Benzo(a) pyrene Level in Indoor Environment due to the Use of Unvented Heating Facilities (煖房器具를 사용하는 室內空氣중의 벤조(a) 피렌 成分濃度에 관한 硏究)

  • Indoor air pollution problems, in fact, have been neglected due to the unawareness of its seriousness and the lack of accumulated data. Recently, some movement of research and regulations, however, have been made for several indoor air pollutants. In this study, one of the carcinogenic polynuclear aromatic hyrocarbon, Benzo(a) pyrene in indoor airborne particulate, was measured and analyzed to estimate emission strength of BaP from an unvented kerosene heater and to know its level in indoor environment with use of a kerosene heater and a wood-burning fireplace. By the measurement of BaP level in a dark room with air exchange rate of app. 3.0, BaP emission strength of a kerosene heater (11,000Btu/hr) was estimated to be 326.6ng/hr (or 29.7 ng/$10^3$ Btu). With 4-hr operation a day of the kerosene heater in an apartment, the BaP level became 2.97ng/m$^3$ from 0.27ng/m$^3$ of background level. The operation of a wood-burning fireplace app. 2hr a day, on the other hand, increased the BaP level from 0.16ng/m$^3$ of background to 3.53ng/m$^3$. So proper ventilation must be considered seriously when unvented heating facilities are used in indoor environment.

  • Mutations of p53 tumor suppressor gene have been associated with exposure to carcinogens. Cultured human breast epithelial cells (MCF10A) were treated with Benzo(a)pyrene(BaP) or N-Methyl-N'-nitro-N-nitrosoguanidine(MNNG).   • This study was implemented as a part of the experiment to develop two kinds of soil-based Benzo[a]pyrene (BaP) proficiency testing materials (PTMs) for soil analysis. A test was carried out for the check of solubility of the reference material (high purity reagent) using several solvents. Another test was also conducted for the evaluation of homogeneity and stability of two kinds of candidate soil reference materials. The test analysis of BaP in terms of the candidate materials was conducted according to the Standard Soil Analytical Methods by Ministry of Environment. Dissolution of the reference material was shown to vary depending on solvent type and was higher in the order of Dichloromethane > Acetone > Acetone/MeOH (9 : 1) > N-hexane. In addition, the slope on calibration curve for BaP standard solutions was largest on BaP standard solutions prepared with dichloromethane of the tested solvents. Such tendency appeared egually in the commercial BaP standard solution. Therefore, it is thought to be reasonable to use dichloromethane as the solvent in case of the standard stock solution that is used for the measurement of BaP concentration in soil. ISO 13528 and IUPAC protocol were used for verification of homogeneity on the two kinds of soil candidate materials, Both candidate materials were sufficiently homogeneous. Stability assessment of the two candidate materials was made according to ISO Guide 35 and the result showed that both batches did not have any long-term and short term stability issues that might occur during shipping. However, monitoring results of BaP concentration in soil showed that BaP concentration of the two batches measured at 15 days after the sample preparation was reduced by about 24~37% compared with that of the samples measured on 0 day of the sample preparation. Identification was done with several treatments such as irradiation and sterilization etc. The major cause was shown to be irradiation to the samples.

Characteristics of Particulate Atmospheric Polycyclic Aromatic Hydrocarbons Pollution in Residential/Roadside Areas of Metropolitan Cities and a Background Area (대도시의 주거/도로변 지역 및 배경 지역의 대기 중 입자상 다환 방향족 탄화수소의 특성)

  • The present study aims to evaluate the characteristics of atmospheric polycyclic aromatic hydrocarbons (PAHs) pollution in roadside and residential areas of two Korean metropolitan cities (Seoul and Incheon) and a background area (Seokmolee). This purpose was established by analyzing temporal and spacial concentration distribution of total and 7 individual PAHs, which were extracted from ambient particulate matters, and by utilizing a multivariate statistical method (principal component analysis, PCA) for the qualitative determination of potential PAH sources. Target PAHs included benzo(a)anthracene (BaA), benzo(a)pyrene (BaP), benzo(b)fluoranthene (BbF), benzo(k)fluoranthene (BkF), chrysene (Chr), dibenzo(a,h)anthracene (DahA), and indeno(1,2,3-cd)pyrene (IcdP). For all surveyed sites, the concentrations of total PAHs were higher in winter season than in other seasons. However, the concentrations of individual PAHs varied with surveyed sites. In both residential and roadside sites of Seoul and Incheon, BbF revealed the highest atmospheric levels. For all 7 target PAHs, the ambient concentrations were higher in Seoul and Incheon than in a background area (Seokmolee). In both residential and roadside areas, the concentrations of 4 target PAHs (BaA, BbF, BkF, DahA) were higher in Incheon than in Seoul. However, both the residential and roadside Chr concentrations were comparable in Seoul and Incheon. In addition, the residential IcdP concentrations were higher in Incheon than in Seoul, whereas the roadside concentrations were higher in Seoul. The roadside and residential BaP concentrations exhibited the reverse result to the IcdP concentrations. An PCA analysis suggested that atmospheric PAHs in both residential and roadside areas would be due to combined effects of several potential sources such as gasoline- and diesel-fueled vehicles, coal/oil combustion, and waste incineration.

