Production of Leptin in E. coli and Its Effect on Glucose and Acetate Transport and Expression of Uncoupling Protein-2 Gene in Adipose Tissues of Korean Cattle (Hanwoo)

  • Leptin has a major role in the regulation of food intake and energy homeostasis. In addition, leptin participates in many physiological functions including regulation of lipid metabolism. Bovine recombinant leptin protein was produced in E. coli cells in order to understand function of leptin in the regulation of lipid metabolism. The leptin expression vector was constructed in pGEX-4T-3 vector and transformed into E. coli BL21 cells. Expression of the GST-leptin fusion protein was induced with IPTG. The fusion protein was purified using glutathione sepharose 4B batch method, and the recombinant leptin was eluted after thrombin protease digestion. The effect of leptin on glucose transport was examined in the differentiated adipocytes of 3T3-L1 cells. Leptin had no effect on basal and insulin-stimulated glucose transport in 3T3-L1 cells (p>0.05). Effect of recombinant leptin on glucose and acetate transport was examined in adipose tissues of Korean cattle (Hanwoo). Insulin stimulated glucose transport in both intramuscular and subcutaneous adipose tissues (p<0.05), but leptin did not affect glucose transport in both adipose tissues (p>0.05). Insulin stimulated acetate transport in bovine adipose tissues (p<0.05), but leptin did not affect acetate transport (p>0.05). Northern and RT-PCR analyses showed that mRNA levels of uncoupling protein-2 were increased by leptin treatment in 3T3-L1 cells without statistical difference (p>0.05). In conclusion, bovine recombinant leptin did not affect glucose and acetate transport in both 3T3-L1 adipocytes and bovine adipose tissues, while it stimulates UCP-2 mRNA expression in 3T3-L1 cells.

Variation in Nutritive Value of Commercial Broiler Diets

  • The classical energy balance method was used to measure the apparent metabolisable energy (AME) of four batches of broiler starter and finisher diets produced by two commercial feed companies. The results showed there was little variation in protein content between batches, but NDF content varied from 13.3% to 15.5% between batches of diet. The batch variation in chemical composition differed between feed manufacturers. While there was no difference in AME and feed conversion ration (FCR) between batches of starter diets produced by company A, FCR and AME ranged from 1.76-1.94 (p<0.001) and 11.38-11.90 MJ/kg air dry (p<0.05), respectively, for diets produced by company B. Similar results were found in a second experiment. There was no difference in AME, dry matter digestibility (DMD) and FCR between batches for finishing diet produced by company B, but a large variation occurred for the finisher diets from company A (p<0.01), where the ranges of FCR, AME and DMD were 1.95-2.30, 10.5-12.3 (MJ/kg air dry) and 58-68%, respectively. FCR was correlated with AME. AME was negatively related to the content of fibre in the diet, but positively related to DMD. The preliminary results based on 24 samples showed that near infrared spectroscopy (NIR) has the potential to predict FCR, intake, AME and DMD of commercial broiler diets, with $R^2$ being 0.93, 0.89, 0.95 and 0.98, respectively. The standard error of cross validation was below 0.2 for AME and only 0.06 for FCR.

Improved Refolding of Recombinant Human Proinsulin from Escherichia coli in a Two-stage Reactor System

  • An improved method of refolding recombinant human proinsulin from E. coli was presented. It was based on a two-stage stirred tank reactor in which denatured proinsulin-s-sulfonate was mixed instantaneously with a reaction buffer in the first stage reactor, and then fed to the second stage reactor. The mixture was stirred further for a total of 30h in the second stage reactor. In this system, unfavorable effects present due to the increase in reaction volume and protein concentration for protein refolding, which becomes significant in a large-scale operation, were avoided. Refolding yields of over 80% was obtained for achieving reaction volume of upto 50 l at protein concentration of 1 mg/ml. The optimum urea concentration was 1M. Refolding yield at the 1-1 reaction volume and protein concentration of 0.5mg/ml was increased about 2.5-fold, compared to that in a batch reactor. By increasing protein concentration in a two-stage refolding reaction, the cost for insulin production could be reduced, therefore, making this process economical.

