A central composite design was applied to investigate the effects of the independent variables of NaOH concentration(X1) and extraction time(X2) on dependent variables such as Yield(Y1), Xyl/Ara ratio(Y2), uronic acid(Y3), $\beta$-glucan(Y4) and total sugars(Y5) of hemicelluloses separated from rice bran. The Coefficients of determination(R2) in various models ranged from 0.8626 to 0.9319. Yield increased with increased NaOH concentration and extraction time. The optimum extraction conditions were NaOH concentration at 2.45M and extraction time of 24.2 h. Predicted values at the optimized conditions were acceptable, compared with experimental values. The structural characteristics of an optimum hemicellulose extract were explored. As a result, it showed that the surfaces of hemicellulose had a highly irregular reticulated structure. And also it was both small and large molecular particle in the hemicelluloses. Their average molecular weights were in the ranges $235{\sim}240$ kDa and $8.0{\sim}9.4kDa$, respectively.
During the pearling process of hull-less barley, protein, lipid, ash and insoluble dietary fiber (IDF) contents decreased, while soluble dietary fiber (SDF) and ${\beta}-glucan$ contents slightly increased. Depending on milling methods and types of grinding mills used, there were differences in particle size distribution of barley flour. Flour particle size was smaller in the following order of Fitz mill, Ball mill, Pin mill, Cyclotec sample mill and Jet mill. Color (brightness) was closely related to the particle size of barley flour. Damaged starch (%) in pearled barley flour was the highest in Jet mill among different mills. Flours prepared with Cyclone mill and Pin mill had a reasonable amount of damaged starch. Flour produced by Fitz mill showed the lowest amount of damaged starch. Scanning electron microscopy (SEM) of the flour samples demonstrated different sizes and shapes of particles consisting of starch granules and cell wall materials. Damaged starch tended to increase water absorption index (WAI), water solubility index (WSI), and water retention capacity (WRC). Pasting viscosity determined by amylograph was relatively high in Pin-milled and Cyclone-milled flours. Viscosity was the lowest in coarsely ground flour by Fits mill.

This study evaluated the functional components of barley bran with different particle sizes and cultivars (Dahan, Hinchalssalbori, Heukgwang, Huknuri, and Boseokchal). Barley bran divided into fractions I (<60 mesh), II (60~100 mesh), and III (>100 mesh) was collected as pearling by-products produced by an industrial process consisting of consecutive barley pearlers. Total ${\beta}-glucan$ contents of all cultivars were especially highest in fraction II. Total arabinoxylan was the highest in barley bran from Boseokchal in fraction II. Total polyphenol contents were the highest in bran from Boseokchal and Hinchalssal in fraction II, and contents ranged of 5.61~7.00 and 4.24~6.58, respectively. Total flavonoid contents of all cultivars were especially highest in fraction II. 2,2'-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities ranged from 2.78~7.53 mg L-ascorbic acid (AA) eq/g and 2.24~4.83 mg AA eq/g, respectively. ABTS and DPPH radical scavenging activities were the highest in barley bran from Dahan in fraction II. In this study, fraction II showed enriched functional components and has the best particle size range for enriched antioxidant activities. These results provide useful data for selection of appropriate cultivars and particle size of bran to achieve high quality barley processing.
Sprassis crispa was cultivated using soybean curd whey, and its antimicrobial activities were examined against those of eight microorganisms that were foodborne pathogens or food-poisoning bacteria. The culture broth of soybean curd whey was superior in mycelium content (17.76 g/L) to that of the defined culture broth, and the ${\beta}$-glucan content was about 10.64 percent (w/w). The antimicrobial activities of the culture broth were confirmed against those of B. cereus, St. aureus, L. monocytogenes and S. typhimurium using the paper disk method. The antimicrobial activity was also maintained after the heat treatment and alcalase treatment. The filtrate with less than 3 kDa M.W. also showed the antimicrobial activity against four strains: B. cereus, St. aureus, L. monocytogenes and S. typhimurium. The minimum inhibitory concentration (MIC) was about 1.26 mg/mL in the B. cereus and 12.6 mg/mL in the St. aureus and L. monocytogenes. The S. typhimurium showed a MIC of 62.8 mg/mL. Thus, the culture of Sparassis crispa using soybean curd whey provides a thermally stable antimicrobial agent that can be used as a natural preservative in the biofood industry.
A new oak mushroom cultivar 'Hwadam' was bred from monokaryotic strains of 'GMLE36062' and 'LE15401'. The optimum temperature for mycelial growth of 'Hwadam' on potato dextrose agar was 25℃. The cultivation period of 'Hwadam' lasted for 129 days until the first harvest of the fruiting bodies, including 35 days for spawn run, 84 days for browning, and 10 days for development of fruiting bodies. The fruiting bodies of 'Hwadam' had a hemispherical, brighter pileus and thicker stipe than the control cultivar (Sanjo 701ho). The three-cycle yield of 'Hwadam' was 554 g, which was higher than that of control cultivar (405 g). In addition, 'Hwadam' contained 79.4 mg/g of beta-glucan and 0.93 mg GAEs/g of phenolic compounds, which was similar to that of the control cultivar but higher than that of 'L808' and 'Chujae 2ho'.
In this study, we analyze the antioxidant and anti-inflammatory activities of various extracts of Cryptoporus volvatus. Three extracts were prepared using hot-water, 70% ethanol, and 70% methanol. Compared to the hot-water and 70% methanol extracts, the 70% C. volvatus ethanol extracts showed significantly higher DPPH radical scavenging activities (76.1%), nitrite scavenging activities (29.5%), and total polyphenol content (9.17 mg GAE/g). The DPPH radical and nitrite scavenging activities of C. volvatus both correlated significantly with the total polyphenol content. The Pearson's correlation coefficient of DPPH radical and nitrite scavenging activities with total polyphenol content were r = 0.744 (p<0.05) and r = 0.921 (p<0.01), respectively. To test their anti-inflammatory effects, nitric oxide production and cell viability were measured by performing nitric oxide (NO) and MTT assays on lipopolysaccharide-treated RAW 264.7 cells. The NO production levels of the 70% ethanol extract-treated cells were slightly lower than those of the others. In our MTT assay, the hot-water extracts showed higher cell viabilities (81.4%) than those of 70% ethanol and 70% methanol extracts. The ${\beta}$-glucan content of C. volvatus was 37.8%. From the results of this study, it is suggested that C. volvatus, an easily accessible wild mushroom, has potential to be used in the development of medicines, cosmetics, and functional health foods.

