Some Properties and Microbial Community Changes of Gul (Oyster) Jeotgal during Fermentation

  • Gul jeotgals (GJs) were prepared using solar salt aged for 3 years. One sample was fermented using starters, such as Bacillus subtilis JS2 and Tetragenococcus halophilus BS2-36 (each $10^6CFU/g$), and another sample was fermented without starters for 49 days at $10^{\circ}C$. Initial counts of bacilli and lactic acid bacteria (LAB) in non-starter GJ were found to be $3.20{\times}10^2$ and $7.67{\times}10^1CFU/g$ on day 0, and increased to $1.37{\times}10^3$ and $1.64{\times}10^6CFU/g$ on day 49. Those of starter GJ were found to be $2.10{\times}10^5$ and $3.30{\times}10^7CFU/g$ on day 49, indicating the growth of starters. The pH values of GJ were $5.93{\pm}0.01$ (non-starter) and $5.92{\pm}0.01$ (starter) on day 0 and decreased to $5.78{\pm}0.01$ (non-starter) and $5.75{\pm}0.01$ (starter) on day 49. Amino-type nitrogen (ANN) production increased continuously during fermentation, and $407.19{\pm}15.85$ (non-starter) and $398.04{\pm}13.73$ (starter) mg% on day 49. Clone libraries of 16S rRNA genes were constructed from total DNA extracted from non-starter GJ on days 7, 21, and 42. Nucleotide sequences of Escherichia coli transformants harboring recombinant pGEM-T easy plasmid containing 16S rRNA gene inserts from different bacterial species were analyzed using BLAST. Uncultured bacterium was the most dominant group and Gram - bacteria such as Acidovorax sp., Afipia sp., and Variovorax sp. were the second dominant group. Bacillus amyloliquefaciens (day 7), Bacillus velezensis (day 21 and 42), and Bacillus subtilis (day 42) were observed, but no lactic acid bacteria were detected. Acidovorax and Variovorax species might play some role in GJ fermentation. Further studies on these bacteria are necessary.

어류병원성 연쇄구균의 생장을 억제하는 Bacillus pumilus CPB-St의 생물학적 특성 (Biological Characters of Bacillus pumilus CPB-St Inhibiting the Growth of Fish Pathogenic Streptococci)

  • 어류병원성 연쇄상구균에 대하여 항 미생물 효과를 갖는 것으로 보고된 Bacillus sp. CPB-St를 수산용 probiotics로 개발하기 위한 기초 조사로서 미생물의 생물학적인 특성을 알아보았다. Bacillus sp. CPB-St는 16S rRNA 유전자 염기서열에 근거하여 Bacillus pumilus로 동정되었으며 B. pumilis CPB-St (이하 CPB-St)로 명명되었다. CPB-St를 배양 온도 및 배지의 종류에 따라 Streptococcus sp.에 대한 생장억제능력을 평가한 결과, $20^{\circ}C$(억제대: 11~29 mm)에서 배양하였을 때가 $30^{\circ}C$ (12~21 mm)에서 배양했을 때보다 생장억제효과가 높았고, NA (29 mm)와 TSA (29 mm)에서는 유사하였으나 BHIA (22 mm)에서는 낮게 나타났다. 또한 생장억제대의 크기는 pH 7.0에서 18 mm로 가장 높게 나타났고, 염분농도 0.5~3%에서는 동일하였다. 한편 CPB-St 의 생물학적 특성을 알아본 결과 $30^{\circ}C$에서 완만한 증식을 보였으며 배양 후 48시간에 최고 생장을 나타내었다. pH 4.0로 조정한 인공위액에서는 5시간까지 약간의 감소만 나타내었다. 넙치에서 채취 한 담즙에서 CPB-St는 농도에 관계없이 동일한 성장을 보였으며 deoxycholic acid가 첨가된 TSA배지에서 $300{\mu}g/ml$의 농도까지 증식 가능하였다. 또한 1%의 염분농도에서 최적의 성장을 보였으며 7%의 농도까지 증식에 큰 차이를 보이지는 않았고 저온 내성의 경우 $-20^{\circ}C$$-70^{\circ}C$에서는 45분경과 시점부터 대조군에 비해 성장 감소가 나타났으나 $4^{\circ}C$에서는 실온과 차이를 보이지 않았다. 따라서 CPB-St는 어류의 소화관 내에서의 생존가능성이 높으며 사료에 첨가하여 저온에 보관하여도 균수가 감소되지 않을 것으로 판단된다.

