Changes in Gut Microbial Community of Pig Feces in Response to Different Dietary Animal Protein Media

  • Beef, pork, chicken and milk are considered representative protein sources in the human diet. Since the digestion of protein is important, the role of intestinal microflora is also important. Despite this, the pure effects of meat and milk intake on the microbiome are yet to be fully elucidated. To evaluate the effect of beef, pork, chicken and milk on intestinal microflora, we observed changes in the microbiome in response to different types of dietary animal proteins in vitro. Feces were collected from five 6-week-old pigs. The suspensions were pooled and inoculated into four different media containing beef, pork, chicken, or skim milk powder in distilled water. Changes in microbial communities were analyzed using 16S rRNA sequencing. The feces alone had the highest microbial alpha diversity. Among the treatment groups, beef showed the highest microbial diversity, followed by pork, chicken, and milk. The three dominant phyla were Proteobacteria, Firmicutes, and Bacteroidetes in all the groups. The most abundant genera in beef, pork, and chicken were Rummeliibacillus, Clostridium, and Phascolarctobacterium, whereas milk was enriched with Streptococcus, Lactobacillus, and Enterococcus. Aerobic bacteria decreased while anaerobic and facultative anaerobic bacteria increased in protein-rich nutrients. Functional gene groups were found to be over-represented in protein-rich nutrients. Our results provide baseline information for understanding the roles of dietary animal proteins in reshaping the gut microbiome. Furthermore, growth-promotion by specific species/genus may be used as a cultivation tool for uncultured gut microorganisms.

Dual Culture Inoculation Enhanced Quality of Silage Produced from Leguminous Plants

  • Ensiling is the most preferred technology to preserve the silage quality with high nutrients by the presence of lactic acid bacteria. In this study, lactic acid bacteria RJ1 and S22 were used to make the silages from different leguminous plants such as alfalfa, hairy vetch and red clover. Experimental groups were divided into control and LAB inoculated groups. LAB inoculated group; all legumes treated with a mixture of RJ1 and S22 and made an anaerobic condition for 45d. Without the addition of LAB considered the control group. The results showed that the lactic acid content was higher in all silages in response to LAB treatment and acetic acid content was slightly increased except red clover by LAB compared to control silages. A poor silage quality marker butyric acid was reduced all legume silages in response to LAB inoculation than control silages. The organic acid is closely associated with microbial population experimental silages. We noted that higher LAB and lower yeast were found in the silage in response to LAB treatment. The contents of crude protein (CP), acid detergent fiber (ADF), neutral detergent fiber (NDF), and total digestible nutrient (TDN) were not altered significantly between control and LAB treated silages. Overall data suggested that the inclusion of additional LAB potentially enhance the silage quality and preserved the nutrients for long period.

Characteristics of Heterotrophic Bacterial Population in the Artificial Lake Geumgang Near Estuary Barrage (금강 하구둑 인근에서 미생물군집의 특성)

  • The monthly variations of physico-chemical and microbiological water quality were investigate in the artificial Lake Geumgang near estuary barrage. Sixty heterotrophic bacteria were isolated and identified by amplification and sequencing of 16S rDNA. Water temperature, pH, and inorganic nutrients($NH_4$-N, $NO_2$-N, $NO_3$-N, $PO_4$-P) were measured. Concentrations of DO, BOD, and inorganic nutrients were lower than in the middle-stream of Geum river The population densities of heterotrophic bacteria and total coliforms varied from $4.1{\pm}1.0\times10^2$ to $6.7{\pm}1.1{\times}10^3\;cfu\;ml^{-1}$, and 0 to $2.3{\pm}0.6{\times}10^2\;cfu\;ml^{-1}$, respectively. Among the measured numbers of physiological groups of bacteria, cellulolytic bacteria showed higher population densities than those of other physiological groups. Bacterial community structure was analysed based on 16S rDNA partial sequencing. Among 60 isolates, dominant genus was Pseudomones (20 strains).

Distribution of Heterotrophic Bacteria and Extracellular Enzyme Activities of Bacteria in the Sediment of South Sea, Korea (남해 퇴적토에서 종속영양 세균의 분포 및 세포의 효소 활성력)

  • In the periods of July 31 to August 10. 1988 and March 9 to 13. 1989. sediment samples were collected from the South Sea stations (010] to 092]) located in the area from $N 32^{\circ}$/30' to $34^{\circ}$/30', of latitude and from E $123^{\circ}$ 30' to $128^{\circ}$30' of longitude. These samples were analyzed for the number of total heterotrophic bacteria and extracellular digesting enzyme activities. In the 1989 spring period the number of heterotrophic bacteria in the sediment surface layer was increased more than 100 times at the maximum compared to that in the 1988 summer period. The proportion of fresh water bacteria to total heterotrophic bacteria was also higher in the spring period than the summer period. The extracellular digesting enzyme activities were higher in spring season than summer. Although the water content of sediment in the spring period was lower than that the summer period. the ash weight indicating organic material content was higher. These results means that the diameters of sediment particles were larger in spring than summer but the input of organic material into the sediment was greater. Based on these results bacterial distributions in the sediment layer of South Sea depend greatly on the season due to the effect of fresh water. During the spring season plankton could grow extensively owing to the inorganic nutrients input by the vertical mixing in the water column, then be precipitated into the sediment. Organic nutrients supplied from enzymatic degradation of polymeric particle from plankton can increase the bacterial number, too.

