생약복합제제(生藥複合製劑)의 약효연구(藥效硏究) 제13보(第13報) -대시호탕(大柴胡湯)이 혈관(血管), 간독성(肝毒性), 이담(利膽) 및 항당뇨효과(抗糖尿效果)에 대(對)하여- (Studies on the Efficacy of Combined Preparation of Crude Drugs (XIII) -Effects of ‘Daeshiho-Tang’ on the Blood Vessels, Activities of the Liver Enzyme, Biliary Secretory and Anti-Diabetic Actions-)

  • These studies were conducted in an attempt to investigate effects of 'Daeshiho-Tang' on the blood vessels, activities of liver enzyme and excretory action of bile juice in serum of $CCl_4-intoxicated$ rabbits, glucose and total cholesterol levels in serum of alloxan-induced diabetic rabbits. The result of these studies were summarized as follows; 1. Hypotensive ind vaso-dilatating actions due to vascular smooth muscle relaxation were noted in frogs, rabbits and dogs. 2. GOT, GPT and total cholesterol in serum of $CCl_4-intoxicated$ rabbits showed significant depressive effects. 3. Significantly increased biliary secretary effect was noted in $CCl_4-intoxicated$ rabbits. 4. Glucose and total cholesterol levels showed markedly decraese in alloxan-induced diabetic rabbits. Considering the experimental studies results, it is suggested that 'Daeshiho-Tang' has therapeutic efficacy to treat febrile diseses and metabolic diseases.

  • An experiment was conducted to establish a large scale production method of anti-serum against chicken IgA and to profile the developmental changes of serum IgA levels during the feeding period(from hatching to 7 weeks of age) in broiler chicks. Blood samples were taken from Hubbard chicken at the age of hatching, 3 days of age, and weekly thereafter till to 7 weeks of age. The pure IgA was isolated from ammonium sulfate treated chicken bile juice by gel filtration chromatography ( Sepharose GL-6B) - The quantitative assay of serum IgA were carried by RID method. Developmental changes of serum IgA concentrations were 0.42 mg /mL at hatching, thereafter dicreased gradually, lowest at 1 week of age(0.17 mg /mL), and gradually increased to 7 weeks of age(2.73 mg /mL). There was no sexual difference in serum IgA level, but female chicks showed higher IgA levels than male chicks during the experimental period.

Salmonella dublin에 의한 소의 살모넬라증의 발생 (An Acute Outbreak of Calf Salmonellosis Caused by Salmonella dublin)

  • An epizootic of calf diarrhea occurred in an extensive dairy farm located in Gangwondo province in December of 1985. The patients showing fever, chills, complete anorexia and watery diarrhea sometimes dysentery were concentrated among the calves ranged from 8 days to 3 months old and many of them died in acute course. The five carcases were examined by pathological and bacteriological means. The predominant gross lesions were edematous swelling of the mesenteric lymph nodes, profuse catarrhal or diphtheritic enteritis and extensive purulent bronchopnenomonia or lobar fibrinous pneumonia. Microscopically the lesions of the liver and spleen were characterized by coagulative necrotic foci and granulomatous nodules. The spleen and lymph node showed also proliferative changes of reticular cells and involution of intestinal lymphatic nodules were noticed. The strains of Salmonella(S) species isolated from the liver, spleen, bile juice, peritoneal fluid and thoracic fluid were identified serologically as S. dublin. These clinical and pathological findings of the disease were those of acute enteritis form of salmonellosis and characterized by high morbidity and mortality among the calf herd. The report also signifies the first description of an epizootic of bovine salmonellosis caused by S. dublin in Korea.

Physiological Characteristics and Immunomodulating Activity of Streptococcus macedonicus LC743 Isolated from Raw Milk

  • To develop a new starter culture for fermented milk, Streptococcus macedonicus LC743 was isolated from raw milk and its physiological characteristics were investigated. S. macedonicus LC743 showed good immunomodulating activity compared to the index LAB starters tested. The optimum growth temperature of S. macedonicus LC743 was $40^{\circ}C$, and it took 18 h to reach pH 4.34 under these conditions. S. macedonicus LC743 showed higher sensitivity to novobiocin in a comparison of 15 different antibiotics and showed the highest resistance to gentamycin. It also showed higher activities of leucine arylamidase and acid phosphatase. Moreover, it was comparatively tolerant to bile juice and acid and displayed high resistance to Escherichia coli, Salmonella Typhimurium, Staphylococcus aureus with rates of 80.0%, 68.42%, and 81.54% respectively. These results demonstrate that S. macedonicus LC743 could be an excellent starter culture for fermented milk with a high level of immunomodulating activity.

