Studies on the physicochemical and biochemical characteristics in sesame seed juice under different roasting conditions (참깨의 볶음 조건이 참깨 착즙액의 이화학적 및 생화학적 특성에 미치는 영향)

  • In this study, we investigated the effect of roasting temperature on nutrient content, digestive enzyme activities, and antioxidative properties of sesame seed juice. The sesame seeds were either roasted at 160, 200, and $240^{\circ}C$ or not roasted, and the juice was extracted using a low-speed juice extractor. Owing to the short duration of roasting, benzo[a]pyrene were not detected and trans fatty acids were negligible detected in all sesame seed juices. The sesame seed juice contained abundant nutrients such as minerals, vitamins, and fatty acids. The contents of minerals, vitamin B1 and B3, and sesamol increased with increase in roasting temperature; however, the levels of fatty acids, vitamin B2, sesamin, and sesamolin decreased. In addition, the antioxidant content and antioxidative activities of sesame seed juice increased with increase in roasting temperature. Therefore, these results suggest that roasted sesame seed juice possesses high antioxidative activities, which may be beneficial for preventing oxidative damage in the body.

Seasonal Concentration of Polycyclic Aromatic Hydrocarbons (PAHs) in Residential Areas Around Petrochemical Complexes and Risk Assessment Using Monte-Carlo Simulation (석유화학단지 주변 주거지역 다환방향족탄화수소(PAHs)의 농도와 Monte-Carlo 모의실험을 통한 위해성평가)

  • Background: Polycyclic aromatic hydrocarbons (PAHs) are generated in petrochemical complexes, can spread to residential areas and affect the health of residents. Although harmful PAHs are mainly present in particle phase, gas phase PAHs can generate stronger toxic substances through photochemical reaction. Therefore, the risk assessment for PAHs around the petrochemical complex should consider both particle and gas phase concentrations. Objectives: This study aimed to investigate the concentration characteristics of particle and gas phase PAHs by season in residential areas around petrochemical complexes, and to assess the risk of PAHs. Methods: Samples were collected for 7 days by seasons in 2014~2015 using a high volume air sampler. Particle and gas phase PAHs were sampled using quartz filter and polyurethane foam, respectively, analyzed by GC-MS. Chronic toxicity and probabilistic risk assessment were performed on 14 PAHs. For chronic toxicity risk assessment, inhalation unit risk was used. Monte-Carlo simulation was performed for probabilistic risk assessment using the mean and standard deviation of measured PAHs. Results: The concentration of particle total PAHs was highest in autumn. The gas phase concentration was highest in autumn. The average gas phase distribution ratio of low molecular weight PAHs composed of 2~3 benzene rings was 85%. The average of the medium molecular weight composed of 4 benzene rings was 53%, and the average of the high molecular weight composed of 5 or more benzene rings was 9%. In the chronic toxicity risk assessment, 7 of the 14 PAHs exceeded the excess carcinogenic risk of 1.00×10-6. In the Monte-Carlo simulation, Benzo[a]pyrene had the highest probability of exceeding 1.00×10-6, which was 100%. Conclusions: The concentration of PAHs in the residential area around the petrochemical complex exceeded the standard, and the excess carcinogenic risk was evaluated to be high. Therefore, it is necessary to manage the air environment around the petrochemical complex.

Atmospheric concentration and mutagenicity of organic pollutants of suspended particulate in Seoul (서울시 대기중 유기오염물질의 농도와 돌연변이원성에 대한 연구)

