IFIT1 Expression Patterns Induced by H9N2 Virus and Inactivated Viral Particle in Human Umbilical Vein Endothelial Cells and Bronchus Epithelial Cells

  • IFIT1 (also known as ISG56) is a member of the interferon-inducible protein with tetratricopeptide repeats (IFITs) family. IFITs are strongly induced by type I interferon (IFN), double-stranded RNA and virus infection. Here, we investigated IFIT1 expression in human umbilical vein endothelial cells (HUVECs) and in human bronchus epithelial cells (BEAS-2Bs) induced by the H9N2 virus and inactivated viral particle at different time points. We also investigated the effect of H9N2 virus and viral particle infection on $IFN-{\alpha}/{\beta}$ production, and assessed whether hemagglutinin or neuraminidase protein induced IFIT1 expression. Results showed that both H9N2 virus infection and viral particle inoculation induced the expression of IFIT1 at mRNA and protein levels in the two cell lines. Hemagglutinin or neuraminidase protein binding alone is not sufficient to induce IFIT1 expression. Surprisingly, the expression patterns of IFIT1 in response to H9N2 virus and viral particles in the two cell lines were opposite, and production kinetics of $IFN-{\alpha}/{\beta}$ also differed. An additional finding was that induction of IFIT1 in response to H9N2 virus infection or viral particle inoculation was more sensitive in HUVECs than in BEAS-2Bs. Our data offers new insight into the innate immune response of endothelial cells to H9N2 virus infection.

  • This study demonstrated the stabilizing role of a cyclodextrin on Keggin $[PW_{12}O_{40}]^{3-}$ via hydrogen bonding complexation unveiled by ESI-MS. The distinctive fragmentation pathways of the $\{PW_{12}\}/{\gamma}$-CD complexes from that of discrete $[PW_{12}O_{40}]^{3-}$ showed that the so-called "weak" non-covalent interactions can effectively change the dissociation chemistry of POM in the gas phase. The influence of different types of solvents and organic additives such as ${\gamma}$-CD on the stability of Keggin $[PW_{12}O_{40}]^{3-}$ was also addressed firstly by ESI-MS.

Fusion Between TMPRSS2 and ETS Family Members (ERG, ETV1, ETV4) in Prostate Cancers from Northern China

  • In this study we evaluated the frequency of fusion between TMPRSS2 and ETS family members (ERG, ETV1, ETV4) in prostate cancers in patients from northern China in order to explore differences in fusion rates among regions in northern and southern China, other parts of Asia, Europe, and North America. We examined 100 prostate cancer patients, diagnosed by means of prostate biopsy; fluorescence in situ hybridization (FISH) was used to detect the expression of TMPRSS2, ERG, ETV1 and ETV4 in cancer tissue. Differences in gene fusion rates among different ethnics groups were also analyzed. Of the 100 prostate cancer patients, 55 (55%) had the fusion gene. Among the patients with the fusion gene, 46 (83.6%) patients had the TMPRSS2:ERG fusion product, 8 (14.8%) patients had TMPRSS2:ETV1 fusion, 1 (1.6%) patient had TMPRSS2:ETV4.

  • Nucleolar and coiled-body phosphoprotein 1 (NOLC1) is a phosphoprotein that transiently associates with the mature nucleolar H/ACA and C/D box small nucleolar ribonucleoproteins (snoRNPs). Several lines of evidence indicate that NOLC1 plays an important role in the synthesis of rRNA and the biosynthesis of ribosomes. In the present study, we examined the transcriptional regulation mechanisms that govern the expression of NOLC1. We first performed functional dissection of the NOLC1 promoter. We demonstrated that transcription factors NF-${\kappa}B$ and CREB could bind to the minimal NOLC1 promoter. This was demonstrated by electrophoretic mobility shift assays and chromatin immunoprecipitation. Mutagenesis and overexpression assays revealed that NF-${\kappa}B$ and CREB positively regulated the NOLC1 promoter. These findings may provide new insight into the mechanisms that regulate NOLC1 expression.

