Genotype Difference of Plant Regeneration from Dormant Bud Culture in Colocasia esculenta Schott.

  • This study was conducted to obtain the basic breeding information of Colocasia esculenta Schott. Effect of supplemental plant growth regulators and genotype difference were investigated on dormant bud tissue for proliferation. The plant regeneration ratio, plant height and root length were the best upon mixed treatment of 0.8mg/L IAA and 2.0mg/L zeatin. Both leaf weight and root weight were heavy upon culture in a dark condition. The leaf and root weights were heaviest in 6Pie sucrose concentrations. In several collected area the heaviest one was Binnangxin and then in the order of Suwon, Wanju and Puan. Genotype differences of tuber diameter and tuber weight were found in Suwon. Tuber weight was found in the order of Suwon (862mg) >Wanju(723mg) >Puan(649mg) >Binnangxin (424mg).

  • The present experiments were conducted to investigate some of the factors affecting the number of haploids derived from anther culture of Nicotiana tabacum. Anther - donor plants grown under controlled environment room at 3$0^{\circ}C$ yielded more haploid than room at 18, 25 and 26-22-18$^{\circ}C$ in anther culture. Donor plants starved of fertilizer yielded more haploids as compared to those of the well fed with fertilizer in anther culture. Pretreatment of exercised flower bud at 5$^{\circ}C$ was shown to be more effective in anther culture than pretreatment at 7 and 1$0^{\circ}C$, and the optimum temperature and period of pretreatment were 4 or 6 days at 5$^{\circ}C$.

Effect of night break treatment using Red LED (660 nm) on flower bud initiation and growth characteristics of chrysanthemum cv. 'Baekma', and cv. 'Jinba' (적색 LED(660nm)의 광중단 처리에 따른 국화 '백마'와 '신마'의 화아분화 및 생육특성)

  • This study was carried out to examine the effect of Red LED (660 nm) and fluorescent lamp for night break (NB) treatments of each 3 hours (22:30-01:30), 4 hours (22:00-02:00) and 5 hours (21:30-:02:30) per day for 53 days on flower bud initiation and growth in Chrysanthemum cv. 'Baekma' and cv. 'Jinba'. The days to flower budding after short-day treatment in 'Baekma' was longer at fluorescent lamp 4 hr (21.0 days) and 5 hr (20.5 days) NB, and it was shorter at Red LED 3 hr (14.2 days). The days to flowering after short-day treatment in 'Baekma' was longer at fluorescent lamp 4 hr (54.0 days), 5 hr (53.5 days) NB, and Red LED 5 hr (53.3 days), and it was shortest at Red LED 3 hr (50.2 days) NB treatment among all treatments. The days to flower budding after short-day treatment of 'Jinba' was longer at fluorescent lamp 4 hr (20.6 days) and was shorter at Red LED 3 hr (14.1 days) among all treatments. Similarly, the days to flowering after short-day treatment of 'Jinba' was longer at fluorescent lamp 4 hr (55.3 days) and was shortest at Red LED 3 hr (50.2 days) among all treatments. Therefore, inhibition of flower bud initiation was the most effective under fluorescent lamp 4 hr treatment. The length of cut flower of 'Baekma' was increased by fluorescent lamp 4 hr, 5 hr, and Red LED 5 hr, but of 'Jinba' was longer at LED 4 hr and 5 hr treatment. The weight of cut flower of 'Baekma' was heaviest at fluorescent lamp 5 hr treatment and was at Red LED 5hr treatment for 'Jinba' even though there was not statistically significant difference between 'Baekma' and 'Jinba'. Consequently, under fluorescent lamp 4 hr for night break was the most effective on flower bud initiation, flowering inhibition and cut-flower characteristics in 'Baekma' and 'Jinba'.

Effects of Extract from Fermented Flower-buds of Panax ginseng C.A. Meyer on Mouse Cytokine IL-6, TNF-α Production (발효 인삼꽃 추출물의 경구 투여가 마우스 사이토카인 IL-6, TNF-α의 생성에 미치는 영향)

  • Panax ginseng C.A. Meyer has been used as a traditional medicinal ingredient and the ginseng flower-buds also proved to have good medicinal properties. In this study, in order to enhance immune activities of ginseng flower-bud, the ginseng flower-bud extract was being fermented by Bacillus subtilis KCTC 1022 (BS), Lactobacillus plantarum KCTC3 and Saccharomyces cerevisiae strain CHY1011 (SC). Mice were orally administered daily for two weeks at two different concentrations (100 and 200 mg/kg B.W.). Treatment samples were water extracts of ginseng flower-buds (FD), water extracts of fermented ginseng flower-buds (FM) and controls for saline solution. Cytokine production (IL-6, TNF-${\alpha}$) either stimulated with LPS or not stimulated with LPS was detected by the ELISA assay when using the cytokine kit. Cytokine was statistically increased at supplemented groups with LPS in both the 100 and the 200 mg/kg B.W. and treatment with FM significantly decreased the LPS-induced TNF-${\alpha}$ and IL-6 production more than the treatment with FD. The results of this study may suggest that supplementation with FM increases the immune function by regulating cytokine production capacity for activated macrophages.

