Development of White Bread Using Fermented Wild Grape Sourdough (머루를 이용한 Sourdough 식빵 개발)

  • The purpose of this study was to manufacture white bread by substituting bakery yeast with natural starter. The general composition of wild grape, was 81.22% moisture, 0.81% ash, 0.74% crude fat, 1.58% crude protein, and 15.65% carbohydrate contents. Fermented wild grape extract was made by culturing for 4 days at $25^{\circ}C$. Flour was added three times to fermented wild grape extract and incubated for 36 hours at $20^{\circ}C$ to manufacture sourdough. We investigated the quality characteristics of white bread with different amounts of fermented wild grape sourdough. The pH of dough decreased with increasing amounts of sourdough, whereas total titratable acidity of dough significantly increased. Fermentation power of dough expansion significantly increased with increasing incubation time, and control showed the highest value of 126.67%. White bread specific volume was highest in, 50% sourdough at 4.76 mL/g. Water content increased with increasing amounts of sourdough, whereas water activity was not significantly different between the samples. For texture, hardness of 50% sourdough was lowest at 19.23 g. In the sensory evaluation, color, flavor, softness and overall acceptability decreased with increasing amounts of sourdough. As a result, 50% sourdough can be considered as a baker's yeast substitute for making natural fermented bread.

Volatile Flavor Components in the Mashes of Takju Prepared Using Different Yeasts (효모종류를 달리한 탁주 술덧의 휘발성 향기성분)

  • In this study, we examined the volatile flavor components in the mashes of takju prepared using different yeasts such as Saccharomyces coreanus, S. ellipsoideus, S. carlsbergensis, S. cerevisiae (Baker's yeast), and S, rouxii by GC and GC-MS. Fourteen alcohols, 13 esters, 5 acids, 3 aldehydes, 7 amines, and 2 other compounds were identified in the mash after 6 days of fermentation. On day 6, the takju fermented by S. coreanus had the greatest variety of volatile flavor components. Fifteen flavor components, including ethanol, isobutyl alcohol, isoamyl alcohol, methyl pentanol, 1,3-butanediol, 3-methylthio-1-propanol, benzeneethanol, ethyl lactate, acetic acid, acetaldehyde, and 1,3-cyclohexane diamine, were typically detected in all the treatments. The relative peak areas of the volatile components were as follows: alcohols (96.758-99.387%), esters (0.081-0.968%), acids (0.040-0.640%), aldehydes (0.266-0.959%), and amines (0.011-0.047%). In particular, 1-propanol, isobutyl alcohol, 3-methyl-1-butanol, 2,3-butanediol, trimethyl benzylalcohol, heptene-2,4-diol, ethyl lactate, diethyl succinate, ethyl nonanoate, methyl hexadecanoate, linoleic acid, hexadecanoic acid, and acetaldehyde were hish in the takju made by S. coreanus. Also, ethyl stearate was high in the takju made by S. carlsbergensis, and hexanoic acid was high in the takju made by S. cerevisiae. Finally, methyl pentanol, 1,3-butanediol, 3-methylthio-1-propanol, benzene ethanol, ethyl octadecanoate, acetic acid, pentanal, and 1,3-cyclohexane diamine were high in the takju made by S. rouxii.

Dietary Value of Neonates from Rotifer, Brachionus plicatilis and B. rotundiformis Resting Eggs for Flounder and Parrot Fish Larvae (넙치 및 돌돔 자어 사육에 있어서 Brachionus plicatilis와 B. rotundiformis 내구란에서 갓 부화한 rotifer의 먹이효율)

  • The dietary value and fatty acids composition for the hatched neonates from the resting eggs of marine roofers, Brachionus plicatilis and B. rotundiformis were compared with that for mass-cultured rotifers (control) by feeding them to larvae of flounder (Paralichthy olivaceus) and parrot fish (Oplegnathus fasciatus). Resting eggs were mass-produced in $1~4m^3$tanks by feeding Chlorella sp. and baker's yeast. The B. plicatilis and B. rotundiformis eggs were preserved at $5^{\circ}C$ in darkness for 3 and 5 months, respectively, and hatched at $28^{\circ}C$ under continuous light. The hatched neonates from the resting eggs and mass-cultured rotifer, which was used as a rontrol were fed to fish larvaes. The growth and survival rates of parrot fish larvae fed on the neonates from the resting eggs of B. rotundiformis were similar to those of fish larvae fed on the control rotifer. And the growth and survival rates of the flounder larvae with neonates from the resting eggs of B. plicatilis were similar or higher than those fed the control rotifer. Also the fatty acids composition of the neonates from the resting eggs of B. plicatilis and B. rotundiformis were similar to those from the control rotifers. This results showed that the hatched neonates from resting eggs of rotifer could be used as an effective diet for flounder and parrot fish larvae.