PAMs enter estuarine and nearshore marine environment via several routes such as combustion of fossil fuels, domestic and industrial effluents and oil spills. PAHs have been the focus of numerous studies in the world because they are potentially carcinogenic, mutagenic, and teratogenic to aquatic organisms and humans from consuming contaminated food. However, one can hardly find any available data on PAM content in marine organisms in Korea. The present study was carried out in order to determine PAM content in oysters from the intertidal and subtidal zones of Chinhae Bay, which is located in near urban communities and an industrial complex, and the bay is considered to be a major repositories of PAHs. 16 PAHs were analyzed by High Performance Liquid Chromatography (HPLC) with uv/vis and fluorescence detectors in oysters: they are naphthalene (NPTHL), acenaphthylene (ANCPL), acenaphthylene (ACNPN), fluorene (FLURN), phenanthrene (PHEN), anthracene (ANTHR), fluoranthene (FLRTH), pyrene (PYR), benzo(a)anthracene (BaA), chrysene (CHRY), benzo(b)fluoranthene (BbF), benzo(k)fluoranthene (BkF), benzo(a)pyrene (BaP), dibenz(a,h)anthracene(DahA), benzo(g,h,i)peryne (BghiP) and indeno(1,2,3,-cd)pyrene (I123cdP). The PAM contents in oysters from the intertidal and subtidal zones of Chinhae Bay ranged from < 0.1 to 992.0 ug/kg (mean $69.8\pm9.8$ ug/kg).

  • Air pollution has been recognized for many years as a factor which heightens the risk of cancer. Extractable polycyclic organic matters in air particulates have been recognized as to have carcinogenic effects. This study examined the health risks posed by organic substances of air particulates in Seoul based on methodelogies that have been developed for conducting risk posed by organic substances of sir particulates in Seoul based on methodologies that have been developed for conducting risk assessment of complex -chemical-mixture. The data used in this study was obtained from air samples collected in a heavy traffic area of Seoul (Shinchon) from 1986 to 1994. The mean concentration of total supended pariculates was 158.0.mu.g/m/msup 3/, 5% of which is consisted of organic matter. The excess cancer risk from benzo(a)pyrene (BaP) was estimated to be 3.48.times.10$^{-6}$ by applying BaP unit risk estimates to the mean concentration. 8.74ng/m BaP equivalents of potency method was 1.0.times.10$^{-3}$ . The calculated risk from EOM were comparably higher than that from benzo(a)pyrene and exceeded the acceptable risk level.

  • Experiments were carried out to investigate the lipid peroxidation (LPO), superoxide dismutase (SOD) activity and histopathological change of hepatic tissue for rockfish, Sebastes schlegeli after feeding sub-chronic dietary Benzo(a)pyrene (BaP) in the concentration of 0 (control), 0.5, 1.0, 1.5, 2.0 mg $kg^{-1}$ dry food of diet for 30 days. In 2.0 mg $kg^{-1}$ dry food group, the significant increase of LPO was observed in all period, and SOD activity was incresed at 30 days significantly in the same concentration. In the histological investigation of liver, there was the swelling of hepatic cells at 10 days over the 1.0 mg $kg^{-1}$ dry food concentration. At 30 days Periodic acid-Schiff (PAS) positive granule was observed in the same group and at 20 days was observed in 2.0 mg $kg^{-1}$ dry food group. And there was necrosis of hepatic cell in some fish of 2.0 mg $kg^{-1}$ dry food group at 30 days.

  • Objectives: Condurango is widely used in various systems of complementary and alternative medicine (CAM) against oesophageal and stomach ailments including certain types of cancer. However, until now no systematic study has been conducted to verify its efficacy and dose with proper experimental support. Therefore, we examined if ethanolic extract of Condurango could ameliorate benzo[a]pyrene (BaP)-induced lung cancer in rats in vivo to validate its use as a traditional medicine. Methods: After one month of scheduled BaP feeding (50 mg/kg body-weight), lung cancer developed after four months. BaP-intoxicated rats were then treated with Condurango (0.06 mL) twice daily starting at the end of the four months for an additional one, two and three months, respectively. Effects of Condurango were evaluated by analyzing lung histology, reactive oxygen species (ROS) and antioxidant biomarkers, DNA-fragmentation, RT-PCR (Reverese Transcriptase-Polymerase Chain Reaction), ELISA (Enzyme linked immunosorbent assay) and western blot of several apoptotic signalling markers and comparing the results against those obtained for controls. Results: A histological study revealed gradual progress in lung tissue-repair activity in Condurango-fed cancer-bearing rats, showing gradual tissue recovery after three months of drug administration. Condurango has the capacity to generate ROS, which may contribute to a reduction in anti-oxidative activity and to an induction of oxidative stress-mediated cancer-cell death. Condurango-activated pro-apoptotic genes (Bax, caspase-3, caspase-9, p53, cytochrome-c, apaf-1, ICAD and PARP) and down-regulated antiapoptotic-Bcl-2 expression were noted both at mRNA and protein levels. Studies on caspase-3 activation and PARP cleavage by western blot analysis revealed that Condurango induced apoptosis through a caspase-3-dependent pathway. Conclusions: The anticancer efficacy of an ethanolic extract of Condurango for treating BaP-induced lung cancer in rats lends support for its use in various traditional systems of medicine.