Physio-Ecological Studies on Stevia(Stevia rebaudiana Bertoni) (스테비아(Stevia rebaudiana Bertoni)에 관한 생리 생태적 연구)

  • Stevia (Stevia rebaudiana Bertoni) is a perennial herb widely distributed in the mountainous area of Paraguay. It belongs to the family Compositae and contains 6 to 12 percent stevioside in the leaves. Stevioside is a glucoside having similar sweetening character to surgar and the degree of sweetness is approximately 300 times of sugar. Since Korea does not produce any sugar crops, and the synthetic sweetenings are potentially hazardous for health, it is rather urgent to develop an economical new sweetener. Consequently, the current experiments are conducted to establish cultural practices of stevia, a new sweetening herbs, introduced into Korea in 1973 and the results are summarized as followings: 1. Days from transplanting of cuttings to the flower bud formation of 6 stevia lines were similar among daylengths of 8, 10 and 12 hours, but it was much greater at daylengths of 14 or 24 hour and varietal differences were noticable. All lines were photosensitive, but a line, 77013, was the most sensitive and 77067 and Suweon 2 were less sensitive to daylength. 2. Critical daylength of all lines seemed to be approximately 12 hours. Growth of plants was severely retarded at daylengths less than 12 hours. 3. Cutting were responded to short daylength before rooting. Number of days from transplanting to flower bud formation of 40-day old cuttings in the nursery bed was 20 days and it was delayed as duration of nursery were shorter. 4. Number of days from emergence to flower bud formation was shortest at short day treatment from 20 days after emergence. It was became longer as initiation of short day treatment was earlier or later than 20 days. 5. Plant height, number of branches, and top dry weight of stevia were reduced as cutting date was delayed from March 20 to May 20. The highest yield of dry leaf was obtained at nursery duration of 40-50 days in march 20 cutting, 30-40 days in April 20 cutting, and 30 days in May 20 cutting. 6. An asymptotic relationship was observed between plant population and leaf dry weight. Yield of dry leaf increased rapidly as plant population increased from 5,000 to 10,000 plants/10a with a reduced increasing rate from 10,000 to 20,000 plants/l0a, and levelled off at the plant population higher than 20,000 plants/l0a. 7. Stevia was adaptable in Suweon, Chengju, Mokpo and Jeju and drought was one of the main factors reducing yield of dry leaf. Yield of dry leaf was reduced significantly (approximately 30%) at June 20 transplanting compared to optimum transplanting. 8. Yield of dry leaf was higher in a vinyl house compared to unprotected control at long daylength or natural daylength except at short day treatment at March 20. Higher temperature ill a vinyl house does not have benefital effects at April 20 transplanting. 9. The highest content of stevioside was noted at the upper leaves of the plant but the lowest was measured at the plant parts of 20cm above ground. Leaf dry weight and stevioside yield was mainly contributed by the plant parts of 60 to 120cm above ground but the varietal differences were also significant. 10. Delayed harvest by the time of flower bud formation increased leaf dry weight remarkably. However, there were insignificant changes of yield as harvests were made at any time after flower bud formation. Content of stevioside was highest at the time of flower bud formation and earlier or later harvest than this time was low in its content. The optimum harvesting time determined by leaf dry weight and stevioside content was the periods from flower bud formation to right before flowering that would be the period from September 10 to September 15 in Suweon area. 11. Stevioside and rebaudioside content in the leaves of Stevia varieties were ranged from 5.4% to 14.3% and 1.5% to 8.3% respectively. However, no definit relationships between stevioside and rebaudioside were observed in these particular experiments.

Influence of the Differences in Altitude during Raising Seedlings on Daughter Plant Characteristics and Subsequent Strawberry Production (육묘 기간중의 고도 차이가 자묘의 특성 및 딸기 수량에 미치는 영향)

  • The nursery plant quality and flower bud induction of new strawberry cultivars, 'Maehyang' and 'Seolhyang' in forcing culture were evaluated in the highland and lowland region. In order to produce daughter plants, the new cultivars were grown in the open field located at both highland (Daekwallyung, above 800 m sea level) and lowland (Gangneung, above 20 m sea level) region, respectively. The average air temperature at highland during nursery plant propagation period was $5.3^{\circ}C$ lower than those at lowland. The number of daughter plants produced at lowland was 2 times as high as those of highland, presumably due to the higher air temperature. Anthracnose incidence rates of runner plants produced at lowland were 4 to 7% as high as those in highland. Mean temperature in the highland ($23.5^{\circ}C$) during flower bud differentiation treatment was $5.1^{\circ}C$ lower than that in the lowland ($28.6^{\circ}C$). Seedlings produced in highland showed higher C/N ratio and lower T/R ratio compared to those produced in lowland. The average flower bud formation date of the daughter plants grown in highland were advanced by 30 days compared to the date in lowland. Accordingly, highland was supposed to be appropriate region for raising seedlings in forcing culture of the new strawberry cultivars of 'Maehyang' and 'Seolhyang'.