  • To evaluate the difference of concentration and mutagenicity of organic pollutants between residential and traffic area of Seoul, air samples were collected in Bulkwang (residential) and Shinchon (traffic) area. Samples were analyzed to measure the concentration of extractable organic matters (EOM) and their subfractions and mutagenicities were tested using Salmonella typhimurium TA 98. The concentrations of polycyclic aromatic hydrocarbons (PAHs) were also measured by gas-chromatography and compared between two areas. The results were as follows ; 1. While the concentration of total suspended particulate (TSP) in residential area was below the environmental standard in annual average, the concentration in traffic area was above the standard and was up to its maximum $256{\mu}g/m^3$ in November. The difference of TSP concentrations in both areas of each month was statistically significant (P<0.05). 2. The concentration of fine particle in traffic area was significantly higher compare to that in residential area and showed statistically significant monthly difference in both areas (P<0.05). The proportion of concentration of fine particle to TSP was 55-68%. 3. Mean concentrations of EOM in residential and traffic areas were $4.3{\mu}g/m^3\;and\;5.3{\mu}g/m^3$ respectively. The proportion of amount of EOM from fine particle to EOM from TSP was 70-88%. 4. While the percentage of polar neutral organic compounds (POCN) of fine particle in Bulkwang's sample was higher compare to Shinchon's sample, the percentage of aliphatic compounds of fine particle in Shinchon's sample was higher compare to Bulkwang's sample. The percentages of PAH fraction were as low as 6-10% in both areas. 5. The mutagenic activity of nit concentration of organic matters extracted from fine particle was higher compare to that of coarse particle and was increased when metabolically activated with S9. Mutagenicities with metabolic activation calculated by unit air volume were significantly different between residential and traffic area, $17\;revertants/m^3$\;and\;22\;revertants/m^3$ respectively. 6. The concentrations of benzo(a)pyrene in fine particle of traffic and residential areas were $3.10ng/m^3\;and\;2.02ng/m^3$ respectively. Sixteen PAHs were higher in samples of traffic area compare to residential area and also concentrations of PAHs in fine particle were higher compare to coarse particle.

Distribution of Persistent Organic Pollutants (POPs) in Different Sizes of Particles in the Ambient Air of the Pyeongteak Area (평택지역 대기 중 먼지 입경별 잔류성유기오염물질 분포특성 연구)

  • Objectives: The concentration distributions of polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs), dioxin-like polychlorinated biphenlys (dl-PCBs), and polycyclic aromatic hydrocarbons (PAHs) in fine particles were investigated to provide basic data on POP behavior and composition analysis. Methods: The concentrations of PCDD/Fs, dl-PCBs, and PAHs by particle size were evaluated for TSP, PM10, and PM2.5. Also, fine dust component analysis and factor analysis were performed to identify the source of PCDD/Fs. Results: The particle size distribution was found to account for 24.3% of >10 ㎛, 14.5% of 2.5-10 ㎛, and 61.2% of <2.5 ㎛. The average contributions of coarse particles (>2.5 ㎛) and fine particles (<2.5 ㎛) were PCDD/Fs 67%, dl-PCBs 66%, benzo (a) pyrene 83% and PAHs 84%, and the contributions of fine particles (<2.5 ㎛) were higher than coarse particles (>2.5 ㎛). However, the contributions of coarse particles increased in April to September with higher temperatures, while those of fine particles increased in February to March with lower temperatures. Conclusions: Low chlorinated (4Cl-5Cl) PCDD/Fs were more adsorbed compared to coarse particles due to the influence of pollutant migration from particulate to gas phase according to temperature rise, whereas high chlorinated (6Cl-8Cl) PCDD/Fs were more adsorbed compared to fine particles. PCDD/Fs sources were assessed to be major sources of emissions, such as incineration facilities and/or open burning.

The Antimutagenic and Anticancer Effect of Taxus cuspidata Extracts (주목 추출물의 발암 억제효과 및 암세포에 미치는 영향)

  • This study was Performed to determine the effects of antimutagenicity and anticancer of Taxus cuspidata produced in Korea. Extracts of Taxus cuspidata were obtained from leaves, barks and roots. All the samples tested had no effects on the mutagenicity by Ames test using Salmonella typhimurium TA98 and TA100 or rec-assay using Bacillus subtilis Hl7 and M45 strains. However the treatment of 50$\mu\textrm{g}$/plate of Taxus cuspidata extracts showed strong antimutagenicity with 98% inhibition against TA100 induced by MNNG and with 98% inhibition against TA98 and TA100 induced by 4NQO whereas 73~89% and 16~60% antimutagenic effect were shown against both strain induced by Trp-P-1 and $Benzo(\alpha)pyrene,$ respectively. The treatment of $0.5$\mu\textrm{g}$/{\mu}\ell$ root extracts had the highest cytotoxicity with 90% against liver cancer cell, Hep 3B, followed by bark extracts(87%) and leave extracts(72%), whereas $0.5$\mu\textrm{g}$/{\mu}\ell$ treatment of Taxus cuspidata extract had only 22~36% cytotoxicity on human normal liver cell WRL 68.