Isolation and Characterization of a Protease-Producing Bacterium, Bacillus amyloliquefaciens P27 from Meju as a Probiotic Starter for Fermented Meat Products

  • This study was performed to select protease-producing Bacillus sp. as a potential probiotic starter for fermented meat products. In order to isolate protease-producing bacterium from meju, measured the diameter of the clear zone on agar plate (TSA, 1% (w/v) skim milk) and analyzed for intracellular protease activity, then 10 Bacillus-like strains were isolated. Three Bacillus-like strains (P19, P27, and P33) among 10 strains were able to tolerate in acidic condition (TSB, pH 2.5, 2 h incubation). These 3 strains were showed antimicrobial activity against food-borne pathogenic bacteria. These vegetative cells of 3 strains were showed a survival rate of 0.04% to 0.08% under the artificial gastric acidic condition (TSB, pH 2.5 with 1% (w/v) pepsin), but spore-forming cells were 56.29% to 84.77%. Vegetative cells of 3 strains were the least bile-resistant, while spore-forming cells of 3 strains showed higher survival rate more than 76% under artificial bile condition (TSB, 0.1% (w/v) oxgall bile). In these strains, P27 strain was finally selected as a good probiotic strain. P27 strain was tentatively identified as Bacillus amyloliquefaciens by API CHB kit and 16S rDNA sequence analysis. The results of this study suggest that B. amyloliquefaciens P27 can be used as a potential probiotic starter for fermented meat product.

Optimization of the Production of Fibrinolytic Enzyme from Bacillus firmus NA-1 in Fermented Soybeans

  • Bacillus strains capable of producing fibrinolytic enzyme were isolated from traditional fermented Korean soybean paste and Japanese fermented soybean (Natto). Among the 16 strains, a selected Bacillus sp. was identified as bacillus firmus, with 80.7% homology, by API kit analysis. Seed starter or B. firmus NA-1 was prepared with 5% soymilk prepared from micronized soybean powder. To produce fibrinolytic enzyme by B. firmus NA-1 the liquid culture was performed with NB broth (pH 7.0) fortified with 1% galactose, 0.1% tryptone, and 0.5% $K_2$HPO$_4$, by shaking with 180 rpm at 37$^{\circ}C$. Fibrinolytic enzyme activity reached the highest value at 7.8 unit/mL (plasmin unit) after fermentation for 72 hr. The crude fibrinolytic enzyme showed higher relative activity in the range of pH 7.0∼9.0. The activity of crude fibrinolytic enzyme was well maintained even after concentration by the vacuum evaporation at 5$0^{\circ}C$ for 1 hr.

발효된 오징어젓갈에서 분리된 Weisella sp. 균주의 특성 분석 및 쌀발효물의 유해세균에 대한 항균활성 검증 (Characterization of Weissella sp. Strains Isolated from Fermented Squid and the Antibacterial Activities of Fermented Rice against Harmful Bacteria)

  • 본 연구의 목적은 전통식품에서 MRSA (Methicillin Resistant Staphylococcus aureus)에 대한 항균 활성을 보이는 유산균을 분리하고 특성을 파악한 후에, 분리된 유산균을 이용한 쌀 발효물의 항균 활성을 확인하는 것이었다. 오징어 젓갈에서 선별된 3종의 균주는 16S rRNA 유전자 염기서열을 기반으로 Weissella sp. ISF-1, ISF-2, ISF-3으로 명명하였다. 이들은 장내 유해세균들인 Bacillus cereus, Staphylococcus aureus 및 MRSA에 대해 우수한 항균 활성을 보였다. 이들의 생장은 0-5% (w/v) NaCl에서 우수하였고 7% (w/v) NaCl까지 비교적 양호하였다. 초기 pH 8이 생육에 최적이었으며, pH 6, 7, 9에서도 우수한 생육을 보였다. 분리 된 Weissella sp. ISF-1 균주를 이용하여 발효한 쌀발효 동결건조물들도 B. cereus, S. aureus 및 MRSA에 대해 항균 활성을 보였다. 본 연구결과로 획득한 균주들은 프로바이오틱스 및 안전한 식품 이용을 위한 생물 보존제 개발이나 MRSA 치료에 유용할 수 있고, 쌀의 부가가치를 높이는 새로운 기능의 제품개발에 활용이 가능할 것으로 기대된다.

