Improvement of Sea Eel Pots (붕장어 통발의 개량)

  • Traditional sea Eel pots can be divided into two groups such as bamboo and plastic pots, however they are nearly same in a shape with one entrance and fishing efficiency, except their materials. Very few yet have been studied on their catching methods or catching mechanisms at the view point of behavior. Accordingly, we have designed tubular pots in order to fill up faults of traditional fishing gear construction and behaviors of sea eel. The suitable tubular pots was decided by comparative experiments in the water tank and the fishing efficiency was compared through the field experiments. The results obtained are as follows : 1. The differences between traditional plastic pots and improved tubular pots are firstly two entrances in both ends of tube without holes, secondly flapper nets are fixed at the end of each cone, and thirdly a bait bag is fixed at the center of pot. 2. The standard size of the suitable tubular pot is: $$Tube\;:\;\phi\;12\~13{\times}L80\;cm$$, $$Cone\;:\;Inside\;ring\;\phi6{\times}D5\;cm$$, Flapper : L10 cm. 3. The fishing efficiency of tubular rots is 2.3 times better than that of plastic pots.

  • There are more than 2,900 different species of termites in the world, and just there are 23 species in Japan. They caused more severe infestation compared with Korea. When a structure has become infested with termites, it is important that appropriate action must be taken: the chemical pest control (fumigation, soil termiticide treatment, bait system etc.) or the non-chemical pest control (low oxygen treatment, carbon dioxide treatment, high/low temperature treatment etc.). Especially, there were attempts to make practical protocol of various alternatives since the fumigant(methyl bromide) had been phased out in 2005 in Japan, and practically non-chemical methods would be effective alternatives for some cases, where the scale of infestation is small and limited and when long treatment is possible. But most of all, it is important that the process of pest control is made according to each species of termites with consideration for different characteristics of termites.

An Analysis of the Frequencies of the Saury Heads(bait) Retained in the Tuna Stomachs (다랑어 위내에 들어있는 꽁치 머리의 빈도 해석)

  • With an object to obtain an indication on the efficiency of the saury baits for tuna longline, frequencies of the saury heads found in the tuna stomachs were analysed by the equations developed from tile binomial distribution. Four factors were introduced into the equations : The hooking rate, p; rate of not being hooked q; rate of the effective baits retained in the stomachs of the captured tuna r; and the rate of tile previously taken baits retained in the tuna stomachs, t. The best estimates of $\frac{p}{p+q^t}$ and r are empirically obtained as follows. Yellowfin tuna: $\frac{p}{p+q^t}$=0.789, r=0.598 Bigeye tuna: $\frac{p}{p+q^t}$=0.810 r=0.608, Albacore tun : $\frac{p}{p+q^t}$=0.838, r=0.621.

Isolation, Identification and Characterization of Phytophthora katsurae, Causing Chestnut Ink Disease in Korea

  • Since July 2005, survey of chestnut ink disease was carried out in chestnut stands located at southern parts of Korea. Dead chestnut trees showing inky ooze on necrotic trunks were found in two different locations. In order to isolate and identify the causal fungus, infected tissues and soil samples around dead or dying trees were collected and placed on Phytophthora-selective medium. Rhododendron and chestnut tree leaves were used as a bait to isolate the fungus from soil samples by attracting zoospores in soil suspensions. On V-8 culture medium, the isolates produced homothallic oogonia with protuberances ($34.0-46.2{\times}21.9-26.7{\mu}m$) abundantly, but did not produced sporangia. Mass production of sporangia was possible by immersing agar plugs with actively growing mycelium in the creek water at $18^{\circ}C$ for 3 days. Sporangia were papillate, and ovoid to obpyriform ($17.0-38.9{\times}14.6-29.2{\mu}m$) in shape. Comparison of the ITS sequences revealed that the isolates had 100% identity to the P. katsurae isolates from Japan and New Zealand and 99.6% identity to other P. katsurae isolates. All of the examined isolates from Korea were completely identical to each other in ITS sequence. Numerous sporangia were formed in filtered as well as unfiltered creek water, but no sporangia formed in sterilized distilled water. Light induced sporangia formation, but has no influence on oospore formation. Amendments of ${\beta}$-sitosterol in culture media have no significant effect on mycelial growth but significantly stimulate oospore and sporangia formation.

