Isolation and Identification of Pathogenic Microorganisms from Soybean Sprouts

  • Raw soybean sprouts were tested for contamination with the following bacteria which have potential for pathogenesis or food spoilage : Salmonella spp., Escherichia coli O157:H7, Yersinia enterocolitica, Vibrio parahae-molyticus, Aeromonas hydrophila, Plesidomonas shigeloides, Pseudomonas aeruginosa, Staphylococcus aureus, Lis-teria monocytogenes, Bacillus cereus, Clostridium perfringens, Campylobacter jejuni, Erwinia spp., and Fusarium spp. Three of the above strains were isolated from the sprouts, and identified by morphological and biochemical methods including an API kit and ATB automated identification system. The isolate cultured in Cereus selective agar, a selective medium, was a Gram-positive, rod shaped, anaerobic spore former. The biochemical and culture tests revealed the following characteristics: catalase-positive, no growth on Simmon's citrate, NO₂ production and requirement of arginine for growth; the ATB automated identification system gave 99.8 % agreement for the identification of Bacillus cereus to the species level. The isolate cultured in Macconkey agar selective medium was Gram-negative, rod shaped and a gas former; the ATB-system gave 99.9% agreement for the identification of Aeromonas hydrophila to the species level. The isolate found in Pseudomonas isolation agar was Gram-negative, rod shaped, cytochrome oxidase-positive, a reducer of nitrates to nitrogen, and pyocyanin producer; the ATB-system gave 99.9 % agreement for the identification of Pseudomonas aeruginosa to the species level. These results indicate that the three bacteria species present in the soybean sprouts were Bacillus cereus, Aero-monas hydrophila, and Pseudomonas aeruginosa. Salmonella spp., Escherichia coli O157:H7, and Yersinia enter-ocolitica, which are associated with serious disease in humans, were not isolated from soybean sprouts examined in this study.

인천지역 대기 환경 중 배양성 세균의 특성 (Characterization of Culturable Bacteria in the Atmospheric Environment in Incheon, Korea)

  • Objectives: This study aims to provide basic data regarding the bacterial total plate count in the atmospheric environment for related studies. Methods: Total plate count and the identification of culturable bacteria in the atmospheric environment in Incheon took place in 2015 using periodic survey. Correlationship analysis was performed between the number of culturable bacteria and environmental elements. In addition, an estimation of novel bacterial species was undertaken using the similarities and phylogenetic tree based on the 16S rRNA gene. Results: The total plate count of culturable bacteria was on average $176CFU/m^3$, and did not exceed $610CFU/m^3$ in the atmospheric environment. Periodic monthly measuring of total plate count was highest in June at $293CFU/m^3$, while the lowest was in July at $125CFU/m^3$. Furthermore, as a result of the identification of culturable bacteria, the genera Arthrobacter and Kocuria were dominant, while novel bacterial taxa that belong to the genera Chryseobacterium and Herbiconiux were separated. Conclusion: The total number of culturable bacteria from the atmospheric environment in Korea is on average $176CFU/m^3$. In addition, the genera Arthrobacter and Kocuria dominate. The presence of novel bacterial taxa are expected in the atmospheric environment, such as belonging to the genera Chryseobacterium and Herbiconiux.

김치로부터 분리한 젖산균의 다양성과 항균성 (Diversity and Antibacterial Activity of Lactic Acid Bacteria Isolated from Kimchi)

