The interaction between ocean and ice shelf is a critical physical process in relation to water mass transformations and ice shelf melting/freezing at the ocean-ice interface. However, it remains challenging to thoroughly understand the process due to a lack of observational data with respect to ice shelf cavities. This is the first study to simulate the variability and circulation of water mass both overlying the continental shelf and underneath an ice shelf and an ice tongue in the Terra Nova Bay (TNB), East Antarctica. To explore the properties of water mass and circulation patterns in the TNB and the corresponding effects on sub ice shelf basal melting, we explicitly incorporate the dynamic-thermodynamic processes acting on the ice shelf in the Regional Ocean Modeling System. The simulated water mass formation and circulation in the TNB region agree well with previous studies. The model results show that the TNB circulation is dominated by the geostrophic currents driven by lateral density gradients induced by the releasing of brine or freshwater at the polynya of the TNB. Meanwhile, the circulation dynamics in the cavity under the Nansen Ice shelf (NIS) are different from those in the TNB. The gravity-driven bottom current induced by High Salinity Shelf Water (HSSW) formed at the TNB polynya flows towards the grounding line, and the buoyance-driven flow associated with glacial meltwater generated by the HSSW emerges from the cavity along the ice base. Both current systems compose the thermohaline overturning circulation in the NIS cavity. This study estimates the NIS basal melting rate to be 0.98 m/a, which is comparable to the previously observed melt rate. However, the melting rate shows a significant variation in space and time.
The integumental secretory structure is exocrine unicellular gland located in the epidermis of goldfish, Carassius auratus, and divided into two groups, mucous and granular cells. By the histochemical studies of integumental secretions the mucos cells reacted for acidic polysaccharides, and the granular cells for neutral glycoprotein. According to concentration of the secretion the integumental mucous are gradually sulphated. The mucous cells are typical form of goblet cell located in the upper region of the epidermis, and membrane bounded vesicles of the mucous are observed several size and electron densities by the cellular differentiation. The granular cells in middle and lower epidermis are present syncitial forms occasionally, and contain electron dense granules sized $1.0{\mu}m$ which are accumulated in cytoplasmic process held the cells to the basal lamina. The precursors of the integumental secretory materials are originated from the rough endoplasmic reticulum and next transported through the Golgi apparatus as a form of membrane bounded vesicles. After accomplish this process mature secretions are extruded to integumental surface by the mechanism of merocrine secretion in response to nerve stimulations respectively.
A sphingid species, Macroglossum corythus Walker, 1856 is newly recorded from Korea. Macroglossum corythus can be characterized by grayish or dark greenish fore- and hindwings, blackish basal, ante- and postmedial lines with a blackish discal dot on the forewing and transverse antemedial line, as well as a strongly dentate postmedial line and a thick, dark brownish tinged subterminal line on the hindwing. The male genitalia of Macroglossum corythus can be characterized by the long saccular process of valva and the phallus with one hooked distal process and the large penshaped cornutus and a notched ridge. The female genitalia of Macroglossum corythus can be characterized by the long tubular ductus bursae and the long ovate corpus bursae with multiple stripes and a long triangular patch of minute signa. To date, six species of the genus Macroglossum have been identified in Korea.
The ultrastructure of germ cells and follicle cells during oogenesis, oocyte degeneration, reproductive cycle, and first sexual maturity in female Cyclina sinensis were investigated for clams collected from Simpo, Jeollabuk-do, Korea, by cytological and histological observations. Vitellogenesis occured by way of endogeous autosynthesis and exogenous heterosynthesis: vitellogensis occurred through a process of autosynthesis, which involves a combined activities of the Golgi complex, mitochondria, and rough endoplasmic reticulum. The process of heterosynthesis involved endocytotic incorporation of extraovarian precursors into the basal region of the early vitellogenic oocytes prior to the formation of vitelline envelope. The follicle cells appear to play an integral role in vitellogenesis and oocyte degeneration, functioning in phagocytosis and digestion of products originating from the degenerated oocytes: these functions can permit the transfer of yolk precursors needed for vitellogenesis. Follicle cells might have a lysosomal system for breakdown and might also resorb phagosomes in the cytoplasm for nutrient storage during oocyte degeneration.