Bioethanol Production from Hydrodictyon reticulatum by Fed-Batch Fermentation Using Saccharomyces cerevisiae KCTC7017

  • The aim of this study was to develop a potential process for bioethanol production from Hydrodictyon reticulatum (HR), a filamentous freshwater alga, using Saccharomyces cerevisiae (KCTC7017). From the sugar solutions prepared by the four different hydrolysis methods, bioethanol production ranged from 11.0 g/100 g dried material (acid hydrolysis) to 22.3 g/100 g dried material (enzymatic hydrolysis, EH). Bioethanol was fermented from a highly concentrated sugar solution obtained by a decompression-mediated (vacuum) enrichment method (VE). As the results, ethanol was more efficiently produced from HR when sugar solutions were concentrated by VE following EH (EH/VE). Using multiple feeding of the sugar solution prepared by EH/VE from HR, ethanol reached up to a concentration of 54.3 g/l, corresponding to 24.9 g/100 g dried material, which attained the economic level of product concentration (approximately 5%). The results indicate that by using HR, it is feasible to establish a bioethanol production process, which is effective for using microalgae as the raw material for ethanol production.

Membrane Diffuser Coupled Bioreactor for Methanotrophic Denitrification under Non-aerated Condition: Suggestion as a Post-denitrification Option

  • Methanotrophic denitrification under a non-aerated condition (without external supply of oxygen or air) was investigated in a bioreactor coupled with a membrane diffuser. Batch experiment demonstrated that both methane consumption and nitrogen production rates were not high in the absence of oxygen, but most of the nitrate was reduced into $N_2$ with 88% recovery efficiency. The methane utilized for nitrate reduction was determined at 1.63 mmol $CH_4$/mmol $NO_3{^-}$-N, which was 2.6 times higher than the theoretical value. In spite of no oxygen supply, methanotrophic denitrification was well performed in the bioreactor, due to enhanced mass transfer of the methane by the membrane diffuser and utilization of oxygen remaining in the influent. The denitrification efficiency and specific denitrification rate were 47% and 1.69 mg $NO_3{^-}-N/g\;VSS{\cdot}hr$, respectively, which were slightly lower than for methanotrophic denitrification under an aerobic condition. The average concentration of total organic carbon in the effluent was as low as 2.45 mg/L, which indicates that it can be applicable as a post-denitrification method for the reclamation of secondary wastewater effluent. The dominant fatty acid methyl ester of mixed culture in the bioreactor was $C_{16:1{\omega}7c}$ and $C_{18:1{\omega}7c}$, which was predominantly found in type I and II methanotrophs, respectively. This study presents the potential of methanotrophic denitrification without externally excess oxygen supply as a post-denitrification option for various water treatment or reclamation.

  • All industrial Products of today are various demand of customers and are switchovered the manufacturing system of a small quantity batch production. Its life cycle was very shortened. An enterprise is improved to satisfaction with external customers and internal customers of infinity competition time. They can exist on achieving customer satisfaction management. In this paper, a new implementation of service information system (SIS) with real time using RFID and X-Internet in Ubiquitous environments is proposed. The proposed SIS system has Web architecture based on XML and has a new X-Internet environment overcoming limitation of Client/server architecture. The service information system analyze its material data and suppress the same CLAIM creation. The implementation method of real time SIS system using RFID and X-Internet is proposed for contribution of business market reducing F-Cost.

