Removal of organic Carbon, Nitrogen and Phosphorus in Wastewater based on tapered Aeration with Bacillus sp. (점감포기에 의한 바실러스 특성을 이용한 폐수의 유기물질 및 질소, 인 처리에 관한 연구)

  • This study was conducted to investigate an aeration tank with RBC process attached Bacillus sp. known as a suitable microorganism for the removing of organic carbon, nitrogen and phosphorus. An aeration tank was based on tapered aeration because Bacillus sp. was well grown in this like environment conditions. The biofilm process with Bacillus sp. as an advanced treatment process could be a best technology for the prominent removal of organic carbon, nitrogen and phosphorus if the mechanism in the process is verified. The operation conditions of DO in the tapered aeration tank were maintained as $1.2{\sim}1.5mg/L$ in aeration tank1, as $0.3{\sim}0.5mg/L$ in aeration tank 2 and less than 0.2 mg/L in aeration tank 3, respectively. Lab-scale experiments were conducted, at room temperature, internal recycle rate was from 200% to 50% and returned sludge rate was from 100% to 50%. As a result, concentration of organic carbons, nitrogen and phosphorus in Period 1 (the time of Bacillus sp. adapted to environment) were decreased gradually. Ultimately, each removal rate in this biological experiment were TCODCr 94%, BOD 87%, T-N 85%, T-P 89% in Period 2. Hence, this process showed an excellent performance of the removal of organic carbon, nitrogen and phosphorus and this is an effective system fur treating of wastewater.

Isolation of Neutral Protease Hyperproducing Bacillus sp. KN103N and Some Properties of the Enzyme (中性 Protease 高 生産性 Bacillus sp. KN103N의 分離 및 酵素의 特性)

  • A bacterial strain KN, which highly produced a protease, was isolated from several soil samples and identified to to belong to the genus Bacillus. We selected mutant strain Bacillus sp. KN103N, which was hyperproducer of protease and was resistant to D-cyclowerine, from the strain KN by several steps of mutagenesis. Neutral protease productivity of mutant strain KN103N was about 55 times as much as that of the original strain KN. The optimum pH and temperature for the enzyme activity were 7.0 and 50$^{\circ}C$, respectively and the enzyme was relatively stable at pH6.0~8.0 and below 40$^{\circ}C$. The enzyme was inactivated by EDTA, but not by DFP. These results indicate that the enzyme from Bacillus sp. KN103N was a neutral (metallo-) protease.

  • A bacterial strain NO. 32 which produced thermostable ${\alpha}$-amylase was isolated from soil and identified to genus of Bacillus. To enhance ${\alpha}$-amylase productivity, a successive mutation of Bacillus sp. No. 32 was attempted with treatment of N-methyl-N'-nitro-N-nitrosoguanidine (NTG). The resulting mutant, Bacillus sp. No. 32H417, which is risistant to refampicin and deficient in spore formation, produced about 90-fold high level of ${\alpha}$-amylase when compared with parental strain. The properties of the enzyme for thermostability were investigated. The optimal temperature and pH for enzyme reaction were 95$^{\circ}C$ and pH6.5, respectively, in the presence of 0.3mM $Ca^{2+}$ as an effective stabilizer.

Isolation and Identification of a Bacteriolytic Enzyme-producing Bacterial Strain from Pusan Coastal Sea (해양에서 용균효소를 분비하는 균주의 분리와 동정)

  • In order to produce the bacteriolytic enzyme, bacterial strains capable of excreting a large amount of the enzyme were screened from the coastal sea water samples in Pusan. The bacterial strain SH-1, which showed the highest activity among 43 bacteriolytic enzyme producing bacteria, was finally selected for further studies. The strain SH-1 was an endospore-forming grampositive rod, and the position of spore was paracentral. These morphological characteristics assigned the isolated strain to the morphological group I classified by Gordon. The fatty acid composition of the bacterial stain was analyzed to be consisted of branched chains of iso-Cn and anteiso-Cn. Based on the percent content of the branched chain (93.85%), the isolates could be identified as a species of Bacillus. According to the experimental results of the API system (API 50CHB & API 20E) the strain was identified as Bacillus subtilis. Numerical texonomy, in which 82 major characters were examined using several species of Bacillus as the standard bacteria, indicated that the strain SH-1 showed 90% similarity to Bacillus subtilis. Thus, the isolated strain SH-1 could be identified as Bacillus subtilis.

  • Various bacterial strains that secret extracellular fibrinolytic enzyme were screened from Doen-Jang, a traditional soybean fermented food in Korea. Five microbes of them were identified to be Bacillus sp. strains according to Bergey's manual of systematic bacteriology. The culture filtrates of B. amyloliquefaciens (2.46 plasmin unit/ml) and B. pantothenticus (3.82 plasmin unit/ml) showed a level of fibrinolytic activity that was about three times higher than that of plasmin 1.0 unit and Bacillus subtilis showed the highest fibrinolytic activity (4.94 plasmin unit/ml). All of the extracellular proteases showing the fibrinolytic activity are confirmed by SDS-PAGE followed by reverse fibrin zymogram activity assay and we proposed that some of the fibrinolytic enzymes from this work are novel enzymes.