For LiMn2O4 based spinel structures the stoichiometric reaction conditions need be considered carefully because the electrical properties depend on the structural stability. Autophagy, which has the literal meaning of self-eating, is a cellular catabolic process executed by arrays of conserved proteins in eukaryotes. Autophagy is dynamically ongoing at a basal level, presumably in all cells, and often carries out distinct functions depending on the cell type. Therefore, although a set of common genes and proteins is involved in this process, the outcome of autophagic activation or deficit requires scrutiny regarding how it affects cells in a specific pathophysiological context. The uterus is a complex organ that carries out multiple tasks under the influence of cyclic changes of ovarian steroid hormones. Several major populations of cells are present in the uterus, and the interactions among them drive complex physiological tasks. Mouse models with autophagic deficits in the uterus are very limited, but provide an initial glimpse at how autophagy plays a distinct role in different uterine tissues. Herein, we review recent research findings on the role of autophagy in the uterine mesenchyme in mouse models.
뇌실을 감싸는 뇌실막층은 대부분이 일반적인 뇌실막 세포로 이루어졌으나, 이들 세포 사이에 간혹 띠뇌실막 세포(tanycyte)가 분포하고 있다. 띠뇌실막세포는 일반적인 뇌실막세포와는 달리 뇌의 실질로 뻗은 매우 긴 기저돌기를 갖고 있다. 특히 제3뇌실의 뇌실막층에서 주로 연구된 띠뇌실막세포는 뇌실과 뇌실질의 혈관 혹은 신경세포와의 사이에 물질교환을 담당하는 것으로 추측되고 있으며, 띠뇌실막세포는 일반적인 뇌실막세포와는 달리 glial fibrillary acidic protein(GFAP)항체에 대해 양성반응을 보이는 것으로 알려져 있다. 본 연구는 면역조직화학 및 면역금표지법을 이용하여 박쥐 맨아래구역을 감싸는 뇌실막층에서 GFAP 항체에 대해 양성반응을 보이는 세포의 분포여부 및 이의 미세 구조를 확인하고자 시행하여 다음과 같은 결론을 얻었다. 세로 절단한 절편을 대상으로 GFAP 항체를 이용하여 면역염색한 후 광학현미경으로 확인한 결과 맨아래구역을 감싸는 뇌실막층에 GFAP 양성반응을 보이는 세포가 관찰되었으며, 특히 맨아래구역의 목부분에서 양성반응 세포가 많이 모여 있었다. GFAP양성반응을 보이는 세포들은 매우 긴 기저돌기를 갖고 있었으며, 기저돌기에서도 매우 강한 GFAP 양성반응을 보였다. 세포체에서는 주변부에서 양성반응을 보였다. 전자현미경하에서 맨아래구역 띠뇌실막세포는 주로 ependymal tanycytes였으며, 자유면에 소수의 미세융모 및 세포질돌기가 관찰되었으나, 섬모는 관찰되지 않았다. 기저부에 특징적으로 긴 기저돌기를 갖고 있었으며 이 돌기에는 중간세사 및 세로로 길게 달리는 사립체가 발달되어 있었고, 세포체와 돌기에 지방방울이 산재되어 있었다. 금입자를 표지한 GFAP항체를 사용하여 면역 염색한 후 전자현미경으로 관찰한 결과 세포체에서 핵을 둘러싸는 중간세사에서 금입자를 관찰할 수 있었으며, 나머지 핵을 비롯한 다른 세포소기관이나 세포질에서는 관찰되지 많았다. 또한 기저돌기에서도 중간세사에서만 금입자를 관찰할 수 있었다. 이와 같은 본 실험의 결과는 박쥐 맨아래구역 뇌실막층에도 띠뇌실막세포가 존재하고 있으며, 이 세포의 발달된 긴 기저돌기는 띠뇌실막세포가 일반적인 뇌실막세포와는 다른 기능을 동면동물인 박쥐의 맨아래구역에서도 수행하고 있음을 암시하고 있으나 이를 확인하기 위해서 더욱 자세한 연구가 필요한 것으로 사료된다.
