HtrA2 Interacts with Aβ Peptide but Does Not Directly Alter Its Production or Degradation

  • HtrA2/Omi is a mammalian mitochondrial serine protease homologous to the E. coli HtrA/DegP gene products. Recently, HtrA2/Omi was found to have a dual role in mammalian cells, acting as an apoptosis-inducing protein and being involved in maintenance of mitochondrial homeostasis. By screening a human brain cDNA library with $A{\beta}$ peptide as bait in a yeast two-hybrid system, we identified HtrA2/Omi as a binding partner of $A{\beta}$ peptide. The interaction between $A{\beta}$ peptide and HtrA2/Omi was confirmed by an immunoblot binding assay. The possible involvement of HtrA2/Omi in $A{\beta}$ peptide metabolism was investigated. In vitro peptide cleavage assays showed that HtrA2/Omi did not directly promote the production of $A{\beta}$ peptide at the ${\beta}/{\gamma}$-secretase level, or the degradation of $A{\beta}$ peptide. However, overexpression of HtrA2/Omi in K269 cells decreased the production of $A{\beta}40$ and $A{\beta}42$ by up to 30%. These results rule out the involvement of HtrA2/Omi in the etiology of Alzheimer's disease. However, the fact that overexpression of HtrA2/Omi reduces the generation of $A{\beta}40$ and $A{\beta}42$ suggests that it may play some positive role in mammalian cells.

Study on the Intensive Catching Method of Anchovy for Live Bait-III Relation Between Variation of Sea Condition and Catch of Anchovy in the Southern Coast of Korea (활멸치의 집약적 생산수단에 관한 연구 -III)

  • This paper was analysed based on the oceanographic and meteorological data complied from 1971 to 1977 for that search relationships among the fluctuation of sea condition and weather condition, and the catch of anchovy. In the year when heat loss from the sea surface in winter was maximum(in 1974, 658 Iy), temperature of midwater in summer was lower 2~4\ulcornerC than normal year. While heat loss was minimum (in1973, 487 Iy), temperature of mid water was higher 2\ulcornerC. When temperature of mid water of southern coast from June to August was higher than normal year, anchovy was caught good deal, but that was lower than normal year was bad fishing. When it had much precipitation (in 1973, 256mm), plankton was checked maximum (12cc) and also the catch of anchovy too (11, OOOm/t). While precipitation was minimum (in 1976, 123mm), plankton (3cc) and anchovy (2, 800m/t) was a litle. If we calcalate heat budget in winter, we can forecast temperature of mid-water in summer of following year. Therefore we may be able to forecast catch anchovy.

Malaria transmission potential by Anopheles sinensis in the Republic of Korea

  • To evaluate the factors that determine the transmission level of vivax malaria using vectorial capacity, entomological surveys were conducted from .lune to August, 2000. From 6 nights of human-bait collection in Paju, the human biting rate (ma) was counted as 87.5 bites/man/night. The parity of Anopheles sinensis from human baiting collections fluctuated from 41% to 71% (average 48.8%) of which the rate gradually increased as time passed on: 35.2% in Jun. ; 55.0% in July; 66.2% in Aug. From this proportion of parous, we could estimate the probability of daily survival rate of An. sinensis to be 0.79 assumed with 3 days gonotrophic cycle and the expectancy of infective life through 11 days could be defined as 0.073. Blood meal analysis was performed using ELISA to determine the blood meal source. Only 0.8% of blood meals were from human hosts. We could conclude that An. sinensis is highly zoophilic (cow 61.8%) Malaria is highly unstable (stability index < 0.5) in this area. From these data, vectorial capacity VC) was determined to be 0.081. In spite of a high human biting rate (ma), malaria transmission potential is very low due to a low human blood index. Therefore, we could conclude that malaria transmission by An. sinensis is resulted by high population density, not by high transmission potential. For this reason, we need more effort to decrease vector population and vector-human contact to eradicate malaria in Korea.

