Cloning of a Gene Involved in Biosynthesis of ${\beta}-1,3-glucan$ in Saccharomyces cerevisiae (베타-1,3-글루칸 생합성에 관여하는 Saccharomyces cerevisiae 유전자의 클로닝)

  • DNA fragment being able to restore in vitro activity of ${\beta}-1,3-glucan$ synthase was cloned by transformation of the Saccharomyces cerevisiae LP353 mutant strain with genomic library constructed in the YCp50. For the selection of transformants which showed no detectable phenotype linked to recovery of the defect in ${\beta}-1,3-glucan$ synthase activity, the colony autoradiography was succesfully applied. The restriction map of the cloned DNA fragment, which is 8.5-kb in length, was constructed. Both the YEplac195 and the YCp50 carrying the 8.5-kb fragment increased ${\beta}-1,3-glucan$ synthase activity of LP353 by two fold. Neither the YEplac195 nor the YCp50 carrying the 8.5-kb DNA fragment, however, complemented the temperature-dependent osmotic sensitivity which is another distinctive phenotype of LP353. Subcloning experiments indicated that a functional region was located in 4.8-kb BglII-KpnI fragment. The 4.8-kb fragment was also able to increase the level of ${\beta}-1,3-glucan$ content in cell wall as well as the resistance of cells to cell wall lytic enzyme, ${\beta}-1,3-glucanase$. The growth rate of the LP353 with 4.8-kb fragment was almost same as that of wild type strain in liquid medium with 1.2 M sorbitol at nonpermissive temperature. Taken these results together, the 4.8-kb fragment seemed to contain the BGS2 gene for ${\beta}-1,3-glucan$ synthase activity in yeast S. cerevisiae.

Immune-Enhancing Alkali-Soluble Glucans Produced by Wild-Type and Mutant Saccharomyces cerevisiae

  • The alkali-soluble glucan of the yeast cell wall contains $\beta-(1,3)-$ and (1,6)-D-linkages and is known to systemically enhance the immune system. In the previous study [6], in order to isolate cell wall mutants, a wild-type strain was mutagenized by exposure to ultraviolet light, and the mutants were then selected via treatment with laminarinase $(endo-\beta-(1,3)-D-glucanase)$. The mass of alkali- and water-soluble glucans produced by the mutant was measured to be 33.8 mg/g of the dry mass of the yeast cell. Our results showed that the mutants generated the amount of alkali-soluble glucan 10-fold higher than that generated by the wild-type. Structural analysis showed that the alkali-soluble glucan from the mutants was associated with a higher degree of $\beta-(1,6)-D-linkage$ than was observed in conjunction with the wild-type. Yeast cell wall $\beta-glucan$ was shown to interact with macrophages via receptors, thereby inducing the release of tumor necrosis factor alpha $(TNF-\alpha)$ and nitric oxide. Alkali-soluble $\beta-glucans$, both from water-soluble and water-insoluble glucan, exhibited a higher degree of macrophage activity with regard to both the secretion of tumor necrosis factor alpha $(TNF-\alpha)$ and nitric oxide and direct phagocytosis, than did the positive control ($1{\mu}g$ of lipopolysaccharide).

The Modulating Effect of β-1, 3/1, 6-glucan Supplementation in the Diet on Performance and Immunological Responses of Broiler Chickens

  • The object of this trial was to investigate the effect of dietary ${\beta}$-1,3/1,6-glucan supplementation on the performance and immunological response of broiler chickens. Two hundred and forty 1-day old male broilers ($39{\pm}1g$) were separated into six treatments which were given six different feeds containing 0 (control), 25, 50, 75, 100 and 125 mg/kg dietary ${\beta}$-1,3/1,6-glucan supplementation. On days 21 and 42, body weight gain, feed consumption and feed conversation rate were recorded as measures of growth performance. The levels of key cytokines in the immuno-regulating pathway: interleukin-1 (IL-1), interleukin-2 (IL-2), interferon $\gamma$(IFN-$\gamma$, tumor necrosis factor $\alpha$(TNF-$\alpha$, and the concentrations of signal molecules: peripheral blood plasma globulin, serum Immunoglobulin G (IgG) and intestinal secretary Immunoglobulin A (sIgA), were measured as indices of the immune response to determine suitable levels of dietary ${\beta}$-1,3/1,6-glucan supplementation. The results indicated that performance was elevated quadratically with dietary ${\beta}$-1,3/1,6-glucan supplementation. Maximal growth performance and an enhanced immunological response were obtained at a supplemented level of 50 mg/kg.