Fast Quadtree Structure Decision for HEVC Intra Coding Using Histogram Statistics

  • The final draft of the latest video coding standard, High Efficiency Video Coding (HEVC), was approved in January 2013. The coding efficiency of HEVC surpasses its predecessor, H.264/MPEG-4 Advanced Video Coding (AVC), by using only half of the bitrate to encode the same sequence with similar quality. However, the complexity of HEVC is sharply increased compared to H.264/AVC. In this paper, a method is proposed to decrease the complexity of intra coding in HEVC. Early pruning and an early splitting strategy are applied to the quadtree structure of coding tree units (CTU) and residual quadtree (RQT). According to our experiment, when our method is applied to sequences from Class A to Class E, the coding time is decreased by 44% at the cost of a 1.08% Bjontegaard delta rate (BD-rate) increase on average.

Using Genetic Algorithm for Optimal Security Hardening in Risk Flow Attack Graph

  • Network environment has been under constant threat from both malicious attackers and inherent vulnerabilities of network infrastructure. Existence of such threats calls for exhaustive vulnerability analyzing to guarantee a secure system. However, due to the diversity of security hazards, analysts have to select from massive alternative hardening strategies, which is laborious and time-consuming. In this paper, we develop an approach to seek for possible hardening strategies and prioritize them to help security analysts to handle the optimal ones. In particular, we apply a Risk Flow Attack Graph (RFAG) to represent network situation and attack scenarios, and analyze them to measure network risk. We also employ a multi-objective genetic algorithm to infer the priority of hardening strategies automatically. Finally, we present some numerical results to show the performance of prioritizing strategies by network risk and hardening cost and illustrate the application of optimal hardening strategy set in typical cases. Our novel approach provides a promising new direction for network and vulnerability analysis to take proper precautions to reduce network risk.

Effect of Deformation Temperature on Crystal Texture Formation in Hot Deformed Nanocrystalline SmCo5 Permanent Magnets

  • In the present study, bulk anisotropic nanocrystalline $SmCo_5$ magnets were prepared by hot deformation. The effect of deformation temperature on the texture and magnetic properties are presented, based on which the mechanism of plastic deformation and texture formation during the hot deformation process is discussed. Our analyses reveal that deformation temperature is one of the most important parameters that determine the texture of $SmCo_5$ grains. We suggest that diffusion creep plastic deformation occurs during hot deformation, which is very sensitive to the energy gain provided by an increase in temperature.

Pounding analysis of RC bridge considering spatial variability of ground motion

  • To investigate the seismic pounding response of long-span bridges with high-piers under strong ground motions, shaking table tests were performed on a 1/10-scaled bridge model consisting of three continuous spans with rigid frames and one simply-supported span. The seismic pounding responses of this bridge model under different earthquake excitations including the uniform excitation and the traveling wave excitations were experimentally studied. The influence of dampers to the seismic pounding effects at the expansion joints was analyzed through nonlinear dynamic analyses in this research. The seismic pounding effects obtained from numerical analyses of the bridge model are in favorable agreement with the experimental results. Seismic pounding effect of bridge superstructures is dependent on the structural dynamic properties of the adjacent spans and characteristics of ground motions. Moreover, supplemental damping can effectively mitigate pounding effects of the bridge superstructures, and reduce the base shear forces of the bridge piers.

  • The study utilizes remote sensing as the main monitoring means. With different spatial high-resolution, multichannel ASTER remote sensing image as the main information in Beijing city zone; with regional border and statistical data as auxiliary factor a study between the thermal space distribution character and the underground medium is analyzed based on the GIS logical algorithm and synthetic analysis technology. Results show thermal forming mechanism and the rule of distribution is mainly related to the underground medium and the change of the city distribution. Different underground medium has different degree and intensity influence on the thermal space distribution. Furthermore, urban greenbelt and water areas can reduce the thermal effect and large-scale greenbelt creates green island effect. In addition, Road net, residential area, population density, heat resources and so on have some positive effect on the thermal distribution, which increase the local temperature and intensity on the other hand. It is important to study the thermal distribution and its related factors, which contributes to the plan, construction and development of the city.

  • Direct implantation of metallic ion species has been employed in surface processing of industrial components and tools with very encouraging improvements in recent years. In spite of high technicla effectiveness, this new surface processing technique has not been extensively accepted by industries mainly because of high cost(capital and operating) compared with other competitive surface processing techniques. High current and large implantation area with eliminating the mass analyzer and the beam-scanning unit make metal vapor vacuum are(MEVVA)source ion implantation versatile, simple and cheap to operate and well suited to commercial surface processing. In this paper, the recent development of MEVVA source ion implantation technique ar Beijing Normal University has been reviewed and the results of production trials of several industrial components and tools implanted by MEVVA source ion implantation have been presented and discussed.
