Changes of Indicator Microorganisms and Pathogenic Bacteria in Spinach during Cook-Chill Process (시금치의 cook-chill 가공 중 오염지표균 및 병원성세균의 변화)

  • Spinach minimally processed using cook-chill and sous vide techniques was vacuum-packed in low gas permeable plastic film, pasteurized at $70^{\circ}C$ for 2 min, cooled rapidly at $3^{\circ}C$, and stored at 3 and $10^{\circ}C$. Contents of mesophilic bacteria, psychrophilic bacteria, anaerobic bacteria, spore-forming bacteria, total coliforms, yeast and molds, fecal Streptococcus, and Enterobacteriacea were measared to identify the degree of food contamination. Number of mesophilic bacteria, detected at $2.2{\times}10^8\;cfu/g$ in raw spinish, decreased to about $6.0{\times}10^3\;cfu/g$ after cook-chill process. During the storage at 3 or $10^{\circ}C$, levels of mesophilic, psychrophilic and anaerobic bacteria increased, whereas total coliforms, yeast and molds, fecal Streptococcus, and Enterobacteriacea were not detected. Twelve strains of Aeromonas hydphila, Escherichia coli O157:H7, Plesiomonas shigelloides, Pseudomonas aeruginosa, Salmonella spp., Shigella spp., Yersinia enterocolitica, Bacillus cereus, Campylococcus spp., Clostridium perfringens, Listeria monocytogenes, and Staphylococcus aureus were examined for detecting the presence of pathogenic bacteria in spinach. B. cereus and C. perfringens were isolated from raw, washed, and cook-chilled spinach, whereas A. hydrophila was isolated only from washed spinach. S. aureus was isolated from raw and washed spinach, but not from cook-chilled spinach. Other pathogenic organisms were not detected in raw, washed, and cook-chilled spinach.

Isolation of Uncultivable Anaerobic Thermophiles of the Family Clostridiaceae Requiring Growth-Supporting Factors

  • Novel groups of uncultivable anaerobic thermophiles were isolated from compost by enrichment cultivation in medium with a cell-free extract of Geobacillus toebii. The cell-free extract of G. toebii provided the medium with growth-supporting factors (GSF) needed to cultivate the previously uncultured microorganisms. Twenty-nine GSF-requiring candidates were successfully cultivated, and 16 isolated novel bacterial strains were classified into three different groups of uncultivable bacteria. The similarity among these 16 isolates and a phylogenetic analysis using 16S rRNA gene sequences revealed that these GSF-requiring strains represented novel groups within the family Clostridiaceae.

  • The anaerobic continuous reactor, which was filled with a sludge of anaerobic digestion from Sooyoung sewage treatment plant, was supplied with synthetic wastewater of various concentration. After changing to substrate concentration, 디 1is research indicated that attached biomass was kept constant after attachment 23 days. In SEM photographs. shape and structure of biofilm could be observed, but bacteria species and methanogens were not identified. A large number of methanogenic bacteria were showed on the surface of PE substratum by fluorescence under 480nm of radiation.

  • The development and industrial application of intestinal anaerbic bacteria is highly important in the munufacture of foodstuffs and medical products. We have conducted studies, screening anaerobic intestinal bacteria for acid and bile tolerance and for pathogens suppression in the intestine of swine. One such study was taxonomic in nature and dealt with the isolate's biochemical responses. The results indicated that the BSA-131 strian ioslated. which suppressed various pathogens (13 typical strains), were identified as Lactobacillus reuterii. The strain, which named L. reuterii BSA-131, was able to tolerate acid condition down to pH2 and could also tolerate bile at 5%. The strain exhibited resistance to a range of antibiotics including, cephalexin, erythromycin, flumequine, furazolidine, gentamycin, penicillin G, norfloxacin, spectinomycin, tetracycline, tiamuline, neomycin, chloramphenicol, kanamycin etc.