PVT(Physical vapor transport)법은 고품질의 대면적 웨이퍼를 생산하기에 이점을 가져 질화물계 반도체의 상용화를 위해 많은 연구가 진행되고 있는 단결정 성장 방법이다. 하지만 복잡한 공정 변수들로 인하여 비평형적인 성장 조건을 갖게 될 경우 수많은 결함들이 발생하게 된다. 결정성장 후 어닐링 공정은 결정성 개선을 위해 널리 사용된다. 효과적인 결정성 개선을 위해서는 적절한 온도, 압력과 시간을 설정하는 게 중요하다. 본 연구에서는 PVT법으로 성장된 AlN 단결정 및 어닐링 조건에 따른 단결정의 결정 미세구조 변화를 X-ray topography, Electron Backscattered Diffraction(EBSD), Rietveld refinement를 통해 분석하였다. Synchrotron Whitebeam X-ray topography 분석 결과 어닐링을 진행하지 않은 단결정에 2차상 및 sub grain, impurity가 존재하였으며 이로 인해 결정성이 저하되는 것을 확인 할 수 있었다. EBSD 결과 어닐링을 진행한 시편의 경우 결정립수가 증가함과 동시에 basal plane의 뒤틀림이 일어나는 것을 관찰할 수 있었다. Rietveld refinement 결과 일부 격자들이 a, b, c축 방향으로 응력을 받아 변형된 것으로 분석되었다. 이는 어닐링 과정 중 hot zone 내의 상하 온도구배에 의해 발생한 응력으로 결정립 방향의 뒤틀림이 일어날 뿐만 아니라 격자 상수가 달라진 것으로 분석된다.
Pongamia pinnata Pierre is a tree legume, having potential in production of raw material for biodiesel. A protocol for in wk propagation of this plant was standardized using seedling explants. Growth regulators (GR) including gibberellic acid $(GA_3),\;N^6-benzylaminopurine(BA)$, thidiazuron (TDZ), and Adenine sulphate (Ads) were tested for optimum germination of seeds. Removal of seed coat prior to germination, controlled fungal growth partially but enhanced bacterial growth. Antibiotic cefotaxime was ineffective in controlling bacterial contamination. Seedling derived nodal explants and cotyledon nodes with attached cotyledons were excised and cultured for induction of shoots. Optimum sprouting and multiplication of shoot buds were obtained in MS medium supplemented with $8.88{\mu}M$ BA. These buds differentiated and rooted on medium devoid of GR. Optimum growth of Pongamia seedling was obtained in cotton plugged culture vessels. Reculturing of the cotyledon node explants produced more shoots from the same site. This process of removing shoots and reculturing of cotyledon node was followed for eight passages yielding 4 to 8 shoots in each cycle. The shoots (75%) rooted on half strength MS basal medium supplemented with 0.22% charcoal. All plants survived on transfer to soil. This is the first report on in vitro regeneration of Pongamia pinnata. This report demonstrates the possibility of coupling more than one parameter in single experiment to hasten the process of standardization. The process of cycling the nodal explant repeatedly for production of large number of shoots from single meristem may find application in genetic transformation experiments wherein meristems are used for transformation.
Rph1 and Gisl are damage-responsive repressors involved in PHR1 expression. They have two $C_{2}$H/ sub 2/ zinc finger motifs as putative DNA binding domains and N-terminal conserved domain with unknown function. They are also found in the human retinoblastoma binding protein 2 and the mouse jumonji- encoded protein. The repressors are able to bind to A $G_{4}$ sequence within a 39-bp sequence called upstream repressing sequence of PHR1 promoter (UR $S_{PHR1}$) responsible for the damage-response of PHR1. We report here that Rph1 is predominantly localized in the nucleus as examined by fluorescence microscopic analysis with GFP-Rph1 fusion protein. On the basis of the fact that the A $G_{4}$ sequence that is recognized by Rph1 and Gisl is also recognized by Msn2 and Msn4 in a process of stress response, we a1so tried to examine the in vivo function of A $G_{4}$ and the role of Msn2 and Msn4 in PHR1 expression. Our results demonstrate that Msn2 and Msn4 are actually required for the basal transcription of PHR1 expression but not for its damage induction. When A $G_{4}$ sequence was inserted into the minimal promoter of the cyc1-LacZ reporter, the increased LacZ expression was observed indicating its involvement in transcriptional activation. The data suggest that the A $G_{4}$ is primarily required for basal transcriptional activation of PHR1 or CYC1 promoter through the possible involvement of Msn2 and Msn4. However, since the A $G_{4}$ is also involved in the repression of PHR1 via Rphl and Gisl, it is proposed that A $G_{4}$ functions as either URS or upstream activating sequence (UAS) depending on the promoter context.t.