The Effects of Bete-Glucan Intake and Strength Exercise on Physical Strength, Blood Lipid and Insulin Resistance in Adult Women (베타글루칸 섭취와 근력운동이 성인여성의 체력, 혈액지질 및 인슐린저항성에 미치는 영향)

  • Purpose : This study aimed to determine the effects of a 12-week program of beta-glucan intake and muscle strengthening exercises on the body composition, physical fitness, blood lipids, insulin and insulin resistance of adult women. Methods : For this study, 45 adult women were selected and divided into two groups : the experimental group (n= 25), which received a combination of beta-glucan and strength exercise, and the control group (n= 20), which received only strength exercise. The program was conducted for 50 minutes three times a week for 12 weeks. Results : Several significant differences were found in both groups pre-to post program. First, in both groups, there were significant differences in hip circumferences (p<.05). Second, the results showed significant improvements in left grip strength and sit-ups achieved (p<.05), right grip strength (p<.001). Third although the results show no significant post-program differences in either group, there was a significant pre-to post-program difference in free fatty acids in both groups (p<.05). Fourth, the results show no significant differences in insulin or insulin resistance in either group, nor do they show any significant differences between the groups. Conclusion : The above results indicate that both groups exhibited decreased hip circumference and increased muscle strength (left grip strength) and muscle endurance. Furthermore, they indicate that when the beta-glucan intake and exercises were combined, the effects were more pronounced. Finally, because both groups exhibited significant increases in fat-free mass, muscle mass, skeletal muscle mass, muscle strength (right grip strength), and coordination as well as decreases in free fatty acids, it can be concluded that both the combined program of beta-glucan intake and exercise and a program of exercise alone can positively affect these outcomes.

Production of Nitric Oxide in Raw 264.7 Macrophages treated with Ganoderan, the ${\beta}-Glucan$ of Ganoderma lucidum (영지의 균사체성 ${\beta}-glucan$에 의한 Raw 264.7 대식세포의 Nitric Oxide생성)

  • Ganoderan (GAN), an immunomodulating ${\beta}-glucan$ of G. lucidum, induces potent antitumor immunity in tumor-bearing mice. This study was set up to elucidate the ability of macrophage activation of GANs. GAN-treated Raw 264.7 macrophages showed enhanced production of nitric oxide (NO). The ability of GANs to produce NO was based on differences in chemical composition of GANs obtained from the mycelium on various carbon sources and mycelial fractionation. The highest NO production was observed in CW-AS-WS polysaccharide which was extracted from the mycelial wall. GAN-treated Raw 264.7 cells gave a 2-to 5-fold (24 hr) formation of NO levels compared with those treated with medium only. Partial removal of the protein in the extracellular GAN by TCA treatment did appreciably reduce its capacity to secrete NO. The mixture effect of GAN and LPS increased the nitric oxide secretion from RAW 264.7. The cell proliferation of GAN-treated Raw 264.7 cell tines inhibited as compared with its control. Of the culture supernatant of macrophage activated by GAN, the percentage of cytotoxicity against mouse leukemia L1210 cells was slightly dependent on the amount of NO in the culture supernatants of the activated-macrophages. These results indicate that the ${\beta}-glucan-related$ polysaccharides of the higher fungus activate macrophage and release nitric oxide. It also suggests that murine macrophages possess certain receptors for ${\beta}-anomeric$ glucans and play a critical role of ${\beta}-glucan-related$ tumor killing mechanism.

Free sugar, amino acid, and beta-glucan content in Lentinula edodes strains collected from different areas (표고 수집종들의 유리당, 아미노산 및 β-glucan 함량)

  • In this study, we analyzed the nutrient composition and ${\beta}$-glucan content in Lentinula edodes fruiting bodies from different collection areas. Four types of free sugars were detected by HPLC, and the range of trehalose prevalence was 0.83% to 9%. The highest total amino acid content was observed from sawdust media cultivation of Lentinula edodes fruiting bodies, collected in China (JMI10050)The highest essential amino acid content, assessed by log cultivation of Lentinula edodes fruiting bodies, collected in Jangheung (JMI10059). Sixteen free amino acids were detected in Lentinula edodes fruiting bodies, and the major free amino acids were histidine, glutamic acid, and arginine. The highest essential amino acid including threonine, valine, methionine, isoleucine, leucine, phenylalanine, histidine, and lysine was fromsawdust media cultivation of Lentinula edodes fruiting bodies collected in China(JMI10052). The ${\beta}$-glucan content from log cultivation of Lentinula edodes fruiting bodies collected in Korea (JMI10059 and JMI10066) was higher than that from sawdust media cultivation of Lentinula edodes fruiting bodies collected in China. The highest ${\beta}$-glucan content was observed from log cultivation of Lentinula edodes fruiting bodies collected in Korea (JMI10066).

Growth Performance and Antibody Response of Broiler Chicks Fed Yeast Derived β-Glucan and Single-strain Probiotics