Treatment of Food Processing Wastewater bearing Furfural by Candida utilis (Candida utilis를 이용한 furfural 함유 식품가공 폐수의 처리)

  • A yeast treatment process was applied to treat food processing organic wastewater containing inhibitory material to anaerobic bacteria. The wastewater contained high concentration of the furfural as a by-product from the food processing. Aerobic yeast (Candida utilis) was selected to remove organics in wastewater. The batch test showed that the wastewater had an inhibition to anaerobic bacteria. The optimum level of temperature for yeast treatment was ranged from 25 to 45$^{\circ}C$. The pH range from 4 to 8 was favorable to yeast growth. The continuous flow reactor was operated at various SRTs. The results were satisfactory with the reduction of COD up to 90% at SRT of more than 1 day. Through the kinetic study of the yeast, the remained COD concentration was mainly caused by the formation of soluble microbial product (SMP).

Effects of Pb, Cu, and Cr on Anaerobic Biodegradation of Diesel Compounds by Indigenous Bacteria (혐기성 토착미생물의 디젤 생분해에 대한 Pb, Cu, Cr의 영향)

  • Anaerobic biodegradation of diesel with coexisting heavy metals (Pb) was monitored in batch mode. Two different groups of the indigenous bacteria from a site contaminated with diesel and lead were used in this research: the first group was composed of a single species and the second group was composed of several species. The effect of heavy metals on the microbial population was monitored and confirmed the biodegradation mechanism in each combined contaminant. Growth of the microorganisms in 21 days was observed Diesel > Diesel + Pb > Diesel + Cu > Diesel + Pb + Cu > Diesel + Cr > Diesel + Pb + Cr. Indigenous microorganisms showed the adaptation in the Pb contaminate. Interactive toxic effect using AMES test observed larger synergistic effect than antagonistic in Diesel + Cr and Diesel + Pb + Cr. Therefore, the main effects of diesel biodegradation in the present of heavy metals are likely to exist other factors as well as toxic of heavy metals. This is a necessary part of the future studies.

Comparative Studies of Steel Wool Method and Gaspak Method for the Culture of Anaerobic Bacteria (염기성 세균 배양을 위한 Steel Wool법과 Gaspak법의 비교)

  • It is a well-known fact that an isolation of non-sporeforming anaerobes, considered normal flora in man ordinarily but causes serious infections sometimes, is a dificult procedure because of their great oxygen sensitivity. Among the many techniques employed in clinical laboratories, despite of its high expenses, the GasPak method has been most widely used because of its relative simplicity. On the other hand, the steel wool method has gained a good reputation recently. This technique makes it possible to treat individual plate so that any single specimen can be promptly cultured anaerobically. The procedure is quite simple and the expenses are negligible. In the present study it is to compare these two methods as to their efficiency of anaerobic cultivation using 13 VPI strains of non-sporeforming amaerobic bacteria. Among the 13 species the following 11, Bacteroides fragilis ss. fragilis, B. fragilis ss. thetaiotaomicron, propionibacterium acnes, Eubacterium limosum, E. lentum, peptococcus asaccharolyticus, Pc. prevotii, Pc. magnus, peptostreptococcus anaerobius, Ps. intermedius nad Veillonella parvula, grew well with the steel wool method whose colony numbers reaching 57 to 119% of those with GasPak method. The remaining two species, Fusobacterium nucleatum and F.necrophorum, did not grow well with the steel wool method showing the colony numbers were only 0.4% of those with GasPak method in the case of Fusobacterium nucleatum. In the case of Fusobacterium necrophorum, very few colonies developed even with a heavy inoculation. As to the size of colonies, there were no significant difference between these two methods.

