Useful genes can be Screened from various environments by construction of metagenomic DNA libraries. In this study, water samples were collected from several lakes in mid Korea, and analyzed by T-RFLP to examine diversities of the microbial communities. The crude DNAs r were extracted by the SDS-based freezing-thawing method, and then further purified using an $UltraClean^{TM}$ kit (MoBio, USA). The metagenomic libraries were constructed with the DNAs partially digested with EcoR I, BamH I, and Sac II in Escherichia coli DH 10B using the pBACe3.6 vector. About 44.0 Mb of metagenomic libraries were obtained with average inserts 13-15 kb in size. The bphC genes responsible for degradation of aromatic hydrocarbons via mets-cleavage were identified from the metagenomic libraries by colony hybridization using the bphC specific sequence as a probe. The 2,3-dihydroxybiphenyl (2, 3-DHBP) dioxygenase gene (bphC ), capable of degradation of 2,3-DHBP, was cloned and its nucleotide Sequences analyzed. The genes consisted of 966 and 897 base pairs with an ATG initiation codon and a TGA termination codon. The activity of the 2,3-DHBP dioxygenase was highly expressed to 2,3-DHBP and Showed a broad substrate range to 2,3-DHBP, catechol, 3-methylcatechol and 4-methylcatechol. These results in-dicated that the bphC gene identified from the metagenomes derived from lake water might be useful in the development of a potent strain for degradation of aromatic pollutants.

This study was done to identify contents of clothing and textile education and to systemize its curriculum from elementary school to high school Every time the official curriculum is updated. the suggestion that the contents of Home Economics Education should be systematized never fails to come forth. Unfortunately. it's a fact that there are still insufficient grounds to base this opinion on. and that adequate research has not yet been done in this particular field. Therefore. basic research must be performed in order to systematize the substance of clothing and textile education. Keeping this in mind. the following study strives to find appropriate methods of conducting clothing and textile education, while attempting to provide basic materials for future organization and selection of contents. In order to achieve these goals. the materials have been sorted into three categories : First of all. what sort of concepts is being taught in the subject of clothing and textile. and are the chapters constructed so that different schools can maintain consistency among them\ulcorner Second, how has clothing and textile education been done so far and is the contents correctly synchronized with the students'development\ulcorner Third. haw is clothing and textile education utilized in real life. and has it actually been systematized in a way that encourages practical utilization\ulcorner This study has mostly been done through the research of various documental resources. Also. the analysis has been performed using the systematic contents analysis method. The main results of obtained this study are as follows : As the concepts of clothing and textile education. which are different from each school, has not completely been systematizied. Also. as the contents of clothing and textile education has not correctly synchronized with the students'development. am it has not actually been systematized in a way that encourages practical utilization. Since this study has touched only limited research on the topic of clothing and textile education. it is essential that further research covering a wider array of themes be done in the future.

In this paper, novel latch-up prevention method that employs power-up sequential switches has been proposed to relieve latch-up problem in liquid crystal display (LCD) driver ICs. These sequential switches are inserted in the 2'nd and 3'rd boosting stages, and are used to short the emitter-base terminals of parasitic p-n-p-n circuit before relevant boosting stages are activated during power-up sequence. To verily the performance of the proposed method, test chips were designed and fabricated in a 0.13-um CMOS process technology. The measurement results indicated that, while the conventional LCD driver If entered latch-up mode at $50^{\circ}C$ accompanying a significant amount of excess current, the driver IC adopting the proposed method showed no latch-up phenomenon up to $100^{\circ}C$ and maintained normal current level of 0.9mA.
Sixteen specimens of Artediellus fuscimentus, belonging to the family Cottidae, were collected from the coastal waters off Samcheok and Donghae, Korea, in December 2010 and February 2015, respectively. The specimens are characterized by four preopercular spines, 12~13 second dorsal fin rays, 10~12 anal fin rays, a first preopercular spine with no minute spine on the inner side, and pectoral fin extending past the origin of the anal fin. We also analyzed 600 base pairs of the mitochondrial DNA cytochrome b sequence to confirm the taxonomic status of the specimens. The sequences of our specimens corresponded completely to those of Japanese A. fuscimentus, but differed from those of A. atlanticus (d=0.057~0.061). We propose the new Korean names "Eom-ji-hoet-dae-sok" for the genus Artediellus and "Eom-ji-hoet-dae" for the species A. fuscimentus.
We examined mRNAs, isolated from the rat brain, to ascertain if there is any polymorphism for S-100 beta protein gene. As templates for polymerase chain reaction (PCR) the reverse-transcribed cDNA from the rat brain or phage DNAs isolated from the rat brain cDNA libraries were used. Although PCR products turned out to be exactly same as the expected size based on the previously reported mRNA sequence a single base substitution (CAT to CAC) was identified at nucleotide level. This change was considered as polymorphism since it did not cause any change of the primary structure for S-100 beta protein. This result should facilitate the understanding of the overall structure of the gene for S-100 beta protein.