Isolation of Polyacrylamide-degrading Microorganisms from Soil

  • Two polyacrylamide degrading bacterial strains, No. 2 and No. 11, were isolated from soil, and identified as Bacillus sphaericus No.2 and Acinetobacter sp. No. 11, respectively. Both strains grew on medium containing polyacrylamide as sole carbon and nitrogen sources. B. sphaericus No. 2 and A. sp. No. 11 reduced by 16% and 19%of the initial polyacrylamide concentration, respectively. Optimal pH and temperature in growth of Acinetobacter sp. No. 11 were 8.0 and $37^{\circ}C$, respectively. After 14-day cultivation of A. sp. No. 11, the average molecular weight of polyacrylamide has been shifted from $2.3{\times}10^6\;to\;0.5{\time}106$.

인삼 근권 토양에서 분리한 Stenotrophomonas sp. 4KR4의 Ginsenoside Rb1 전환능 및 분류학적 특성 (Conversion of Ginsenoside Rb1 and Taxonomical Characterization of Stenotrophomonas sp. 4KR4 from Ginseng Rhizosphere Soil)

  • 인삼 근계(근권, 근면, 근내부)로부터 ginsenoside Rb1 전환효소인 ${\beta}$-glucosidase 생산 균주(BGB)를 분리하였다. 인삼 근계부터 분리된 BGB 28균주의 계통학적 특성을 확인한 결과, 근권에서 Stenotrophomonas 속(3균주), Pseudoxanthomonas 속(1균주), Bacillus 속(1균주)로 확인되었다. 근면로부터 분리된 BGB는 Stenotrophomonas 속(16균주), Streptomyces 속(1균주), Microbacterium 속(1균주)이며, 근내부는 Stenotrophomonas 속(3균주), Lysobacter 속(2균주)를 포함하는 다양한 계통군이 확인 되었다. 특히 인삼 근계로부터 분리된 BGB 균주의 90%가 Stenotrophomonas 계통군에 속하는 특징을 나타내었다. 근권으로부터 분리된 4KR4 균주는 108.17 unit의 ${\beta}$-glucosidase 활성을 나타내었으며, ginsenoside Rb1을 Rd, Rg3 그리고 minor ginsenoside Rh2로 전환되었다. 4KR4 균주는 Stenotrophomonas rhizophila e-$p10^T$ (AJ293463)와 99.65%의 높은 상동성을 나타내었다. 본 연구에서 분리된 ginsenoside 전환세균 4KR4 균주의 계통학적 위치와 표현형적 특징, 균체 지방산조성, 생리 생화학적 특성을 검토한 결과, Stenotrophomonas sp. 4KR4 (=KACC 17635) 균주로 확인되었다.

지렁이로부터 분리한 Bacillus pumilus JS-01 균주의 유기물 분해능 및 응집능 (Degrading and Flocculating Property of A Bacterium Isolated from the Extract of Earthworm)

  • 음식물류폐기물의 친환경적 처리를 위한 소재를 개발하기 위하여 지렁이로부터 유기물 분해 및 응집능을 갖는 기능성 미생물을 분리하였다. 분리된 균주 중 음식물류폐기물을 처리하는데 적합한 것으로 판단되는 균주를 최종 선별하여 JS-01균주로 명명하였으며 16s rDNA 염기서열분석에 의한 최종결과, Bacillus pumilus로 동정되었다. 0.5% Kaolin clay을 대상으로 JS-01균주의 응집능을 조사한 결과 응집을 위한 최적 생장조건은 MRS 액체배지에서 온도 $37^{\circ}C$ 및 pH 7.0으로 나타났다. 유기물분해능 중에서는 특히, 음식물류폐기물의 대표적 성분인 섬유소에 대한 분해능을 확인하였다. 따라서 JS-01균주는 음식물류폐기물의 친환경적 처리 및 재활용을 위한 미생물 소재 및 음식물 침출수의 효과적인 응집 처리를 위한 생체량 생성에 이용할 수 있을 것으로 사료된다.