Distribution of heterotrophic bacteria and physico-chemical characteristics of sediments in Kum river estuary (금강 하구 퇴적토의 이화학적 성질과 종속영양세균의 분포에 관하여)

  • Vertical distribution of heterotrophic bacteria and physico-chemical characteristics were measuted in Kum River estuarine sediments. And interrelationship between heterotrophic bacterka and environmental factors was also studied. The type of sediment of Site 1 was silty clay, and sand at Site 2. Annual pH ranges were between 7.1 and 7.7 in the clay type sediment (Site 1) and 6.9-7.2 in the sand type sediment (Site 2). It was shown that organic matter contents were higher in the clay type sediment than those of sand type sediment. Redox potential values of sediments were decreased rapidly with depth at Site 1, but those of Sete 2 showed vertical fluctuation. Nitrogens(ammonia+amino acid-N, nitrate-N, nitrite-N) and phosphate in the clay type sediment showed higher values than those of sand type sediment. Annual distribution of heterotrophic bacteria were ranged $6.71{\times}10^4$ cells/g dry wt. $-2.50{\times}10^6$ cells/g dry wt. In the clay type sediment and $2.67{\times}10^3$ cells/g dry wt. $-1.94{\times}10^6$ cells/g dry wt. in the sand type sediment. Distribution of proteolytic, lipolytic, and amylolytic bacteria were decreased with the depth and the highest density was found in April and the lowest in January. Bacterial populations in sediments were closely correlated with such environmental factors as pH, redox potential, moisture content, organic matter contents, and inorganic nutrients such as nitrite-N and phosphate-P.

Changes in Antioxident Nutrients and Vitamin $B_1$ Contents of Gamma Irradiated Foods Consumed most Frequently in Korea (한국인 다소비 섭취 식품의 방사선 조사후 항산화 영양소와 비타민 $B_1$의 변화)

  • Food irradiation has been steadily increased in many countries concomitantly with increasing international trades and concerns about naturally occurring harmful contaminants of food. Although, irradiation provides an excellent safeguard for the consumer by destroying harmful bacteria, it would be necessary to investigate nutritional adequacy of irradiated foods with a special emphasis on such foods as red pepper powders, cold-water fishes (mackerel), sesame seeds that are widely consumed in Korea. These food items are of special concern due to the fact that most of them are imported and that several nutrients contained in them are sensitively reacting to irradiation. We observed changes in the antioxidant nutrients contents, such as vitamin A, $\beta$-carotene, vitamin C and total phenol, and also vitamin B$_1$ in the foods that were irradiated at different dose levels. The nutrients contents in red pepper were respectively 98.6% and 92.3% for total phenol, and 97.8% and 94.5% for $\beta$-carotene, right after irradiation at the dose of 10 and 20 kGy. The vitamin C contents decreased with irradiation doses up to 76.4% (10 kGy) and 68.5% (20 kGy) and B$_1$ contents decreased to 54.9% and 80.7% of non irradiated red pepper. When the mackerel was irradiated at the applied dose of 3 and 7 kGy, total phenol concentration of the irradiated fish was respectively 91.3% and 94.2% of non irradiated fish. The amounts of vitamin A were 82.6% (3 kGy) and 78.0% (7 kGy) of unirradiated sample and vitamin B$_1$ contents reduced to 30.5% and 51.5%. $\alpha$-Tocopherol content was 33.9% of the non-irradiated one when it was irradiated at 7 kGy. Vitamin A and ${\gamma}$-tocopherol contents in sesame seeds linearly decreased with the applied doses (5 and 10 kGy) Vitamin A contents decreased up to 87.8% (5 kGy) and 82.9% (10 kGy) and that of ${\gamma}$-tocopherol were 72.4% and 66.5% of the unirradiated sample.

Studies on the Biological Treatment of Dye Waste Water and Degration of Polyvinyl Alcohol (염색공장 폐수중 PVA 분해세균의 분리 및 생물학적 처리효과)

  • As a research for treatment of waste water by biological method, we investigated general characteristics of waste water and isolated some useful bacteria which effectively treated waste water. Compositions of waste water were analyzed to give COD 2060 ppm, PVA 560 ppm, T-N 50 ppm, T-P 3.3 ppm and PH 12. Also, we inverstigated optimum nutrients requirement and growth conditions by mixed culture as well as the effect of coagulants. The COD removal rate reached maximum state for 48 hrs culture at pH 7.0 and 30$^{\circ}$C. Alum as the coagulated was the most effective. The COD removal rate was also increased by supplementing 10 ppm phosphorous sources as additional nutrients. The COD of waste water was reduced to 10% of its initial value by the continuous culture. As a result of overall experiments the COD of effluents became about 100 ppm and final pH 7.