Structure and Function of NtCDPK1, a Calcium-dependent Protein Kinase in Tobccco

  • We have isolated a cDNA encoding a calcium-dependent protein kinase (CDPK) in Nicotiana tabacum, which was designated NtCDPK1. Accumulation of the NtCDPK1 mRNA was stimulated by various stimuli, including phytohormones, CaCl$_2$ wounding, fungal elicitors, chitin and methyl jasmonate. The NtCDPK1 gene encodes a functional Ser/Thr protein kinase of which phosphorylation activity is strongly induced by calcium. By analyzing expression of the NtCDPK1-GFP fusion protein and by immunoblotting with antibody which reacts with NtCDPK1, we found that NtCDPK1 is localized in membrane and nucleus in plant cells. Silencing expression of the NtCDPK1 transgene resulted in marked decrease of lateral root development in the transgenic tobacco plants. Yeast two hybrid screening using NtCDPK1 as a bait identified a tobacco homologue of proteasome regulatory subunit 21D7, designated Nt21D7. The 21D7 mRNA has been shown to be predominantly expressed in proliferating tissues in the cell cycledependent manner in carrot. The recombinant NtCDPK1 protein associated with Nt21D7 in vitro, and could phosphorylate the Nt21D7 protein in vitro in the presence of calcium, suggesting that Nt21D7 protein is a natural substrate of NtCDPK1 in tobacco. These results suggest that NtCDPK1 may regulate tell proliferation processes, such as lateral root formation, by regulating specificity and/or activity of proteasome-mediated protein degradation pathway.

A Development on Assessment Method of PVC Gloves Used in Pest Control Program (해충제어프로그램에서 사용되는 PVC장갑의 평가방법 개발)

  • Following a Mediterranean fruit fly outbreak in South Australia, a bait spray program involving the pesticides like malathion(MAL) was carried out. During the application, dermal exposure was considered for the pest controllers wearing PVC gloves. However there is a lack of information about PVC glove performance like break through times and permeation rates with MAL, therefore, a new analytical method for HPLC-UV was developed. A standard permeation test cell was used in this study. From the results of this study, more than 96% solubility of MAL was provided at 30% isopropyl alcohol in distilled water as a collecting media. However, there was significant decomposition of MAL when the solutions were kept at over $50^{\circ}C$ for 2-3 hours. As a mobile phase, 50% acetonitrile water solution (pH 6.0) gave the greater sensitivity compared with other compositions of acetonitrile solution. The arm section of the gloves had shorter breakthrough times and higher permeation rates compared with the palm. There was no malathion solution breakthrough up to 24 hours using the 1% MAL working strength solution. When the temperature was changed from $22{\pm}1^{\circ}C\;to\;37{\pm}1^{\circ}C$, the breakthrough times were decreased by 14.5% on palm and 37.5% on arm, and permeation rates were increased significantly. The findings of this study indicate that further investigations on used gloves, periods of use and varying working conditions like tasks and seasons should be carried out to assess potential worst case scenarios.

Interaction between IgE-Dependent Histamine-Releasing Factor and Triosephosphate Isomerase in HeLa Cells (HeLa 세포에서 IgE-dependent Histamine-Releasing Factor와 Triosephosphate Isomerase의 상호작용 규명)

  • IgE-dependent histamine-releasing factor (HRF) is found extracellularly to regulate the degranulation process of histamine in mast cells and basophils and known to play a predominant role in the pathogenesis of chronic allergic disease. HRF has been also identified in the intracellular region of the cell. Previously, we reported that HRF interacts with the 3rd cytoplasmic domain of the alpha subunit of Na,K-ATPase. To understand the molecular mechanism of the regulation of Na, K-ATPase activity by HRF, we investigated the interaction between HRF and TPI since TPI was obtained as HRF-interacting protein in HeLa cDNA library, using yeast two hybrid screening. Domain mapping study of the interaction between HRF and TPI revealed that the C-terminal region of the residue 156-249 of TPI is involved in the interaction with HRF. The interaction between HRF and TPI was confirmed by immunoprecipitation from HeLa cell extracts. Our results suggest that TPI is a HRF-binding protein and the interaction between HRF and TPI nay thus affect Na, K-ATPase activity.

Insecticidal Activity of Metarhizium anisopliae FT83 against the Different Stages of Beet Armyworm, Spodoptera exigua (곤충병원성 곰팡이 Metarhizium anisopliae FT83의 파밤나방 생육단계별 살충활성)

  • The beet armyworm, Spodoptera exigua is pest which is difficult to control. For eco-friendly beet armyworm managements, we isolated entomopathogenic fungi from soil samples by insect-bait method using Tenebrio molitor and conducted bioassay to larvae of beet armyworm. The result of bioassay, a selected strain Metarhizium anisopliae FT83 caused 100% mortality against first ~ third instar larva of S. exigua at $1{\times}10^8conidia/ml$ and medial lethal time ($LT_{50}$) were 0.5 days, 2.6 days and 2.5 days respectively. Mortality against fourth and fifth larvae were $83.3{\pm}6.2%$ and $86.0{\pm}5.7%$ and medial lethal time ($LT_{50}$) were 4.2 days and 3.6 days respectively. Mortality against pupae and eggs of S. exigua were 100%. M. anisopliae FT83 showed high virulence at all developmental stages of S. exigua.