We cultured a tubular marine green alga, originally identified as Capsosiphon groenlandicus (J. Agardh) K.L. Vinogradova, from Amaknak Island, Alaska. The alga had an alternation of heteromorphic generations in which tubular monoecious fronds produced quadriflagellate zoospores and/or biflagellate isogametes. The gametes fused to produce cysts or Codiolum-like zygotes with long, tortuous stalks. Cysts and codiola produced 8-16 aplanospores, which germinated in situ to yield upright fronds. Fronds arising from both aplanospores and zoospores displayed a distinctive development in which non-septate colorless rhizoids from the base of the initially uniseriate, Ulothrix-like filament were transformed into septate uniseriate Ulothrix-like photosynthetic filaments. These transformed filaments then developed new basal non-septate rhizoids. This pattern of rhizoids becoming filaments, which then produced new rhizoids, was repeated to yield a tuft of up to 50 fronds. Periclinal and longitudinal divisions occurred in each filament, starting basally, until the mature tubular thallus was achieved. Pyrenoid ultrastructure revealed several short inward extensions of chloroplast lamellae, each of which was surrounded by pyrenoglobuli. Analysis of ribosomal SSU and ITS sequences placed this alga in the family Ulotrichaceae, order Ulotrichales, together with but as a distinct species from North Atlantic Capsosiphon groenlandicus. Analysis of a partial ITS sequence from authentic Capsosiphon fulvescens, the current name of the type of the genus Capsosiphon, indicated that neither our material nor C. groenlandicus belongs in that genus, and we propose a new genus, Pseudothrix, to accommodate both species. We propose P. borealis for the North Pacific entity formerly called C. groenlandicus and make the new combination P. groenlandica for the Atlantic species.
Small Tail Han Sheep has significant characteristics of high prolificacy and non-seasonal ovulatory activity and is an excellent local sheep breed in P. R. China. Recently a novel member of the transforming growth factor $\beta$ (TGF$\beta$) superfamily termed bone morphogenetic protein 15 (BMP15) was shown to be specifically expressed in oocytes and to be essential for female fertility. Therefore, BMP15 is a candidate gene for reproductive performance of Small Tail Han Sheep. The whole genomic nucleotide sequence of BMP15 gene in Small Tail Han Sheep was searched for polymorphisms by PCR-SSCP and direct sequencing, and only one polymorphism was found. The polymorphism was a result of a 3 base pair deletion, which eliminated a single Leu codon (CTT). The allelic frequencies for A (without deletion) and B (with a codon deletion) are 0.73 and 0.27 respectively. The effects of BMP15 genotype on litter size were evaluated using the least squares model. This indicated that there was a significant association between litter size of Small Tail Han Sheep and a deletion in BMP15 gene (p=0.02<0.05). Small Tail Han Sheep ewes with AA and AB genotype produce on average 0.5 and 0.3 more lambs per litter than those ewes with BB genotype.
This paper is concerned with the performance evaluation and comparison analysis fur several kinds of LIPCA (Lightweight Piezo-Composite Actuator) device system. LIPCA device system is composed of a piezoelectric ceramic layer and fiber reinforced light composite layers, typically a PZT ceramic layer was sandwiched by a top fiber layer with low CTE (coefficient of thermal expansion) and base layers with high CTE. To investigate the effect of lay-up structure of the LIPCA on the actuating performance, four kinds of actuator with different lay-up stacking sequence were designed, manufactured, and tested. The performance of each actuator was evaluated using an actuator test system consisted of an actuator supporting jig, a high voltage actuating power supplier, and a non-contact laser measuring system. From the comparison of the performance of the LIPCA prototypes, it was found that the actuator with higher coefficient of unimorph actuator can generate larger actuating displacement.
Objectives : Genetic analysis and taste pattern were performed to identify species between Glycyrrhiza uralensis and G. glabra which are officially listed in Korean Pharmacopoeia IX as origin of Gamcho(g$\={a}$nc$\v{a}$o, licorice root, Glycyrrhizae Radix et Rhizoma). Methods : Genetic analysis showed that identification between two species was done by comparing base sequence of ITS(intergenic transcribed spacer) and trnH-psbA regions from eleven Gamchoes sold in market. There was different taste pattern using by taste sensor in Glycyrrhiza uralensis and G. glabra. Results : Genetic analysis showed that six Gamchoes from China were identified as Glycyrrhiza uralensis and five Gamchoes from Uzbekistan were G. glabra. From the results of taste pattern, sourness and astringency of Glycyrrhiza uralensis from China were significantly higher than G. glabra from Uzbekistan, and aftertaste of astringency, aftertaste of umami, and saltiness of Glycyrrhiza uralensis were signicantly low as compared to G. glabra. There is no significant difference between two species in terms of bitterness, aftertaste of bitterness, and umami. Conclusions : Taken together, Glycyrrhiza uralensis from China and G. glabra from Uzbekistan were identified by taste sensor, and this technic could be applied to establishment of taste pattern marker for identification of different species located in various regions.
Although Pterocladiella is a well-known agar-producing red algal genus, its taxonomy in Korea appears to be confused. In the present study, we demonstrate the occurrence of Pterocladiella nana and P. tenuis, as well as P. capillacea in Korea on the basis of morphological and plastid rbcL sequence comparisons. P. capillacea commonly occurs along the coasts of Korea and has regular pinnate branches with abundant second-order branches, growing up to 10 cm in length. P. nana occurs in the intertidal zone of Jeju and is distinguished by the small size of its thalli (up to 5 cm) and regular branches with up to third-order branches. P. tenuis is distributed mostly subtidally on the southern coast of Jeju and has irregular branches with rarely second-order branches, growing up to 19 cm in length. We determined rbcL sequences from 19 specimens (15 from Korea and four from France) and downloaded 28 sequences from GenBank. Analyses of all 47 rbcL sequences revealed that each of three species was consistently resolved. P. capillacea and P. tenuis always formed a sister clade with P. nana at the base. Given that 12 rbcL haplotypes from 28 specimens of P. capillacea have been found to date, analysis of a fast-evolving gene from across the range of the species should highlight its genetic diversity.