"Nulichal" was developed by the National Institute of Crop Science, RDA, in 2010. The initial cross was conducted in 2000, and an elite line (HB15305-B-B-31-2) was selected in 2005. Subsequently, preliminary yield and advanced yield trials have designated it as "Iksan 95." It showed good agronomic performance in regional yield trials from 2008 to 2010 and was released as "Nulichal," with resistance against the barley yellow mosaic virus (BaYMV) and high yields with a waxy endosperm. The average heading and maturation dates of "Nulichal" were April 24 and May 30, respectively in paddy fields, which were 1 day later than those of the check cultivar "Saechalssalbori." It had culm and spike lengths of 82 and 4.0 cm, respectively. It showed $774spikes/m^2$, 63 grains/spike, 25.0 g of 1,000-grain weight, and 793 g of test weight. "Nulichal" showed higher levels of resistance to BaYMV than the naturally occurring Iksan (type III strain), Naju (type I), and Jinju (type II). The expansion rate was higher than that of "Saechalssalbori" and its hardness, gumminess, and chewiness were lower than those of "Saechalssalbori." Its average pearled grain yield in the regional yield trial was 4.00 MT/ha in paddy fields, which was 10% higher than that of the check cultivar. The combined availability of "Nulichal" was improved by its increased culm length compared with that of "Hinchalssal."

A new black barley variety, "Heuksoojeongchal," was developed as a cross between "Keunalbori 1" and "Masankwamaek/Mortoni" at the National Institute of Crop Science, RDA in 2014. Heuksoojeongchal had a longer culm length (89 cm) and a later average heading date and maturity than Seodunchalbori. It also had a spike length of 5.0 cm, $543spikes/m^2$, and 54 grains/spike. The 1000-grain weight of Heuksoojeongchal (36.0 g) was heavier than that of Seodunchalbori. Heuksoojeongchal showed resistance to barley yellow mosaic virus (BaYMV). The yield potential of Heuksoojeongchal was approximately 7% higher than that of Seodunchalbori upland, but the average yield potential was similar in upland and paddy fields. Among the quality characteristics, the ${\beta}-glucan$ content (6.7%) was higher and the amylose content (5.5%) was lower than those of Seodunchalbori. The whiteness of the black barley was low, and its absorption rate and spreadability were similar to those of Seodunchalbori.

Flammulina velutipes, known as winter mushroom in the family of Physalacriaceae, is the main edible and export mushroom with the third highest production after oyster and king oyster mushroom in Korea. However, as normal consumers regard F. velutipes as a simple subsidiary material, there is a limitation to increasing mushroom demand. In order to overcome the consumption limit and increase the differentiation of new varieties, it is necessary to breed varieties with enhanced functionality in consideration of consumer preferences. Therefore, the study was performed to analyze nutrient components and several useful functional substances with 26 genetic resources of F. velutipes. Analyses of inorganic compound(Ca, K, Mg) and 15 amino acids revealed that Strain 4148 had the highest content among the 26 strains. Beta-glucan, which increases immune activity and polyphenol, which exert antioxidant effects were higher in non-white strains than in white strains with a small number of exceptions. Among the five fatty acids, linoleic acid(an omega-6 fatty acid) and α-linolenic acid(an omega-3 fatty acid), were detected in six mushroom strains. α-linolenic acid, which was not found in five major mushrooms including oyster mushrooms, was identified in F. velutipes. The results of HPLC analysis showed that 'Auram' (Strain 4232) and 'Baekseung'(Strain 4230) had the highest content of the stabilizing neurotransmitter GABA(15.38 ㎍/ml and 20.56 ㎍/ml, respectively) among non-white and white strains, respectively. Our findings provide useful information for breeding F. velutipes to obtain strains with enhanced functionality.

Lentinula edodes is one of the most produced mushrooms in the world. In this study, the effects of L. edodes water extracts and lentinan, a beta-glucan from this mushroom, on the proliferation of Bos taurus Hanwoo myosatellite cells were studied. The betaglucan content of the L. edodes water extract was approximately 15.20% at 85 ℃ for 4 h, 13.64% at 100 ℃ for 4 h, 9.48% at 40 ℃ for 8 h and 8.21% at room temperature for 24 h. L. edodes water extract was added to the culture of Hanwoo myosatellite cells. The expression of the MyoD gene increased in the addition of the extract at 40 ℃ for 8 h and 100 ℃ for 4 h, and the expression of the Myogenin gene increased in the addition of the extract at 40 ℃ for 8 h, but proliferation and activity did not increase compared to no addition. However, the addition of lentinan to the culture of Hanwoo myosatellite cells increased the expression of Myogenin gene related to muscle formation increased and the proliferation and viability of the cells. This study proved that the components of L. edodes can affect the proliferation of Hanwoo myosatellite cells, and further research will help develop the mushroom industry and cultured meat industry in the future.