유산균을 안정화시킨 마이크로캅셀의 제조 및 평가 (Microcapsules for Stabilization of Lactic Acid Bacteria)

  • A new technique has been developed for the preparation of Lactobacillus microcapsules to enhance the stability against high temperature, humidity, gastric acid and bile acid. Employing fluidized bed coating, primary sub-coating was processed in non-organic solvent system, so that Lactobacillus did not directly contact with organic solvent. Secondary enteric-coating was processed in organic solvent with low temperature $(below\;33^{\circ}C)$ technique, which minimized the heat labilability of Lactobacillus. Survival rate of Lactobacillus within microcapsule was not less than 95% and acid tolerance was above 30% in the artificial gastric acid. Further more it was dissolved in the artificial intestine juice within 2-3 hr. Average size of Lactobacillus microcapsules was $450\;{\mu}m$(25-50 mesh) and its viability was above 90% in the direct tableting.

김치 유래 유산균의 항균적 특성과 이용 (Chacteristics & Applications of Lactobacillus sp. from Kimchi)

  • Earlier in Korea Kimchi was made in every family and every province has own taste and specialties. These days almost of the Kimchis are manufactured. We collected variable Kimchis, which were made for private use and isolated microorganisms. Some interesting micobial cells were identified and studied for its application as food and drinks. One of them was identified as Lactobacillus sakei KJ123. This strain is known as producing interesting aromatic components during Sakei fermentation like Kimchi in variable conditions. We tried to develop a health beverage with fermentation process. The Cucurbita maxima has been known as a traditional healthy food and variable positive effects on the human body were already reported. In this study we tried to develop a production process for a healthy fermented drink on this substrate with strains originated from Kimchi. Many kinds of lacctobacilli species existed in the fermented food cannot survive in the acidic conditions like human stomach. So we selected resisting strains in this conditions. The survival rate of Lactobacillus sakei cells in the artificial gastric juice and bile acid and other physiological characteristics at the variable conditions have been tested. After fermentation process some sensory tests on the product with panels were tried.

Physiological Characteristics and Immunomodulating Activity by Lactobacillus paracasei subsp. paracasei BFI46 Isolated from New-Born Infant Feces

  • In order to develop a new starter for fermented milk, Lactobacillus paracasei subsp. paracasei BFI46 (BFI46) obtained from new-born infant feces was investigated for physiological characteristics. Good immunomodulating activity was evident compared with commercial lactic acid bacteria starter cultures. The optimum growth temperature of BFI46 was $40^{\circ}C$ with 12 h required to reach pH 4.3. Testing with 13 different antibiotics revealed greatest sensitivity of BFI46 to penicillin- G and chloramphenicol, and heightened resistance to neomycin, kanamycin and polymyxin. BFI46 displayed higher esterase activities compared to 18 other enzymes, was comparatively tolerant to bile juice and able to survive at pH 2 for 3 h, and displayed high resistance against Escherichia coli and Salmonella Typhimurium with a survival rate of 57.14% and 96.36%, respectively. The results indicate that BFI46 could be an excellent starter culture for fermented milk with high level of immunomodulating activity.

김치에서 분리한 저온성 젖산균의 배양특성 (Cultural Characteristics of Psychrotrophic Lactic Acid Bacteria Isolated from Kimchi)

  • 저온발효된 김치에서 분리되어 Leuconostoc mesenteroides subsp. mesenteroides, Leu. mesenteroides subsp. dextranicum, Leu. paramesenteroides, Leu. lactis, Lactobacillus bavaricus 및 Lac. homohiochii로 각각 동정된 저온성 젖산균 60주에 대하여 여러 조건에서의 배양특성들을 조사하였다. 모든 균주들이 $5^{\circ}C,\;10^{\circ}C$$37^{\circ}C$에서 증식하였고, $45^{\circ}C$에서는 증식하지 못하였다. 최적 증식 온도는 Leu. mesenteroides의 균주들은 $33{\sim}34^{\circ}C$이었고, Lactobacillus속의 균주들은 $34{\sim}36^{\circ}C$이었다. Leu. mesenteroides와 Lactobacillus속의 균주들은 모두 초기 pH 4.8 및 4.2에서 증식하였으나 4.0에서는 증식하지 못하였다. Glucose broth에서의 최종 pH는 Leu. mesenteroides의 균주들은 $3.85{\sim}4.10$이었고, lactobacillus속 균주들은 $3.82{\sim}3.99$이었다. 60균주 모두 litmus milk에서 우유를 응고시키지 못하였고, litmus를 환원시키지도 못하였다. Leu. mesenteroides와 Lactobacillus속의 균주들은 모두 에탄올 7% 함유배지와 NaCl 6.5% 함유배지에서 증식하였고, 에탄올 15% 함유배지와 NaCl 10% 함유배지에서는 증식하지 못하였다. 모든 균주들이 pH 3.5인 인공위액과 40%의 담즙액에서 내성을 나타내었다. 특히 Leu. mesenteroides의 균주들은 모두 pH 3.0인 인공위액에서도 내성을 나타내었다. 본 젖산균들의 생리적인 특성은 여러 점에서 Bergey's Manual of Systematic Bacteriology에 기재된 내용들과 일치하지 않으므로 더욱 정밀한 조사를 실시하여 분류학상의 위치를 정확히 밝혀 볼 필요가 있을 것으로 생각되었다.