Changes in Physicochemical Properties of Packaged Backsulgi during Storage (포장한 백설기의 저장과정 중 이화학적 변화)

  • Packaged Backsulgi cooked by steam/convection oven and then rapidly chilled was examined by research of microbiological test and sensory evaluation while storing them at the temperatures of 3$^{\circ}C$ and 3$0^{\circ}C$ for 0, 2, 4, 6, 10 days . The pH and reducing sugar content were seemed to change little at 3$^{\circ}C$. However the pH was rapidly reduced until 4 days and then decreased a little at 3$0^{\circ}C$, the reducing sugar content was inclosed little by little. In the microbiological test, any microbial growth in total aerobic, psychrophilic, anaerobic, spore forming bacteria, yeast and molds was not observed until 10 days at 3$^{\circ}C$, but microbial changes of aerobic, psychrophilic and anaerobic bacteria increased to 6 logCFU/g until 10 days at 3$0^{\circ}C$. However microbial changes of them decreased from 6 logCFU/g to 5 logCFU/g. As a result of the sensory evaluation, appearance, taste, color, softness, chewiness and overall Quality were significantly decreased during storage times(p<0.05), but scores of taste and overall quality on 6th days were 7.38${\pm}$1.06, 7.00${\pm}$0.93. Therefore we concluded that there was no problem about stability of storage 6 days at 3$^{\circ}C$.

Investigation of Microbial Communities in the Anammox Reactor Seeded with Sewage Sludge and Anaerobic Granule (하수 슬러지와 혐기성 입상슬러지를 식종한 혐기성 암모니아 산화 반응기의 미생물 탐색)

  • Anammox reactor seeded with sewage sludge from RBC reactor and anaerobic granule from full-scale UASB reactor treating distillery wastewater was operated. Mixed granule and suspended sludge in the ammonium oxidizing process were taken and analyzed to investigate microbial community structure by molecular methods such as gene cloning and phylogenetic tree analysis after 250 days of continuous cultivation. The average nitrogen removal rate showed $0.9kg\;N/m^3-day$ after 250 days of continuous operation, then the maximum nitrogen removal rate showd $1.9kg\;N/m^3-day$ when $2.1kg\;N/m^3-day$ of nitrogen loading rate was applied. As results of gene cloning and phylogenetic tree analysis, Three kinds of phylum were found to be Proteobacteria, Acidobacteria and Planctomycetes (anammox bacteria) in mixed granule. Five kinds of phylum were found to be Proteobacteria, Chlorobi, Planctomycetes, Nitrospirae and Verrucomicrobia in suspended sludge. We found planctomycete KSU-1 and putative new anammox bacteria in the reactor. Microbial structure represented different consortia depending on the types of sludge in the anammox reactor.

SUPPRESSION OF HYDROGEN CONSUMING BACTERIA IN ANAEROBIC HYDROGEN FERMENTATION

  • Severe loss or hydrogen occurred in most anaerobic hydrogen fermentation reactors. Several selected methods were applied to suppress the consumption of hydrogen and increase the potential of production. As the first trial, pH shock was applied. The pH of reactor was dropped nearly to 3.0 by stopping alkalinity supply and on]y feeding glucose (5 g/L-d). As the pH was increase to $4.8{\pm}0.2,$ the degradation pathway was derived to solventogenesis resulting in disappearance of hydrogen in the headspace. In the aspect of bacterial community, methanogens weren't detected after 22 and 35 day, respectively. Even though, however, there was no methanogenic bacterium detected with fluorescence in-situ hybridization (FISH) method, hydrogen loss still occurred in the reactor showing a continuous increase of acetate when the pH was increased to $5.5{\pm}0.2$. This result was suggesting the possibility of the survival of spore fanning acetogenic bacteria enduring the severely acidic pH. As an alternative and additive method, nitrate was added in a batch experiment. It resulted in the increase of maximum hydrogen fraction from 29 (blank) to 61 % $(500\;mg\;NO_3/L)$. However, unfortunately, the loss of hydrogen occurred right after the depletion of nitrate by denitrification. In order to prevent the loss entangled with acetate formation, $CO_2$ scavenging in the headspace was applied to the hydrogen fermentation with heat-treated sludge since it was the primer of acetogenesis. As the $CO_2$ scavenging was applied, the maximum fraction of hydrogen was enhanced from 68 % to 87 %. And the loss of hydrogen could be protected effectively.