  • 본 연구는 김치로부터 분리한 젖산균의 다양성과 항균성을 조사하기 위하여 수행되었다. 희석한 김치 국물을 CaCO$_3$가 첨가된 MRS 고체 배지에 도말 한 후, $25^{\circ}C$에서 2일 동안 배양하여 여러 가지 김치로부터 27가지의 젖산균을 분리하였다. 이렇게 분리한 젖산균들을 Bergey 분류법과 BPB 배지를 통한 생화학적인 방법과 16S rDNA PCR을 이용한 분자 생물학적인 방법으로써 분석하였다. 그러나 이들 방법으로 분석한 결과가 일치하지 않은 경우가 많았다. 이들 분리 젖산균은 16S rDNA 부분 염기서열을 결정한 결과 Leuconostoc mesenteriodes(11균주), Leu. carnosum(3균주), Lactobacillus curvatus(8균주), Lac. pentosus(2균주), Weisselia kimchi(1균주), W. cibaria(1균주), Pediococcus pentosaceus(1균주) 등의 것과 각각 매우 유사한 것으로 분석되었다. 이들중 Leu. carnosum은 이전에 김치에서 분리되었다고 보고된 바가 없다. 그리고 이들 김치에서 분리 동정한 균들의 항균작용 활성을 검사 해 본 결과 균주에 따라서 활성의 정도는 다르게 나타났으나 Bacillus subtilis, Escherichia coli, Salmonella enteritidis, S. paratyphica, S. typhi, Staphylococcus aureus, Shigella boydii, S. sonnei 등에 대해 뚜렷한 항균효과를 나타내었다.

냉면육수에서 분리한 대장균군의 동정 및 저온 증식성 (Identification of Coliform Bacteria Isolated from Nangmyun-Broth in Korea and Psychrotrophic Character)

  • The nangmyun is a Korean iced noodle made by putting buckwheat-noodles into cold broth. Seven samples of nangmyun-broths were collected from Korean restaurants in Bucheon during July 1994. The contamination levels of nangmyun-broths by coliform bacteria were determined, and then the 40 colonies of coliform bacteria, isolated randomly from 4 samples of nangmyun-broths, were identified at genus or species level with the additional test for psychrotrophic character The coliform counts in nangmyun-broths were 6.0x102-6.5$\times$104/ml(average 2.3$\times$104%). Among the 40 strains of isolates, 27 strains(67.5%) were identified as the genus Klebsiella, 9 strains(22.5%) as the genus Enterobacter, 2 strains(5.0%) as the genus Citrobacter and 2 strains (5.0%) as the genus Escherichia. Among 27 strains of Klebsiella, 11 strains (40.8%) were identified as K planticola, 4 strains(14.8%) as K. pneumoniae, 2 strains(7.4%) as K. ozaenae and 2 strains(7.4%) as K. terrigena, but 8 strains(29.6%) of a typic\ulcorner1 Klebgiella could not be Identified at species level. All the 40 strains of coliform bacteria were psychrotrophs showing slow growth at 1$0^{\circ}C$, and 18 strains(45%) grew at 5$^{\circ}C$. It was thought to be a good basic data In describing the reason for too high coliform counts in nangmyun-broths that all coliform bacteria tested In this study were psychrotrophs.

느타리 재배에서 버섯파리의 생물학적 방제를 위한 곤충병원성 세균의 분리 및 동정 (Isolation and Identification of Entomopathogenic Bacteria for Biological Control of the Mushroom Fly, Lycoriella mali)

  • 느타리버섯 재배에 있어서 주요해충인 버섯파리 Lycoriella mali의 생물학적 방제를 위하여 전국 느타리버섯 재배지와 버섯파리의 병사충에서 버섯파리 유충에 병원성을 갖는 세균 8균주를 분리하였다. 이 중 Bti-D 및 Bti-U 2균주에 의한 사충율이 각각 82.3%와 87.3%로 가장 높았다. 버섯파리 유충의 발육단계에 따른 살충 효과 검정에서는 두 균주 모두 3령 층에서 높은 살충효과를 보였다. 이들 세균으로 감염된 유충에 나타나는 병징으로서 감염초기에는 중장의 앞부분이 연한 갈색을 띄고, 차츰 중장의 뒷부분까지 진행되어 전 부분이 흑갈색으로 변색되면서 치사되었다. 이들 두 균주를 동정하기 위하여 배양적, 생화학적 및 생리적 특성을 조사하고 Bergey's manual과 Biolog system을 동정에 이용하였으며, 주사전자현미경으로 세포형태를, 위상차현미경으로 내생포자와 내독소를 관찰하여 판정한 결과, 두 균주 모두 Bacillus thuringiensis로 동정되었으며, 편모항원성을 조사한 결과, B. l. subsp. israelensis로 동정되었다.