Effects of Temperature, Salinity and Diet on the Productivity of the Cyclopoid Copepod, Apocyclops royi (수온, 염분 및 먹이에 따른 기수산 cyclopoid 요각류, Apocyclops royi의 생산력)

  • The productivity of cyclopoid copepod, Apocyclops royi fed by various diets (Isochrysis galbana, Tetraselmis suecica, Phaeodactylum tricornutum, concentrated freshwater Chlorella and baker's yeast) was investigated at tile different temperatures ($16-36^{\circ}C$) with different salinities (5-34 ppt). A. royi was cultured in 6 ml vessels (12 wells culture plate). Total production (188 inds.) and daily production (13.4 inds.) of nauplii by A. royi female at $32^{\circ}C$ were significantly higher than those of nauplii at the different temperatures (P<0.05). Development time from nauplii to copepodite and from nauplii to adult tended to increase with increasing water temperature up to 32. And total production (169 inds.) and daily production (9 inds.) of nauplii by A. royi female at 10 ppt were significantly higher than those of nauplii at the different salinities (P<0.05). The fastest development time from nauplii to copepodite and from nauplii to adult was observed at 10 ppt and 15 ppt, respectively (P<0.05). The highest total production of A. royi nauplii and fastest development time from nauplii to adult were obtained in females fed Isochrysis galbana (P<0.05). These results may indicate that the optimum culture temperature and salinity for A. royi are $32^{\circ}C$ and 10 ppt, respectively, and Isochrysis galbana is one of the suitable diets for this copepod.

Relative Effectiveness of Replacement Diets for Mass Production of the Brackish Water Flea Diaphanosoma celebensis (기수산 물벼룩(Diaphanosoma celebensis)의 대량생산을 위한 대체 먹이원의 효과)

  • This study examined the effect of formulated diets (composed of condensed freshwater $Chlorella$ or $Schizochytrium$ sp., $Crypthecodinium$ sp., and EPA oil sources derived from baker's yeast) on the population growth and fatty acid composition of the brackish water flea ($Diaphanosoma$ $celebensis$) to determine diet effectiveness for mass production. Results indicated that the highest maximum density, fecundity, and population specific growth rate (SGR) of the flea were observed using a diet of $Tetraselmis$ $suecica$ (control). However, results using condensed freshwater $Chlorella$ and the mixed treatment of $Schizochytrium$ sp.+$Crypthecodinium$ sp.+EPA oil did not significantly differ from the control treatment ($P$>0.05). Thus, $T.$ $suecica$ could be replaced with a combination diet of $Schizochytrium$ sp., $Crypthecodinium$ sp., and EPA oil in condensed freshwater $Chlorella$ without growth interruption of $D.$ $celebensis$. As the percentage of combination diet (mix of $Schizochytrium$ sp., $Crypthecodinium$ sp., and EPA oil) in condensed freshwater $Chlorella$ increased, the level of ARA, EPA, DHA, and n-3 HUFA in $D.$ $celebensis$ also increased. However, the population density of $D.$ $celebensis$ sharply decreased with the use of treated supplements C5M5 (CHL [50%] and $Crypthecodinium$ sp.+ $Schizochytrium$ sp. + EPA oil [MIX 50%]). The optimum proportions of CHL and MIX for $D.$ $celebensis$ within the mixture were 70% and 30%, respectively. Based on the results, the combination of $Schizochytrium$ sp., $Crypthecodinium$ sp., and EPA oil in condensed freshwater $Chlorella$ appears to be a stable and effective food source for mass production of $D.$ $celebensis$.

Growth of rotifer by the air, oxygen gas-supplied and the pH-adjusted and productivity of the high density culture (공기 및 산소 공급과 pH 조절에 따른 rotifer의 성장과 고밀도 배양의 생산성)

  • The growth of rotifer, Brachionus rotundiformis was evaluated at different culture conditions. Rotifer was fed on condensed freshwater Chlorella. The productivity of rotifer in the high density culture system was compared to that of rotifer in the batch culture system, in which rotifer was fed on baker's yeast. The growth rate of rotifer increased as temperature increased in the culture system supplied with air or oxygen gas. The maximum density of rotifer in the culture systems supplied with air was in range of 16,300$\~$17,000 ind./ml at $24^{\circ}C$. In the culture systems supplied with oxygen gas, it ranged 26,300$\~$30,500 ind/ml at $28^{\circ}C$. When the concentration of dissolved oxygen in the culture system supplied with air reached to below 1 ppm or when the concentration of undissolved ammonia in the culture system supplied with oxygen gas reached 16.6$\~$22.6 ppm, the growth of rotifer decreased. When oxygen gas was supplied and pH was adjusted to 7, the maximum density of rotifer reached to 43,000 ind/ml at $32^{\circ}C$. The production costs for 10 billion rotifer in the high density culture and batch culture were 693,000 and 961,000 won, respectively. Therefore, it was concluded that the productivity of rotifer in the high density culture was higher than that in a batch culture.

