Ultrasonic relaxation associated with the complex formation of benzoic acid derivative and β-cyclodextrin (벤조산 유도체와 베타 사이클로덱스트린의 복합체 형성반응에 의한 초음파 완화)

  • The dynamic interactions between benzoic acid derivative ($pH{\approx}7.0$)(guest) and ${\beta}$-cyclodextrin (${\beta}$-CD)(host) were investigated in an aqueous solutions in terms of ultrasonic absorption in the frequency range 0.2 MHz ~ 50 MHz with emphasis on the low-frequency range below 1 MHz at $25^{\circ}C$. We show that the interaction of ${\beta}$-CD and benzoic acid derivative complies with a typical spectrum of a single relaxation process around a few MHz. The ultrasonic relaxation observed in these solutions was due to a perturbation of a chemical equilibrium related to a reaction of an inclusion complex formed by the host and guest. The rate constant ($k_b=7.48{\times}10^6M^{-1}s^{-1}$) and equilibrium constant ($K=68.6M^{-1}$) were determined from the concentration dependences of benzoic acid on the relaxation frequency. The standard volume change (${\Delta}V=10.6{\times}10^{-6}m^3mol^{-1}$) of the reaction was also computed from the maximum absorption per wavelength. It was found that the hydrophobicity of guest molecules played an important role in the formation of the inclusion complex.

  • The Synthesis of azobenzene containing long chain fatty acid and poly vinyl alcohol by esterification reaction($C_{n}-Azo-PVA$) was optimized, starting from P-(P'-hydroxy phenyl azo)-benzoic acid and the product of reaction containing azobenzene chromophores was investigated by ultraviolet spectrophotometery in toluene solvent at room temperature. In addition, UV absorption spectra of Langmmuir Blodggett (LB) film deposited on quartz plate have been measured and the structure of these compounds were ascertained by means of Ultraviolet and FT-IR. Recrystallization of reaction product in the solvent results the experimental yield obtained about 22.27% P-(P'-octadecyloxy phenyl azo)-benzoic acid-poly vinyl alcohol. Long chain azobenzene derivative-poly vinyl alcohols are induced phtoisomerization by u, v, and visible light irradiation. The LB film of azobenzene containing long chain fatty acids($C_{18}-Azo-PVA$) are possible of being applied to functional molecular devices such as photomemory and light switching.

  • The chloroform extract of Pulicaria gnaphalodes yielded flavonoids (1-2), sesquiterpenoid (3), sterol (4), benzoic acid derivative (5) and fatty acids (6-7). Their structures were elucidated with the aid of NMR spectroscopy. The described compounds have not been reported so far from this source.

Chemical constituents from the culture filtrate of a Himalayan soil fungus, Preussia sp. and their anti-inflammatory activity (히말라야의 토양 곰팡이, Preussia sp. 배양액으로부터 추출된 화학 성분들 및 항 염증 활성)

  • A new naturally occurring benzoic acid derivative, benzyl 2,4-di(benzyloxy)benzoate (1) and six known compounds (2-7) were isolated from the fungus, Preussia sp. found in frozen soil of the Himalaya Mountain. The structures of the new compound, together with the known compounds were determined by 1D-and 2D-NMR experiments, as well as comparison with published values. In addition, to the best of our knowledge, the known compounds 2-7 were isolated for the first time from the genus Preussia and the family Sporormiaceae. The isolates were evaluated for cancer chemopreventive potential based on their ability to inhibit nitric oxide (NO) production induced by lipopolysaccharide (LPS) in mouse macrophage RAW 264.7 cells in vitro. Compounds 1 and 2 inhibited NO production by 50.7% and 88.5% at a concentration of 100 mg/ml, respectively.