  • A study was conducted to evaluate the effects of dietary yeast derived ${\beta}$-glucan and single-strain probiotics on the growth performance and antibody response in broiler chicks. Six hundred and thirty 1-d-old male broiler chicks were divided into seven groups, placed into three pens per group (30 birds per pen) and fed one of seven non-medicated corn-SBM based experimental diets containing 0.025, 0.05 or 0.1% Saccharomyces cerevisiae ${\beta}$-glucan and 0.05, 0.1 or 0.2% Bacillus amyloliquefaciens (BA-pro, $1.3{\times}10^9/g$) or devoid of them for 5 wk. The body weight gains in groups fed diets containing 0.025 or 0.1% ${\beta}$-glucan, 0.1% or 0.2% BA-pro were significantly higher (p<0.05) than the control over 1-35 d. Feed conversion rates of groups fed ${\beta}$-glucan and BA-pro tended to be improved compared to the control group. There were no significant differences in the relative weights of liver, abdominal fat and breast muscle. No significant differences were observed in the activities of serum enzymes and concentrations of various cholesterol fractions. The antibody titers against Newcastle disease or infectious bronchitis virus in the chicks fed diets containing ${\beta}$-glucan and BA-pro were significantly higher (p<0.05) than in the control. The concentrations of cecal lactic acid bacteria in all groups fed BA-pro were significantly increased (p<0.05) compared to the control. These results indicated that dietary yeast derived ${\beta}$-glucan and BA-pro exerted growth-promoting and immune-enhancing effects in broiler chickens. In addition, BA-pro added to the diets modulated the profiles of cecal microflora, reflecting a potential to be beneficial microorganisms in chickens.

The Improvement Effects of β-Glucan on Adiposity and Serum Lipids Levels in High Fat Diet-Induced Obese Rats (베타-글루칸의 고지방 식이 유도 비만쥐에서 체지방 및 혈청지질 개선효과)

  • This study was aimed to investigate the effect of dietary ${\beta}$-glucan obtained from bacterial fermentation on the adiposity and serum lipids level in rats. Sprague-Dawley rats fed high fat diet for 6 weeks to induce obesity, and subsequently fed with 0% (high fat control group), 0.1% or 0.5% ${\beta}$-glucan supplemented high-fat diets (w/w) for another 5 weeks. For comparison, normal control groups fed AIN-76A diet. Supplemented with 0.1% ${\beta}$-glucan resulted in a significant reduction of high-fat induced peritoneal fat and visceral fat development by 16%, 19%, and 28%, respectively(P<0.05). Serum free fatty acid levels were reduced(by 19%), whereas the HDL cholesterol level was increased(by 50%) by 0.1% dietary ${\beta}$-glucan(P<0.05). In conclusion, dietary ${\beta}$-glucan reduced adiposity and improved serum lipids in obese rats fed high fat diet. The present study suggest that ${\beta}$-glucan supplementation to the diet is beneficial in suppressing diet-induced obesity and dyslipidemia.

Effects of Beta-glucan from Coriolus versicolor on Scavenger Receptor B1 Expression and their Involvement of Dectin-1 and Casein Kinase 2

  • The mushroom Coriolusversicolor contains biologically active polysaccharides, most of which belong to the ${\beta}$ glucan group. Diverse physicochemical properties, due to different sources and isolated types of ${\beta}$-glucans, may induce distinct biological activities. Here, we examined the effects of ${\beta}$-glucan from Coriolusversicolor (CVG) on the scavenger receptor B1 (SR-B1) expression and the role of SR-B1 in CVG-induced phagocytosis regulation by using SR-B1-specific shRNA transfected cells. We also examined whether Dectin-1 and CK2 are involved in SR-B1 expression in CVG-treated cells. Our study results showed that CVG increased the SR-B1 expression via Dectin-1 and CK2 in macrophages. However, the inhibition of SR-B1 expression by shRNA did not completely eliminate the effect of CVG on the increase of phagocytosis suggesting that SR-B1 is not essential for CVG-stimulated phagocytosis. This study will contribute to identify CVG's mechanism of action and its use in the development of functional foods.

Method Validation for the Determination of Eleutherosides and β-Glucan in Acanthopanax koreanum (탐라오가피의 Eleutheroside B, E 및 β-Glucan 함량 분석 및 분석법 검증)

  • The aim of this study was to investigate the method validation for the determination of eleutherosides (B and E) and ${\beta}$-glucan in Acanthopanax (A.) koreanum. This medicinal plant reportedly mainly included eleutherosides which exhibit the pharmacological effects, and ${\beta}$-glucan substantially enhances the function of the immune system by activating macrophages. The specificity, linearity, precision, accuracy, limit of detection (LOD, S/N=3), and limit of quantification (LOQ, S/N=10) were measured by HPLC and enzymatic methods. Our results showed that the coefficient of calibration correlation ($R^2$) for eleutheroside B and E were 0.9997 and 0.9999, respectively. The limits of detection (LOD) for eleutheroside B and E were $0.050{\mu}g/mL$ and $0.025{\mu}g/mL$, respectively. The recovery rate of eleutheroside B and E were revealed in the high range of 100.66~110.04% and 94.26~111.62%, respectively. The inter-day precision of eleutheroside B and E in the root and stem in A. koreanum were 1.4~5.0% and 1.1~2.5%, respectively. The intra-day precision of eleutheroside B and E in the root and stem in A. koreanum were 2.8~2.9% and 0.4~1.1%, respectively. Furthermore, the inter-day and intra-day precision of ${\beta}$-glucan in the stem, leaf, and fruit of A. koreanum were 1.32~5.67% and 8.01~11.76%, respectively. In conclusion, the methods were validated for the detection of eleutherosides and ${\beta}$-glucan in A. koreanum.