Rapid Purification of Glucose-6-Phosphate Dehydrogenase by Affinity Chromatography (Affinity Chromatography를 이용한 Glucose-6-Phosphate Dehydrogenase의 신속한 정제방법 개발)

  • An improved procedure for the rapid purification of glucose-6-phosphate dehydrogenase from extracts of Saccharomyces cerevisiae was developed by using affinity chromatography. Among six affinty media tested, $NADP^+ -agarose$ and Affi-gel Blue were more effective than others (i.e., Affi-gel Red, AMP-agarose, ATP-agarose, and $NAD^+ -agarose$). Conditions to desorb the enzyme bound to the affinity media were examined to increase the purity as well as yield. The best result was obtained when the column was developed with a linear gradient of KCl (0-1.0M). In case of Affi-gel Blue, introduction of $NAD^+$ (15mM) washing step prior to the salt gradient was most effective to remove $NAD^+ -binding$ proteins. For a large scale preparation of G-6-P dehydrogenase higher recovery was obtained by Affi-gel Blue than $NADP^+ -agarose$, however, the purity of the enzyme was decreased by 10 times if the former was used as the affinity medium. The capacity of Affi-gel Blue for G-6-P dehydrogenase was found to be 5 times higher than that of $NADP^+ -agarose$. Furthermore Affi-gel Blue could be reused repeatedly and its preparation is relatively easier and less expensive than $NADP^+ -agarose$.

A 4 week Randomized, Double-blind Human Trial to Compare the Efficacy and Safety of Aureobasidium pullulans Cultured Solution and Placebo on Improvement of Immune in Subjects (흑효모배양액 분말의 면역관련 사이토카인에 미치는 영향에 대한 무작위 배정 임상연구)

  • $\beta$-glucan is a fiber-type complex sugar (polysaccharide) derived from the cell wall of baker's yeast, oat and barley fiber, and many medicinal mushrooms, such as maitake. The primary uses of $\beta$-glucan are to enhance the immune system, to lower blood cholesterol levels and to treat tumor. $\beta$-glucan has no systemic toxicity in mice, therefore it needed clinical trail to prove efficacy and safety for human. The subjects total 56 healty volunteers were divided into two groups including taken $\beta$-glucan tablet group and placebo group. Subjects were taken two tablets per oral for 4 weeks. They had agreed to take part in this experiment, and didn't take any other clinical trail products. After 4 weeks blood of subjects were checked. The check list are TNF-$\alpha$, INF-$\gamma$, IL-2, IL-4, total WBC, differential WBC, RBC, hemoglobin, platelet, MCV, MCH, MCHC, HCT, Na, K, Ca, Cl, AST, ALT, ALP, $\gamma$-GTP, total protein, triglyceride, total cholesterol, total bilirubin, albumin, uric acid, creatinine, BUN, pH, protein, glucose, ketone body, blood, bilirubin. We evaluated efficacy by cytokines that compare before and after taking. Collected data were analyzed as two sample t-test, chi-square test and ANOVA using SAS V.9.1.This study results are that in TNF-$\alpha$ of $1^{st}$ efficacy measurement item, all of two groups figure were increased significantly compare to before figure. In IL4 of $2^{nd}$ efficacy measurement item, experimental group figure were decreased significantly but placebo group figure were increased. The conclusions show that based on the above results, $\beta$-glucan has favorable effect to enhance immune system, especially IL4 results showed that it has effect to improve the allergic immune system.

Effects of Sourdough on the Quality Characteristics of Rye-Wheat Mixed Bread (Sourdough 대체가 호밀-밀 혼합빵의 품질 특성에 미치는 영향)

  • Rye-wheat mixed bread samples made with substitutions of 20, 40, 60 and 80% sourdough, and control made with the addition of naturally fermented raisin extract, were examined for quality characteristics such as pH, total titratable acidity, fermentation power of dough expansion, specific volume, baking loss, water activity, color, texture, external and internal surface appearance, and sensory qualities, in order to determine the optimal ratio of sourdough in the formulation. As the incubation time of sourdough increased, pH decreased, while total titratable acidity increased. The pH of rye-wheat mixed doughs decreased with increasing sourdough content, but total titratable acidity increased. The rye-wheat mixed breads prepared with substitutions of sourdough had higher pH and lower total titratable acidity than the control group. Fermentation power of dough expansion of rye-wheat mixed doughs increased with increasing incubation time. Specific volume was maximum on the 20% substitution sourdough. Baking loss was inferior with the 80% substitution sourdough. Water activity and lightness was minimum on the control group. The higher amounts of sourdough showed the higher tendency of lightness, redness and yellowness. In the texture characteristics, hardness, gumminess, and chewiness was minimum on the control group and tended to increase with higher substitution of sourdough. Substitution of sourdough showed decrease adhesiveness. Fracturability, cohesiveness, and resilience of all rye-wheat mixed breads were not significantly different. Springiness was maximum on the 20% substitution sourdough and minimum on the 40%. A side of loaf of rye-wheat mixed breads, except for 20%, had a poor break and shred. In sensory evaluation, as substitution amount of sourdough increased, the scores of color and consistency of crumb, uniformity of crumb pore, gumminess, and overall acceptability decreased; while the density of crumb pore, springiness of crumb, aroma of rye flour, sourness, and bitterness showed the reverse effect; the 20, 40, and 60% sourdough samples obtained fairly good scores. In conclusion, these results indicated that $20{\sim}60%$ of sourdough could be very useful as a substitute for baker's yeast in developing rye-wheat mixed bread.