  • Gentisic acid (GA), a benzoic acid derivative present in various food ingredients, has been shown to have diverse pharmaceutical activities such as anti-carcinogenic, antioxidant, and hepatoprotective effects. In this study, we used a co-culture system to investigate the mechanisms of the anti-inflammatory and anti-adipogenic effects of GA on macrophages and adipocytes, respectively, as well as its effect on obesity-related chronic inflammation. We found that GA effectively suppressed lipopolysaccharide-stimulated inflammatory responses by controlling the production of nitric oxide and pro-inflammatory cytokines and modulating inflammation-related protein pathways. GA treatment also inhibited lipid accumulation in adipocytes by modulating the expression of major adipogenic transcription factors and their upstream protein pathways. Furthermore, in the macrophage-adipocyte co-culture system, GA decreased the production of obesity-related cytokines. These results indicate that GA possesses effective anti-inflammatory and anti-adipogenic activities and may be used in developing treatments for the management of obesity-related chronic inflammatory diseases.

Development of a Single Chain Antibody Using a Phage Display Cloning Method for the Detection of 2,4-Dinitrotoluene

  • Single-chain variable fragments of antibodies (scFv) specific to 2,4-dinitrotoluene (DNT) were isolated from a phage library displaying synthetic human scFv fragments with 6 diversified complementary determining regions (CDRs). A DNT derivative that contained an extended amine group was synthesized and conjugated to the NHS-group that was linked to magnetic beads. Phages specific to the immobilized DNT derivatives were isolated from the library after 4 rounds of sequential binding and elution processes. The displayed scFv fragments from the isolated phages showed consensus CDR sequences. One DNT-specific scFv was expressed in E. coli and purified using Ni-affinity chromatography. The purified DNT-specific scFv binds specifically to the immobilized DNT-derivative with $K_D$ value of $6.0{\times}10^{-7}$ M. The scFv and DNT interaction was not disrupted by the addition of 4-nitrotoluene or benzoic acid. These data demonstrate that the screened scFv from the phage displayed library could be used for selective and sensitive detection of explosives such as TNT.

Preparation of Benzoyloxy Benzophenone Derivatives and Their Inhibitory Effects of ICAM-1 Expression

  • Benzoyloxy benzophenone derivatives were prepared in 3 steps including DCC coupling, Fries rearrangement and esterification from benzoic acids in 24-89% total yields. Among the prepared 12 benzophenone analogues 1a-1l, the compound 1b having three chloro groups at the para position showed maximum inhibitory effects of ICAM-1 expression but, 1a which have no substituents at all showed no inhibitory activity. This study provides the evidences that benzoyloxy benzophenone derivative, 1b may exert its anti-inflammatory activity by suppressing IFN-${\gamma}$-induced ICAM-1 expression.

  • A new azocalix[4]arene, 5,14,17,23-tetra[(2-benzoic acid)(azo)phenyl] calix[4]arene (2), has been prepared by hydrolysis of its ester derivative and characterized by 1H NMR, IR, UV-Vis spectroscopy and elemental analysis. Based on UV spectral changes, we found that 2 exhibits Pb2+ ion selectivity. In basic media, Pb2+ forms a 1:1 complex with the ligand. Beer's law is obeyed in the range of 2.0 × 10?6 ? 2.4 × 10?5 mol L?1 of Pb2+. The molar absorptivity (ε ) of 2-Pb2+ complex is 1.89 × 104 L mol?1 cm?1 at 440 nm, and the detection limit is 1.6 × 10?6 mol L?1.

Isolation of secondary metabolites from an Arctic bacterium, Pseudomonas aeruginosa and their antimicrobial activities (북극유래 박테리아, Pseudomonas aeruginosa로 부터 대사산물들의 분리 및 항진균 활성)

  • Chemical study of an Arctic bacterium, Pseudomonas aeruginosa (Pseudomonadaceae) led to the isolation of two diketopiperazines 1 and 2, two phenazine alkaloids 3 and 4, and an indole carbaldehyde 5, along with a benzoic acid derivative 6. The structures of the compounds were confirmed by 1D and 2D NMR, and MS experiments, as well as by comparison of their data with published values. Among the isolates, compounds 5 and 6 were isolated for the first time from P. aeruginosa of the seawater of Arctic Chuckchi Sea. Antimicrobial activities of compounds 1‒6 against a Staphylococcus aureus and Candida albicans were evaluated.