Measurement of Determination Time of In-Vitro Flowering in Ginseng (Panax ginseng) (인삼의 기내 개화 결정시기의 측정)

  • To measure the time required for ginseng explants to become determined to form flower buds, we cultured zygotic embryos, seedlings, and cotyledonary nodes on MS medium supplemented with BA and GA$_3$of 5 ${\mu}$M each (flower inducing medium, FIM) for various periods and transferred to the basal medium. The explants required a minimum of 10 days on FIM to be determined. Histological observations revealed that the axillary meristem to be fated to develop into flower bud remained in a state of shoot meristem during the first 10 days of culture and differentiated into flower bud after 15 days of culture. We suggest that the in-vitro flowering system described in this study is useful in investigating (a) regulatory element(s) to cause the phase change from the vegetative to reproductive state by comparing predetermined explants with determined ones at the molecular level.

  • Adventitious buds were obtained from isolated cotyledons cultured on MS medium with various concentrations of 6-benzylamino purine (BA) and thidiazuron (TDZ). The highest numbers of adventitious buds were obtained on MS medium supplemented with 0.2 mg/L BA. Experimental culturing with half the petiole portion and half with the terminal segments were grown on MS medium contained with 0.2 mg/L BA. Frequency of the adventitious bud induction was variable accordingly to the type of cultured explants. Explants with the half petiole showed the highest adventitious bud induction rate (80%) compared to explants of half with terminal segment (20%). An elongated shoot from the buds and growth of advent roots were both possible on the 1/2 MS medium without a plant growth regulator. These results offer an effective way in which clonal propagation can be accomplished.

  • The growth pattern of bud-shoot-needle of isolated 15-year-old trees, and seansonal changes in litter-falls and fine root dry weights in the unthinned 28-year-old plantation were investigated to understand the growth phenology of Pinus koraiensis. Shoot growth was continued by 7th June when buds appeared, while current needle growth was by 19th July when the bud growth started. Most of the litter-falls occurred in October but many of them were fallen in July and August due to storms, Fine roots were produced mostly in autumn(1,004 kg/ha), and were dead during winter (583 kg/ha) and spring(1,331 kg/ha).

Studies on the Control of Bud Bright of Mulberry Tree when Intermediate-Cutting Harvest in Autumn (추기 중간벌채수확 뽕나무의 아고병방제에 관할 연구)

  • These studies aimed to establishment of controlling measure of the bud blight of mulberry tree when intermediate cutting harvest in autumn. The results obtained as following. 1. Effect of chemicals a) Results of 1972 (1) Of the seventeen chemicals tested. according to in vitro, the mycelial growth of Gibberella lateritium f. sp. mori was highly inhibited by PTAB and Benlate. PTAB and Benlate were also highly effective under the conditions of pathogen inoculated to the piece of mulberry tree and fungicidal action of these chemicals were stronger than the others. (2) Topsin-M. organic fungicide and plant growth regulator were not effective to inhibition of mycelial growth. (3) The effectiveness of fungicides were reduced gradually as the time after treatment increased. PTAB and Benlate retained approximately 50 percent of the original activity even after 15 days. (t) Of the twelve chemicals tested in fold, Benlate combination of PTAB with Apion, Apion and PTAB were the most effective fungicides for the control of bud blight of mulberry tree. (5) There is a sigificant difference between the timing on spraying, but it is not greatly effective for the control of bud blight of mulberry tree. b) Results of 1975 (1) Of the ten chemicals tested, according to in vitro RH 893, combination of Benlate with TMTD and TMTD were the most effective to inhibition of mycelial growth, followed by Benlate and combination of Benlate with Topsin M. (2) Of the eleven chemicals tested, Benlate, combination of Benlate with Kinondu, combination of Benlate with Topsin-M and Topsin M were highly effective under the renditions of pathogen inoculated to the piece of mulberry tree. (3) Of the eleven chemical evaluation tests were conducted in 3 experiment fold of Seoul, Chuncheun and Pusan, Benlate, Topsin-M, Apion, TMTD. combination of Benlate with Topsin-M, combination of Benlate with TMTD, combination of Benlate with Apion were highly effective chemicals for the control of bud blight of mulberry tree. (4) There is no difference between the timing on the chemical spraying after intermediate cutting harvest mulberry tree in autumn. 2. Ecological Control. When the mulberry shoot were cut off one to two buds from the top of intermediate cutting from 15th to 30th on October, bud blight of mulberry tree was greatly reduced in next spring compared with the effective chemical spraying. 3. Economical analysis on the results of chemical and ecological tests. When the expenses of ecological control for bud blight of mulberry tree per one ha was 100. thats of chemical control was from 159 to 254 according to spraying machines.
