis of Purchasing Moderating Effect on Perfume Purchasing Propensities & Behavioral Attitudes

  • Purpose: This study looked at perfume buying patterns among 328 adolescents. Research design, data and methodology: The purchasing behaviors and attitudes of adolescents were divided into clusters and whether the purchasing behaviors and attitudes of each cluster had a moderating effect on purchasing behavior factors was analyzed. Results: Group classification according to attitude toward perfume purchasing behavior was divided into group 1, which purchases perfume according to one's own subjective opinion, and group 2, who purchases perfume according to external factors. Among the six purchasing behavior factors, the internal pleasure-seeking (p.<001) and fashion-seeking (p.<001) factors were statistically significant in both clusters 1 and 2, and in cluster 2, economic feasibility (p.<001)) internal product. It was found to be statistically significant other than pleasure and trend-seeking, indicating that there is a difference between the two groups. Conclusion:Adolescents consider economic feasibility when purchasing perfume, so it is necessary to set low prices and diversify products for marketing plans for perfume products

Morphology of Metal Salt of Carboxylic Acid: Metal and Acid Dependency on Branched Round Cluster Images

  • Metallic salts of C10-18 aliphatic carboxylic acids were prepared and their scanning electron microscopic images were analyzed for the morphology dependency with the metal and the carboxylic acid. Regardless of metal ion, metal salts of dicarboxylic acids showed a high crystallinity with a fiber image (SuA-Na). The aromatic dicarboxylates also represented a morphology of a rectangular-rod or board shapes (IA-Li, IA-Ba). With Na ion, most aliphatic carboxylate (MA, PA, SA) showed a fiber-like crystallinity. However, other monovalent Li, K and multivalent Mg, especially Al ion resulted a glassy-amorphous in the metallic salts of acids (MA, PA, SA). With divalent Ba and Ca ions, the metal salt of aliphatic acids expressed a branched round cluster shape as in SA-Ca, SA-Ba. Both Li and Mg ions with a similar size showed a strong morphological similarity in the metallic salts of aliphatic acids MA, PA, SA. In the case of Na and Ca ions with a similar size (98, 106 pm), both ionic salts of stearic acid gave a branching effect for a fiber or round granular image. In the case of hydroxyl-aliphatic acids (HLA, HPA, HSA), the fiber images in HLA-Na and HSA-Na was appeared about 100 nm thicker than those of nonhydroxycarboxylates (LA-Na, SA-Na). The metallic salts of unsaturated C-18 carboxylic acids (OlA, LeA and LnA) showed an amorphous glassy image due to a kinked carbon chain.

Synthetic Biology Tools for Novel Secondary Metabolite Discovery in Streptomyces

  • Streptomyces are attractive microbial cell factories that have industrial capability to produce a wide array of bioactive secondary metabolites. However, the genetic potential of the Streptomyces species has not been fully utilized because most of their secondary metabolite biosynthetic gene clusters (SM-BGCs) are silent under laboratory culture conditions. In an effort to activate SM-BGCs encoded in Streptomyces genomes, synthetic biology has emerged as a robust strategy to understand, design, and engineer the biosynthetic capability of Streptomyces secondary metabolites. In this regard, diverse synthetic biology tools have been developed for Streptomyces species with technical advances in DNA synthesis, sequencing, and editing. Here, we review recent progress in the development of synthetic biology tools for the production of novel secondary metabolites in Streptomyces, including genomic elements and genome engineering tools for Streptomyces, the heterologous gene expression strategy of designed biosynthetic gene clusters in the Streptomyces chassis strain, and future directions to expand diversity of novel secondary metabolites.

HPC 환경을 위한 워크플로우 기반의 바이오 데이터 분석 시스템 (Workflow-based Bio Data Analysis System for HPC)

  • 인간 게놈 프로젝트의 완성 이후 유전체 분석 비용은 매우 빠르게 감소하고 있다. 이에 따라 인간 유전체 분석 요구가 급증할 것으로 예상된다. 인간 유전체 분석과 같은 대규모 바이오 데이터 분석을 고속으로 수행하기 위해서는 비IT 전문가들이 다양한 특성의 바이오 응용들을 고성능컴퓨팅 시스템을 통해 효과적으로 실행할 수 있어야 한다. 이를 위해서는 여러 응용들이 조합되어 순서를 갖고 실행되어야 하는 바이오 응용들을 워크플로우 형태로 쉽게 정의할 수 있어야 하며, 이 워크플로우를 HPC 클러스터 시스템에서 최적 자원을 할당 받아 분산 병렬 수행시켜야 한다. 이를 통해 바이오 데이터 분석 성능과 응답시간의 개선을 기대할 수 있다. 본 논문에서는 HPC 환경에 익숙하지 않은 비IT 바이오 연구자들이 쉽게 바이오 데이터 분석을 할 수 있도록 바이오 워크플로우를 쉽게 정의하고 실행할 수 있는 바이오 특화된 워크플로우 기반 대규모 데이터 분석 시스템을 제안한다.

수박의 착과절위가 종자 발아 및 유묘활력에 미치는 영향 (Influence of Fruit Set Internode on Seed Germination and Seedling Vigor in Watermelon)

  • This study was conducted to investigate the effects of fruit set internode on the germination of watermelon seeds. Generally fruits setted in higher than 20 internode were high percent of germination, fruit setted in low internode(5-10) is low. The higher fruit-set internode, fruit weight was significantly increased, but 1,000 seed weight was decreased the more fruit set internode. However number of seed was about 300 seeds irrespective of fruit-set internode. Investigating viability of seed by BP test, general percent germination was recorded highest at 5 flower cluster above 20 internode of fruit-set internode. In contrast, condition of fruit setted on 3, 4, 6 flower cluster were high percent of germination, regarded as unnormal germination not to be as normal seedling by 2~3%. Seed harvested at 5 flower-cluster had high viability on hypocotyl height and diameter of seedling through early growth test, but were not significant. Therefore 5 flower-cluster was optimum fruit-set internode to obtain high-quality seed.