  • We isolated and cultured bacteria that inhabited marine biofilms, and identified them by phylogenetic analysis using 16S rDNA sequences. In the marine environment, biofilms cover most subtidal and intertidal solid surfaces such as rocks, ships, loops, marine animals, and algae. The bacteria in most biofilms are embedded in extracellular polymeric substances that comprise mainly of exopolysaccharides. The exopolysaccharides are excreted from multiple bacterial species; therefore, biofilms are a good source for screening exopolysaccharide-producing bacteria. Thirty-one strains were cultured, and a total of 17 unique strains were identified. Phylogenetic analysis using 16S rDNA sequences indicated that the 17 strains belonged to ${\alpha}$-Proteobacteria (Ochrobactrum anthropi, Paracoccus carotinifaciens); ${\gamma}$-Proteobacteria (Pseudoalteromonas agarovorans, P. piscicida, Pseudomonas aeruginosa, Shewanella baltica, Vibrio parahaemolyticus, V. pomeroyi); CFB group bacteria (Cytophaga latercula, Tenacibaculum mesophilum); high GC, Gram-positive bacteria (Arthrobacter nicotianae, Brevibacterium casei, B. epidermidis, Tsukamurella inchonensis); and low GC, Gram-positive bacteria (Bacillus macroides, Staphylococcus haemolyticus, S. warneri).

침출수를 이용한 미생물 제제의 활성에 관한 연구 (Study on the Activation of Microbial Products by Using the Leachate)

  • Activation bacteria, identified from commercial microbial products, were applied to leachate treatment. Total seven strains of bacteria Enterobacteriaceae spp. (5), Bacillus sp. (1), Aeromonas sp. (1) were seeded in the leachate and cultured in the shaking incubator at 25$^{\circ}$C and 250 rpm. While cultured, they were sampled in given time intervals and the removal rates of SS, COD, BOD, T-N.and T-P were measured an indicators of leachate treatment. Through the screening test, four of 7 strains of bacteria were considered to be effective and they were named as "effective group". The capability of leachate treatment was observed on three different groups of bacteria single, effctive, and total mixed. The result showed that the removal rates of COD and SS for the total mixed group were 64 and 71% respectively. BOD removal rate was reached nearly 99% by seeding of effective griup and removal rates of T-P and T-N were 83 and 82% respectively. However seeding of single strain was less effective than that of any mixed group in leachate treatment.

간장의 미생물학적 연구 (Microbiological Studies on Soysauce Isolation and Identification of Bacteria from soysauce to brew by Conventional Procesa)

  • Ten(10) strains of aerobic bacteria and two(2) strains of microaerobic bacteria were isolated out of soysauce which was brewed by the conventional process. The following bacteria were identified by studying their morphorogies and physiological characters. Bacillus pumillus-R-2; Bacillus subtilis var., aterimus-S-1; Bacillus licheriformis var-S-2; Bacillus subtilis-T-1; Sarcina maxima-T-2; Pediococcus acidi lactici-Z-2; Bacillus citreus var, . soyaB-Z-5. T-2 and Z-5 of the isolated bacteria were found good in growth even in the 24%-salted density, and Z-5 was more vigorous than T-2 though stinking. S-1 produced black-brown pigment from the medium containing various kinds of carbohydrate and the medium of soysauce which are available to S-1.

Isolation and identification of blacken spoilage inducing bacteria from Korean dried persimmon

  • Blacken spoilage is one of the common problems during the cold storage of dried persimmon in Korea. We collected the spoiled dried persimmon in the refrigerator and classified them to 4 types depending on their appearances. Furthermore we isolated blacken spoilage inducing bacteria from type D of dried persimmons. Among the isolates we identified the seven blacken spoilage inducing bacteria. They are Aeromonas hydrophila DP1, Cedecea davisae DP2, Ewingella americana DP3, Flavimonas oryzihabitans DP4, Providencia rettgeri DP5, Providencia rustigianii DP6 and Serratia plymuthica DP7. Strains were identified based on their morphological, cultural and physiological properties. We also found that Ewingella americana DP3, Flavimonas oryzihabitans DP4 were the major blacken spoilage inducing bacteria during dried persimmon storage.
