Objectives : This study was conducted to carry out Purification of Melittin and other peptide components from Bee Venom using gel filtration chromatography and propionic acid/urea polyacrylamide gel electrophoresis Methods : Melittin and other peptide components were separated from bee venom by using gel filtration chromatography on Sephadex G-50 column in 0.05M ammonium acetate buffer. Results : Melittin and other peptide components were separated from bee venom by using gel filtration chromatography on Sephadex G-50 column in 0.05M ammonium acetate buffer. The fractions obtained from gel filtration chromatography was analyzed by using SDS-PAGE and propionic acid/urea polyacrylamide gel electrophoresis. The melittin obtained from the gel filtration contained residual amount of phospholipase $A_2$ and a protein with molecular weight of 6,000. The contaminating proteins were removed by the second gel filtration chromatography. Conclusion : Gel filtration chromatography and propionic acid/urea polyacrylamide gel electrophoresis are useful to separate peptide components including melittin from bee venom.
Bee Venom acupuncture has anti-inflammatory and analgesic effect that facilitates natural healing. So it is used in treatment of several musculoskeletal diseases. Recently, we experienced a case of De Quervain's disease that was not responded to steroid injection. It was successfully treated with intra-synovial Bee Venom acupuncture for two weeks. It is thought that Bee Venom acupuncture can be used in tenosynovitis. And Bee Venom injection techniques like intra-joint and subcutaneous method are need to be developed.
Mastitis is an inflammatory condition of the mammary gland, most often caused by bacterial infections, resulting in significant economic losses to the dairy industry. Antimicrobial resistance has been of great concern because of the extensive clinical use of antibiotics. For this reason, the development of new compounds as an alternative treatment to bovine mastitis is needed. Bee venom has been widely used as an oriental treatment for several inflammatory diseases and bacterial infections. The aim of the present study was to evaluate the antimicrobial activity of bee venom on bacteria isolated from bovine mastitis. A total of 107 isolates from bovine mastitic milk samples collected in 2019 and 2020 in Jeonbuk province. All bacterial isolates were tested for susceptibility to bee venom of the honey bee (Apis mellifera). In order to obtain comprehensive antibacterial activities of the bee venom, we measured the minimal inhibitory concentration (MIC) of the bee venom against bacterial strains. Bee venom showed significant inhibition of bacterial growth of Gram-negative bacteria Citrobacter spp., Escherchia coli, Klebsiella spp., Pseudomonas spp., Serratia spp. and Raoultella with MIC values of 96, 81, 72, 230, and 85 ㎍/mL, respectively, and Gram-positive bacterial Enterococcus spp., Staphylococcus spp. and Streptococcus spp. with MIC values of 29, 21 and 16 ㎍/mL, respectively. The results indicated that the MIC values were different depending on the bacterial strains, and those of Gram-positive bacteria were lower than those of Gram-negative bacteria for bee venom. These findings suggested that bee venom could be an effective antimicrobial treatment for bovine mastitis; however, further research is necessary to evaluate the mechanism underlying the antimicrobial action, its effectiveness/safety in vivo and effective application for therapeutic use.
The purpose of this study is to investigate the effects of Bee Venom Herb-Acupuncture on the jaw opening reflex evoked by tooth pulp stimulation. Rats were anesthetized with thiopental sodium given intraperitoneally in an initial dose of 80mg/kg. Maintenance doses of 5mg/kg thiopental sodium were given through a cannular in the femoral vein as required to maintain light anesthesia. To apply noxious stimuli, a pair of enameled wires were inserted into the tooth pulp of the lower incisor. The effects of conditioning stimuli were estimated as an indicator of the degree of suppression of the digastric muscle electromyogram(dEMG) in the jaw opening reflex. Bee Venom Herb- Acupuncture(0.2% solution 0.1ml/rat) was injected to Hapgok(LI4) loci. In addition, Normal Saline (0.1ml/rat) was injected to Hapgok loci so as to compare the degree of suppression elicited from Bee-Venom. By administration of Bee Venom Herb-Acupuncture, the amplitude of dEMG was maximally suppressed to $67.5{\pm}3.38%$ ipsilaterally, 73.33{\pm}8.00%$ contralaterally. Generally, the dEMG activities caused by electrical stimulation were gradually suppressed during the stimulation and maximal suppressive effect showed at 15min after its onset. However the dEMG activities by Be Venom Herb-Acupuncture were immediately suppressed after its onset and the suppressive effect continued for a long time compared to electrical stimulation. In conclusion, Bee Venom Herb-Acupuncture may have a different mechanism of analgesia from that of electro-acupuncture and contribute to the modulation of pain analgesia.