${\beta}-Galactosidase$ 생산 유산균 선별 및 특성 조사 (Characterization and Selection of Lactic Acid Bacteria Producing ${\beta}-Galactosidase$)

  • [ ${\beta}-Galactosidase$ ] (lactase)를 생산하는 유산균을 선별하고 그들이 생산하는 ${\beta}-galactosidase$의 특성을 조사하였다. X-gal이 포함된 MRS배지에서 푸른색을 나타내는 약 100여개의 콜로니를 김치로부터 분리하였다. 그들 중 ET-1과 LA-12 두 균주를 최종 선별하였으며, 16S rDNa 염기서열 분석을 통해 각 각 Lactobacillus fermentum과 L. acidophilus와 상동성이 높은 Lactobacillus 속으로 동정되었다. 선별된 두 균주는 높은 ${\beta}-galactosidase$ activeity와 우수한 생존력을 나타내었으며, 이들이 생산하는 ${\beta}-galactosidase$의 최적 활성조건을 조사한 결과, 반응온도 $55^{\circ}C$에서 가장 높은 활성을 나타내었으며, 반응 pH의 경우 ET-1은 pH 5.5에서 최고 활성을 나타내었고, LA-12는 pH7.0에서 가장 높은 활성을 보였다. 선별 유산균 ET-1과 LA-12는 위액과 담즙산액에 대해 우수한 내성을 나타내었다. 두 개의 선별 균주는 인공위액에 3시간 배양 후 초기 생균수와 비교하여 생균수 변화가 거의 없었고, 담즙산액으로 사용한 0.3% oxgall에서 24시간 반응 후에도 1 log cycle 정도 감소된 $10^8CFU/ml$의 생균수를 유지하였다. 이러한 결과를 바탕으로 유산균 ET-1과 LA-12는 유제품 산업에 이용 가능성이 크다고 판단된다.

담즙분비촉진 식물추출물을 이용한 심근관류영상 검사법 개선 (A Modified Protocol for Myocardial Perfusion SPECT Using Natural Plant Extracts for Enhancement of Biliorg Excretion)

  • 목적: 심근관류 영상은 관상동맥 질환을 진단하고 환자의 예후를 평가하는데 널리 사용되고 있다. $^{99m}Tc$ 제재를 이용한 심근관류 영상은 간 방사능을 줄이고 좋은 영상을 얻기 위해 주사 후 일정시간을 대기하고, 지방식을 섭취하는 검사방법을 사용하고있다. 이번 연구에서는 식물 추출물 혼합제재를 이용하여 간 방사능의 배출을 촉진함으로써 검사시간을 단축시키고, 심근영상의 질을 향상시킬 수 있는지 알아보고자 하였다. 대상 및 방법: 프로토콜 1은 식물추출물의 환약제재(이하 혼합제재)를 투여한 개선법이 간 방사능의 배출을 촉진하고 영상의 질을 향상시키는 지 알아보기 위해 건강하고 젊은 자원자 10명(남: 녀=7: 3, 평균 연령 $24.1{\pm}2.4$세)을 대상으로 혼합제재를 복용하고 15분 후 MIBI를 주사하였으며, 주사 후 30분, 60분, 90분, 그리고 120분째 영상을 얻어 분석하였다. 프로토콜 2는 $^{99m}Tc$ 심근관류 영상법에서 널리 이용되고 있는 1일 휴식-부하방법에 혼합제재를 사용하였을 때의 개선법의 효과를 알아보기 위해 다른 자원자 10명(남: 녀=8: 3, 평균 연령 $26.1{\pm}2.9$세)을 대상으로 개선법과 고식적 방법(우유와 아이스크림 등 지방식 투여)을 적용한 영상을 얻어 분석하였다. 결과: 프로토콜 1에서 개선법과 고식적 방법의 심근/폐 방사능 비는 90분 영상까지 유의한 차이를 보이지 않았으며, 간/폐 방사능 비는 120분 영상까지 개선법이 고식적 방법에 비해 유의하게 낮은 값을 보였다. 심근/간 방사능 비는 개선법에서 지속적으로 높은 값을 보였다. 프로토콜 2에서 첫 번째 MIBI를 주사한 후 심근/페 방사능은 유의한 차이가 없었으나 심근/간 방사능비는 45분과 60분 영상에서 개선법이 유의하게 높은값을 보였다. 두 번째 MIBI를 주사한 후에는 20분 영상부터 지속적으로 개선법의 심근/간 방사능 비가 높은 값을 보였으나, 첫 번째 주사와 마찬가지로 심근/폐 방사능 비는 차이가 없었다. 결론: 담즙 배출을 촉진하는 천연식물 추출 혼합제재를 심근관류 영상을 획득하는데 적용하면 지방식 섭취없이 검사 소요 시간을 2시간대로 단축시킬 수 있어 환자의 불편을 최소화시키면서 영상의 질을 향상시킨 수 있을 것으로 판단된다.