Microbes causing abnormal Takju fermentation in traditional wheat-based Nuruk: A case study (밀누룩으로 제조한 전통탁주의 이상발효 원인 규명: 사례분석)

  • In this article, we report a case study on the cause of abnormal fermentation of Takju based on a microbial community analysis of wheat-based Nuruk and Takju using next-generation sequencing. The microbial community showed notable changes during alcoholic fermentation. Members of the genera Aspergillus and Staphylococcus were dominant in Nuruk, whereas Saccharomyces, Pediococcus, Bacillus, Lactobacillus, and Enterobacter were predominant during Takju fermentation. In particular, members of the Enterobacter and Bacillus genera that were present were identified as the opportunistic pathogens B. wiedmannii and E. cloacae. The relative abundance of two species increased in Mitsul-3, wherein the concentration of organic acid rapidly decreased, and in Deotsul-1, in which raw materials were added. Furthermore, the relative abundance of lactic acid bacteria (LAB) in both the Nuruk and Takju was very low. Therefore, this result showed that the abnormal proliferation of E. cloacae and B. wiedmannii in Takju was due to the absence of LAB.

Effect of microbial product made of Bacillus stearothermophilus DL-3 on microorganisms in soil and growth of lettuce and Chinese cabbage. (Bacillus stearothermophilus DL-3 미생물 제재의 처리가 토양 미생물상 및 상추와 배추의 생장에 미치는 영향)

  • Effect of the microbial product made of Bacillus stearothermophilus DL-3, which was isolated from the soil and identified in this study, and rice bran on microorganisms in soil and growth of lettuce (Red skirt lettuce) and Chinese cabbage (Ga rack new No.1 Chinese cabbage) was investigated. Total numbers of microorganisms in the pot with untreated soil, treated with standard amount of microbial product and treated with double amounts of microbial product for growth of lettuce after 6 weeks were 2.78${\times}$10$^{7}$ CFU/g, 2.72${\times}$10$^{8}$ CFU/g and 3.63${\times}$10$^{8}$ CFU/g. Total numbers of microorganisms in the soil without treatment of microbial product and treated with standard amount of microbial product were 2.06${\times}$10$^{8}$ CFU/g and 5.49${\times}$10$^{8}$ CFU/g. Total numbers of microorganisms in the pot with untreated soil, treated with standard amount of microbial product and treated with double amounts of microbial product for growth of Chinse cabbage after 6 weeks were 1.43${\times}$10$^{7}$ CFU/g, 3.42${\times}$10$^{8}$ CFU/g and 7.22${\times}$10$^{8}$ CFU/g. Total numbers of microorganisms in the soil without treatment of microbial product and treated with standard amount of microbial product were 5.75${\times}$10$^{8}$ CFU/g and 7.96${\times}$10$^{8}$ CFU/g. On basis of leaf length, leaf width, leaf number, wet weight and dry weight, the growth of lettuce and Chinese cabbage on the soil treated with microbial product was faster than that on the untreated soil. The treatment of microbial product in the soil resulted in the increase of useful microorganisms, which seemed to enhance the growth of lettuce and Chinese cabbage.

Comparison of Microbial Community Compositions between Doenjang and Cheonggukjang Using Next Generation Sequencing (차세대 염기서열 분석법을 이용한 전통 된장과 청국장의 미생물 분포 분석)

  • To profile the microbial compositions of Korean traditional fermented paste made from whole soybeans, Doenjang and Cheonggukjang, and compare their taxonomic differences, we analyzed the V3-V4 region of 16S rRNA of naturally fermented foods by using next generation sequencing. α-Diversity results showed that values indicating bacterial community abundances (OTUs) and richness (ACE, Chao1) were statistically significant (p=0.0001) in Doenjang and Cheonggukjang. Firmicutes was the most common phylum in both groups, representing 97.02% and 99.67% in the Doenjang and Cheonggukjang groups, respectively. Bacillus was the most dominant genus, accounting for 71.70% and 59.87% in both groups. Linear discriminant (LDA) effect size (LEfSe) analysis was performed to reveal the significant ranking of abundant taxa in different fermented foods. A size-effect threshold of 2.0 on the logarithmic LDA score was used for discriminative functional biomarkers. On the species level, Bacillus subtilis, Tetragenococcus halophilus, and Clostridium arbusti were significantly more abundant in Doenjang than in Cheonggukjang, whereas Bacillus thermoamylovorans, Enterococcus faecium, and Lactobacillus sakei were significantly more abundant in Cheonggukjang than in Doenjang. Permutational multivariate analysis of variance (PERMANOVA) showed that the statistical difference in microbial clusters between the two groups was significant at the confidence level of p=0.001. This research could be used as basic research to identify the correlation between the biochemical characteristics of Korean fermented foods and the distribution of microbial communities.

  • 1. A Kind of Bacillus, which was seemed to be pathogenic, was isolated from Pinus rigida planting. 2. The bacillus were grown well on the Agar-Bacto-Beef media (pH 7.4) at $30^{\circ}C$. C for 24 hours. 3. The soil infection for the one year old seedling was the best inoculating method in this experiment. 4. DNA-DNA hybridization에 의한 Bacillus coagulans의 분류학적 연구

  • 서로 다른 11주의 Bacillus coagulans와 13종의 Bacillus 속 14주를 deoxyribonucleic acid (DNA) -DNA hybridization method에 의해서 분류학적인 연구를 하였다. 사용한 B. coagulans 11주종 6주는 흙에서(일본 오사까교회) 분리했고 나머지 5주는 ATCC, IFO에서 authentic strains을 얻어서 사용했다. 사용된 B. coagulans는 Bergey's Manual(8 thed)에 의거 Grodon씨들의 방법으로 동정한 결과 B. coagulans로서 확인되었다.(중략)