Identification of CATHEPSIN B as a Novel Binding Protein of the Cell Death Regulating Protein IEX-1 (세포사멸 조절 단백질인 IEX-1의 새로운 결합단백질로서의 CATHEPSIN B의 발견)

  • Previously, we identified that the overexpression of IEX-1 induces apoptosis in ovarian cancer cells. Herein we report a new binding partner of IEX-1, CATHEPSIN B, as a lysosomal enzyme which contributes to the various apoptotic signaling in tumor cells. To investigate how IEX-1 regulates cellular survival and death event, we performed yeast two-hybrid screening of rat ovarian cDNA library using IEX-1 as the bait and found CATHEPSIN B. In the present study, CATHEPSIN B and IEX-1 proteins were overexpressed in 293T cells and their specific association was determined by immunoprecipitation and immunoblot analysis. In addition, the endogenous interaction between CATHEPSIN B and IEX-1 was confirmed in HeLa cells. The current finding of lysosomal CATHEPSIN B as the IEX-1-binding partner implies that IEX-1 may involve in lysosome-mediated apoptotic signaling pathways.

Selective Interaction Between Chloroplast β-ATPase and TGB1L88 Retards Severe Symptoms Caused by Alternanthera mosaic virus Infection

  • The multifunctional triple gene block protein 1 (TGB1) of the Potexvirus Alternanthera mosaic virus (AltMV) has been reported to have silencing suppressor, cell-to-cell movement, and helicase functions. Yeast two hybrid screening using an Arabidopsis thaliana cDNA library with TGB1 as bait, and co-purification with TGB1 inclusion bodies identified several host proteins which interact with AltMV TGB1. Host protein interactions with TGB1 were confirmed by biomolecular fluorescence complementation, which showed positive TGB1 interaction with mitochondrial ATP synthase delta' chain subunit (ATP synthase delta'), light harvesting chlorophyll-protein complex I subunit A4 (LHCA4), chlorophyll a/b binding protein 1 (LHB1B2), chloroplast-localized IscA-like protein (ATCPISCA), and chloroplast ${\beta}$-ATPase. However, chloroplast ${\beta}$-ATPase interacts only with $TGB1_{L88}$, and not with weak silencing suppressor $TGB1_{L88}$. This selective interaction indicates that chloroplast ${\beta}$-ATPase is not required for AltMV movement and replication; however, TRV silencing of chloroplast ${\beta}$-ATPase in Nicotiana benthamiana induced severe tissue necrosis when plants were infected by AltMV $TGB1_{L88}$ but not AltMV $TGB1_{L88}$, suggesting that ${\beta}$-ATPase selectively responded to $TGB1_{L88}$ to induce defense responses.

The Isolation of Keratinophilic Fungi from Soils in Chon-ju Area (전주근교의 토양에서 분리된 호각화질성 진균의 오염에 관한 조사)

  • The purpose of this study was to investigate the contamination rates of Keratinophilic fungi in soils In Chonju. From september to November 1986, 98 soil samples were collected at the school and children's play ground, dog barn, pig barn, cow barn, horse barn and fowl barn( total 33 sites ). The samples were collected at the different depths(0~2cm, 30cm, 50cm ) in each of the sites, respectively. Each sample was cultured at 25$^{\circ}C$ according to Modified hair baiting method by using horse mane hair as a bait. The results obtained were summerized as follows ; 1. Eighty one of the 98 soil samples were found to be positive for Keratinophilic fungi. 2. In the examination of keratinophilic fungi in 98 soil samples, the organisms isolated weie as follows; Microsporum gypseum 20, Trichophyton ajelloi 15, Chrysosporium tropicum 40, C. Keratinophilum 24, C. tuberculatum 6 and Chrysosporium spp. like organism 4 strains. 3. The positive rates of keratinophilic fungi in each of the depths were 87.9%(29/33) in surface layer, 90.9%(30/33) in middle layer and 68.8%(22/32) in deep layer. 4. The positive rates of Keratinophilic fungi in each of the sites were 100%(3/3) in horse barn 91.7% (l1/12) in dog barn, 88.9%(16/18) in pig barn, 86.7%(13/15) in cow barn, 76.2%(16/21) in fowl barn and 75.9%(22/29) in school and children's play ground. 5. The isolation rates of M. gypseum, Pathogenic fungi for human and animals, were as follows ; 58.3% in dog barn(surface layers 100%, middle layers 50%, deep layers 25%) and 16.7% in pig barn(surface 33.3%, middle 16.7%, deep 0%). In the cow barn, the isolation rates were 13.3%(surface 40.0%, middle 0%, deep 0%) and 10% in horse barn(surface l00%, middle 100%, depp 100%). In the fowl barn, the isolation rates was 19.1%(surface 28.8%, middle 14.3%, deep 14.3%) and 3.5% in school and children's play ground(surface 0%, middle 0%, deep 11.1%).