Behavioural Characteristics of Walleye Pollack Theragra chalcogramma by Acoustic Sound Conditioning (음향 순치에 의한 명태의 행동 특성)

  • It is most important to understand the behaviour of fish in case of the betterment of the current fishing gear and methods or the development of the conditioning by acoustic sound in marine ranching. This investigation has been attempted to provide for the prediction of the response action of fish to acoustic sound. The experimental fish was conditioned with sound and bait. As the acoustic sound for stimulus, the pure tone of sine waveform at the frequency of 200Hz was used. This pure tone was determined from previous investigation about hearing ability of walleye pollock Theragra chalcogramma. The fork length of walleye pollock used in this experiment was 385~450mm. The conditioning proceeding was recorded in the video tape recorder. Frequency of appearance in the feeding area was analyzed with computer and video tape recorder. The position of fish was tracked using the mouse cursor and picture mixed on the superimpose board. The response of conditioned fish to sound stimulus was appeared in the 8th day firstly. The conditioned fish remembered the stimulus sound for 4 days. Average frequency of appearance in the feeding area during the 30 seconds sound projection or 1 minute after the sound stimulus was 51%, and was higher than before it.

Octopus fisheries in the coastal waters of Gangneung- II - Octopus drift-line fishery - (강릉시 연안 문어어업에 관한 연구- II - 문어흘림낚시어업 -)

  • A survey on the fishing gear for octopus drift-line fishing was done in Gangwon-do, Sacheon, Gyeongnam and Hokkaido, Japan, while a survey on the environments of fishing grounds used data from January to August from the Korea east coast farming forecast system of Donghae Fisheries Research and Development Institute. The present situation of fishing was examined with boats engaging in drift-line fishing from March to August in the coastal waters along Gangneung. The fishing tackle for octopus drift-line fishing was made manually, and the size, shape, and weight of the hook and number of shooting used vary according to the fishing time and region. Lead is used as the material for sinkers. As bait, pork fat with skin is mostly used in Gangwon-do. The temperature of the bottom water layer in the coastal fishing ground of Gangneung from April to June ranges from $3.2-12.4^{\circ}C$, which is the optimal temperature for octopuses. During July and August, the temperature ranges from $5.0-20.6^{\circ}C$. The maximum difference between day and night temperatures reached up to $9.2^{\circ}C$. Salinity is generally stable at $33.2-35.324.6%_{\circ}$, which does not affect the inhabitation of octopuses. The octopus catch reached its peak from May to July, while most octopuses weighed less than 1 kg (76.7%). The results show that it is effective to carry out octopus drift-line fishing up to a depth of 40 meters; and that the maximum number of octopuses per line is obtained with an operation time of six hours.

Determination factors for catch rate of the target species between circle hook and straight shank hook in the Korean tuna longline fishery (우리나라 다랑어연승어업에 있어서 환형낚시와 재래식낚시를 사용하여 목표종의 어획률을 결정하는 요인 분석)

  • We conducted experiments to compare the catch rate of bigeye tuna and yellowfin tuna between circle hooks and straight shank hook in the Korean tuna longline fishery at the eastern and central Pacific Ocean from 2005 to 2007. We analyzed difference of fork length, survival and hooking location between a circle hook and a straight shank hook for both tunas, respectively. There was no difference in the mean fork length size of yellowfin tuna caught on the two type of hook but bigeye tuna was significant. In case of survival, there was no difference between two hook type, but the difference of hooking location was significant for both species. We also analyzed to find determinants of both tunas catch rate using generalized linear models (GLMs) which were used latitude, longitude, year, month, depth, hook type, bait type and so on as independent variables. Spatial factors, latitude and longitude, and temporal factors, year and month, affected catch rate of bigeye tuna and yellowfin tuna. And also, depth such as a marine environment factor was influenced on catch rate.