The therapeutic effect of honeybee venom collected using be venom collector on bovine mastitis was investigated. Mastitic cows from four farms were selected in the Yang-pyeong areas. Chronic mastitic cows were injected with the various concentrations of honey-bee venom per day. There was a significant difference in the reduction rates of somatic cell counts (SCC) according to treatment concentration and method of bee venom. The milk SCC were significantly decreased in all concentrations of bee venom 3 days after treatment. The reduction rates of SCC in treatment of 3, 6, 12 and 24mg honeybee venom were 20, 43, 63.3 and 65.8% respectively. Honeybee venom treatment consisted of two methods, a syringeful and a Bovivet Spenstift. The treatment with Bovivet Spenstift was more effective in the reduction rates of SCC compared with the syringeful. Thirty two out of 53 quarters were cured by Bovivet Spenstift with 12mg bee venom per day for 14 days. The venom cure rates of bovine mastitis by Escherichia coli, Stapylococcus aureus, Gram positive bacteria and Gram negative bacteria were 33.3, 75, 75 and 43.8% respectively. These results suggested that bee venom treatment (by Bovivet Spenstift with 12mg) might be effective for treatment of bovine mastitis.
Purpose: The purpose of this study is to analyze the clinical characteristics of anaphylaxis and anaphylactic shock caused by bee venom. Methods: We retrospectively collected the data of the patients who experienced anaphylaxis caused by natural bee sting or acupuncture using bee venom from January 1999 to December 2008. Seventy subjects were divided into the shock and non-shock groups. The clinical characteristics, sources of bee venom, treatments and outcomes were compared between the two groups. Results: The mean age of the subjects was $45.5{\pm}16.3$ years old and the number of males was 44 (62.9%). There were 25 patients in the shock group and 45 in the non-shock group. The age was older (p=0.001) and females (p=0.003) were more frequent in the shock group. Transportation to the hospital via ambulance was more frequent in the shock group (p<0.001). No difference was found in species of bee between the two groups. The cephalic area, including the face, was the most common area of bee venom in both groups. Anaphylaxis caused by bee sting commonly occurred between July and October. Cutaneous and respiratory symptoms were the most frequent symptoms related to anaphylaxis. Cardiovascular and neurologic symptoms were more frequent in the shock group. The amount of intravenously administered fluid and subcutaneous injection of epinephrine were much more in the shock group than that in the non-shock group. Conclusion: Older age was the factors related to anaphylactic shock caused by bee venom. Further validation is needed to evaluate the gender factor associated with shock.

Background: This study investigated current research trends in bee venom acupuncture in Korea. Materials and methods: The literature in Korea was reviewed using the search engines Science and Technology Society Village, Korean Studies Information Service System, National Discovery for Science Leaders, and Oriental Medicine Advanced Searching Integrated System. The keywords searched were "bee venom," "apitoxin," "apitherapy," and "bee sting." We selected 412 studies, which included Korean literary studies, experimental studies, and clinical studies. Results: We classified them by publication year, research type, disease and topic, and quality assessment. Among bee venom-related studies, clinical studies (235 studies) outnumbered experimental studies (149 studies) and literary studies (28 studies). Nineteen experimental studies concentrated on the theme of "anticancer." In the type analysis of clinical studies, lumbar disorders comprised 45 of 235 studies, followed by the effect on body. Forty-two randomized controlled trials (RCTs) have been published from 2003 to date (2015). Twenty-two studies used appropriate randomization methods. Fourteen studies had a Jadad score of 4-5 points; 15 studies, 1-3 points; and 13 studies, 0 points. Conclusions: Bee venom treatment is based on Korean traditional medicine theory. Numerous research studies suggest its effectiveness. Effort and academic approach on bee venom are expected to receive a positive evaluation.