  • Rats and Cockroaches are the most problematic pests of our living environment, Rats generally fall under three categories, namely, the Norway rats, the roof rats and the mouses, Among these three types, the roof rats are increasing rapidly in number and their habitats are spreading quickly in the city area, where as the Norway rats are decreasing both in number and habitat. There are numerous damages that these pests create in our society, The rat induce damages to computers and other information processing equipments, sometimes cause light-out, and other financial losses and other sanitary problems. The cockroaches, apart from inducing serious sanitary problems, is an insect that creates insectophobia. decently, it has been revealed that cockroaches are also the main carriers of allergic diseases. Among the pesticides employed against rats in Korea, Beta - fluoro acetate(1080) was initially used after the National Liberalization. Later, the Korean government imported anticoagulant rodenticide for its control. This was followed by a brief, unsuccessful utilization of Zinc Phosphide. Since the begin of 1980s, with the advent of pest control companies, first generation anticoagulant rodenticide were developed in Korea and recently followed by a widespread use of pesticide that utilizes second generation anticoagulant material, the "Brodifacoum" . Among insecticides employed against cockroaches, "Fenitrothion" "Dusban" "DDVP" "Diazinon" and others have been used. They were gradually being replaced by the use of synthesised pyrethroid insecticides until recently. Today there is a wide spread use of bait poisons, the "hydramethylnon" Nevertheless, full prevention and control can not be realized with the sole use of the pesticides. In the future, hygine and proofing, combined with affective pest management, must be incorporated in order to bring about more satisfactory and fruitful preventive measures.

Applicability Study on Reticulitermes speratus kyushuensis (Isoptera: Rhinotermitidae) Colony Eliminator to Preserve Wooden Cultural Heritage (일본흰개미 아종 모니터링 및 군체제거를 위한 예찰제어기 개발 연구)

  • This study is conducted to develop new termite colony elimination system that can control termite colony much faster, and to eco-environmentally prevent termite damage occurred in wooden cultural heritage. As a result of laboratory test, we developed a component system, of which fipronil 0.001% (w/w) treated bait was used as a suitable termite colony eliminator. This system can be monitored without taking off underground, and it makes regular monitoring much more efficient. The result of field test showed that 36 termite baiting devices among 367 installed devices were damaged by foraging termites. After baiting, all of termite colonies attracted to devices were eliminated or their activity clearly decreased.

Elasticity of the Funnel Ribs and Hydrodynamic Characteristics on the Sea Eel Pots (장어통발의 깔대기 탄성과 유체역학적 특성)

  • The basic experiments on the plastic sea eel pots used in fishing were carried out in order to investigate the elasticity of the funnel ribs, hydrodynamic resistance, sinking time and diffusion of the bait from June to October, 1987. The elasticity of the bamboo funnel ribs was higher than that of the polypropylene ribs up to the load 150g. The hydrodynamic resistance R (kg) of the pots towing to the head direction horizontally in relation to towing velocity V (m/sec) was expressed as following formula; R=0.36V super(2.01) and coefficient of drag C sub(D) was 0.52. The sinking times of the covered pots by tape fully or partially were late 1-2 second than the typical pots within the water depth 7.5m. The diffusion tendency through the covered pots using dye and sardine extracts solution was concentrated to the entrance more than the typical pots. However, fishing efficiency as number and weight of fish per pots for nine times fishing operation was revealed no difference between the covered pots and the typical pots.