Influence of Roasting Conditions on Polycyclic Aromatic Hydrocarbon Contents in Ground Coffee Bean (원두커피의 로스팅 조건이 polycyclic aromatic hydrocarbons 생성에 미치는 영향)

  • Roasting may lead to the formation of undesired compounds, such as polycyclic aromatic hydrocarbons (PAHs). In this study, green coffee beans were roasted under controlled conditions and the formation of PAHs during the roasting process was monitored. Roasting was performed in a hot air roaster, with an inlet air temperature varying from 150 to $250^{\circ}C$ for 5, 10, and 20 min. The PAH content of the roasted coffee was then evaluated by HPLC-FLD. The levels of total PAHs in Arabica (Colombia, Brazil) and Robusta (India) coffee samples were 1.26-215.07, 1.85-178.14, and 0.18-2.61 ${\mu}g$/kg, respectively.

Distribution and Human Risk Assessment of Polycyclic Aromatic Hydrocarbons and Polychlorinated Biphenyls in Sediments and Fish from Suyoung Estuary of Busan, Korea (부산 수영강 하구역의 퇴적물과 어류에서 다환방향족탄화수소 및 폴리염화비페닐의 농도분포와 인체 위해도 평가)

  • Distributions of 15 PAHs and 82 PCBs were investigated in sediment and fish samples collected from the Suyoung Estuary of Busan, one of the most urbanized and newly contaminated areas in Busan. The concentrations of $S_{15}PAH$ in sediments ranged from 17.9 to 777 ng/g-dry and were mainly originated from combustion processes. The concentrations of $S_{82}PCB$ in sediments ranged from 0.55 to 12.5 ng/g-dry and were significantly correlated with those of 12 dioxin-like PCBs and 6 non-dioxin like PCBs. Higher concentrations of PAHs and PCBs were found in the upper river and the inner Suyoung Bay than in marina and the outer Suyoung Bay. No sites exceeded the ecotoxicological values of PAHs and PCBs in marine sediments. Benzo[a]-pyrene was not detected in fish samples and the concentrations of $S_4PAH$ (0.15~0.45 ng/g-wet) were lower than the maximum level set by EU. The levels of PCBs in fish samples were at less than 1% of the Korean maximum level and 2% of the EU maximum level. The concentrations of dioxin-like PCBs were $0.01{\sim}0.32pg-TEQ_{WHO-2005}/g-wet$, and the highest concentration was found in eels, which were at less than 5% of the EU maximum level. Dietary intakes of PAHs and PCBs through fish consumption were estimated, and their lifetime cancer risk and non-cancer risk were much lower.

Risk Analysis and Safety Assessment of Microbiological and Chemical Hazards in the Raw Short-Neck Clams Ruditapes philippinarum Distributed in the Yeongnam and Honam Area During the Spring Season (봄철 영·호남 지역에서 유통되는 생바지락(Ruditapes philippinarum)의 미생물학적·화학적 위해요소 분석 및 안전성 평가)

  • For the safety assessment of microbiological and chemical hazards in raw short-neck clam Ruditapes philippinarum distributed in the Yeongnam and Honam areas during the spring season, the contamination levels of total viable bacteria, coliforms, Escherichia coli, and nine pathogenic bacteria (Staphylococcus aureus, Salmonella spp., Vibrio parahaemolyticus, Clostridium perfringens, Listeria monocytogenes, enterohemorrhagic Escherichia coli, Yersinia enterocolitica, Bacillus cereus, Campylobacter jejuni) as microbiological hazards, and heavy metals (lead, cadmium, total mercury), benzopyrene, shellfish poison (paralytic, diarrhetic, amnesic), and radioactivity (131I, 134Cs+137Cs) were also analyzed in 15 samples based on the methods of the Korean Food Code. The average contamination levels of total viable bacteria were 3.11 (1.40-4.49) log CFU/g, and coliforms were detected in 5 out of 15 samples (1.18-1.85 log CFU/g). E. coli and S. aureus were not detected in all samples. Furthermore, the presence of 8 pathogens were not detected in all samples. The average contamination levels of lead, cadmium, and total mercury were 0.155 (0.079-0.264), 0.160 (0.040-0.287), and 0.017 (0.008-0.026) mg/kg, respectively. Benzo(a)pyrene, shellfish poison, and radioactivity were not detected in all samples. The results of this study suggest that the safety against all microbiological and chemical hazard factors in raw short-neck clams distributed in markets has been assured.