Hydrolysis of Agricultural Residues and Kraft Pulps by Xylanolytic Enzymes from Alkaliphilic Bacillus sp. Strain BK

  • An alkaliphilic bacterium, Bacillus sp. strain BK, was found to produce extracellular cellulase-free xylanolytic enzymes with xylan-binding activity. Since the pellet-bound xylanase is eluted with 2% TEA from the pellet of the culture, they contain a xylan-binding region that is stronger than the xylan-binding xylanase of the extracellular enzyme. The xylanases had a different molecular weight and xylan-binding ability. The enzyme activity of xylanase in the extracellular fraction was 6 times higher than in the pellet-bound enzyme. Among the enzymes, xylanase had the highest enzyme activity. When Bacillus sp. strain BK was grown in pH 10.5 alkaline medium containing xylan as the sole carbon source, the bacterium produced xylanase, arabinofuranosidase, acetyl esterase, and $\beta$-xylosidase with specific activities of 1.23, 0.11, 0.06, and 0.04 unit per mg of protein, respectively. However, there was no cellulase activity detected in the crude enzyme preparation. The hydrolysis of agricultural residues and kraft pulps by the xylanolytic enzymes was examined at 50$^{\circ}C$ and pH 7.0. The rate of xylan hydrolysis in com hull was higher than those of sugarcane bagasse, rice straw, com cop, rice husk, and rice bran. In contrast, the rate of xylan hydrolysis in sugarcane pulp was 2.01 and 3.52 times higher than those of eucalyptus and pine pulp, respectively. In conclusion, this enzyme can be used to hydrolyze xylan in agricultural residues and kraft pulps to breach and regenerate paper from recycled environmental resources.

Bioconversion of Soybean Curd Residues into Functional Ingredients with Probiotics

  • Soybean curd residues (SCR) obtained from hot and cold manufacturing processes were fermented by indigenous microorganisms, Lactobacillus rhamnosus LS and Bacillus firmus NA-l for 15 h at 37$^{\circ}C$. The pH, acidity, viable cell counts, and tyrosine content were evaluated in samples with variations in sugar, starter and type of SCR. The raw Doowon SCR (D-SCR, cold-processed) fermented by indigenous microorganism had a 0.9% acidity and 6.7 ${\times}$ 10$^{7}$ CFU/g viable cell counts, compared with the 0.11 % acidity and 6.7 ${\times}$ 10$^{6}$ CFU/g viable cell counts of raw fermented Pulmuwon SCR (P-SCR, hot-processed). After fermentation of raw P-SCR with 1 % glucose and 1 % L. rhamnosus LS starter, the viable cell counts, tyrosine content and acidity were 4.7 ${\times}$ 10$^{8}$ CFU/g, 16.3 mg% and 0.9%, respectively. In addition, the raw P-SCR fermented with Bacillus firmus NA-l as co-starter had a 0.45% acidity, 2.4 ${\times}$ 10$^{8}$ CFU/g lactic acid bacteria, and 3.3 ${\times}$ 10$^{6}$ CFU/g Bacillus sp. In particular, the tyrosine content was increased 5 fold. The drying of fermented SCR was completed by hot-air drying (5$0^{\circ}C$) within 12 h; the dried P-SCR and D-SCR had 1.8 ${\times}$ 10$^{7}$ CFU/g and 5.3 ${\times}$ 10$^{6}$ CFU/g viable cell counts, respectively. The concentrate of methanol extract from fermented D-SCR inhibited the initial cell growth of E. coli, Staphylococcus aureus and Pseudomonas aeruginosa in liquid culture.