An Alternative Approach to the Traditional Mixotrophic Cultures of Haematococcus pluvialis Flotow (Chlorophyceae)

  • In traditional mixotrophic cultures of microalgae, all the inorganic nutrients and organic carbon sources are supplied in the medium before inoculation. In this study, however, an alternative approach was adopted in Haematococcus pluvialis Flotow, a microalga capable of growing mixotrophically on sodium acetate (Na-Ac). First, the cells were grown under 75 ${\mu}Mol$ photons $m^{-2}s^{-1}$ phototrophically without Na-Ac until the stationary phase and then exposed to five different light regimes by the addition of Na-Ac (e.g., dark, 20, 40, 75, and 150 ${\mu}Mol$ photons $m^{-2}s^{-1}$). Dry weight (DW), pigments, and especially cell number in alternative mixotrophy (AM) were higher than traditional mixotrophy (TM). Cell number in AM almost doubled up from 21.7 to $42.9{\times}10^4$ cells/ml during 5-day exposure to Na-Ac, whereas the increase was only 1.2-fold in TM. Maximum cell density was reached in 75 ${\mu}Mol$ photons $m^{-2}s^{-1}$ among the light intensities tested. We propose that Na-Ac in TM of H. pluvialis can not be utilized as efficiently as in AM. With this respect, AM has several advantages against TM such as a much higher cell density in a batch culture period and minimized risk of contamination owing to the shorter exposure of cells to organic carbon sources. In consequence, this method may be used for other strains of the species, and even for the other microalgal species able to grow mixotrophically.

Two-Step Process Using Immobilized Saccharomyces cerevisiae and Pichia stipitis for Ethanol Production from Ulva pertusa Kjellman Hydrolysate

  • We established a two-step production process using immobilized S. cerevisiae and P. stipitis yeast to produce ethanol from seaweed (U. pertusa Kjellman) hydrolysate. The process was designed to completely consume both glucose and xylose. In particular, the yeasts were immobilized using DEAE-corncob and DEAE-cotton, respectively. The first step of the process included a continuous column reactor using immobilized S. cerevisiae, and the second step included a repeated-batch reactor using immobilized P. stipitis. It was verified that the glucose and xylose in 20 L of medium containing the U. pertusa Kjellman hydrolysate was converted completely to about 5.0 g/l ethanol through the two-step process, in which the overall ethanol yield from total reducing sugar was 0.37 and the volumetric ethanol productivity was 0.126 g/l/h. The volumetric ethanol productivity of the two-step process was about 2.7 times greater than that when P. stipitis was used alone for ethanol production from U. pertusa Kjellman hydrolysate. In addition, the overall ethanol yield from glucose and xylose was superior to that when P. stipitis was used alone for ethanol production. This two-step process will not only contribute to the development of an integrated process for ethanol production from glucose-and xylose-containing biomass hydrolysates, but could also be used as an alternative method for ethanol production.

  • We have fabricated SmBCO coated conductor on IBAD-MgO substrates using unique co-evaporation method. The batch type co-deposition system was specially designed and named as EDDC(evaporation using drum m dual chamber) that is possible to deposit superconducting layer with different composition ratio at low temperature of $700^{\circ}C$. In this study, we have investigated the influence of SmBCO phase composition and texture of IBAD-MgO template on the critical current density. We have changed the deposition rates of Sm, Ba and Cu during co-evaporation to examine the optimal composition ratio shown better critical current density. The composition ratio and surface morphology of SmBCO coated conductors were analyzed by the EDX and SEM, respectively. A higher critical current density was measured at superconducting phase composition ratio of Ba deficiency, Sm or Cu rich compared to the Sm1Ba2Cu3Ox stoichiometry.

  • The adsorption of nickel(Ni) from sulfuric acid solution was carried out by ion exchange method. A series of batch tests in synthetic solutions were carried out using Lewatit Monoplus TP 220 resin. The following experimental parameters, such as temperature, shaking rate, reaction time, pH, resin dosage and concentration of nickel ions etc. were investigated to establish the effective optimum conditions of nickel adsorption. The solution pH(2.0~5.0) and shaking rate had little effects on the adsorption of nickel and adsorption time of 72hours was required to reach equilibrium. The experimental results show a good agreement with Feundlich isotherm and pseudo-second order reaction. The adsorption behavior of Ni obtained from synthetic solution was compared with that of waste electroplating solution. Elution of nickel from loaded resin increased with increase in $H_2SO_4$ concentration.