  • Background : Lung cancer is frequently cited as an example of a disease caused solely by exposure to environmental carcinogens. However, there is a growing realization that the genetic constitution is also important in determining individual's susceptibility to lung cancer. This genetic susceptibility may result from functional polymorphims of the genes involved in carcinogen metabolism. In this study, the association between GSTM1 and CYP1A1 polymorphisms and the lung cancer risk in Korean males was investigated. Materials and Method : The study population consisted of 153 male lung cancer patients and 143 healthy male controls. The GSTM1 and CYP1A1 genotypes were determined by multiplex PCR and PCR-RFLP analysis. Result : There were no significant differences in the frequency of the GSTM1 null genotype between the cases and the controls. When the cases were categorized by their histologic type, the frequency of the GSTM1 null genotype in the small cell carcinoma group was higher than those of the controls(67.2% vs 55.9%), but the difference was not statistically significant(OR=1.772 ; 95% CI=0.723-4.340). The distribution of the CYP1A1 MspI genotypes among the cases were similar to those among the controls. When the cases were grouped by their histologic type, the ml/m1, ml/m2, m2/m2 genotypes frequencies among the small cell carcinomas(23.0%, 38.5%, and 38.5%, respectively) were significantly different from those of the controls(36.4%, 46.2%, and 7.4%, respectively, p<0.05). When the m1/m1 genotype was used as a reference, the ml/m2 and m2/m2 genotypes were associated with an increased risk for small cell lung cancer(ml/m2 genotype : OR=1.337, 95% CI=0.453-3.947 ; m2/m2 genotype : OR=3.374, 95% CI=1.092-10.421). Conclusion : These results suggest that the GSTM1 and CYP1A1 genotypes may be a genetic determinant of the risk for lung cancer, particlulary small cell carcinoma. Further investigation is needed to confirm these results.

Antimutagenic and Cytotoxic Effects of Extracts of Kalopanax pictus NAKAI Endodermis (음나무 내피 추출물의 항돌연변이원성 및 세포독성 효과)

  • This study was performed to determine the antimutagenic and cytotoxic effect of ,Kalopanacis cortex endothelium. Methanol extract was used on Salmonella typhimurium TA98, TA100 and three cancer cell lines. In the Ames test, methanol extract of Kalopanacis cortex endothelium alone did not exhibit any mutagenicity but showed substantial inhibitory effects against mutation induced by 4-nitroquinoline-l-oxide(4NQO), N'-methyl- N'-nitro-N-nitrosoguanidine(MNNG) and benzo(a)pyrene(B(a)P). The methanol extract of Kalopanacis cortex endothelium showed approximately 79.29% and 75.38% inhibitory effect on the mutagenesis induced by 4NQO and B(a)P, respectively, against TA98 strain. Whereas 79.49%, 89.3% and 68.85% inhibitions were observed on the mutagenesis induced by 4NQO, MNNG and B(a)P against TA100 strain. Methanol extracts from Kalopanacis cortex endothelium showed high antimutagenic effects against 4NQO, MNNG and B(a)P. The anticancer effects of methanol extract from Kalopanacis cortex endothelium against human lung carcinoma(A549), human breast adenocarcinoma (MCF-7) and human hepatocellular carcinoma(Hep3B) were investigated. The treatment of 0.5mg/ml Kalopanacis cortex endothelium methanol extracts had the highest cytotoxicity against A549(81.54%), followed by MCF-7(81.92%) and Hep3B (78.57%). In contrast 0.5mg/ml treatment of methanol extracts from Kalopanacis cortex endothelium had only $4{\sim}25%$ cytotoxicity on normal human liver cell(293).