DISTRIBUTION CHARACTERISTICS OF NUTRIENTS IN CHINESE BOHAI SEA

  • Nutrients are key environmental factors in marine ecosystem. They limit algal growth when at low concentrations and cause algal bloom when at high contents. They also control the growth and succession of many other biota including bacteria and zooplankton, either directly or indirectly. Nutrient contents therefore affect both the structure and functions of marine ecosystem. To study the contents and distribution of nutrients in Chinese Bohai Sea, two cruise surveys were undertaken in August 2000 (summer) and January 2001 (winter), respectively. A total of 595 water samples were collected from 91 sites. After collection the samples were transported to the laboratory and five nutrients, i.e., nitrate, nitrite, ammonia, phosphate and silicate, were analyzed. The results showed that tile average concentration of total inorganic nitrogen (TIN) in Bohai Sea in winter (6.5293.717 ${\mu}$mol$.$l$\^$-1/) was significantly higher than that in summer (3.717 ${\mu}$mol$.$l$\^$-1/). The phosphorus concentration in winter (0.660 ${\mu}$mol$.$l$\^$-1/) was also significantly higher than that in summer (0.329 ${\mu}$mol$.$l$\^$-1/). Mean silicate concentration in winter (7.858 ${\mu}$mol$.$l$\^$-1/) was not significantly different from that in summer (7.200 ${\mu}$mol$.$l$\^$-1/). Nutrients also varied considerable among different areas within Bohai Sea. TIN concentration in Laizhou Bay (4.444 ${\mu}$mol$.$l$\^$-1/), for example, was significantly higher than those in Bohai Bay (2.270 ${\mu}$mol$.$l$\^$-1/) and Bohai Straight (2.431 ${\mu}$mol$.$l$\^$-1/), which probably reflects tile discharge of large amounts of nitrogen into Laizhou Bay via Yellow River. The nutrients also showed vertical distribution pattern. In summer, nutrients in bottom layer were generally higher than those in surface and medium layers. In winter, however. nutrients in different layers were not significantly different Compared with historic data, TIN contents increased continuously since early 1980s, phosphorus arid silicone contents, nevertheless, fell down to some degree. Based on atomic ratios of different nutrients, nitrogen is still the main limiting factor for algal growth in Bohai Sea.

Study on Microorganism Multiplication Behavior and Efficiency of Chlorine Disinfection in the Sewage Effluent from J Municipal Waste Water Treatment Plant (J 하수 처리장 방류수 중 세균의 성장 거동 및 염소 소독 효율 고찰)

  • We evaluated the relationship between the multiplication of heterotrophic microorganisms and physicochemical factors in the final discharged sewage water from J municipal waste water treatment plants. Dissolved organic carbon (DOC) was the most crucial factor influencing multiplication of heterotrophic plate counts (HPC) among the water quality variables selected. Degrading bacteria, such as proteolytic bacteria, lipholytic bacteria, starch degrading bacteria, cellulolytic bacteria, and pectinolytic bacteria, were monitored to understand the condition of nutrients in finished sewage effluent. The percentages of lipid and protein combined occupied 81% in finished sewage water. The multiplication of HPC showed the highest value in August. The formation of trihalomethane (THM) was low in the finished discharge water during chlorine disinfection, which was $71{\mu}/L$ (which was less than $100{\mu}/L$- the standard of drinking water quality) with 10 mg/L of chlorine during 15 min.

Microbial Reduction of Iron(III) Oxides: Implication for Permeable Reactive Barriers. (철환원 미생물을 이용한 3가 철의 환원에 관한 연구)

  • Remediation of groundwater using zero valent iron filings has received considerable attention in recent years. However, zero valent iron is gradually transformed to iron(III) oxides at permeable reactive barriers, so the reduction of iron(III) oxides can enhance the longevity of the reactive barriers. In this study, microbial reduction of Fe(III) was performed in anaerobic condition. A medium contained nutrients similar to soil solution. The medium was autoclaved and deoxygenated by purging with 99.99% $N_2$ and pH was buffered to 6, while the temperature was regulated as 2$0^{\circ}C$. Activity of iron reducing bacteria were not affected by chlorinated organics but affected by iron(III) oxide. Although perchloroethylene(PCE) was not degraded with only ferric oxide, PCE was reduced to around 50% with ferric oxide and microorganism. It shows that reduced iron can dechlorinate PCE.
